Norwegian coastal waters are subject to PAH contamination from electrochemical industries such as aluminium smelters. Evidence of PAH exposure has been established in fish and bivalves. The present study tests the applicability of a novel crustacean PAH exposure biomarker to a PAH contamination gradient in the field (Karmsund Strait, SW Norway). Fluorescence analysis of urine samples collected from crabs at each site revealed 1-OH pyrene "equivalent" levels (indicative of pyrogenic PAH contamination) decreased with increasing distance from a point source of pyrogenic PAH (a large aluminium works). The assay was shown to be suitable for the detection of PAH exposure in wild crustacean populations, for discriminating between contaminated and clean sites and is also sufficiently sensitive to detect gradients of PAH contamination. The method provides a rapid, inexpensive and non-destructive measure of biologically available PAH in crustaceans.  相似文献

A population of killifish (Fundulus heteroclitus) inhabiting a creosote-polluted inlet of the Elizabeth River demonstrates tolerance to the acute toxic effects exerted by contaminated sediments on reference site killifish. Previous data have suggested that upregulated antioxidant defenses contribute to short-term tolerance in killifish exposed to Elizabeth River sediments. This study investigated population differences in antioxidant defenses from wild caught Elizabeth River and reference population killifish in different seasons, and after being held in the laboratory. Parameters measured in the killifish were total glutathione concentrations (GSH(T)), activities of glutathione reductase (GR), glutathione peroxidase (GPx), and lipid peroxidation (LPO), all in adult hepatic tissues. The Elizabeth River population exhibited greater GSH(T), higher GPx activities, and increased LPO as compared to the reference population. Sex specific population differences were also observed in GSH(T) and GPx. Both populations displayed decreased GSH(T) and increased GR from early to late summer, as well as after being held in the laboratory. This study indicates that there are many factors that may contribute to differences in levels of antioxidant defenses in addition to exposure to contaminants, including reproductive status and environmental conditions.  相似文献

Mummichogs, Fundulus heteroclitus, an estuarine fish with a relatively small home range found along the eastern coast of the United States are well-suited to monitoring contaminant effects, including those of polycyclic aromatic hydrocarbons (PAHs). One of the common PAHs in estuaries is pyrene. We report here on efforts to develop multiple biomarkers of pyrene exposure in this species. Adult male mummichogs were exposed in the laboratory to the weak aryl hydrocarbon receptor (AhR) agonist pyrene at 0, 30, or 50 microg/L in 7-day static renewal exposures. The RNA was extracted from livers and alterations in mRNA expression were assessed by subtractive hybridization and differential display in order to produce multiple biomarkers of pyrene exposure. Genes demonstrating differential expression were confirmed by quantitative-PCR (Q-PCR) and include cytochrome P-450 1A (CYP1A), a putative hepatocyte growth factor activator, a X-ray inducible retrotransposon, and several expressed sequenced tags (ESTs). Some of these genes represent new biomarkers of pyrene exposure and potential biomarkers of PAH exposure. Therefore, similar changes were investigated at a Superfund site in Charleston, SC. Mummichogs from a creosote contaminated site and from a reference site (North Inlet National Estuarine Research Reserve near Georgetown, SC) were trapped, RNA extracted from the livers, and Q-PCR performed. Many of the genes differentially expressed following pyrene exposure were not altered at the creosote contaminated site in comparison to the reference site. However, CYP1A and an EST were induced. CYP1A induction at Diesel Creek indicates that this population of fish does not demonstrate refractory CYP1A phenotypes observed at several sites with high levels of AhR agonists. Ultimately, we anticipate that the use of multiple biomarkers of PAH exposure will provide useful information on the potential effects of toxicants.  相似文献

Feral Atlantic cod (Gadus morhua) and corkwing wrasse (Symphodus melops) were investigated for polyaromatic hydrocarbon (PAH) contamination in the Karmsund strait, western Norway. This strait is highly contaminated with PAHs, and a main source is the chronic release of gas-scrubbing effluents from a local aluminium works. In both species, the level of biliary PAH metabolites and hepatic DNA adducts were higher in fish collected near the aluminium works. Interestingly, a significantly higher level of both biliary PAH metabolites and hepatic DNA adducts was found in corkwing wrasse as compared to cod, indicating a higher potential for genotoxic effects in this species. Hepatic cytochrome P4501A (CYP1A) in cod estimated by ethoxyresorufin-O-deethylase and an immunoassay technique (ELISA), seemed to be weakly induced at the contaminated sites. At the most contaminated site, skin ulcers and fin erosion were detected in about 70 and 45% of the cods, respectively. The data demonstrated that both cod and corkwing wrasse may be suitable target species for PAH pollution monitoring.  相似文献

As part of a multinational workshop on marine environmental quality, sediments were collected from seven sites in Vancouver Harbour and analyzed for polynuclear aromatic hydrocarbons (PAHs) and organochlorines (OCs), and for trace metals. English sole were collected from five sites, and muscle was analyzed for trace metals and liver for OCs. As expected, sediment PAH and OC concentrations and tissue OC concentrations were higher at sites east of the First Narrows, compared to the outer harbor and reference sites. Sediment PAH concentrations east of the First Narrows were similar to concentrations at moderately contaminated sites in Puget Sound, south of Vancouver Harbour. In contrast, concentrations of OCs in sediments and tissue were low to moderate, even at relatively contaminated sites within Vancouver Harbour. Although several trace metals in sediments were higher than in contaminated sediments from Puget Sound, trace metals measured in fish muscle were lower.  相似文献

Previous studies showed that adult and 6-month-old young-of-year Fundulus heteroclitus from a polluted site have enlarged thyroid follicles and (in adults) higher thyroxine levels compared to fish from an unpolluted reference site. This study's purpose was to discover when in development these differences begin to appear. For 15-20 mm fish, the average follicle size of the polluted population was significantly larger than that of the reference population, similar to the pattern found in older fish. However, for the smaller size (12-14.5 mm), the average follicle size of fish from the polluted site was significantly smaller. Although not statistically significant, radioimmunoassay measurements of thyroxine levels were consistent with follicle differences: in larger sizes (20-25 and 15-20 mm) fish from the polluted site had higher thyroxine levels but the smaller size (12-14.5 mm) had lower levels. Therefore, the thyroid abnormalities seen in adults begin when the fish are approximately 15-20 mm long. However, the follicle size reversal for the 12-14.5 mm range may also indicate abnormality at that developmental stage.  相似文献

Sewage treatment works (STWs) are a common source of chemicals entering into the aquatic environment. In order to assess effects of these effluents on oxidative stress parameters in aquatic organisms, we caged rainbow trout at five sites: upstream, near an STW effluent, and three sites downstream in the river Viskan in western Sweden for 14 days during autumn, 2006. We then measured protein carbonyls in plasma as well as 20S proteosome activity and lipid peroxidation products, i.e. MDA and 4-HNE, in liver samples. Levels of both lipid and protein oxidative damage products were elevated in fish caged near the STW effluent while 20S activity showed no differences. This argues that complex mixtures of chemicals entering into the aquatic environment do have deleterious effects on fish. Additionally, oxidative stress parameters can serve as a biomarker in aquatic organisms.  相似文献

Fixed wavelength fluorescence (FF) of bile was used as a biomarker for PAH exposure in fish caged adjacent to an aluminium works. After 30 days of caging, a 20–50 fold increase of pyrene fluorescence was found in groups caged adjacent to the works as compared to two control groups. The PAH uptake occurred mainly from suspended particles rather than from PAH contaminated sediments at the caging sites. Our results demonstrates the combined use of fish caging and bile-fluorescence measurements as a powerful field model for the monitoring of coastal waters subjected to PAH pollution.  相似文献

Halogenated aromatic hydrocarbons (HAHs) and polynuclear aromatic hydrocarbons (PAHs) are major environmental contaminants. Fish species that are chronically exposed to these compounds can develop resistance to their toxic effects. In all fish species studied to date, toxicant resistance has been accompanied by decreased inducibility of the xenobiotic metabolizing enzyme, cytochrome P450 1A (CYPIA). CYP1A induction is mediated through the Aryl Hydrocarbon Receptor (AHR). Although these compounds mediate their effects through this pathway, there have been resistant populations in which one chemical class cannot induce CYPIA expression (HAHs) while the other (PAHs) can. Resistance to PAHs was examined in a HAH-resistant population of Fundulus heteroclitus collected from a site contaminated with both compound classes (Newark Bay, NJ). Fish were injected intraperitoneally with the HAH 3,4,3',4'-tetrachlorobiphenyl (PCB77), benzo[a]pyrene (B[a]P, a PAH) or vehicle and sacrificed after 2 (B[a]P) or 5 days (PCB77, vehicle). We found no significant increase in CYP1A mRNA levels in resistant Newark Bay F. heteroclitus treated with either B[a]P or PCB77, while there was a 3.9 fold (PCB77) and 4.2 fold (B[a]P) increase in CYP1A mRNA in Flax fish relative to controls. AHR labeling studies revealed significantly (P < 0.05) lower levels of hepatic AHR in Newark fish (1,770 +/- 1,693.2 DPM) relative to Flax fish (6,082.5 +/- 1,709.9 DPM). Overall, these data suggest Newark F. heteroclitus are resistant to both PAHs and HAHs at the level of CYP1A mRNA, which might be mediated, in part, though lower expression of AHR. We are currently studying the promoter sequence to determine its role in chemical resistance.  相似文献

Mummichogs (Fundulus heteroclitus) from Piles Creeks (PC), New Jersey (a polluted site), are sluggish and show poorer prey capture and predator avoidance than reference fish from Tuckerton (TK). The behavioral dysfunction of the PC fish may be associated with thyroid impairment due to endocrine disruption. In this study, we compared thyroid histology and thyroid hormones in the two populations and determined experimentally whether the polluted environment could alter thyroid hormone levels. PC fish had larger thyroid follicles, greater follicle cell heights, and contained higher plasma thyroxine (T4) levels than TK fish. However, there were no significant differences in either plasma or tissue triiodothyronine (T3). TK fish held in simulated PC environments had higher plasma T4 and lower plasma T3 than field-sampled fish. PC fish held in clean water had lower plasma T4 and T3 than field-sampled fish. In either case, there was no significant difference in tissue T3 content. The contaminants in PC alter thyroid structure and function, which may relate to the behavioral differences between fish from the polluted and reference sites.  相似文献

Mytilus edulis digestive gland microsomes were prepared from indigenous populations sampled from a clean reference site (Port Quin) and an urban-industrial contaminated site (Blackpool) in the UK. Samples were collected in March/April, May, August and December 1998. Western blot analysis was performed using polyclonal antibodies to fish CYP1A and rat CYP2E using partially purified M. edulis CYP as a positive control, to aid identification. CYP1A- and CYP2E-immunopositive protein levels showed different site-specific seasonal variation with higher levels of CYP2E determined in May (P < 0.05). At both sites, lower levels of CYP1A-immunopositive protein but not CYP2E-immunopositive protein were observed in the samples collected in December (P < 0.05). This correlated with lower levels of nuclear DNA damage (Comet assay expressed as per cent tail DNA) observed in December compared to August (P < 0.05).  相似文献

This study was conducted to test whether live cells from the liver of fish could be used to detect early changes that are indicative of pollutant-induced liver damage. Low-molecular-weight fluorescent molecular probes were inserted into isolated hepatocytes from dab (Limanda limanda) from a contaminated site and a reference site in the North Sea. These included bioprobes for endoplasmic reticulum (ER), cytochrome P-450-associated 7-ethoxyresorufin-O-deethylase (EROD), oxyradicals, reduced glutathione (GSH) and microtubules (MT). Endocytosis of Texas Red-albumin was used as an integrated indicator of hepatocyte performance. Findings showed increases in ER-associated fluorescence, EROD and oxyradical generation, with a marked decrease in endocytosis in hepatocytes from fish caught at the contaminated site. These results indicate that fish from the historically contaminated site were impacted by organic xenobiotics which induced the biotransformation system, radical production and cell injury.  相似文献

Physiological acclimation, genetic adaptation and genetic differentiation were examined in darter gobies (Gobionellus boleosoma) inhabiting a coastal marsh with a long history of PAH contamination. No acclimation was detected; a 2-week pre-exposure at the polluted site resulted in a decreased rather than an increased resistance in a subsequent laboratory exposure to polluted sediment. Similarly, fish collected from sites with elevated sediment PAH levels did not exhibit an increased resistance in bioassays with polluted sediment, confirming the lack of acclimation and indicating a lack of adaptation to the pollutants. No differences were detected in frequencies of allozyme genotypes when comparing gobies from the polluted area to those from a nearby control site, and overall levels of heterozygosity were similar in the two populations. While lack of UV-induced toxicity in the laboratory exposures could have played a role, at least four other factors may explain the apparent lack of adaptive responses and genetic differences. First, bioavailability of the contaminants to the darter goby could have been low, though other evidence contradicts this. Second, the contaminated marsh contains a large number of different chemicals and acclimation, adaptation and genetic differentiation are expected to be less likely when more contaminants are involved. Third, the hydrocarbon distribution at the contaminated marsh is very patchy, such that fish may avoid exposure to the highly-contaminated sediment. Fourth, gene flow may be sufficiently high in this mobile species to prevent local adaptation.  相似文献

This work evaluates the effects of caging, a known confinement stress, in Nile tilapia (Oreochromis niloticus) during an environmental study in Cubatão river, southern Brazil. Caging animals for 7 days, regardless of being at the reference or at a contaminated site, resulted in lower levels of antioxidant-related defenses (glutathione, glutathione S-transferase, glucose-6-phosphate dehydrogenase) in liver and physiological parameters (blood glucose and lactate) as compared with free-swimming animals. Higher hepatic glutathione reductase activity and elevated Hb content could be associated to contaminant exposure. In conclusion, the confinement stress in caged Nile tilapia biochemical and physiological disturbances, acting as a confounding factor in field studies.  相似文献

Mytilus edulis were collected from a reference site (Port Quin) and an urban/industrial contaminated site (New Brighton) in the UK during June 1999. Levels of PCBs (sigma7 congeners) and CB-138 were determined to be, respectively, 21 fold and 16 fold higher in the mussel digestive glands from New Brighton. Levels of CYPIA-immunopositive protein were 1.5 fold higher (P < 0.05) at the polluted site but the levels of DNA strand breaks were 1.3 fold higher (P<0.05) at the reference site. Mussels from Port Quin were placed in cages at both sites and both transplanted and indigenous populations sampled in September (13 weeks). Mussels transplanted from the reference site to the industrial site, reported elevated levels of CYP1A-immunopositive protein (1.4 fold; P < 0.05) and higher levels of DNA damage (1.2 fold; P < 0.05) compared to caged populations at the reference site and a PCB loading similar to the populations from the polluted site. Moreover, transplanted mussels had DNA damage 1.8 fold greater (P < 0.05) than indigenous mussels at the transplant site. These changes were small but significant when compared to the observed temporal changes in the indigenous populations.  相似文献

Acute (4 days) and chronic (14 days) effects of copper were evaluated on the antioxidant defenses of Laeonereis acuta (Polychaeta) collected in unpolluted (UP) and polluted (P) sites. In the acute assay (125 and 250 micro g Cu/l) superoxide dismutase (SOD) and glutathione S-transferase (GST) activities did not change, whereas catalase (CAT) increased in worms from both the sites. Lipid peroxidation was higher in copper exposed worms from the P site. In the chronic assay (62.5 micro g Cu/l) polychaetes from the P site showed enhanced activities of SOD, GST and CAT and higher contents of metallothionein-like proteins and sulfhydrils compared to worms from UP. Differences in responses between polychaetes from UP and P sites suggest that organisms from the polluted site, P, are more susceptible to oxidative stress conditions.  相似文献

DNA single-strand breaks were analysed in the blue mussel (Mytilus edulis L.) deployed in intertidal and subtidal zones in the PAH contaminated Reykjavík harbour and at a reference site, Hvalfj?rethur, Iceland. DNA strand breaks were analysed by Comet assay in isolated gill and haemocyte cells from six mussels from each site and depth. Increased DNA damage in both gill cells and haemocytes were observed in mussels deployed in Reykjavík harbour compared to the reference site. Intertidal mussels from Reykjavík harbour had higher DNA damage in haemocytes compared to subtidal mussels. The Comet assay seems to be useful for measuring genotoxic exposure in mussels from the field, and that DNA damage might be higher in the intertidal zone either due to higher exposure to contaminants or because of physiological and biochemical responses to variations in oxygen availability.  相似文献

The in vivo response of freshwater fish exposed to pollutants was assessed using two biomarkers, 7-ethoxyresorufin-O-deethylase (EROD) induction and DNA single strand breaks. Chub (Leuciscus cephalus) were caught in spring and in fall at various locations in the river Rhône watershed. EROD activity was measured in the liver while DNA damage was evaluated in chub erythrocytes using the recently developed Comet assay. Chemical contamination was evaluated both in fish muscle (PCBs) and in sediment (PCBs, PAHs, heavy metals) collected at each sampling station. Sex of individuals was shown to influence the level of EROD activity but not the level of DNA damage. The EROD activity as well as the DNA damage were found to be higher in the mostly contaminated stations compared to the reference one. This study shows that multibiomarker-based approach provides complementary informations about early effects in feral fish exposed to complex chemical pollution and highlights the interest of the Comet assay in genotoxicity assessment.  相似文献

As part of a joint workshop organised by the International Council for the Exploration of the Sea (ICES) and the Intergovernmental Oceanographic Commission (IOC) on biological effects monitoring techniques. dad (Limanda limanda) were examined from six spaced stations along a 200 km transect extending from near the mouth of the Elbe River out to the Dogger Bank in the North Sea. Based on historical data, differences in contaminant concentrations in sediments exist along the transect (lipophilic organic xenobiotics at the inshore sites and heavy metals offshore over the Dogger Bank). The most contaminated site sampled was the most inshore, the least contaminated was approximately midway along the transect, with contamination building up again over the farthest point along the transect, over the Dogger Bank. Multiple organs and tissues were examined for full pathology from each fish sampled. Only liver data are presented here (11–20 livers per station). The most significant lesions were considered to be well-developed foci of cellular alteration, high mitotic activity and high neutral lipid accumulation in livers from dab sampled from the most inshore site examined. Livers from the least-contaminated station showed minimal evidence of such changes. Foci of cellular alteration and neutral lipid accumulation were also seen in dab liver sampled from fish from the Dogger Bank site. Thus far, the hepatic changes seen correlate well with the most contaminated sites along the transect. The value of comprehensively examining the histopathology of an organ of toxicological significance, such as the liver in a European species of flatfish, is demonstrated.  相似文献
