The solubility of meat protein of croaker and jack mackerel was significantly affected by pH shifting. The protein yield of alkali-aided processing is higher than those of conventional and acid-aided processing. The addition of sarcoplasmic protein increases the breaking force and deformation value. The breaking force of recovered protein gel from alkali-aided processing is decreased by the addition of NaCI. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) revealed that fish protein of alkali is similar to that at pH7.0. Alkali-aided processing for recovering fish protein is a valuable method for increasing the utilization of frozen and pelagic fishes and for making kamaboko products.  相似文献

应用间接酶联免疫吸附试验(ELISA)技术测定不同水温下鳜经嗜水气单胞菌GYK1株全菌灭活疫苗浸泡免疫后皮肤黏液中抗体,以研究抗体动态变化,揭示温度对抗体产生的影响。结果表明:27.0~29.8℃水温(下称"较高温")下,鳜皮肤黏膜抗体滴度于第7天达到峰值211,而水温介于19.8~23.9℃(下称"较低温")时,抗体滴度于第11天达到峰值26,说明温度较高时,抗体形成快,峰值出现时间早,并且抗体滴度明显升高。另取不同温度下经上述疫苗浸泡免疫的鳜皮肤黏液,采用硫酸铵盐析、离心、浓缩等生物化学技术粗提鳜皮肤黏液中的免疫球蛋白,SDS-PAGE检测发现,较高水温下,高渗免疫组(加10 mg/mL食盐)鳜皮肤黏液蛋白中目标条带(72×103和29×103)深度较其他3个实验组(同温度下全菌组、较低水温下高渗及全菌组)有所增加,ELISA检测该组抗体效价为211。  相似文献

Changes in tissue structure, rheological properties and water content of raw and heated sea cucumber meat were studied. Sea cucumber Stichopus japonicus was heated at 25℃, 70℃ and 100℃ water for 5 min. The structural changes were observed using a light microscope and the rheological parameters (rupture strength, adhesive strength and deformation) determined using a texture meter. Microscopic photograph revealed that the structural change of heated meat was greater than that of raw meat. The rupture strength, adhesive strength and deformation of raw meat were smaller than those of the heated meat. Meanwhile, rheological parameters showed positive correlation with heating temperature. These changes are mainly caused by thermal denaturation and gelatinization of collagen during heating. These changes were also evidenced in observations using a light microscope and differential scanning calorimetry.  相似文献

The ability of cell to modulate the fluidity of plasma membrane was crucial to the survival of microorganism at low temperature. Plasma membrane proteins, fatty acids and carotenoids profiles of Antarctic psychrophilc yeast Rhodotorula sp. NJ298 were investigated at -3 ℃, 0 ℃ and 8 ℃. The results showed that plasma membrane protein content was greater at -3 ℃ than that at 8 ℃, and a unique membrane polypeptide composition with an apparent molecular mass of 94.7 kDa was newly synthesized with SDS-PAGE analysis; GC analysis showed that the main changes of fatty acids were the percentage of unsaturated fatty acids (C18∶ 1 and C18∶ 2) and shorter chain saturated fatty acid (C10∶ 0) increased along with the decrease of the culture temperature from 8 ℃ to -3 ℃; HPLC analysis indicated that astaxanthin was the major functional carotenoids of the plasma membrane, percentage of which increased from 54.6±1.5% at 8 ℃ to 81.9±2.1% at -3 ℃. However the fluidity of plasma membrane which was determined by measuring fluorescence anisotropy was similar at -3 ℃, 0 ℃ and 8 ℃. Hence these changes in plasma membrane's characteristics were involved in the cellular cold-adaptation by which NJ298 could maintain normal plasma membrane fluidity at near-freezing temperature.  相似文献

According to the three-dimensional contour maps showing the gel-forming properties of surimi derived from freshwater fish, 8 species of surimi were classified into two types. The-valley type surimi (silver carp, big-head carp, Chinese snake head and blunt snout bream) shows easy setting, low resistance to gel collapse, high enhancement ability with two-step heating, and narrow optimum heating temperature and time area, which are of the same characteristics as the walleye pollack surimi. In contrast, the Plateau type surimi (tilapia, grass carp, mud carp and common carp) exhibits difficult setting, high resistance to gel collapse, no enhancement ability with two-step heating, and wide optimum heating temperature and time area. There are seasonal changes of gelling properties of silver carp surimi, and the setting ability of surimi gel is higher in winter and lower in summer. The marine fish meat gels and the freshwater fish meat gels have the same acceptability for inland Chinese according to the sensory evaluation results. A slight increase in sensory scorings of kamaboko gels occurred when the extract from walleye pollack muscle was added, especially in the odor scoring of silver carp kamaboko gels.  相似文献

施氏獭蛤(LutrariasieboldiiReeve)属暖水性贝类,分布于热带—亚热带海区,在我国东海和南海都有。据在广西和海南沿海的调查,它生活在潮下带至水深78m的沙或沙泥底质中。营埋栖生活,深度达21cm。生活海区的水温为8~32℃,盐度11 .50~33 00,pH值8. 2~8 .4,透明度4~7m。食物以底栖硅藻和有机碎屑为主。繁殖期在5~9月。其肉供食用,是较珍贵的食品,可以发展人工增养殖。  相似文献

Crossbreeding is an effective approach to manage the genetic decline in aquaculture.One-way hybrids of Crassostrea sikamea(♀)and Crassostrea gigas(♂)have advantages in growth traits and adaptation to high temperature.Here,we used high-throughput sequencing to analyze the molecular processes in the hybrids under and after thermal stress.The hybrids were cultured in the seawater with an increasing temperature from 25℃to 40℃during 10 hours,which is regarded as the thermal stress stage.Then the temperature decreased from 40℃to 25℃within 2 h,which is regarded as the recovery stage.In this study,1293 significant diffe-rentially expressed genes(DEGs)were obtained under thermal stress,of which 576 were upregulated and 717 were downregulated,and 740 significant differentially expressed genes(DEGs)were obtained in the recovery stage,of which the number of upregulated and downregulated genes was 409 and 331,respectively.The antigen processing and presentation,NOD-like,and NF-kappa B path-ways were significantly enriched during the thermal stress stage.The MAPK and PPAR signaling pathways were significantly enrich-ed during the recovery stage.The HSP70,HSP90,and CANX genes were strongly and rapidly upregulated in the control/thermal stress groups but were slightly less upregulated in the thermal stress/recovery group.These results indicate that the innate immune system or nonspecific immunity was deployed to protect interior tissues from thermal stress.In addition,85%of the mutual DEGs were involved in bidirectional regulation(up/down or down/up)when the oysters were removed from the thermal stress to recover.This study provides preliminary insight into the molecular response of C.sikamea(♀)and C.gigas(♂)hybrids to thermal stress and provides a basis for future studies on temperature-adaptation and the possible expansion of hybrid breeding.  相似文献

Two-dimensional electrophoresis(2-DE) of protein extracted and purified from Alexandrium sp. LC3 was conducted. In the SDS-PAGE study, the relative molecular weights of the proteins were mainly in the range of 14 kDa?31 kDa and 43 kDa?66 kDa, and more proteins were detected between 14 kDa and 31 kDa. With the improved protein preparation, the two-dimensional electrophoresis patterns indicated that the relative molecular weights of the proteins were between 14 kDa and 100 kDa, and most of them ranged from 14 kDa to 31 kDa. This was consistent with the result of the SDS-PAGE analysis. The isoelectric points were found to lie between 3.0 and 8.0, and most of them were in the range of 3.0?6.0. Better separation effect was acquired with pre-prepared immobilized gradient (IPG) strip (pH 3?5.6), and about 320 protein spots could be visualized on the 2-DE map by staining. Within pH 3?10 and pH 3?5.6 strips, the protein samples of Alexandrium sp. LC3 could be separated well.  相似文献

设置5个温度梯度(14、19、24、29、34℃),海水盐度为32,研究温度和规格对管角螺Hemifusustuba(Gelin)耗氧率和排氨率的影响。结果表明:14~29℃范围内,管角螺耗氧率随温度的升高,29℃时达到最大值;14~34℃范围内,排氨率随温度的升高而增加。在相同温度下,耗氧率和排氨率随单位软体部干重的增加而下降,管角螺软体部干重(W)与单位体重耗氧率(OR)、排氨率(NR)之间的关系分别符合幂函数方程OR=aW-b、NR=cW-d,W与OR、NR呈负相关关系。  相似文献

在室内控制条件下,研究了几种环境因素对西施舌稚贝生长发育的影响。结果表明,西施舌稚贝最适生长温度为25.50~27.40℃,最适盐度为24.16~25.66,适应pH为7.5~8.5。壳长1 000~1 200μm的稚贝,在气温25℃与30℃时,干露时间不宜超过25 min;中砂有利于稚贝附着与生长。立体培育的西施舌稚贝,活动强、生长快。  相似文献

Juvenile silver pomfret, Pampus argenteus, was grown in culture tanks for 9 weeks on four different diets, and their effects on fish growth, digestive enzyme activity, and body composition were assessed. The feeding regime was as follows: Diet 1: fish meat; Diet 2: fish meat+artificial feed; Diet 3: fish meat+artificial feed+Agamaki clam meat; Diet 4: fish meat+artificial feed+Agamaki clam+copepods. The greatest weight gain was associated with Diet 4, while the lowest weight gain was associated with Diet 1. No significant difference was observed in weight gain between fish receiving Diet 2 and Diet 3. Specific growth rate followed similar trends as weight gain. The feed conversion ratio (FCR) of fish fed Diet 1 was significantly higher than the other fish groups, but no significant differences were observed in FCRs of fish fed Diet 2, Diet 3 or Diet 4. There was also no significant difference in the hepatosomatic index (HSI) between the four diets. For fish that received Diets 2-4, containing artificial feed, higher protease activities were detected. A higher lipid content of the experimental diets also significantly increased lipase activities and body lipid content. No significant differences in amylase activity or body protein content were found between Diets 1-4. In conclusion, a variety of food components, including copepods and artificial feed, in the diet of silver pomfret significantly increased digestive enzyme activity and could improve growth performance.  相似文献

为找寻光温敏核不育水稻幼穗与育性调控有关的蛋白质,采用固相pH梯度-SDS聚丙烯酰胺双向凝胶电泳对光温敏核不育水稻(培矮64S)育性转换期幼穗总蛋白进行了分离,获取凝胶图像并进行重复性分析,通过微量测序-Edman降解方法和原位酶解及MALDI-TOF质谱的方法鉴定了60个蛋白质点。结果表明:本方法可得取分辨率和重复性较好的凝胶图谱。蛋白质点在2D胶上的重复性为:沿等电聚焦方向偏差为1.49±0.22 mm,沿SDS-PAGE方向偏差为1.24±0.18 mm。37个蛋白质点得到了准确的鉴定结果,其中16个是差异蛋白质点。在不育变化为可育的过程中,明显表达上调的蛋白质点包括几丁质酶,丙酮酸脱氢酶等;明显下调的蛋白质包括超氧化物歧化酶,硝酸还原酶脱辅基酶蛋白等;金属硫蛋白RicMT只在可育中存在。  相似文献

在4.9hm2养殖池的一角围成面积0.13hm2的小塘,形成半封闭围隔池,内设投饵台,灌注21~23℃的地下水,维持围隔中水温在13~16℃,研究吉富罗非鱼成鱼低温越冬技术。结果表明,罗非鱼越冬成活率98%,未出现罗非鱼的低温损伤,溶氧从5.8mg/L降为3.2mg/L、透明度从50cm降为35cm、总氨氮低于0.3mg/L、亚硝酸氮低于0.5mg/L,COD低于23.6mg/L,水质良好。在同样气温条件下,大棚越冬对照池保持水温15~16℃,罗非鱼存活率95%,溶氧从5.8mg/L降为1.2mg/L、透明度35cm降为25cm、总氨氮0.9mg/L升为2.3mg/L、亚硝酸氮0.6mg/L升为2.5mg/L,COD为43.6mg/L,水质变差。半封闭围隔池可保持水温在罗非鱼的近临界伤害温度13℃或13℃以上,罗非鱼成鱼在野外不搭棚能安全越冬。  相似文献

研究了温度、盐度、pH值的变化对曼氏无针乌贼幼体成活率和活力的影响。结果表明,曼氏无针乌贼幼体的生存盐度为11.73~31.43,适宜盐度为19.61~26.18;生存水温为14~30℃,适宜水温为22~28℃;生存pH值为6.0~9.5,适宜pH值为7.5~8.5。说明环境因子变化对幼体存活有较大影响。乌贼幼体对渐变的适应性强,幼体成活率渐变大于突变,但温、盐度的耐受值宽度并未增大。  相似文献

以凡纳滨对虾为原料,以ACE抑制率为指标,利用响应面法对虾肉蛋白自溶制备ACE抑制肽的工艺条件进行了优化,即在酶解条件(pH值、温度、虾头虾肉质量比)和ACE(Angiotensin I-converting Enzyme,ACE)抑制率之间建立了数学模型Y=23.59-0.21X1+0.84X2+0.85X3-0.71X12-0.94X22-1.06X32+0.088X1X2-0.46X1X3-0.87X2X3。分析表明,在3个因素中,虾头与虾肉比例对ACE抑制率的影响最为显著。优化后的工艺参数为:pH7.35,温度57.2℃,虾头与虾肉比例为1∶1。根据回归方程的预测结果,反应时间为4 h,其ACE抑制率达41.9%。  相似文献

An 8-week feeding experiment was conducted in an indoor recirculation seawater system to investigate the effects of partial replacement of dietary fishmeal with proteins from five sources on the growth performance and feed utilization of Sebastes schlegeli. Six isonitrogenous and isoenergetic diets were formulated using fishmeal (FM, the control) as sole protein source, or proteins from five sources including poultry by-product meal (PBM), meat and bone meal (MBM), soybean meal (SBM), cottonseed meal (CSM) and canola meal (CNM). Fifteen percent of the crude protein provided by fish meal was replaced, respectively. The results showed that the differences in specific growth rate (SGR) and survival rate (SR) among fish fed PBM, MBM, SBM, CSM and whole FM diets were not significant. However, SGR and SR of fish fed CNM diet was significantly lower than that of other treatments. Feeding rate, feed conversion, nutrient retention showed similar patterns to that of growth. Fish fed CSM and CNM showed significantly lower apparent digestibility coefficient (ADC) of dry matter and gross energy than those fed others while fish fed CNM showed lower ADC of crude protein than those fed others (P<0.05). These results suggested that it was feasible to substitute 15% dietary protein provided by fishmeal with PBM, MBM, SBM and CSM, respectively, but not with CNM as the replacement with CNM reduced fish growth and feed utilization.  相似文献

Activities of hexokinase(HK),pyruvate kinase(PK) and levels of HSP70 were measured to evaluate the response of Litopenaeus vannamei to rapid temperature changes under controlled laboratory conditions.Shrimps were subjected to a quick temperature change from 27℃ to 17℃ for the summer case(Cold temperature treatment),or from 17℃ to 27℃ for the winter case(Warm temperature treatment).After 0.5,1,3,6,12,24,48,and 72 h of exposure time,shrimps were sampled and prepared for further analysis.The results showed that the effect of acute temperature changes on activities of HK was significant.Patterns of variations of the two glycolytic enzymes suggested that enzymes in the glycolysis cycle could adjust their activities to meet the acute temperature change.The HSP70 level increased in both cold and warm temperature treatments,suggesting that the rapid temperature changes activated the process of body's self-protection.But the difference in expression peak of HSP70 might be related to the different body size and the higher thermal sensitivity to temperature increase than to temperature decrease of L.vannamei.  相似文献
