Heavy oil contamination is one of the most important environmental issues. Toxicities of polycyclic aromatic hydrocarbons (PAHs), including immune toxicities, are well characterized, however, the immune toxic effects of heavy oil, as a complex mixture of PAHs, have not been investigated. In the present study, we selected Japanese flounder (Paralichthys olivaceus) as a model organism, and observed alteration of immune function by the exposure to heavy oil. To analyze the expression profiles of immune system-related genes, we selected 309 cDNAs from our flounder EST library, and spotted them on a glass slide. Using this cDNA array, alteration of gene expression profiles was analyzed in the kidneys of flounders exposed to heavy oil. Six Japanese flounders (mean body weight: 197 g) were acclimated to laboratory conditions at 19–20 °C. Three fish were exposed to heavy oil C (bunker C) at a concentration of 3.8 g/L for 3 days, and the others were kept in seawater without heavy oil and used as the control. After the exposure period, the fish were transferred into control seawater and maintained for 4 days, and then they were dissected and their kidneys were removed. Total RNA was extracted from the kidney samples to use in gene expression analyses. The microarray detected alteration of immune system-related genes in the kidneys of heavy oil-exposed flounders, including down-regulation of immunoglobulin light chain, CD45, major histocompatibility complex class II antigens and macrophage colony-stimulating factor precursor, and up-regulation of interleukin-8 and lysozyme. These results suggest that pathogen resistance may be weakened in heavy oil-exposed fish, causing a subsequent bacterial infection, and then proinflammatory genes may be induced as a defensive response against the infection. Additionally, we found candidate genes for use as biomarkers of heavy oil exposure, such as N-myc downstream regulated gene 1 and heat shock cognate 71 kDa proteins.  相似文献

As basic research for the effect of heavy oil on the fish immune system, in this study, the number of leukocyte was counted in Japanese flounder Paralichthys olivaceus, after exposure to heavy oil at a concentration of 30 g/8 L for 3 days. To compare the numbers of bacteria in the skin mucus between oil-exposed and control fish, viable bacteria were enumerated by counting colony forming unit (CFU). Compared with 5.79 ± 1.88 × 107 leukocytes/mL in the controls, the exposed fish demonstrated higher counts, averaging 1.45 ± 0.45 × 108 cells/mL. The bacterial numbers of control fish were 4.27 ± 3.68 × 104 CFU/g, whereas they were 4.58 ± 1.63 × 105 CFU/g in the exposed fish. The results suggest that immune suppression of the fish occurred due to heavy oil stressor, and bacteria could invade in the mucus, resulting in the increasing leukocyte number to prevent infectious disease.  相似文献

Sea microlayer (SML) and subsurface water (SSW) samples were collected around Xiamen Island to study the enrichment and partitioning of polycyclic aromatic hydrocarbons (PAHs). Total PAH concentrations ranged from 93.43 to 411.05 ng L−1 in the SML and 49.29–279.42 ng L−1 in the SSW. Compared with the results of previous studies before pollution control measurements, PAHs levels decreased significantly. The enrichment factors (EFs) of dissolved and particulate PAHs varied from 0.68 to 2.71 and 0.43–3.56. EFs showed the consistent enrichment trends with sites and exhibited different enrichment characteristics between 2 and 3 ring PAHs and 4 ring PAHs. Furthermore, the much higher concentrations of BaP (strong carcinogenicity) were accompanied by higher EFs in the SML samples from the Western Xiamen Harbour, which together indicated the risk of impacts to the fish eggs that usually float on the SML water after exposure to oil spills and combustion, contributed directly by the port and shipping activities.  相似文献

The concentrations of 16 polycyclic aromatic hydrocarbons (PAHs) were determined in the sediment from the Hormuz strait, Persian Gulf. The sum of 16 PAHs (ΣPAH) concentrations varied from 72.17 to 277.77 ng g−1 dry weight, with an average value of 131.20 ± 59.29 ng g−1 dry weight. An ecological risk assessment of PAHs, indicated that adverse biological effects caused by acenaphthene and acenaphthylene occasionally may take place in the sediment of Hormuz strait. PAH source identification showed that the PAHs in the sediments come from pyrogenic and mixed origin. Based on classification of pollution levels, sediments from Hormuz strait could be considered as low to moderately polluted with PAHs.  相似文献

The concentrations of 16 polycyclic aromatic hydrocarbons (PAHs) were determined in mullet (Liza klunzingeri), water and sediment from northern part of Hormuz strait (Persian Gulf). The concentration levels of total PAHs in L. klunzingeri, water and sediment were 133.99–268.57 ng g−1 dry weight, 3.12–5.88 ng l−1 and 42.29–228.9 ng g−1 dry weight, respectively. Based on isomer ratios, analysis of the PAHs source in the sediment demonstrated that the PAHs come from pyrogenic and petrogenic origin. Risk assessment showed PAHs threshold concentrations to occasionally be exceeded in the study area.  相似文献

The molecular distribution and compound specific stable carbon and hydrogen isotope ratios were measured on solvent extractable hydrocarbons from the Late Paleoproterozoic McArthur River, or “Here's Your Chance” (HYC) Pb/Zn/Ag ore deposit in the Northern Territory of Australia. Five samples were collected from the McArthur River mine on a northeast–southwest transect in order to sample a gradient of hydrothermal alteration. One sample was taken from the unmineralized W-Fold Shale unit immediately below the HYC ore deposit. δD of n-alkanes, branched alkanes and bulk aromatic fractions were measured, and δ13C of n-alkanes, polynuclear aromatic hydrocarbons (PAHs) and bulk kerogen were measured to assess the isotopic effects of a varying degree of hydrothermal alteration on different components of HYC organic matter (OM). Relative to n-alkanes in Barney Creek Formation sediments that did not undergo mineralization, HYC n-alkanes are enriched in deuterium (D) by 50–60‰. This is likely to be a result of equilibrium hydrogen exchange during ore genesis with a highly D-enriched fluid that originated in an evaporitic basin. Trends with distance along the sample transect are ambiguous, but from the northernmost to southernmost point, n-alkanes are less D-enriched, and PAHs are less abundant and less 13C-enriched. This could be due to decreasing hydrothermal alteration effects, decreasing delivery of highly altered OM by the mineralizing fluid, or both. The carbon isotopic composition of HYC PAHs is inconsistent with a Barney Creek Fm source, but consistent with an origin in the underlying Wollogorang Formation. PAHs are 13C-depleted relative to n-alkanes, reflecting a kerogen source that was 13C-depleted compared to n-alkanes, typical for Precambrian sediments. PAHs are more 13C-depleted with increasing molecular weight and aromaticity, strengthening the case for a negative isotopic effect associated with aromatization in ancient sediments. Together, these data are consistent with a an ore deposition model in which fluids originated in an evaporitic deposit lower in the basin and interacted with metals and OM in the Tawallah Group at temperatures above 250 °C and a depth of ~ 6 km before ascending along a flower structure associated with the Emu Fault and cooling to 200 ± 20 °C before reaching Barney Creek sediments.  相似文献

Polybrominated diphenyl ethers (PBDEs) are now found ubiquitously in the aquatic environment and biota, and there is a growing concern that PBDEs may disrupt endocrine systems, leading to reproductive impairments of aquatic animals. In our study, zebrafish (Danio rerio) were exposed to the 5 ng/L, 1 μg/L and 50 μg/L of DE-71 for the duration of the whole life cycle (120 days, from eggs to adults). The expression of selected genes along the brain–pituitary–gonadal (BPG) axis and liver, and the levels of plasma sex hormones were examined.In male fish, up-regulation of GnRH in brain, FSHβ and LHβ in pituitary, FSH-receptor, LH-receptor, and CYP19a in testis was clearly evident, while down-regulation of CYP11a and 3β-HSD was found in testis. In female fish, a 2.4-fold up-regulation of 3β-HSD was found in ovary upon exposure to 50 μg/L of DE-71. GnRH in brain, FSHβ and LHβ in pituitary were also up-regulated, while ERβ, TH and TPH in brain and GnRH-receptor in pituitary were significantly down-regulated. Hepatic ERα, AR and VTG in males were all down-regulated, while hepatic ERα and AR in female were up-regulated.Serum estradiol (E2) was reduced in both male and female upon exposure to DE-71, while significant increases in serum testosterone (T) and 11-keto-testosterone (11-KT) were only found in male but not female fish. The ratio of T/E2 as well as the ratio of 11-KT/E2 in male fish increased in a dose-dependent manner upon exposure to DE-71. Our overall results showed that whole life exposure of DE-71 altered the expression of regulatory genes and receptors at all three levels of the BPG axis in zebrafish, and the responses are sex dependent. The observed disruption of GnRH and GtHs can be further related to the subsequent disruption in both levels and balance sex steroid hormones.  相似文献

Polychlorinated biphenyls (PCBs) are known as neurotoxic chemicals and possibly alter animal behavior. We previously reported that PCB-exposure induced abnormal schooling behavior in Japanese medaka (Oryzias latipes). This abnormal behavior might be caused by the functional alteration of central or terminal nervous system. To understand the mechanism(s) of behavioral change by PCB-exposure, we analyzed the gene expression profiles in the brain of medaka exposed to 3,3′,4,4′,5-pentachlorobiphenyl (PCB126) or a PCB mixture (Kanechlor-400: KC-400) using a cDNA microarray that we constructed. Twelve FLF-II strain medaka (six individuals per treatment) were dietary exposed to PCB126 (0.01 μg/g b.w./day) or KC-400 (1 μg/g b.w./day) for three weeks. For the control, six fish were fed a control diet. After the exposure period, fish were dissected, and the brain samples were collected. The samples from control fish were pooled and used as a common reference in the microarray experiment. Microarray data were normalized by the LOWESS method, and we screened the genes whose expression levels were altered more than 1.5-fold. Gene expression profiling showed 97 down-regulated and 379 up-regulated genes in the brain of medaka exposed to PCB126. KC-400 exposure suppressed 15 genes and induced 266 genes in medaka brain. Among these genes, the expression levels of 7 and 188 genes were commonly down- or up-regulated, respectively in both treatment groups. On the other hand, 31 gene expressions were significantly different between PCB126 and KC-400 treatment groups, and three out of 31 genes were received opposite effects. In addition, the microarray data showed that thyroid hormone-responsive genes were up-regulated by PCB-exposure, which may imply that PCBs or their metabolites mimic thyroid hormone effects in the brain of PCB-exposed medaka.  相似文献

Sediment samples were collected from 12 locations of Kaohsiung Harbor, Taiwan and analyzed for polycyclic aromatic hydrocarbons (PAHs). Total PAH concentrations varied from 472 to 16,201 ng g−1 dry weight. The highest PAH concentrations were from the industrial zone docks situated in south Kaohsiung Harbor. Diagnostic ratios showed that the possible source of PAHs in the industrial zone dock could be coal combustion while in the other zones it could be petroleum combustion. The toxic equivalent concentrations (TEQcarc) of PAHs varied from 55 to 1964 ng TEQ g−1 dry weight. Higher total TEQcarc values were found at industrial zone docks (from 1404 to 1964 ng TEQ g−1 dry weight). As compared with the US Sediment Quality Guidelines (SQGs), the observed levels of PAHs at industrial zone docks exceeded the effects range low (ERL), and could thus cause acute biological damage. However, the lower levels of PAHs at the other zones would probably not exert adverse biological effects.  相似文献

A comparative study of 23 PAH congeners in sediment of the Caspian Sea coast and Anzali Wetland was conducted in 2010. Surface sediment was analyzed using chromatography and mass spectrometry. Total PAH concentrations ranged between 212 and 9009 ng g1 dw. Spatial distribution maps revealed that PAH levels were higher in the coastal areas of the Caspian Sea where oil related activities have been common since 1800’s. Diagnostic ratios analysis indicated that PAHs largely originated from petrogenic processes. PAH toxicity level was assessed using sediment quality guidelines and toxic equivalent concentrations to determine toxic effects on marine organism. Based on these investigations, in our study areas, the probability of toxicity for benthic organisms is “low to medium”. The toxic equivalent concentrations of carcinogenic PAHs varied between 11 and 231 ng TEQ/g; higher total toxic equivalent concentrations values were found in the coastal areas of the Caspian Sea.  相似文献

The contamination of surface sediments in Guan River Estuary, China, by polycyclic aromatic hydrocarbons (PAHs) has been fully investigated. Total concentrations of 21 PAHs ranged from 90 to 218 ng/g with an average of 132.7 ng/g, which is relatively low in comparison with other estuaries around the world. PAH concentrations appeared to be positively correlated with clay content and negatively correlated with sediment grain size. Source identification implied that the PAHs originated mainly from pyrolytic sources. However, source patterns may be continuously changed to a petrogenic origin due to the heavy ship traffic and continuous discharge of oily sewage in this area. The PAH levels were also compared with international Sediments Quality Guidelines and Sediments Quality Criteria, and the results indicated low negative effects for most individual PAHs. However, toxic effects related to FLO would occur occasionally in most locations in the estuary.  相似文献

It has been well known that oil spills cause serious problems in the aquatic organisms. In particular, some species of teleosts, which develop on the sea surface thought to be affected by heavy oil (HO). During the embryogenesis, the nervous system is constructed. Therefore, it is important to study the toxicological effects of HO on the developing neurons. We exposed HO to eggs of Japanese flounder (Paralichthys olivaceus) and investigated the neural disorder. In larvae exposed by HO at the concentration of 8.75 mg/L, the facial and lateral line nerves partially entered into the incorrect region and the bundle was defasciculated. Furthermore, in the HO-exposed larvae, Sema3A, a kind of axon guidance molecule, was broadly expressed in second pharyngeal arch, a target region of facial nerve. Taken together, we suggested the possibility that the abnormal expression of Sema3A affected by HO exposure causes disruption of facial nerve scaffolding.  相似文献

In January 2006, 25 tonnes of heavy fuel oil spilled into the Port of Gladstone in Queensland, Australia, from the breached hull of a bulk carrier ship. Over the following days, approximately 18 tonnes of the oil was recovered, however a certain amount of oil was deposited in the intertidal areas of Port Curtis leaving a highly visible, viscous residue. The objectives of this research were to assess the immediate impacts on the intertidal habitat and to gain baseline information for future comparative assessments.Sediment PAH and metal concentrations, mangrove communities and intertidal macroinvertebrates were assessed within one month post-spill at oil-impacted sites; adjacent sites which were not visibly impacted; and reference sites which were located outside the recorded distribution of the oil spill. Highest PAH concentrations were found at the impacted sites, with concentrations of some PAHs exceeding Australian and New Zealand Sediment Quality Guidelines (ANZECC/ARMCANZ, 2000). These sites contained very few or no crab holes in the high intertidal area, indicating a low crab density in comparison to reference sites. Little immediate impact was evident on the mangrove and macrobenthic communities, however future surveys may show evidence of longer-term impacts on these communities.  相似文献

Seawater samples (including surface water and bottom water) were collected from the Western Taiwan Strait (WTS) during June 24–25, 2009; polycyclic aromatic hydrocarbons (PAHs) in dissolved phase and particulate phase were analyzed, respectively. The results showed that the total concentrations of PAHs in the dissolved phase and particulate phase were ranged from 12.3 to 58.0 ng L−1, and 10.3–45.5 ng L−1, which showed a low-middle contamination level in the China Seas. The spatial variability of PAHs may be related to the complicated currents of WTS, especially the Min-Zhe coastal current. PAHs diagnostic ratios suggested that PAHs mainly originated from the inputs of pyrolytic (combustion) sources, which might be contributed to land-based atmospheric deposition. The particle-water partition coefficients of individual PAH showed that partitions were not correlated with suspended particulate matter content, dissolved organic carbon or salinity, similar to the Yangtze coastal area.  相似文献

Dietary intake is one of the important routes of human exposure to brominated flame retardants (BFRs) such as polybrominated diphenyl ethers (PBDEs). The use of PBDEs may also result in exposure to polybrominated dibenzo-p-dioxins and dibenzofurans (PBDDs/DFs), as these compounds are impurities in technical mixtures of BFRs and can also be formed unintentionally by the same processes that generate chlorinated dioxins. This study determined the concentrations of polybrominated compounds in common seafood in Guangzhou and Zhoushan, and assessed the health risks of these chemicals via consumption of contaminated seafood. Seafood samples (fish, bivalves, shrimp, crab, and cephalopods) purchased from local markets in 2003 and 2004 were analyzed for PBDEs and PBDDs/DFs. The highest concentration of total PBDEs (46.3 ng g−1 lipid wt.) was detected in fish from Guangzhou, in which BDEs 47 and 209 were the two predominant congeners. The total daily intakes of PBDEs, PBDDs, and PBDFs were, 946, 6.39, and 6.54 pg kg−1 body weight (bw) in Guangzhou, and 489, 4.99, and 7.65 pg kg−1 bw in Zhoushan, respectively. The hazard ratios for PBDDs and PBDFs were both greater than unity, indicating that these compounds may pose some health risks to the local population.  相似文献

The up-to-date concentration of polycyclic aromatic hydrocarbons (PAHs) in sediment materials of Victoria Harbour was investigated so as to evaluate the pollution potential associated with the reclamation projects in Hong Kong. A total of 100 sediment samples were collected at 20 locations. Except the control point in reservoir, the PAHs concentrations were detectable levels all sites (131–628.3 ng/g, dw) and such values were higher than Dutch Target and Intervention Values (the New Dutch standard in 2016). The PAHs concentration indicating that construction waste and wastewater discharges were the main pollutant sources. Results of correlation in single cell gel electrophoresis assay (comet assay) studies also revealed that the PAHs concentration was highly correlated (< 0.01) with DNA migration (i.e. the length of tail moment of fish cells) in 5 mg/ml of PAHs. The above observation indicates that the PAHs present in the sediment may substantially effect the marine ecosystem. Although the dredged sediment can be a useful sea-filling material for land reclamation; however, the continuing leaching of PAHs and its impact on the aquatic environment need to be studied further.  相似文献

Raindrops falling on the sea surface produce turbulence. The present study examined the influence of rain-induced turbulence on oil droplet size and dispersion of oil spills in Douglas Channel in British Columbia, Canada using hourly atmospheric data in 2011–2013. We examined three types of oils: a light oil (Cold Lake Diluent - CLD), and two heavy oils (Cold Lake Blend - CLB and Access Western Blend - AWB). We found that the turbulent energy dissipation rate produced by rainfalls is comparable to what is produced by wind-induced wave breaking in our study area. With the use of chemical dispersants, our results indicate that a heavy rainfall (rain rate > 20 mm h? 1) can produce the maximum droplet size of 300 μm for light oil and 1000 μm for heavy oils, and it can disperse the light oil with fraction of 22–45% and the heavy oils of 8–13%, respectively. Heavy rainfalls could be a factor for the fate of oil spills in Douglas Channel, especially for a spill of light oil and the use of chemical dispersants.  相似文献
