The toxicity of seven major HAB (harmful algal bloom) species/strains, Prorocentrum donghaiense, Phaeocystis globosa, Prorocentrum micans, Alexandrium tamarense (AT-6, non-PSP producer), Alexandrium lusitanicum, Alexandrum tamarense (ATHK) and Heterosigma akashiwo were studied against rotifer Brachionus plicatilis under laboratory conditions. The results show that P. donghaiense, P. globosa, P. micans, A. tamarense (AT-6), or A. lusitanicum could maintain the individual survival and reproduction, as well as the population increase of the rotifer, but the individual reproduction would decrease when exposed to these five algae at higher densities for nine days; H. akashiwo could decrease the individual survival and reproduction, as well as population increase of the rotifer, which is similar to that of the starvation group, indicating that starvation might be its one lethal factor except for the algal toxins; A. tamarense (ATHK) has strong lethal effect on the rotifer with 48h LC50 at 800 cells/mL. The experiment on ingestion ability indicated by gut pigment change shows that P.donghaiense, P. globosa, P. micans, A. tamarense (AT-6) and A. lusitanicum can be taken by the rotifers as food, but A. tamarense (ATHK) or H. akashiwo can be ingested by the rotifers. The results indicate that all the indexes of individual survival and reproduction, population increase, gut pigment change of the rotifers are good and convenient to be used to reflect the toxicities of HAB species. Therefore, rotifer is suggested as one of the toxicity testing organisms in detecting the toxicity of harmful algae.  相似文献

We present results on the effect of modified clay on cyst formation of Scrippsiella trochoidea in harmful algal bloom (HAB). Modified clay (in concentration of 0, 0.1, 0.5, and 1.0 g/L) were added to cultures, and observations were made on cysts of S. trochoidea under controlled laboratory conditions. Results indicate that the removal rate of algal cells reached 97.7% at the clay concentration of 1.0 g/L. The cyst formation rate increased from 4.6% to 24.6% when the concentration of clay was increased from 0 to 1.0 g/L. Two cyst metamorphs were observed: spinal calcareous cysts and smooth noncalcareous ones. The proportion of the spinal cysts decreased from 76.9% to 24.1% when clay concentration increased from 0 to 1.0 g/L. In addition, modified clay affected cyst germination. The germination rate decreased with the increases in the clay concentrations. Non-calcareous cysts had a lower germination rate with a longer germination time. We conclude that modified clay could depress algal cell multiplication and promote formation of temporal cysts of S. trochoidea, which may help in controlling HAB outbreaks.  相似文献

Fractal dimensions of flocs between clay particles and HAB organisms   总被引:1,自引:0,他引:1  
The impact of harmful algal blooms (HABs) on public health and related economics have been increasing in many coastal regions of the world. Sedimentation of algal cells through flocculation with clay particles is a promising strategy for controlling HABs. Previous studies found that removal efficiency (RE) was influenced by many factors, including clay type and concentration, algal growth stage, and physiological aspects of HAB cells. To estimate the effect of morphological characteristics of the aggregates on HAB cell removal, fractal dimensions were measured and the RE of three species of HAB organism, Heterosigma akashiwo, Alexandrium tamarense, and Skeletonema costatum, by original clay and modified clay, was determined. For all HAB species, the modified clay had a higher RE than original clay. For the original clay, the two-dimensional fractal dimension (D2) was 1.92 and three-dimensional fractal dimension (D3) 2.81, while for the modified clay, D2 was 1.84 and D3 was 2.50. The addition of polyaluminum chloride (PACl) lead to a decrease of the repulsive barrier between clay particles, and resulted in lower D2 and D3. Due to the decrease of D3, and the increase of the effective sticking coefficient, the flocculation rate between modified clay particles and HAB organisms increased, and thus resulted in a high RE. The fractal dimensions of flocs differed in HAB species with different cell morphologies. For example, Alexandrium tamarense cells are ellipsoidal, and the D3 and D2 of flocs were the highest, while for Skeletonema costatum, which has filamentous cells, the D3 and D2 of flocs were the lowest.  相似文献

A bstract Cadmium(Cd) is one of the most common and widespread heavy metals in the environment. Cd has adverse effects on photosynthesis that are countered by photosystem I(PSI) and photosystem II(PSII); however, the protective responses of these photosystems to heavy metal stress remain unclear. Using the model diatom P haeodactylum tricornutum, a biological indicator that is widely used to assess the impact of environmental toxins, we simultaneously measured the effects of Cd on PSI and PSII and examined the levels of pigments in response to high light treatments before and after Cd exposure. Cd significantly reduced the quantum yield and electron transport rates of PSI and PSII. The quantum yield of non-photochemical energy dissipation in PSI due to donor side limitation increased faster than the quantum yield due to acceptor side limitation. The Cd treatment activated the P. tricornutum xanthophyll cycle under artificial light conditions, as indicated by an increased diatoxanthin content. Xanthophyll is important for photoprotection; therefore, the accumulation of diatoxanthin may down-regulate PSII activities to reduce oxidative damage. Together, our results suggest that the rapid reduction in PSII activities following Cd exposure is an adaptive response to heavy metal stress that reflects the variable exposure to external stressors in the native P. tricornutum environment.  相似文献

Blooms of Akashiwo sanguinea frequently break out around the world, causing huge economic losses to the aquaculture industry and seriously damaging coastal ecosystems. However, the formation mechanisms of A. sanguinea blooms remain unclear. We investigated the allelopathic effects of A. sanguinea on multiple phytoplankton species, explored the mode of allelochemicals action and the way of nutrient factors regulation of the allelopathic activity. Results show that strains of A. sanguinea could inhibit the growth of co-occurring phytoplankton including Scrippsiella trochoidea, Phaeocystis globosa, and Rhodomonas salina, but inhibition of Prorocentrum micans was not obvious. The inhibition rates on phytoplankton were positively correlated with the cell densities of A. sanguinea. The highest inhibition rate of 94% on R. salina was for A. sanguinea CCMA256 culture of 2 000 cells/mL at 72 h. We observed that cells of S. trochoidea, Ph. g lobosa, and R. sali na were lysed when co-cultured with A. sanguinea, with the shortest time for S. trochoidea. Additionally, the growth rates of A. sanguinea were promoted by coculturing with S. trochoidea, Ph. globosa, and R. salina. Four components of A. sanguinea culture were all able to inhibit growth of R. salina:the strongest inhibitory effect was found in the sonicated culture, followed by whole-cell culture, filtrates of sonicated culture, and filtrate culture. The crude extract of A. sanguinea culture also lysed cells of R. salina, and the inhibition rates on R. salina increased with the increasing dose of crude extract. It was shown that both nutrient enrichment and nitrogen:phosphorus ratio imbalance enhanced remarkably the allelopathic activity of A. sanguinea. The highest inhibition rate on R. salina of 70% occurred in A. sanguinea JX13 treatment at 2 000 cells/mL under high nutrient condition in 48 h. In JX14 treatment at 2 000 cells/mL for N:P of 10:1, the inhibition rate increased by 1.7 times of that for N:P of 20:1. In addition, the allelopathy of A. sanguinea could not only be a competitive strategy but also a nutrition strategy, playing an important role in formation and/or maintenance of blooms of the mixotrophic dinoflagellate A. sanguinea.  相似文献

Phaeocystis globosa is an important harmful algal bloom causative species distributing widely in temperate and tropical coastal waters in the world.The morphological,physiological,and biochemical characte ristics are different amo ng geographic strains,which can not be distinguished with nuclear ribo somal DNA markers at present.Therefore,the genetic distance and phylogeographic relationships of nuclear 28 S rDNA D1-D2 and ITS regions,and three chloroplast intergenic spacers(petN-trnS1,trnM1-psbA,and rbcS-rpl27) were analyzed and compared among 13 strains of P.globosa isolated from the Pacific Ocean and Atlantic Ocean in this study.In addition,the nucleotide polymorphisms of 28 S rDNA D1-D2,ITS,and rbcS-rpl27 regions were evaluated in two P.globosa strains.The various levels of nucleotide polymorphism were in the nuclear 28 S rDNA D1-D2 region and ITS region,but no polymorphism was in the chloroplast rbcS-rpl27 intergenic spacer.A reasonable intraspecific phylogeographic relationship was presented by rbcS-rpl27 intergenic spacer,which had the strongest distinction to geographic strains compared to those of 28 S rDNA D1-D2 and ITS regions.In the phylogenetic tree of rbcS-rpl27 intergenic spacer,the two strains from the North Sea of the Atlantic Ocean were divided firstly from the species of P.globosa,and then formed an independent clade,while the other Atlantic strains and all of Pacific strains j oined up to build the other clade.It was implied that at least two genetically distant populations of P.globosa existed in the Atlantic coastal regions.This study provided a high-resolution chloroplast marker to analyze intraspecific phylogeographic populations of P.globosa,and preliminarily clarified the genetic relationships of the Pacific and Atlantic strains of P.globosa.  相似文献

Cells of Haematococcus pluvialis Flot. et Will were collected in four different growth phases. We quantified the initial and total enzyme activity of ribulose-1,5-bisphosphate carboxylase (Rubisco) in crude extracts, and the relative expression of large-subunit ribulose-1,5-bisphosphate caboxylase / oxygenase (rbcL) mRNA. We measured the ratio of photosynthetic rate to respiration rate (P/R), maximal effective quantum yield of photosystem II (Fv/Fm), electron transport rate (ETR), actual photochemical efficiency of PSII in the light (ΦPSII), and non-photochemical quenching (NPQ). Green vegetative cells were found to be in the most active state, with a relatively higher P/R ratio. These cells also displayed the lowest NPQ and the highest Fv/Fm, ETR, and ΦPSII, indicating the most effective PSII. However, both Rubisco activity and rbcL mRNA expression were the lowest measured. In orange resting cysts with relatively lower P/R and NPQ, Rubisco activity and rbcL expression reached a peak, while Fv/Fm, ETR, and ΦPSII were the lowest measured. Taking into account the methods of astaxanthin induction used in industry, we suggest that Rubisco may participate in astaxanthin accumulation in H. pluvialis. A continuous and sufficient supply of a carbon source such as CO2 may therefore aid the large scale production of astaxanthin.  相似文献

Using modified clay is one of the most promising methods for the mitigation of harmful algal blooms(HABs). However, the environmental impact of modified clay has become a subject of global concern. In this study, turbot(Scophthalmus maximus L.) embryos were used as a model to assess the effect of modified clay on this sensitive stage of fish development. Results show that the 24 and 48 h LC_(50)(median lethal concentrations) of the modified clay were 1.70 and 1.65 g/L, and the safe concentration was 0.47 g/L, which is much higher than the concentration used to treat HAB. The modified clay did not affect significantly the hatchability of turbot embryos but when the concentration exceeded 0.50 g/L, the deformity rate of newly hatched larvae increased significantly. The total length, specific growth rate(SGR)and yolk sac absorption rate of larvae reached their peaks at 0.50 g/L and then gradually decreased as the concentration of modified clay increased. Therefore, a moderate amount of modified clay does not harm the survival and hatching of turbot embryos, or the growth and development of newly hatched larvae.  相似文献

Harmful algal blooms(HABs),caused by the overgrowth of certain phytoplankton species,have negative effects on marine environments and coastal fisheries.In addition to cell-counting methods using phytoplankton nets,a hydroacoustic technique based on acoustic backscattering has been proposed for the detection of phytoplankton blooms.However,little is known of the acoustic properties of HAB species.In this study,as essential data to support this technique,we measured the acoustic properties of two HAB species,Akashiwo sanguinea and Alexandrium affine,which occur in the South Sea off the coast of Korea.Due to the small size of the target,we used ultrasound for the measurements.Experiments were conducted under laboratory and field conditions.In the laboratory experiment,the acoustic signal received from each species was directly proportional to the cell abundance.We derived a relationship between the cell abundance and acoustic signal received for each species.The measured signals were compared to predictions of a fluid sphere scattering model.When A.sanguinea blooms appeared at an abundance greater than 3 500 cells/mL,the acoustic signals varied with cell abundance,showing a good correlation.These results confirm that acoustic measurements can be used to detect HAB species.  相似文献

We investigated the effects of dried macroalga Gracilaria lemaneiform (Rhodophyta) on photosynthesis of the bloom-forming microalga Chaetoceros curvisetus. C. curvisetus was cultured with different amounts of dried G. lemaneiformis under controlled laboratory conditions. We measured the photosynthetic oxygen evolution rate and established the chlorophyll a fluorescence transient (OJIP) curve coupled with its specific parameters. We observed concentration-dependent and time-dependent relationships between dried G. lemaneiformis and inhibition of photosynthesis in C. curvisetus. Co-culture with dried G. lemaneiformis also resulted in a decrease in the light-saturated maximum photosynthetic oxygen evolution rate (P~ax) in C. curvisetus, and a decrease in the OJIP curve along with its specific parameters; the maximum photochemical efficiency of PSII (FJFm), the amount of active PSII reaction centers per excited cross section at t=0 and t=--tFM (RC/CS0 and RC/CSm, respectively), the absorption flux per excited cross section at t=0 (ABS/ CS0), and the efficiency with which a trapped exciton moves an electron into the electron transport chain (~u0). The dark respiration rate (Rd) increased in C. curvisetus co-cultured with dried G. lemaneiformis. The JIP-test and the oxygen evolution results indicated that dried G. lemaneiJbrmis decreased the number of active reaction centers, blocked the electron transport chain, and damaged the oxygen-evolving complex of C. curvisetus. This result indicated that dried fragments of G. lemaneiformis could effectively inhibit photosynthesis of C. curvisetus, and thus, could serve as a functional product to control and mitigate C. curvisetus blooms.  相似文献

In recent years, harmful algal blooms (HABs) have occurred frequently along the coast of China, and have been exhibiting succession from diatom- to dinoflagellate-dominated blooms. To examine the effects of different diatom and dinoflagellate HABs, the life history parameters of rotifers (Brachionus plicatilis Müller) were measured after exposure to different concentrations of HAB species. The HAB species examined included a diatom (Skeletonema costatum) and four dinoflagellates (Prorocentrum donghaiense, Alexandrium catenella, Prorocentrum lima and Karlodinium veneficum). Compared with the control treatment (CT), the diatom S. costatum showed no adverse impacts on rotifers. Exposure to dinoflagellates at densities equivalent to those measured in the field resulted in a reduction in all the life history parameters measured. This included a reduction in: lifetime egg production (CT: 20.34 eggs/ind.) reduced to 10.11, 3.22, 4.17, 7.16 eggs/ind., life span (CT: 394.53 h) reduced to 261.11, 162.90, 203.67, 196 h, net reproductive rate (CT: 19.51/ind.) reduced to 3.01, 1.26, 3.53, 5.96/ind., finite rate of increase (CT: 1.47/d) reduced to 1.16, 1.03, 1.33, 1.38/d, and intrinsic rate of population increase (CT: 0.39/d) reduced to 0.15, 0.03, 0.28, 0.32/d, for the dinoflagellates P. donghaiense, A. catenella, P. lima and K. veneficum, respectively. The results showed that the diatom S. costatum had no detrimental consequences on the reproduction and growth of B. plicatilis, however, the four dinoflagellates tested did show adverse effects. This suggests that dinoflagellate HABs may suppress microzooplankton, resulting in an increase in algal numbers.  相似文献

Water samples were collected in the coastal area of the Changjiang Estuary on four cruises from August 2002 to May 2003. The seasonal variations of dissolved inorganic arsenic (DIAs) distributions were analyzed. The results showed that the distributions of DIAs were mainly influenced by Water (KSSW). The concentration of the total dissolved the terrestrial input and the intrusion of the Kuroshio Subsurface inorganic arsenic (TDIAs) decreased consecutively from winter to summer, while it increased in autumn. The distributions of TDIAs showed some relationships with salinity and suspended particulate matter (SPM). The relationships between DIAs speciation (including arsenite [ As( Ⅲ ) ] and arsenate [ As( Ⅴ ) ]), biological activity and the availabilities of the phosphate were investigated in the study area for the cruise August 2002. The ratio of As (Ⅲ)/TDIAs increased with the decrease of phosphate concentrations. In the bottom water, the As( Ⅲ )/TDIAs ratio decreased with the increasing of N/P. The concentration of TDIAs decreased 28.7% approximately after the occurrence of harmful algal blooms (HAB) because of the uptake of arsenate by algae. Further study is needed about the arsenic source/sink relationships in their vertical or horizontal profiles and the uptake mechanism during the occurrence of harmful algal blooms.  相似文献

Harmful algal blooms (HABs) have led to extensive ecological and environmental issues and huge economic losses. Various HAB control techniques have been developed, and biological methods have been paid more attention. Algicidal bacteria is a general designation for bacteria which inhibit algal growth in a direct or indirect manner, and kill or damage the algal cells. A metabolite which is strongly toxic to the dinoflagellate Alexandrium tamarense was produced by strain DH46 of the alga-lysing bacterium Alteromonas sp. The culture conditions were optimized using a single-factor test method. Factors including carbon source, nitrogen source, temperature, initial pH value, rotational speed and salinity were studied. The results showed that the cultivation of the bacteria at 28°C and 180 r min?1 with initial pH 7 and 30 salt contcentration favored both the cell growth and the lysing effect of strain DH46. The optimal medium composition for strain DH46 was determined by means of uniform design experimentation, and the most important components influencing the cell density were tryptone, yeast extract, soluble starch, NaNO3 and MgSO4. When the following culture medium was used (tryptone 14.0g, yeast extract 1.63g, soluble starch 5.0 g, NaNO3 1.6 g, MgSO4 2.3 g in 1L), the largest bacterial dry weight (7.36 g L?1) was obtained, which was an enhancement of 107% compared to the initial medium; and the algal lysis rate was as high as 98.4% which increased nearly 10% after optimization.  相似文献

Overthelastseveraldecades,harmfulalgalblooms(HABs)havebecomeaseriousenvironmental problem in many parts of the world. A rapid and accurate detection process for HAB algae has yet to be developed. Heterosigma akashiwo is one of the most important HABs species in China. The objective of this study was to develop an immunologic technique that can rapidly and sensitively count H. akashiwo cells. Five HABs species (Alexandrium catenella, Thalassiosira sp., Cryptomonas sp., Alexandrium tamarense and Symbiodinium sp.,) were used in this study to evaluate the analysis process we developed. A polyclonal antibody with high titers against H. akashiwo was obtained by injecting H. akashiwo cells into rabbits. Immuno-magnetic beads (IMB) were produced via conjugated polyclonal antibodies with magnetic beads and applied to isolate and count H. akashiwo cells from the culture. Results show that 66.7%-91.6% of the cells were captured from unialgal culture by IMBs, and only 5.3%-12.5% of the four other HAB microalgae species were captured, indicating that the constructed IMBs combined specifically with the H. akashiwo cells. At the same time, flow cytometry (FCM) sorting was exploited to screen H. akashiwo cells after labeling with FITC conjugated polyclonal antibodies. Using the FCM technique, 91.7% of the targeted cells were sorted out from mixed microalgae samples in just a few minutes. These results indicate that both antibody-involved IMB and antibody-based FCM techniques are highly effective at detecting and quantifying HAB species. These techniques, especially immuno-magnetic separation, have low associated cost, and are fast and simple processes compared with other techniques currently in use.  相似文献

In contrast, Heterosigma akashiwo had an adverse effect on B. plicatilis population and its growth was largely unaffected by rotifer grazing. Although biofilm formation may promote growth, biofilms are not always beneficial to their hosts. The biofilm formation characteristics of Bacillus cereus WPySW2 and its changes at different temperatures were studied. Results show that B. cereus WPySW2 promoted the growth of Neoporphyra haitanensis (an economically cultivated seaweed) at 20 °C but accelerated algal rot at 28 °C. Thicker B. cereus WPySW2 biofilms covered the surface of N. haitanensis thalli at 28 °C, which hindered material exchange between the algae and surrounding environment, inhibited algal photosynthesis and respiration, and accelerated algal decay. Compared with planktonic bacteria, mature biofilm cells had lower energy consumption and metabolic levels. The biofilm metabolic characteristics of B. cereus WPySW2 changed significantly with temperature. High temperature accelerated biofilm maturation, which made it thicker and more stable, allowing the bacteria to easily adapt to environmental changes and obtain greater benefits from their host. High temperature did not affect the production or increased the abundance of toxic metabolites, indicating that the negative effects of B. cereus WPySW2 on algae were not caused by toxins. This study shows that increased temperature can transform a harmless bacterium into a detrimental one, demonstrating that temperature may change the ecological function of phycospheric bacteria by affecting their morphology and metabolism.
