The effects of nutrients on the fluorescence characteristics and biochemical composition of marine diatom Thalassiosira pseudonana 3H in light and dark cycles were investigated with continuous culture. The results show that with the increase of nutrient deficiency, the ratio of enhanced fluorescence to fluorescence (Fd/F), cellular chloropyll-a and protein content of the algae decline, but the fluorescence yield (F/Chl), DCMU enhanced fluorescence yield (Fd/Chl), cellular carbohydrate content, carbohydrate/Chl, protein/Chl, carbohydrate/protein increase. The changing amplitude of each parameter is different at different nutrition status, sampling time and different light intensity.  相似文献

Laminaria japonica, Undaria pinnatifida, Ulva lactuca, Grateloupia turuturu and Palmaria palmata are suitable species that fit the requirements of a seaweed-animal integrated aquaculture system in terms of their viable biomass, rapid growth and promising nutrient uptake rates. In this investigation, the responses of the optimal chlorophyll fluorescence yield of the five algal species in tumble culture were assessed at a temperature range of 10 - 30℃. The results revealed that Ulva lactuca was the most resistant species to high temperature, withstanding 30℃ for 4 h without apparent decline in the optimal chlorophyll fluorescence yield . While the arctic alga Palmaria palmata was the most vulnerable one, showing significant decline in the optimal chlorophyll fluorescence yield at 25℃ for 2 h. The cold-water species Laminaria japonica, however, demonstrated strong ability to cope with higher temperature (24 -26℃ ) for shorter time (within 24 h) without significant decline in the optimal chlorophyll fluorescence yield . Grateloupia turuturu showed a general decrease in the optimal chlorophyll fluorescence yield with the rising temperature from 23 to 30℃ , similar to the temperate kelp Undaria pinnatifida. Changes of chlorophyll fluorescence yields of these algae were characterized differently indicating the existence of species-unique strategy to cope with high light. Measurements of the optimal chlorophyll fluorescence yield after short exposure to direct solar irradiance revealed how long these exposures could be without significant photoinhibition or with promising recovery in photosynthetic activities. Seasonal pattern of alternation of algal species in tank culture in the Northern Hemisphere at the latitude of 36°N was proposed according to these basic measurements.  相似文献

-The detritus from the decomposition of dead plant materials of Spartina with great biomass provides abundant food for the heterotrophs in estuarine and nearshore coastal waters. It is for this reason that the decomposition rates of S. anglica and S. alterniflora near the estuary of obsolete Huanghe River were studied. The changes in nutritive components during decomposition are also studied.Results showed that the decomposition rate of dead leaves of S. anglica on ultra-low marsh was about 90% for the first year. In the eighth month, it was 80% and 76 % respectively for S. anglica and S. alterniflora on low marsh. Protein content of the two species of Spartina increased in the course of decomposition, with a maximum increment of 100% or more. The energy value also increased, whereas cellulose content decreased markedly. The percentage content of ash and lipid varied significantly with zonal differences.  相似文献

- The behavior of the reinforced concrete members subjected to combined torsion and shear is studied in this paper. Based on the skew bending failure pattern observed in tests, and according to the gradual increase in strain on the concrete surface, a nonlinear full range analysis is performed for predicting the torque-twist relationship of members under combined torsion and shear for the ratio of torsion to shear is chosen bigger than 0.5 (T/ V b> 0.5). The investigation on sea-ice biology in combination with physics, chemistry and ecology was carried out in the northwestern Weddell Sea, Antarctica, during the cruise ANT/XX III-7 on board POLARSTERN in the austral winter (August-October) in 2006. The distribution of chlorophyll a was measured and related to sea ice texture. The mean concentrations of chlorophyll a in the sea ice varied considerably with ice texture. The concentration of chlorophyll a per core ranged from 2.10– 84.40 μg/dm 3 with a mean of 16.56 μg/dm 3 . And the value of R (chlorophyll a / gross chlorophyll) ranged from 0.79–0.83. These high winter chlorophyll values indicate that primary production is considerable and confirms that there is significant primary production in Antarctic sea ice during winter. Thus this constitutes a major proportion of southern ocean primary production and carbon flux before the sea ice retreats.  相似文献

The effect of sonication pretreatment condition on Eucheuma cottonii and Gelidium amansii seaweed towards agar extraction wae studied.Four parameters were changed during sonication to investigate the effects on agar yield and quality.These parameters include the time interval,concentration ratio,frequency,and intensity.The highest amount of agar extracted from Eucheuma cottonii species could be obtained from the time interval of 30min,seaweed weight to solvent volume ratio of 1:20,the frequency of 35Hz,and the sonication power intensity of 30%.For Gelidium amansii species,the best agar yield also could be obtained from the time interval of 30 min,1:20 of seaweed weight to water volume ratio,the frequency of 35Hz,and power intensity of 30%.From the experiment,sonication pretreatment signifi cantly influenced the yield and properties of extracted agar.The sonication with autoclaved seaweed produced agar containing less sulfate content,which is an excellent chemical property for gel electrophoresis applications.The gelstrength of sonication with autoclaving for both seaweeds,Eucheuma and Gelidium species was the highest among those by sonication with direct heating,which proved that sonication pretreatment with autoclaving could enhance the physical properties of the agar.  相似文献

Microalgae,a sustainable source of multi beneficial components has been discovered and could be utilised in pharmaceutical,bioenergy and food applications.This study aims to investigate the sugaring-out effect on the recovery of protein from wet green microalga,Chlorella sorokiniana CY1 which was assisted with sonication.A comparison of monosaccharides and disaccharides as one of the phaseforming constituents shows that the monosaccharides,glucose was the most suitable sugar in forming the phases with acetonitrile to enhance the production of protein(52% of protein).The protein productivity of microalgae was found to be significantly influenced by the volume ratio of both phases,as the yield of protein increased to 77%.The interval time between the sonication as well as the sonication modes were influencing the protein productivity as well.The optimum protein productivity was obtained with 10s of resting time in between sonication.Pulse mode of sonication was suitable to break down the cell wall of microalgae compared to continuous mode as a lower protein yield was obtained with the application of continuous mode.The optimum condition for protein extraction were found as followed:200g/L glucose as bottom phase with volume ratio of 1:1.25,10s of resting time for ultrasonication,5s of ultrasonication in pulse mode and 0.25g of biomass weight.The high yield of protein about 81% could be obtained from microalgae which demonstrates the potential of this source and expected to play an important role in the future.  相似文献

Seasonal variations and distributions of dissolved carbohydrate concentrations at the ?zmir Bay were investigated with salinity, chlorophyll a(Chl a), and dissolved organic carbon(DOC) levels to understand their relationships.Samples were collected from surface, subsurface and bottom depths at seven stations. DOC concentrations ranged from 32.2 to 244.2 μmol/L, and in general, DOC levels increased from winter to summer, then slightly decreased in autumn. Monosaccharide(MCHO), polysaccharide(PCHO) and total dissolved carbohydrate(TDCHO) levels were found between 0.7–8.3, 0.7–19.5, and 2.6–24.6 μmol/L. DOC, MCHO, PCHO and TDCHO levels were found higher in middle-inner bays, under the influence of anthropogenic inputs, compared to outer bay. Seasonal changes of MCHO/DOC, PCHO/DOC and TDCHO/DOC ratios were statistically significant(p0.05)and the ratios showed decrease trends from winter to summer-autumn seasons. Distributions of TDCHO/DOC ratios at wide ranges(2.5%–42.3%) indicated the presence of newly forming and degrading fractions of DOM.According to results of factor analysis, Chl a, MCHO and TDCHO were explained in the same factor groups. In conclusion, the results showed that dissolved carbohydrate levels in the ?zmir Bay might be influenced by biological processes and terrestrial/anthropogenic inputs.  相似文献

The distributions of DMS and its precursor dimethylsulfoniopropionate, in both dissolved (DMSPd) and particulate fractions (DMSPp) were determined in the sea-surface microlayer and corresponding subsurface water of the Jiaozhou Bay, China and its adjacent area in May and August 2006. The concentrations of all these components showed a clear seasonal variation, with higher concentrations occurring in summer. This can be mainly attributed to the higher phytoplankton biomass observed in summer. Simultaneously, the enrichment extents of DMSPd and DMSPp in the microlayer also exhibited seasonal changes, with higher values in spring and lower ones in summer. Higher water temperature and stronger radiant intensity in summer can enhance their solubility and photochemical reaction in the microlayer water, reducing their enrichment factors (the ratio of concentration in the microlayer to that in the corresponding subsurface water). A statistically signi.cant relationship was found between the microlayer and subsurface water concentrations of DMS, DMSP and chlorophyll a, demonstrating that the biogenic materials in the microlayer come primarily from the underlying water. Moreover, our data show that the concentrations of DMSPp and DMS were signi.cantly correlated with the levels of chlorophyll a, indicating that phytoplankton biomass might play an important role in controlling the distributions of biogenic sulfurs in the study area. The ratios of DMS/chlorophyll a and DMSPp/chlorophyll a varied little from spring to summer, suggesting that there was no obvious change in the proportion of DMSP producers in the phytoplankton community. The mean sea-to-air .ux of DMS from the study area was estimated to be 5.70 μmol/(m2 ·d), which highlights the e.ects of human impacts on DMS emission.  相似文献

Intertidal marine macroalgae experience periodical exposures during low tide due to their zonational distribution. The duration of such emersion leads to different exposures of the plants to light and aerial CO2, which then affect the physiology of them to different extents.The ecophysiological responses to light and CO2 were investigated during emersion in two red algae Gloiopeltis furcata and Gigartina interrnedia, and two brown algae Petaloniafascia and Sargassum hemiphyllum, growing along the Shantou coast of China. The light-saturated net photosynthesis in G. furcata and P. fascia showed an increase followed by slightly desiccation, whereas that in G.interrnedia and S. hemiphyllum exhibited a continuous decrease with water loss. In addition, the upper-zonated G. furcata and P. fascia,exhibited higher photosynthetic tolerance to desiccation and required higher light level to saturate their photosynthesis than the lower-zonated G. interrnedia and S. hemiphyllum. Desiccation had less effect on dark respiration in these four algae compared with photosynthesis. The light-saturated net photosynthesis increased with increased CO2 concentrations, being saturated at CO2 concentrations higher than the present atmospheric level in G. furcata, G. intermedia and S. hemiphyllum during emersion. It was evident that the relative enhancement of photosynthesis by elevated CO2 in those three algae increased, though the absolute values of photosynthetic enhancement owing to CO2 increase were reduced when the desiccation statuses became more severe. However, in the case of desiccated P. fascia (water loss being greater than 20 %), light saturated net photosynthesis was saturated with current ambient atmospheric CO2 level. It is proposed that increasing atmospheric CO2 will enhance the daily photosynthetic production in intertidal macroalgae by varied extents that were related to the species and zonation.  相似文献

Antarctic ice microalga can survive and thrive in channels or pores containing high salinity in Antarctic ice layer. In this study, it was found that cell membrane permeability of green microalga Chlamydomonas sp. L4 from Antarctic sea ice was high in cells treated with hypersalinity due to the induction of active oxygen and radicals. However, increased super oxide dismutase (SOD) scavenged harmful free radicals effectively to keep cell membrane integrity. Also, the analysis of membrane fatty acids demonstrated the content of saturated fatty acids and monounsaturated fatty acids increased and polyunsaturated fatty acids decreased under the high-salt treatment for 14 d, which effectively reduced the membrane fluidity and minimized the injury to cell membrane. The morphological changes showed that hypersalinity induced the increase of cell volume and the consumption of starch granules. However, because of the increase in detoxification of vacuoles, electron-dense deposits and SOD activity under high-salt stress, the complete noninterference thylakoids, mitochondria and cell nucleus maintained cellular fundamental metabolism. Global-expression profiling of proteins showed eight protein spots disappeared, 18 protein spots decreased and 18 protein spots were enhanced after the high-salt shock obviously (P<0.05). One new peptide (pI 6.90; MW 51 kDa) was primarily confirmed as the processor of light reaction center protein CP43 in photosystem Ⅱ, which increased photosynthesis ability of Chlamydomonas sp. L4 treated with high salinity.  相似文献

Light is an important parameter in algal culturing.In this work,the effects of different light qualities on growth of the marine diatom Chaetoceros gracilis are evaluated.The cells were cultured under light quantum flux density of 60μmol photons/(m2·s)with nine light qualities:LED red light(LR),LED blue light(LB),LED red plus LED blue light(LR+LB),LED white light(LW),fluorescent white light(FW),and combinations of LW and FW lights with increased proportions of red or blue light(FW+LR,FW+LB,LW+LR,and LW+LB).Blue light promoted the growth of C.gracilis largely.Three light qualities,FW+LR,LW+LR,and LR,resulted in the lowest growth rate.Both chlorophyll and carotenoids reached the highest level under LB and the lowest level under LR among monochromatic light sources;however,increasing of the proportion of blue or red light in the white light induced an increase of the chlorophyll and carotenoid concentrations.Light absorption ability of microalgae was critical for the growth of these organisms,as we observed a positive correlation between the extinction coefficient at different wavelengths and the specific growth rate.These findings are of importance to improve the culturing of this alga in bioreactors.  相似文献

According to two cruises investigation information in summer and winter during 1998 and1999, the phosphorous concentration distribution and changes of summer and winter were discussed pri-marily in the South China Sea. The results show that the phosphate concentration of surface seawater insummer is distinctly lower than that in winter, averaging 0.04 μmol/dm3 in summer and 0.35 μmol/dm3 in winter. The organic phosphorous concentration of surface seawater in summer is higher than thatin winter, averaging 0.12μmol/dm3 in summer and 0.04 μmol/dm3 in winter respectively. The season-al changes of total phosphorus are similar to phosphate, averaging 0.22 μmol/dm3 in summer and 0.61μmol/dm3 in winter respectively. In vertical direction, phosphate, TDP and TP content are the lowestin upper 50 m water column, and increase in linearity rapidly with water depth, increasing slowly under500 m, reach to maximum about 1 000 m, then decrease slightly with water depth increasing. The ver-tical distribution is typical in summer, and there is small dispersed for phosphorus concentration for thesame depth of different stations. However, in winter there is a large disperse for phosphate, TDP andTP, specifically for phosphate at 200 m at which the concentration is maximum. This result indicatesthat there are large differences in hydrology and biology conditions that affect largely the chemical envi-ronment of the South China Sea. The organic phosphorus is the predominant in surface seawater of theSouth China Sea, but the inorganic phosphorus is the predominant in layers below depth of 150 m. Theorganic phosphorus concentration in deep water usually decreases with water depth increasing. The or-ganic phosphorus in summer is remarkably more than that in winter because of the strong biology activi-ties in summer.  相似文献

Enhancement effects of rare earth elements on the growth of Chaetoceros mulleri is studied in this paper. The results show that all of the light, middle and heavy rare earth elements have similar enhancement effect on the growth of Chaetoceros mulleri, with the beneficial concentrations of La, Gd and Yb being 7.28~87.40 μ mol/L, 6.36~57.23 μ mol/L and 5.78~17.34 μ mol/L. The optimum concentrations of La, Gd and Yb are 7.28 ~50.98 μ mol/L,31.80~44.52 μ m ol/L and 5.78~17.34 1μ mol/L, respectively. When the concentrations of La, Gd and Yb are 7.28~87.40 μ mol/L, 6.36~57.23 μ mol/L and 5.78~ 17,34 μ mol/L, the concentrations of chlorophyll have increased by 9.3~47.0%, 33.4~44.3%, and 36.5~40.3%, respectively as compared with the control group. The mechanism of enhancement of rare earth elements on the growth ot Chaetoceros mulleri is also discussed in this paper.  相似文献

The influence of macronitrogen (NO - 3 and NH + 4 ) addition with Ulva pertusa on dissolved inorganic carbon system in seawater was studied. The results indicate that p(CO 2 ) and HCO 3 concentration decrease significantly, while pH and CO 2- 3 concentration increase significantly. When the concentration of NO 3 was less than 71 μmol/dm 3 or NH + 4 was less than 49.7 μmol/dm 3 , dissolved inorganic carbon (DIC) absorption rates by Ulva pertusa generally increased with the increasing of nitrogen concentration. The DIC decreased 151 μmol/dm 3 with the addition of 71 μmol/dm 3 NO 3 and decreased 232 μmol/dm 3 with the addition of 49.7 μmol/dm 3 NH + 4 after the experiment compared with DIC measured without nitrogen addition. A significant negative-correlation was found between c(DIC) and growth rate (μ) of Ulva pertusa (r = -0.91, P <0.000 1, n=11). NH + 4 had more influence on the species of inorganic carbon system than NO 3 .  相似文献

The effects of ambient CO2/ambient UV-B, enriched CO2/ambient UV-B, ambient CO2/enhanced UV-B, and enriched CO2/enhanced UV-B on the ultrastructure of Dunaliella salina were investigated. (1) The ultrastructure of D..salina cell in the control experiment showed that the arrangement of thylakoid lamellae was regular, and there were many large starch grains among the thylakoid lamellae. A prominent well-developed pyrenoid was found in the middle of the chloroplast. Nucleus envelope and nucleolus were clearly observed. The Golgi apparatus accompanied by numerous vesicles with a compact arrangement of cisternae and the peripheral tips of the cisternae were swollen to a size comparable to that of some of the associated vesicles. (2) The ultrastructure of D. salina cell in enriched CO2 showed that the arrangement of thylakoid was regular and the lamellae were vivid. Developed pyrenoids were found in the low-CO2-grown cells, but not in the high-CO2-grown cells. The mitochondria cristae were vivid. The arrangement of Golgi apparatus was compact. (3) The ultrastructure of D. salina cell in enhanced UV-B showed that the thylakoid was dissolved and the cells had a less developed pyrenoid or no detectable pyrenoid. Part of the nucleus envelope was dissolved. The number ofmitochondria was increased and some mitochondria cristae were disintegrated. The starch grains were broken apart into many small starch grains. The Golgi apparatus with a loose arrangement ofcistemae and the peripheral tips of the Golgi cistemae were not especially swollen, with several large associated vesicles. (4) The ultrastructure of D. salina cell in the enriched CO2/enhanced UV-B showed that part of the thylakoid and nucleus envelopes of some cells were dissolved. The pyrenoid was larger than that of the enhanced UV-B. There were many mitochondria between stroma and chloroplast membrane, but mitochondria cristae were partly dissolved. Many small starch grains were accumulated in cells. The starch sheath was broken into several discontinuous starch grains with different sizes. The arrangement of Golgi apparatus was loose. Above all, although the enriched CO2 can alleviate the damage induced by the UV-B radiation, the effects of experimental UV-B radiation were larger than the effects of actual UV-B radiation, the damage induced by the UV-B radiation was so severe, therefore, CO2 enrichment could not restore the ultrastructure to the control level.  相似文献

Three harmful algal bloom (HAB) species, Phaeocystis globosa, Thalassiosira rotula, and Proro-centrum donghaiense were isolated from the coast of China and cultured in batches at three light intensities (40, 70 and 150 μmol photons·m-2·s-1). The variation patterns of cell numbers and growth rates with light intensity during growth process were different among species. In P. globosa and T. rotula, maximum growth rates were found at 150 μmol photons·m-2·s-1 and ranged from 0.60 divisions per day in T. rotula, to 1.17 divisions per day in P. globosa. The highest growth rate of P. donghaiense, however, was found at 70 μmol photons·m-2·s-1 (0.36 divisions per day). In general, all the three HAB species showed adaptation to increasing light intensity by decreas-ing cellular concentrations of chlorophyll a (Chl a), but the variation patterns during the growth process were species-specific. The cellular concentrations of Chl a in P. donghaiense and T. rotula increased gradually with incubation time, but the opposite trend was found in P. globosa. Most of the pigment ratios and pigment indices of these three species were nearly constant during the growth process and showed small changes at different light intensities illustrating the applicability of chemotaxonomy during the initial and developing stages of HAB events, which is very important to study the ecological issues related to HAB species. Ratios of photoprotective carotenoids, such as diadinoxanthin, diatoxanthin and β,β-carotene to total chlorophylls a (Tchl a) showed the trend of increasing with the increase of light intensity during growth process. The species-specific and pigment-specific variations in pigment ratios/indices at different light intensities during growth pro-cess probably reflected the differences in the pigment composition as well as the adaption capabilities of different species to the changes of physical conditions.  相似文献

Seasonal and inter-annual variability of hydrological parameters and its impact on chlorophyll distribution was studied from January 2009 to December 2011 at four coastal stations along the southwest Bay of Bengal. Statistical analysis(principal component analysis(PCA), two-way analysis of variance(ANOVA) and correlation analysis)showed the significant impact of hydrological parameters on chlorophyll distribution in the study area. The ranges of different parameters recorded were 23.8–33.8°C(SST), 4.00–36.00(salinity), 7.0–9.2(p H), 4.41–8.32 mg/L(dissolved oxygen), 0.04–2.45 μmol/L(nitrite), 0.33–16.10 μmol/L(nitrate), 0.02–2.51 μmol/L(ammonia),0.04–3.32 μmol/L(inorganic phosphate), 10.09–85.28 μmol/L(reactive silicate) and 0.04–13.8 μg/L(chlorophyll).PCA analysis carried out for different seasons found variations in the relationship between physico-chemical parameters and chlorophyll in which nitrate and chlorophyll were positively loaded at PC1(principal component1) during spring inter-monsoon and at PC2(principal component 2) during other seasons. Likewise correlation analysis also showed significant positive relationship between chlorophyll and nutrients especially with nitrate(r=0.734). Distribution of hydrobiological parameters between stations and distances was significantly varying as evidenced from the ANOVA results. The study found that the spatial and temporal distribution of chlorophyll was highly dependent on the availability of nutrients especially, nitrate in the southwest Bay of Bengal coastal waters.  相似文献
