The intertidal copeopod Tigriopus japonicus, which is abundant and widely distributed along the coasts of Western Pacific, has been suggested to be a good marine ecotoxicity testing organism. In this study, a series of experiments were conducted to investigate the reproducibility and variability of copper (Cu) sensitivity of T. japonicus so as to evaluate its potential to serve as an appropriate test species. To understand the seasonal variation of Cu sensitivity, individuals of T. japonicus were collected from the field in summer and winter, and subjected to standard 96 h acute (static renewal) toxicity tests. 96 h-LC50 values of T. japonicus collected from the two seasons were marginally different (p = 0.05), with an overall coefficient of variation (CV) of 33%. Most importantly, our results indicated that chronic Cu sensitivity of T. japonicus was highly reproducible. The CVs of intrinsic rates of increase in the population of the control and Cu treatment (10 μg Cu l−1) groups were only 10–11% between 10 runs of a standardised complete life-cycle test. Moreover, different Cu(II) salts generally resulted in a similar 96 h-LC50 value while Cu(I) chloride was consistently slightly less toxic than Cu(II) salts. Given such a high reproducibility of toxic responses, it is advocated to use T. japonicus as a routine testing organism.  相似文献

Intertidal harpacticoid copepods are commonly used in eco-toxicity tests worldwide. They predominately live in mid-high shore rock pools and often experience a wide range of temperature and salinity fluctuation. Most eco-toxicity tests are conducted at fixed temperature and salinity and thus the influence of these environmental factors on chemical toxicity is largely unknown. This study investigated the combined effect of temperature and salinity on the acute toxicity of the copepod Tigriopus japonicus against two common biocides, copper (Cu) and tributyltin (TBT) using a 2 × 3 × 4 factorial design (i.e. two temperatures: 25 and 35 °C; three salinities: 15.0‰, 34.5‰ and 45.0‰; three levels of the biocide plus a control). Copper sulphate and tributyltin chloride were used as the test chemicals while distilled water and acetone were utilised as solvents for Cu and TBT respectively. 96h-LC50s of Cu and TBT were 1024 and 0.149 μg l−1 respectively (at 25 °C; 34.5‰) and, based on these results, nominal biocide concentrations of LC0 (i.e. control), LC30, LC50 and LC70 were employed. Analysis of Covariance using ‘concentration’ as the covariate and both ‘temperature’ and ‘salinity’ as fixed factors, showed a significant interaction between temperature and salinity effects for Cu, mortality increasing with temperature but decreasing with elevated salinity. A similar result was revealed for TBT. Both temperature and salinity are, therefore, important factors affecting the results of acute eco-toxicity tests using these marine copepods. We recommend that such eco-toxicity tests should be conducted at a range of environmentally realistic temperature/salinity regimes, as this will enhance the sensitivity of the test and improve the safety margin in line with the precautionary principle.  相似文献

In Korea, the new permission criteria for industrial effluents based on Daphnia magna acute toxicity tests will be gradually implemented starting from 2011. Thus, in this study, toxicity assessment and identification using a marine species (Tigriopus japonicus) and the freshwater species (D. magna) was comparatively investigated. Effluent from an acid mine drainage treatment plant showed acute toxicity toward both organisms due to low pH, which was removed by neutralization of the effluent. Additionally, evaluation of the effluent of an electronics company revealed that Cu was attributable to the observed toxicity, and the effluent was more toxic toward T. japonicus than D. magna. Moreover, effluents from a metal plating factory were acutely toxic toward D. magna (6.50 TU), while they were not toxic against T. japonicus. Toxicity identification revealed that the high level of Cl (12,841 mg L−1) was the cause of toxicity. Thus, the effluents had no effect on the marine species, T. japonicus. These findings suggest that a marine species rather than a freshwater species is more desirable for toxicity assessment of industrial effluent discharged into the saltwater, and thus should be considered in the legislation of toxicity-based discharge limits in Korea.  相似文献

We investigated the effects of salinity (5‰, 15‰, 25‰ and 35‰) on metal ion (Cu and Zn) and nanoparticle (NP) CuO and ZnO toxicity to Tigriopus japonicus. Increasing the test media volume without renewal increased the 96-h LC50 for Cu (32.75 mg L−1) compared to the reported value (3.9 mg L−1). There was no significant difference in acute toxicity at different salinities between acclimated and unacclimated T. japonicus (> 0.05). Increasing salinity decreased the dissolved concentrations of Cu and Zn ions due to the precipitation of the metal ions, consequently reducing the acute toxicity to T. japonicus. The effect of salinity on acute CuO and ZnO NP toxicity was similar to that on metal ion toxicity. Since the aggregation of NPs generally enhanced at higher salinities, both the dissolution and aggregation of CuO and ZnO NPs may control the effect of salinity on acute toxicity to T. japonicus.  相似文献

The effects of Cu, Cd, Zn and Cr on the survival and feeding behaviour of the sandy shore scavenging gastropod Nassarius festivus were compared. The 96-h LC50 for Cu, Cd, Zn and Cr were 0.36, 1.52, 1.76 and 36.9 mg l−1, respectively. Four sublethal concentrations of each metal plus a control were prepared and the snails were exposed to experimental solutions for 96 h. Feeding behaviour was studied after the snails were starved for five days. As compared with the control, the number of individuals feeding was significantly reduced by exposure to 0.05 mg l−1 Cu, 0.2 mg l−1 Zn, 0.5 mg l−1 Cd and 5 mg l−1 Cr. The time spent feeding was greater for individuals exposed to greater concentrations of Zn and Cd but no effect was found for Cu and Cr. Chemoreception of food was studied by placing the snails at a fixed distance of 15 cm from the bait. The success rate of reaching the bait was less for individuals exposed to Cr but no effect was found for Zn, Cu or Cd. The time required for an individual to reach the bait decreased as the concentration of Zn increased. In contrast, a longer time was required for individuals exposed to Cr whereas the effect of Cd and Cu was insignificant. The potential of using feeding behaviour and chemoreception in contaminant evaluations is discussed.  相似文献

In recent years, there has been a decline in the wild populations of river pufferfish, Takifugu obscurus. Besides overexploitation for commercial purposes, environmental pollution is believed to have contributed to its decline. However, almost no information exists about genes involved in metabolism of xenobiotics by this species. Nevertheless, there is interest in fugu fishes, since they possess the smallest genome among vertebrates. We cloned and characterized the full-length cDNA sequence of a cytochrome P450 1A (CYP1A) gene from T. obscurus. Phylogenic relationship of T. obscurus CYP1A was also compared to other fish species. The tissue distribution and time-dependant induction of CYP1A mRNA were studied by real-time PCR in T. obscurus exposed to an aryl hydrocarbon receptor (Ahr) agonist, β-naphthoflavone (BNF). The greatest basal expression in livers of control as well as BNF-treated individuals. However, brain, gill, gonad, intestine, and kidney also expressed CYP1A. Muscles expressed the least CYP1A. The results of the time-course study revealed induction in brain and gills after 6 h and at 12 h in most tissues. Except for gills, all other organs retained induced expression of CYP1A mRNA up to 96 h.  相似文献

We have designed and calibrated a piston-cylinder cell assembly suitable for conductingin situ measurements of enthalpies of phase transitions at elevated pressures by heat-flux differential scanning calorimetry (DSC). The high-pressure DSC detector consists of a Pt-Pt13%Rh thermopile wrapped around a frame of fired pyrophyllite. Four thermocouple junctions, arranged radially around the sample capsule, are connected in series, with four reference thermocouple junctions located 3–4 mm above the sample and embedded in thermally inert ceramic. A W-W25%Re control thermocouple is situated directly above the top of the sample; the whole detector assembly is enclosed in a 1.5 mm thick cylindrical ceramic sleeve located at the center of a 8–10 mm long hot-zone in the tapered graphite furnace. Using this detector design and cell assembly, we have observed the thermal signal associated with the fusion of Au at 0.5 and 1.2 GPa, and have calculated a calibration factor (K) for this detector based on the gold melting curve ofMirwald andKennedy (1979). Detector sensitivity decreases by a factor of four over this pressure-temperature interval. The reproducibility of the enthalpy of fusion of gold at 0.5 GPa suggests that detector geometry is reproducible from one experiment to the next, and thus confirms the viability of this particular detector design for quantitative DSC measurements. Subsequent experiments will assess the dependence of (K) on temperature and pressure by measuring the enthalpies of fusion of additional metals (e.g., Ag, Cu, Al, Ge) and salts (e.g., NaCl, CsCl).  相似文献

Soil samples are collected from 74 sites in Shenyang city, northeast China, and the concentrations, spatial distribution, enrichment factors (EF), and potential ecological and human health risk (Er) of potentially toxic metals (Sr, Zn, Cr, Pb, Cu, Ni, and As) are investigated. The mean concentrations of potentially toxic metals in topsoil follow the order Sr > Zn > Cr > Pb > Cu > Ni > As. Compared with 2011, the contamination magnitude has improved in 2017, but the pollution status still exists, especially for As and Cr. EF and the mean Er of potentially toxic metals decrease following the order As > Pb > Cu > Ni > Zn > Cr > Sr and As > Cu > Zn > Pb > Sr > Ni > Cr, suggesting that the potentially toxic metals in the topsoil of Shenyang city mainly come from natural sources, and the potential ecological risk for potentially toxic metals is low. There are potentially non‐carcinogenic and carcinogenic health risks, but they are not considered significant. For non‐carcinogenic health risks, oral ingestion is the main exposure pathway, and Cr and Pb are the major pollutants. Finally, As is found to be the main carcinogenic contamination metal.  相似文献

Pressure–volume–temperature relations have been measured to 32 GPa and 2073 K for natural magnesite (Mg0.975Fe0.015Mn0.006Ca0.004CO3) using synchrotron X-ray diffraction with a multianvil apparatus at the SPring-8 facility. A least-squares fit of the room-temperature compression data to a third-order Birch–Murnaghan equation of state (EOS) yielded K0 = 97.1 ± 0.5 GPa and K′ = 5.44 ± 0.07, with fixed V0 = 279.55 ± 0.02 Å3. Further analysis of the high-temperature compression data yielded the temperature derivative of the bulk modulus (∂KT/∂T)P = −0.013 ± 0.001 GPa/K and zero-pressure thermal expansion α = a0 + a1T with a0 = 4.03 (7) × 10−5 K−1 and a1 = 0.49 (10) × 10−8 K−2. The Anderson–Grüneisen parameter is estimated to be δT = 3.3. The analysis of axial compressibility and thermal expansivity indicates that the c-axis is over three times more compressible (KTc = 47 ± 1 GPa) than the a-axis (KTc = 157 ± 1 GPa), whereas the thermal expansion of the c-axis (a0 = 6.8 (2) × 10−5 K−1 and a1 = 2.2 (4) × 10−8 K−2) is greater than that of the a-axis (a0 = 2.7 (4) × 10−5 K−1 and a1 = −0.2 (2) × 10−8 K−2). The present thermal EOS enables us to accurately calculate the density of magnesite to the deep mantle conditions. Decarbonation of a subducting oceanic crust containing 2 wt.% magnesite would result in a 0.6% density reduction at 30 GPa and 1273 K. Using the new EOS parameters we performed thermodynamic calculations for magnesite decarbonation reactions at pressures to 20 GPa. We also estimated stability of magnesite-bearing assemblages in the lower mantle.  相似文献

Polycyclic aromatic hydrocarbons (PAHs) are widespread persistent pollutants that readily undergo biotic and abiotic conversion to numerous transformation products in rivers, lakes and estuarine sediments. Here we characterize the developmental toxicity of four PAH transformation products each structural isomers of hydroxynaphthoic acid: 1H2NA, 2H1NA, 2H3NA, and 6H2NA. Medaka fish (Oryzias latipes) embryos and eleutheroembryos were used to determine toxicity. A 96-well micro-plate format was used to establish a robust, statistically significant platform for assessment of early life stages. Individual naphthoic acid isomers demonstrated a rank order of toxicity with 1H2NA > 2H1NA > 2H3NA > 6H2NA being more toxic. Abnormalities of circulatory system were most pronounced including pericardial edema and tube heart. To determine if HNA isomers were AhR ligands, spatial-temporal expression and activity of CYP1A was measured via in vivo EROD assessments. qPCR measurement of CYP1A induction proved different between isomers dosed at respective concentrations affecting 50% of exposed individuals (EC50s). In vitro, all ANH isomers transactivated mouse AhR using a medaka CYP1A promoter specific reporter assay. Circulatory abnormalities followed P450 induction and response was consistent with PAH toxicity. A 96-well micro-plates proved suitable as exposure chambers and provided statistically sound evaluations as well as efficient toxicity screens. Our results demonstrate the use of medaka embryos for toxicity analysis thereby achieving REACH objectives for the reduction of adult animal testing in toxicity evaluations.  相似文献

The fractional advection–dispersion equation (FADE) known as its non-local dispersion, has been proven to be a promising tool to simulate anomalous solute transport in groundwater. We present an unconditionally stable finite element (FEM) approach to solve the one-dimensional FADE based on the Caputo definition of the fractional derivative with considering its singularity at the boundaries. The stability and accuracy of the FEM solution is verified against the analytical solution, and the sensitivity of the FEM solution to the fractional order α and the skewness parameter β is analyzed. We find that the proposed numerical approach converge to the numerical solution of the advection–dispersion equation (ADE) as the fractional order α equals 2. The problem caused by using the first- or third-kind boundary with an integral-order derivative at the inlet is remedied by using the third-kind boundary with a fractional-order derivative there. The problems for concentration estimation at boundaries caused by the singularity of the fractional derivative can be solved by using the concept of transition probability conservation. The FEM solution of this study has smaller numerical dispersion than that of the FD solution by Meerschaert and Tadjeran (J Comput Appl Math 2004). For a given α, the spatial distribution of concentration exhibits a symmetric non-Fickian behavior when β = 0. The spatial distribution of concentration shows a Fickian behavior on the left-hand side of the spatial domain and a notable non-Fickian behavior on the right-hand side of the spatial domain when β = 1, whereas when β = −1 the spatial distribution of concentration is the opposite of that of β = 1. Finally, the numerical approach is applied to simulate the atrazine transport in a saturated soil column and the results indicat that the FEM solution of the FADE could better simulate the atrazine transport process than that of the ADE, especially at the tail of the breakthrough curves.  相似文献

Irgarol 1051 (2-methythiol-4-tert-butylamino-6-cyclopropylamino-s-triazine) is an algaecide commonly used in antifouling paints. It undergoes photodegradation which yields M1 (2-methylthio-4-tert-butylamino-6-amino-s-triazine) as its major and most stable degradant. Elevated levels of both Irgarol and M1 have been detected in coastal waters worldwide; however, ecotoxicity effects of M1 to various marine autotrophs such as cyanobacteria are still largely unknown. This study firstly examined and compared the 96 h toxicities of Irgarol and M1 to the cyanobacterium Chroococcus minor and two marine diatom species, Skeletonema costatum and Thalassiosira pseudonana. Our results suggested that Irgarol was consistently more toxic to all of the three species than M1 (96 h EC50 values: C. minor, 7.71 μg L−1 Irgarol vs. >200 μg L−1 M1; S. costatum, 0.29 μg L−1 Irgarol vs. 11.32 μg L−1 M1; and T. pseudonana, 0.41 μg L−1 Irgarol vs. 16.50 μg L−1 M1). Secondly, we conducted a meta-analysis of currently available data on toxicities of Irgarol and M1 to both freshwater and marine primary producers based on species sensitivity distributions (SSDs). Interestingly, freshwater autotrophs are more sensitive to Irgarol than their marine counterparts. For marine autotrophs, microalgae are generally more sensitive to Irgarol than macroalgae and cyanobacteria. With very limited available data on M1 (i.e. five species), M1 might be less toxic than Irgarol; nonetheless this finding warrants further confirmation with additional data on other autotrophic species.  相似文献

Three sediment cores were sampled at Sepetiba Bay and four cores at Ribeira Bay, Rio de Janeiro State, Brazil. Sediment accumulation rates and chronologies were obtained from 210Pb activity-depth profiles. Sediment accumulation rates in Ribera Bay ranged from 1.2 mm y−1 in the inner bay to 2.6 mm y−1 close to its entrance. In Sepetiba Bay two sediment accumulation rates were observed: a lower rate of 0.35 cm y−1 before the 1960s and 0.76 cm y−1 since then. The cause of this change is due to the construction of the Santa Cecília impoundment (1955) that brings water from the Paraíba do Sul Basin into the Guandu River, which increased its flow from the original 20 m3 s−1 to 160 m3 s−1. Concentration of 44 elements was obtained by ICP-MS after total dissolution of samples from two selected cores. The relative differences between the concentrations of crustal elements, such as Al, Fe and Ti are only about 20% (p < 0.05). Cd and Zn are 15 and four times larger in Sepetiba Bay than in Ribeira Bay, respectively. Other major and minor elements show lower statistically significant differences. The enrichment factors and metal inventories show that Sepetiba Bay can be considered polluted with Bi, Cd, Cr, Cu, Sb and Zn. Particularly, Cd and Zn present concentrations three and four times higher than the Brazilian existing limits.  相似文献

Concentrations of 14 different dissolved free amino acids (DFAA), dissolved inorganic nitrogen (DIN), and chlorophyll a were determined in the surface microlayer and subsurface waters at 41 stations in the Yellow Sea, China in April 2006. The concentrations of DFAA in the subsurface water ranged from 0.13 to 1.62 μM, with an average of 0.57±0.05 μM, while those in the surface microlayer varied between 0.22 and 2.6 μM, with an average of 0.94±0.08 μM. Major constituents of DFAA present in the study area were glycine, alanine, glutamic acid, serine and histidine. One-way analysis of variance (ANOVA) showed no significant difference in average mol fractions of DFAA between microlayer and subsurface water (F=0.0440, P=0.8355). Hierarchical cluster analysis of the station similarity based on the DFAA composition in both the surface microlayer and subsurface water separated three clusters of stations at the 70% Bray–Curtis similarity level. The average concentrations of chlorophyll a and DIN were 1.18 (0.34–4.44) μg L−1 and 16.57 (3.98–49.59) μM in the subsurface water, and those in the surface microlayer were 1.30 (0.24–3.97) μg L−1 and 18.56 (5.77–48.93) μM, respectively. Our results showed that concentrations of chlorophyll a (r2=0.7940, n=41, p<0.0001), DIN (r2=0.6939, n=41, p<0.0001) and DFAA (r2=0.2416, n=41, P=0.0011) in the microlayer were, respectively, correlated with their subsurface water concentrations, implying that there was a strong exchange effect between the microlayer and subsurface water. The enrichment factor of DFAA in the microlayer ranged from 0.47 to 2.24 with a mean of 1.88±0.16.  相似文献

By modifying a previous method with constant elements, we developed a quadratic element method for more accurately estimating groundwater flow by the inversion of tilt data. In this method: (1) a region of groundwater flow is divided into quadratic elements in which the change in groundwater volume per unit volume of rock (Δv) and the Skempton coefficient (B) vary in a quadratic manner with the coordinates, (2) the values of Δv are set to zero at the boundaries of the region of groundwater flow and (3) the sum of the squared second derivatives of Δv is adopted as a constraining condition that is weighted and added to the sum of the squared errors in tilt. First, analyses were performed for a flow model to determine the accuracy of this method for estimating groundwater flow and also to clarify the effect of the assumed size of a region of groundwater flow. These analyses showed that the quadratic element method proposed in this study gives a much better estimation of Δv than the constant element method and that a large region of groundwater flow should be assumed, rather than a small region, since the values of Δv at points outside of the actual region of groundwater flow are estimated to be nearly zero when a large region is assumed while these values are greatly overestimated when an excessively small region is assumed. Finally, the quadratic element method was applied to the site of the Mizunami Underground Research Laboratory in the Tono area, Japan. Inverse analyses were performed for tilt data measured by four tiltmeters with a resolution of 10−9 radians during the excavation of two shafts under the assumption that the rock mass is an isotropic and homogeneous half- space. The results showed that the method proposed in this study reproduced the tilt data very accurately. Thus, the distribution of Δv was estimated without sacrificing the reproducibility of the tilt data. The contour maps of B(1 + ν)Δv (ν: Poisson’s ratio) showed that the heterogeneous flow of groundwater occurred at the site and that groundwater volume decreased mainly in the area surrounded by two faults. The latter result is consistent with the finding obtained by previous investigations that these faults have low permeability in the direction perpendicular to the strike and may act as a flow barrier.  相似文献

Since it is often difficult to estimate possible adverse effects due to contamination in selected ecosystems, multi-species biomonitoring may provide more information, taking into account different routes of exposure, ecological roles and metabolic capabilities of animals. In this context, we exposed for 7 days the mussel Mytilus galloprovincialis and the crab Carcinus aestuarii to 4-nonylphenol (NP), a well-known xenoestrogen. In mussels (0–0.2 mg NP l−1), we measured NP bioaccumulation in soft tissues and vitellogenin (Vg)-like protein levels in digestive glands from both males and females by the alkali-labile phosphate assay (ALP). As no reference data were available for crab exposure, the NP 96-h LC50 value was previously determined. Then, in sublethally exposed (0–1.0 mg NP l−1) male crabs, NP bioaccumulation and Vg levels were measured in hemolymph, gonads and digestive gland. Bioaccumulation of NP increased from 43 to 371 μg g−1 d.w. in mussels, and from 3.6 to 37 μg g−1 d.w. in crabs, depending on the NP concentration in water. Dose-dependent Vg-like protein induction was observed in both species, appearing to be related to NP bioaccumulation, although a partial decrease was recorded at the highest concentration tested. A similar trend was observed in both digestive gland and gonad of exposed crabs; Vg increased to a lesser extent, although significantly, in hemolymph. Results demonstrated that NP induces Vg synthesis both in male and female mussels, as well as in male crabs. On the basis of the responsiveness of both species investigated, a multi-species approach is indicated in biomonitoring programmes.  相似文献

The Laccadive Ridge (L-R), trending roughly parallel to the west coast of India, is an intriguing segment of the northernmost Chagos-Laccadive Ridge (C-L-R) system. Although crustal nature and isostatic response of the southern C-L-R is well known, there are no similar studies on the L-R. In the present study, the isostatic response of the lithosphere beneath the L-R is estimated so as to characterize its crustal nature, total crustal as well as effective elastic plate thickness and mode of compensation. Twelve gravity and bathymetry profiles across the ridge were analyzed using linear transfer function and forward model techniques. The observed admittance function within the diagnostic waveband of 250 < λ > 80 km (0.025 < k > 0.080 km−1) fits well with (i) the Airy model whose average crustal thickness (Tc) and density are 17 ± 2 km and 2.7 × 103 kg m−3, respectively, and (ii) the thin plate flexure model of isostasy with an effective elastic plate thickness (Te) of 2–3 km. The estimated average crustal thickness and density are in good agreement with published seismic refraction results over the ridge. The results of the present study support an Airy model of isostasy for the L-R. The low Te value, in view of other published results in the study area, suggests stretched and loaded continental lithosphere of the L-R during the evolution of the western continental margin of India.  相似文献

In the eustigmatophycean Nannochloropsis limnetica the content of polyunsaturated fatty acids (PUFA) is extremely high in comparison to different planktonic green algal taxa in freshwater ecosystems. The sums of n-6 and n-3 fatty acids in N. limnetica were ten-fold higher than in the other picoplankton Choricystis minor and Pseudodictyosphaerium jurisii, and higher than in the nanoplanktonic green algae Chlorella vulgaris, Monoraphidium neglectum and Scenedesmus obtusiusculus. The content of fatty acids in N. limnetica was highly variable under different culture conditions. The highest concentrations of PUFA in N. limnetica were found in non-aerated suspension cultures, with a high content of phosphate (40 mg l−1 K2HPO4) in the culture medium: linoleic acid 22.19 mg g−1 DW, arachidonic acid 10.52 mg g−1 DW, and eicosapentaenoic acid 55.56 mg g−1 DW. N. limnetica represent a high-quality food resource in freshwater food chains. Furthermore, cultures of this eustigmatophycean alga have a high potential for use in biotechnology and aquaculture.  相似文献
