免疫遗传算法及其在VRP中的应用   总被引:2,自引:0,他引:2  
物流配送车辆路径问题(VRP)是一类典型的NP问题。在基本遗传算法的基础上,根据生物的免疫系统原理,提出一种改进的算法——免疫遗传算法。在算法中构造一种基于抗体浓度的群体多样性保持策略,引入免疫算子和免疫记忆库。将该算法应用于求解VRP问题,实验结果表明算法可以实现解的多样性,避免出现早熟收敛,可以有效防止进化过程中最优解退化的可能,是求解车辆路径问题的一种有效的算法。  相似文献

抗菌肽对凡纳滨对虾生长和机体免疫的影响   总被引:1,自引:0,他引:1  
基础饲料(对照组)中添加10g/kg抗菌肽制剂喂养体重0.10±0.01g、体长2.17±0.11cm的凡纳滨对虾(Penaeus vannamei)30d,进行10d的水浴攻毒(鳗弧菌10~8/mL),探讨饲料中添加抗菌肽对凡纳滨虾生长、成活率、免疫保护率,以及肌肉中溶菌酶活性、超氧化物歧化酶活性、总抗氧化能力的影响。结果表明:饲料中添加抗菌肽可显著提高凡纳滨对虾相对增重率、成活率和对致病菌的免疫保护率(P<0.05),但对饲料系数无显著影响(P>0.05);未受外界微生物刺激时,饲料中添加抗菌肽对凡纳滨对虾总抗氧化能力有促进作用(P<0.05),受外界微生物刺激后,饲料中添加抗菌肽喂养凡纳滨对虾可提高对虾超氧化物歧化酶活性(P<0.05)和总抗氧化能力(P<0.01)。因此饲料中添加抗菌肽喂养凡纳滨对虾可提高其机体非特异性免疫。  相似文献

【目的】研究复方中草药对虹鳟(Oncorhynchus mykiss)肝脏免疫功能及肠道免疫相关基因的影响。【方法】将红枣提取物、山药提取物和黄芪提取物按照质量比1∶1∶1混合,分别按质量分数0(对照)、0.3%、0.6%和1.2%添加到虹鳟基础饲料中,饲喂体长7.6~8.9 cm的虹鳟幼鱼56 d。用副溶血弧菌(Vibrio parahemolyticus)对幼鱼进行攻毒试验(5.48×106 cfu/尾),4 d后,取其肝脏和肠道,测定肝脏的生理生化指标,用半定量PCR及荧光定量PCR测定肠道免疫相关基因的表达。【结果】与对照组比较,肝脏的总抗氧化能力(T-AOC)显著升高(P<0.05),溶菌酶(LZM)水平极显著升高(P<0.01),酸性磷酸酶(ACP)、过氧化氢酶(CAT)活性显著降低(P<0.05),碱性磷酸酶(ALP)活性极显著降低(P<0.01),0.6%剂量组的丙二醛(MDA)极显著降低(P<0.01),而SOD无显著差异(P>0.05);各复方中草药组肠道的肿瘤坏死因子(TNF-α)及1.2%剂量组转化生长因子-β(TGF-β)转录水平极显著降低(P<0.01),0.3%和0.6%剂量组的补体3(C3)转录水平显著升高(P<0.05)。【结论】复方中草药(红枣+山药+黄芪)可提高感染副溶血弧菌的虹鳟非特异性免疫功能。  相似文献

溶藻弧菌疫苗对凡纳滨对虾免疫功能的影响   总被引:3,自引:0,他引:3  
应用溶藻弧菌疫苗饲喂凡纳滨对虾,测定了弧菌疫苗对凡纳滨对虾免疫指标的影响以及 疫苗的免疫保护作用。结果发现,投喂弧菌疫苗后,溶菌活力在12~20d内迅速增强,在20d时 活力达到最高,VF1组高于VF2和VF3组,但VF2和VF3之间差异不显著;凝集效价在投喂后的 第8天开始升高,在14d时达到最高,实验组的凝集活力均高于对照组,但实验组之间无明显的差 异;而超氧化物歧化酶(SOD)活力和溶血活力变化不明显。以每克饲料含疫苗细胞1.0×109、 1.0×108和1.0×107个的浓度投喂,对凡纳滨对虾的免疫保护率分别为66.7%、46.7%和13.3%。 可见弧菌疫苗对凡纳滨对虾的免疫保护率随疫苗浓度的增大而升高。  相似文献

从溶藻弧菌中提取肽聚糖,以0.05、0.1、0.15 mg/mL 3种浓度为注射凡纳滨对虾后,测定血淋巴中的血细胞数目、酚氧化酶(PO)、超氧化物歧化酶(SOD)、一氧化氮合成酶(NOS)的活性和虾青素的含量,探讨溶藻弧菌肽聚糖对凡纳滨对虾虾青素含量、免疫指标、免疫保护率的影响。结果表明:注射不同质量浓度的溶藻弧菌肽聚糖都对血清中的血细胞数目、PO、SOD、NOS活性影响显著(p值﹤0.05),对虾青素的含量影响极显著(p值﹤0.01);虾青素的含量与血细胞数目、PO、NOS活性变化趋势无明显的相似性,与SOD活性变化有相似的变化趋势;0.05 mg/mL组的免疫保护率为40%,0.1 mg/mL组为53.33%,0.15 mg/mL组为66.67%。溶藻弧菌肽聚糖可有效地增强凡纳滨对虾非特异免疫力,提高对溶藻弧菌的抗病力,虾青素含量与SOD活性的高低变化趋势相似。  相似文献

提出一种基于非劣排序的多目标优化免疫遗传算法。算法基于非劣排序对种群进行分类来评价个体的价值,在选择操作中引入个体浓度保持种群多样性。采用免疫克隆操作为产生新种群和算法实现全局搜索提供了基础,采用遗传种群与父代群体锦标赛竞争的方式保留最优解。仿真实验结果表明:算法在收敛性和分布性方面要优于NSGA-II算法。  相似文献

Effects of hypoxia on the dopamine concentration and the immune response of White Shrimp Litopenaeus vannamei were studied. The results showed that hypoxia had significant effects on the concentration of dopamine (DA) in the haemolymph, haemocyte count, phenoloxidase activity, phagocytic activity of haemocytes and bacteriolytic and antibacterial activity in the haemolvmph (P<0.05). The concentration of the dopamine in haemolymph reached its maximum in the 3.0 and 1.5mgL-1 DO groups at 12h and 6h, and then returned to normal after 24h and 12h, respectively. All immune parameters decreased with the reduction of dissolved oxygen. Total haemocyte count (THC), the hyaline cells and semi-granular cells in the 3.0mgL-1 DO group became stable after 12h,while granular cells did so after 24h, The THC and different haemocyte count (DHC) in the 1.5mgL-1 DO group became stable alter 24h. Phenoloxidase activity and bacteriolytic activity in the 3.0 and 1.5mgL-1 DO groups reached their stable levels after 24 h and 12 h respectively, while phagocytic activity and antibacterial activity became stable after 24 and 12, and 36 and 24 It, respec-tively. It was also indicated that the changes of dopamine concentrations in haemolymph, haemocyte count and phenoloxidase activ-ity were obviously related to the exposure time under hypoxic conditions.  相似文献

为研究不同添加量的包被壳寡糖对刺参生长和免疫的影响,在基础饲料中分别添加质量分数0.00%、0.75%、1.50%、2.25%及3.00%包被壳寡糖配制成实验饲料,养殖初始质量(5.03±0.03)g刺参(Apostichopus japonicus)60 d。结果表明:1.50%添加组刺参增重率和特定生长率高于对照组(P0.05),各添加组体壁水分、粗蛋白、粗脂肪、粗灰分含量差异不显著(P0.05),1.50%添加组吞噬活性显著高于对照组(P0.05),1.50%添加组的呼吸爆发活力在各时间点较对照组有显著提高(P0.05),其体腔上清液LZM活性于实验60 d较对照组有显著提高(P0.05),体腔上清液SOD活性于实验50、60 d较对照组有显著提高(P0.05);各组ACP活性、NO活性、补体C_3活性和PO活性无显著性差异(P0.05)。饲料中添加1.50%包被壳寡糖能提高刺参生长性能,对刺参的免疫能力具有一定促进作用。  相似文献

采用水体中常见污染物——苯酚对奥尼罗非鱼进行刺激,研究不同浓度苯酚对奥尼罗非鱼非特异性免疫功能的影响。奥尼罗非鱼分别暴露于0.005、0.025、0.125、0.625、3.125 mg/L等5个浓度的苯酚中8周,每两周采集抗凝血液和血清,用于检测非特异性免疫指标。结果表明:暴露于低浓度苯酚组(0.005 mg/L和0.025 mg/L)的奥尼罗非鱼NBT(氯化硝基四氮唑蓝)阳性细胞数等非特异性免疫指标相对于对照组无显著性差异(P>0.05),中浓度苯酚组(0.125 mg/L和0.625 mg/L)和高浓度苯酚组(3.125mg/L)对罗非鱼非特异性免疫指标的影响存在显著的时间效应和剂量效应(P<0.05),可减少NBT阳性细胞数、抑制超氧化物歧化酶活力和抗菌活力;促使溶菌酶活力和溶菌酶含量先升后降。表明中高浓度苯酚组对奥尼罗非鱼具有明显的免疫毒性。  相似文献

To study immune mechanism of fish lymphocyte we performed a proliferation assay and ELISA using monoclonal antibody against human IL-2. The result showed that an interleukin-2 (IL-2)-like factor was detected in the supernatant of plant haemoglutinin (PHA)-stimulated lymphocyte culture from peripheral blood, spleen and head kidney of olive flounder,Paralichthys olivaceus. The quantities of IL-2-like factor in the supernatant from different lymphoid tissues were quite different. The IL-2 like factor in the supernatant from cultured head kidney lymphocytes was much higher than those of peripheral blood lymphocytes and spleen lymphocytes (P<0.01). The IL-2 activity was found in either mouse thymocyte proliferation assay or flounder head kidney lymphocyte proliferation assay and shown to have obvious enhancing effect on proliferation of the above two types of cell. The recombinant human IL-2, (rhIL-2) was able to stimulate flounder thymocyte proliferation and used to detect the IL-2 receptor (IL-2R) on the surface of flounder lymphocyte. The cross-reaction between the lymphocytes of flounder peripheral blood and CD25(IL-2R) was detected with flow cytometry and shown that the percentage of CD25-positive cell in peripheral blood was 7.74±0.67%. Extracts of 21 marine invertebrates belonging to Coelenterata, Mollusca, Annelida, Bryozoa,Echiura, Arthropoda, Echinodermata and Urochordata were screened for the studies on their antitumor and immune regulation activities. Antitumor activity was determined by MTT method and immune regulation activity was studied using T- and B-lymphocytes in mice spleen in vitro. It was found that the n-butanol part of Asterina pectinifera, the acetic ether part of Tubuaria marina, 95% ethanol extract of Acanthochiton rubrolineatus have a high inhibition rate of 96.7%, 63.9% and 50.5% respectively on tumor cell line HL-60 at the concentration of 0.063 mg/ml. The inhibition rate of the acetic ether part of Tubuaria marina on the tumor cell line A-549 is 65,4 % at concentration of 0.063 mg/mL. The 95% ethanol extract of Meretrix meretrix has so outstanding promoting effect on T-lymphocytes that their multiplication increases 25% when the sample concentration is only 1μg/ml. On B-lymphocytes, the 95% extract of Rapana venosa, at concentration of 100μg/ml, has a promotion percentage of 60%. On the other hand, under the condition of no cytotoxic effect, the 95% ethanol extracts of Acanthochiton rubrolineatus and Cellana toreum can reach 92% inhibition rate on T lymphocyte at concentration of 100μg/ml, while the inhibition rate on B lymphocyte of the 95% extract of Acanthochiton rubrolineatus reaches 92% at the same concentration.  相似文献

采用溶藻弧菌ATCC17749菌体作为抗原免疫新西兰大白兔,制备兔抗溶藻弧菌多克隆抗体;利用柠檬酸钠还原氯金酸法制备胶体金及胶体金-抗体结合物,并将其喷涂在玻璃纤维膜上冷冻干燥;在硝酸纤维素膜上包被兔抗溶藻弧菌多克隆抗体和羊抗兔IgG分别作为检测线和控制线;将处理后的玻璃纤维素膜、硝酸纤维膜、样品垫及吸水纸组装成双抗夹心试纸条,并对试纸条灵敏度、特异性、稳定性进行评价。结果表明,制备的兔抗溶藻弧菌多克隆抗体的效价可达1∶512 000;胶体金溶胶的最佳制备条件为:加入1.8 mL 10 mg/mL的柠檬酸三钠,500 W微波炉中火加热10 min;试纸条检测线和质控线的抗体最佳包被量分别为8.0 mg/mL和1.0 mg/mL;试纸条检测灵敏度为1×106cfu/mL,特异性试验显示,试纸条与哈氏弧菌、副溶血弧菌、霍乱弧菌、拟态弧菌、美人鱼弧菌、弗氏弧菌、河流弧菌、创伤弧菌等8株菌无交叉反应;稳定性试验表明,试纸在4℃干燥条件下能稳定保存5个月以上。制备的溶藻弧菌多克隆检测试纸条可灵敏、较特异地检测源自病鱼的溶藻弧菌,整个检测过程可在10 min内完成,适用于溶藻弧菌的快速检测。  相似文献

Using western immunoblotting, we obtained heat-shock protein 70 (HSP70) induction data and distribution in different tissues from shrimp Fenneropenaeus chinensis during thermal and immune-challenged stresses. This is probably the first report of the effects of various stressors on the expression of HSP70 in shrimp.HSP70 was prominently induced in hepatopancreas and gills, but not in muscle, eyestalk and hemolymph, when the shrimp were exposed to heat shock and Vibrio anguillavium-challenged stresses. Cold shock and WSSV treatment had no significant effects on the levels of HSP70 expression in all tissues examined. HSP70 induction was greatest after 2 h exposure to heat shock stress, which was elevated after acute heat shock exposure of 10℃ above ambient temperature.  相似文献

制备无乳链球菌(Streptococcus agalactiae)临床分离株ZJ02的灭活疫苗,采用注射和浸泡2种方法,研究疫苗对吉富罗非鱼(Oreochromis niloticus)的免疫保护效果和血清抗体效价。注射组以4个不同浓度的白油佐剂灭活疫苗免疫罗非鱼,浸泡组以相同免疫剂量的灭活疫苗分别与消旋山莨菪碱、脂肪醇聚氧乙烯醚和氯化钠3种佐剂免疫罗非鱼。结果显示,无乳链球菌疫苗对罗非鱼具有较强的免疫原性,与对照组相比,免疫组血清中的抗体效价水平均显著性提高(p<0.05),注射组和浸泡组的抗体效价最高分别达到1∶256和1∶128;注射组中不同剂量的疫苗免疫组免疫保护率较高,高浓度组可达到95%,浸泡组中使用3种佐剂的免疫组免疫保护率分别为59%、64%、55%。表明无乳链球菌灭活疫苗对罗非鱼具有很好的免疫保护性,免疫剂量在5×108~4×109cfu/mL之间对注射免疫效果有显著性影响,消旋山莨菪碱、脂肪醇聚氧乙烯醚和氯化钠对浸泡免疫效果具有不同的免疫保护率。  相似文献

探索含CpG基序的未甲基化寡脱氧核苷酸(ODNs)对鱼类的免疫调节,将人工合成的鱼免疫刺激剂寡脱氧核苷酸序列1670(含一个CpG基序)、1670-D(含两个CpG基序)加入到离体培养的异育银鲫的头肾巨噬细胞和外周血白细胞中,采用MTT法测定其对鲤外周血白细胞增殖的影响,NBT还原法和Griess试剂显色法测定对头肾巨噬细胞的呼吸爆发的影响。结果显示,1670可显著(浓度为20.0μg·mL-1)或极显著(浓度为0.1、1.0、10.0μg·mL-1)提高异育银鲫巨噬细胞的氧呼吸爆发活性,但对氮呼吸爆发活性和外周血白细胞的增殖无显著作用;1670-D可显著(浓度为0.1μg·mL-1)或极显著(浓度为1.0、10.0、20.0μg·mL-1)增强异育银鲫巨噬细胞的氧呼吸爆发活性和外周血白细胞的增殖,且可极显著(浓度为1.0、10.0、20.0μg·mL-1)提高异育银鲫巨噬细胞的氮呼吸爆发活性。表明特定序列、特定作用剂量的CpG-DNA可增强体外培养的异育银鲫巨噬细胞和外周血白细胞的非特异性免疫应答。  相似文献
