The Ria Formosa lagoon is an ecosystem whose water quality reflects the anthropogenic influence upon the surrounding areas. In this lagoon, the clam Ruditapes decussatus has a great economical importance and has been widely used as a biomonitor. A multibiomarker approach (δ-aminolevulinic acid dehydratase, metallothionein, lipid peroxidation, acetylcholinesterase, alkali-labile phosphates, DNA damage) was applied to assess the environmental quality of this ecosystem and the accumulation of contaminants and their potential adverse effects on clams. Clams were sampled in different shellfish beds in the period between July 2007 and December 2008 and abiotic parameters (temperature, salinity, pH and dissolved oxygen of seawater and organic matter in the sediment), condition index, metals (Cd, Cu, Zn, Ni, Pb), TBTs and PAHs concentrations were measured in clam tissues. Data was integrated using Principal Component Analyses and biomarker indices: IBR (Integrated Biomarker Response) and HSI (Health Status Index). This multibiomarker approach enabled discrimination of a time and space trend between sites with different degrees of anthropogenic contamination, identifying one of them (site 2) as the most stressful and summer months as the most critical period for clams due to an increase of environmental stress (anthropogenic pressure along with extreme environmental conditions, e.g. temperature, dissolved oxygen, organic matter in the sediments, etc). The selected biomarkers provided an integrated response to assess the environmental quality of the system, proving to be a useful approach when complex mixtures of contaminants occur.  相似文献

During the last 15 years of the 20th century, several researchers carried out extensive sampling of benthic macrofauna communities in the Ria Formosa lagoon. The main objective of this paper is to discern the main communities of benthic fauna of the Ria Formosa and the leading stressors that limit them based on this large data set.The high species richness and high diversity values observed at most of the stations or groups of stations indicate a good ecological status, with the exception of the sewage-impacted areas and the physically stressed areas. It was possible to define three main stressors acting on the Ria Formosa. The most important is the physical stress imposed by the limited water renewal in some areas of the Ria Formosa. The second is the nutrient and organic matter pollution that is limited to the vicinity of the sewage outfall stations and to some locations of the Ria Formosa. This is due to the dual effects of the low residence time of water, but also due to a degradation of water quality in places of relatively restricted water circulation but with high primary production, symptoms of eutrophication. The third stressor is the natural land–ocean gradient, denoting the influence of seawater exchanges and emersion time, attenuated by the high water exchange rates of Ria Formosa.It was possible to differentiate the anthropogenic stress from the natural physical stress through the application of multivariate analysis. However, both structural (species richness and diversity indices) and functional indicators (trophic composition) indicate the same trend – low species richness, low diversity indices, and a community dominated by detritivores species at the locations characterized by high anthropogenic impact or by physical stress. The physically stressed areas were dominated by chironomid larvae and hydrobiid gastropods and the most polluted areas were dominated by oligochaetes, mainly tubificids.  相似文献

Brackish water ecosystems are often exposed to wide variations in environmental variables, including temperature and salinity, which may cause strong selective pressures on organisms modifying the genetic patterns of species. The aim of this work was to test whether there is a ‘divergence-with-gene flow’ in coastal lagoon populations of white seabream (Diplodus sargus) (Ria Formosa, S Portugal and Mar Menor, SE Spain) respect to four marine populations, by using partial sequences of cyt b mitochondrial gene and information from nine microsatellite loci. Genetic diversity was highest in both coastal lagoons (Mar Menor and Ria Formosa) considering mitochondrial and nuclear markers. Although some of FST population pairwise comparisons were not significant, analyses of molecular variance (AMOVAs) detected differences between groups (coastal lagoon and marine) close to significance. Also, only two haplotypes (Cytb-17 and Cytb-18) were detected in both coastal lagoon sampling sites and these localities (Mar Menor and Ria Formosa) showed the highest number of singletons, some of them with a high number of mutations, as has been already described for other Mar Menor populations (Pomatochistus marmoratus and Holothuria polii). Also, several tests detected significant positive and balancing selection considering mtDNA and microsatellite data. These data support the hypothesis of selection as one of the drivers of the genetic differences found between coastal lagoon and marine populations. The life strategy adopted by Diplodus sargus in coastal lagoons allows it to decrease its mortality rate and improve the heritability of its genes. Also, the increase time spent in coastal lagoons with different temperatures and salinities favours the fitness selection and the maintenance of exclusive haplotypes and genotypes in coastal lagoon inhabitants favouring the ‘divergence-with-gene-flow’.  相似文献

The plant reproductive effort, the seed germination rate and the seedling survival and development of Zostera marina (eelgrass) were assessed in four populations (Fuzeta, Culatra, Barrinha and Armona) at the species’ southern distribution limit in the Eastern Atlantic, the Ria Formosa lagoon. Germinated seeds were individually placed in Petri dishes with natural sandy sediments and kept in a culture chamber at the same temperature and salinity conditions as the natural environment. In addition, seeds from three different depths of Fuzeta population were cultivated in outdoor mesocosms. The populations of Fuzeta and Barrinha showed higher seed production and the seeds produced were heavier than the other populations. The germination of the seeds both in the laboratory and in the outdoor tanks began c. 8–12 weeks after the collection of the flowering shoots at a water temperature of 22 °C. The spontaneous germination in the laboratory (2.4–5.3%) and in the mesocosm experiment (5.6–8.9%) was low and from all the germinated seeds (n = 20) only three reached the seedling stage. The spontaneously germinated seeds from Fuzeta survived for a longer period than those from Barrinha, but only the germinated seeds of Barrinha reached the seedling stage (one‐leaf seedling stage). In outdoor tanks, higher seed germination and earlier seedling emergence (2 weeks after seeding) and survival (for 208 days) occurred for the seeds obtained from the shallow meadow. The reproductive effort of Z. marina populations of Ria Formosa showed that flowering shoots and seed traits are site‐specific. The low reproductive success indicated by the low germination and seedling survival suggests a bottleneck in the species’ reproductive cycle that may account for the scarce presence of the species in Ria Formosa lagoon. The high water temperatures of Ria Formosa in winter may partly explain this bottleneck. Increased temperatures due to climate change may reduce even further the sexual reproduction of Z. marina in its southern distributional limit in the Eastern Atlantic.  相似文献

The abiotic disturbance of urban wastewater discharge and its effects in the population structure, plant morphology, leaf nutrient content, epiphyte load and macroalgae abundance of Zostera noltii meadows were investigated in Ria Formosa coastal lagoon, southern Portugal using both univariate and multivariate analysis. Four sites were assessed, on a seasonal basis, along a gradient from a major Waste Water Treatment Works (WWTW) discharge to a main navigation channel. The wastewater discharge caused an evident environmental disturbance through the nutrient enrichment of the water and sediment, particularly of ammonium. Zostera noltii of the sites closest to the nutrient source showed higher leaf N content, clearly reflecting the nitrogen load. The anthropogenic nutrient enrichment resulted in higher biomass, and higher leaf and internode length, except for the meadow closest to the wastewater discharge (270 m). The high ammonium concentration (158–663 μM) in the water at this site resulted in the decrease of biomass, and both the leaf and internode length, suggesting a toxic effect on Z. noltii. The higher abundance of macroalgae and epiphytes found in the meadow closest to the nutrient source may also affect the species negatively. Shoot density was higher at the nutrient-undisturbed site. Two of the three abiotic processes revealed by Principal Component Analysis were clearly related to the WWTW discharge, a contrast between water column salinity and nutrient concentration and a sediment contrast between both porewater nutrients and temperature and redox potential. A multiple regression analysis showed that these abiotic processes had a significant effect on the biomass-density dynamics of meadows and on the overall size of Z. noltii plants, respectively. Results show that the wastewater discharge is an important source of environmental disturbance and nutrients availability in Ria Formosa lagoon affecting the population structure, morphology and N content of Z. noltii. This impact is spatially restricted to areas up to 600 m distant from the WWTW discharge, probably due to the high water renewal of the lagoon.  相似文献

Though there is some information on cytotoxicity of copper nanoparticles and silver nanoparticles on human cell lines, there is no information on their genotoxic and cytotoxic behaviour in bivalve molluscs. The aim of this study was to investigate the genotoxic impact of copper oxide and silver nanoparticles using mussels Mytilus galloprovincialis. Mussels were exposed to 10 μg L−1 of CuO nanoparticles and Cu2+ and Ag nanoparticles and Ag+ for 15 days to assess genotoxic effects in hemocytes using the comet assay. The results obtained indicated that copper and silver forms (nanoparticles and ionic) induced DNA damage in hemolymph cells and a time-response effect was evident when compared to unexposed mussels. Ionic forms presented higher genotoxicity than nanoparticles, suggesting different mechanisms of action that may be mediated through oxidative stress. DNA strand breaks proved to be a useful biomarker of exposure to genotoxic effects of CuO and Ag nanoparticles in marine molluscs.  相似文献

Acoustic telemetry and standard tag-recapture were used to determine the home range and residency of juveniles and sub-adults of Diplodus sargus and Diplodus vulgaris in the Ria Formosa (Portugal) coastal lagoon. Maximum time between recaptures for the standard tag-recapture method was 128 days for D. sargus and 30 days for D. vulgaris. The majority of the fish were recaptured in the vicinity of the tagging location. Fish tagged with acoustic transmitters had a maximum period of time between first and last detections of 62 days for D. sargus and 260 days for D. vulgaris. Minimum convex polygons areas ranged between 148?024 m2 and 525?930 m2 for D. sargus and between 23?786 m2 and 42?134 m2 for D. vulgaris. Both species presented a high residency index between first and last detections. Two D. sargus tagged with acoustic tags were recaptured by fishermen outside the coastal lagoon at distances of 12 km and 90 km from the tagging position, providing evidence that this species leaves the Ria Formosa during the winter time for the adjacent coastal waters. The results of this study reinforce the importance of Ria Formosa as a nursery for D. sargus and D. vulgaris in the south coast of Portugal.  相似文献

Fish communities of the Ria Formosa coastal lagoon (south Portugal) were sampled on a monthly basis with a beach seine at 4 sites, during two different time periods: 1980–1986 and 2001–2002. Community indices, species ranking and multivariate analysis were used in order to identify changes in the fish community between the two time periods. A total of 153,511 fish representing 57 taxa were recorded. Although species composition was very similar for both sampling periods, multivariate analysis performed on annual species abundance in number and weight revealed differences in fish community structure between the two periods. Structural changes in fish community were related mostly to a sharp decrease in the abundance of Mugilidae from 1980–1986 to 2001–2002. These changes were probably associated to a decrease in organic matter contents and nutrients concentrations due to improvements in sewage treatment and better water circulation inside the lagoon. The changes in fish community structure are more evident in the inner areas of the lagoon than near the inlet. The association between changes in sewage patterns and changes in the ichthyofaunal community structure reinforces the importance of fish communities as a biological indicator of human induced changes in marine systems.  相似文献

We explore the use of the clam Tapes semidecussatus Reeves 1864 as an indicator for the presence of potentially genotoxic substances in estuarine sediments. The limitations associated with the interpretation of Comet assay data (expressed as % DNA in tail) in terms of clam reproductive state, size (age) and thermal exposure history following laboratory acclimation are discussed. Hatchery-reared clams, subjected to ambient temperature fluctuations during growth, were exposed in vivo under laboratory conditions for three weeks to sediment samples collected from a polluted site and a "clean" reference site. The DNA damage observed in haemocytes, gill and digestive gland cells was significantly higher in animals exposed to contaminated sediment compared to those exposed to sediment from the reference site. The extent of DNA damage recorded was not correlated with size (age). Spawning was not observed during the experiment. Nevertheless, clams with well-developed gonads showed a statistically higher degree of DNA damage in gill and digestive gland cells- but not haemocytes, demonstrating an increased sensitivity to potential genotoxic compounds, possibly caused by impaired DNA repair capacity due to reproductive activity. Furthermore, the degree of DNA damage in clams exposed to contaminated sediments was higher in autumn and winter compared to spring and summer, suggesting an effect of seasonal priming.  相似文献

The faunal communities of four intertidal habitats namely sand, mud, seagrass (Zostera noltii) and seagrass patches (mixSM) of a temperate coastal lagoon, Ria Formosa (southern Portugal), were sampled. A total of 47 species were taken in 428 bottomless drop sampler samples, with the highest number of species and the more commonly occurring species belonging to the Mollusca phylum. The dominance of these gastropod species underlines the importance of the grazing food chain in these habitats. Bittium reticulatum was the most abundant species, being especially abundant in the seagrass habitat. The most frequent and highest biomass species in the community was Carcinus maenas, a predator that makes use of the available resources and that is adapted to the highly variable intertidal environment. Pomatoschistus microps was the most abundant fish species, with highest densities in the mud habitat, which demonstrates an ability to occupy a low depth area. The seagrass habitat had the highest diversity, abundance and biomass, followed by the mixSM habitat and was different from all the others. Assemblages were highly influenced by the presence of vegetation, providing forage and refuge from predation. A well defined summer group was identified in all habitats. These results highlight the importance of seagrass beds and the idea that their decrease implies the decrease of lagoon production through the impoverishment of the trophic structure of the lagoon.  相似文献

The population dynamics of the seagrass Cymodocea nodosa was investigated in three meadows differently affected by the artificial relocation of an inlet in the western sector of the Ria Formosa lagoon, southern Portugal. One meadow was subjected to increasing currents that caused erosion, another one was subjected to an initial episode of burial and a third meadow was undisturbed by the inlet opening. The hydrodynamic/sedimentary condition of meadows was assessed through comparative sediment analysis between the meadow and the adjacent unvegetated area.  相似文献

A numerical model was used to predict toxic microalgal transport and dispersion in Ria de Aveiro in Portugal. A previously developed Lagrangean particle tracking model coupled to a calibrated two-dimensional hydrodynamic model of Ria de Aveiro was used. Microalgae were regarded as passive particles and the methodology used allowed the determination of their trajectories, as induced by the tidal currents predicted by the hydrodynamic model. The model assumes Ria de Aveiro as vertically homogeneous and does not take into account the vertical distribution patterns of microalgae. Simulations were carried out during extreme spring and neap tides, with microalgal released at the mouth of the lagoon at the local flood. The maximum and minimum areas affected during the occurrence of toxic microalgal blooms were estimated to evaluate the suitability of the distribution of the sampling stations included in the local monitoring program. It was found that the tidal currents greatly determine the microalgal horizontal distribution and dispersal in the lagoon. The results confirmed that the locations of water and bivalve shellfish sampling stations, postulated by INIAP/IPIMAR, in the context of the local harmful algal bloom (HAB) program, were appropriate, although some possible refinements were identified.  相似文献

Although mudflats seem relatively planar, closer inspection reveals a succession of meso-topographical features, including consecutive convex and concave meso- and micro-topographical features. The objective of this study was to determine the influence of meso-scale surface sediment morphology on the dynamics of the macroalgae Ulvales (Chlorophyta) and associated macroepifauna in the Ria Formosa tidal lagoon (southern coast of Portugal). Four sites in the Ria Formosa were sampled monthly. Two were located on convex sections (mounds) of the mudflat and the other two on concave sections (depressions). Macroalgae and related macroepifauna were sampled at each station. Biomass was quantified by determination of the ash-free dry weight (AFDW). Data were analysed using the software package ‘PRIMER’ (Plymouth Routines In Multivariate Ecological Research). Results show a clear distinction between convex and concave areas. In convex sections, Enteromorpha dominated, to the point of being the only algal species present during part of the year. Conversely, biomass and dynamics of Enteromorpha and Ulva were almost the same in concave sections. The associated macroepifauna was also different in protruding or depressed sections of the mudflat. In the convex areas, the macroepifauna population showed less diversity and was dominated by the snail Hydrobia ulvae. In concave areas, the species diversity was larger, but dominated in terms of biomass by the amphipod Melita palmata and the gastropod Nassarius pfeifferi. Results of the study indicate that the benthic communities associated with concave or convex features were different. No relevant differences in texture and sediment physico-chemical characteristics were found between convex and concave sections. The inference is that the morphological nature of the bottom in tidal mudflats can act as a structuring agent of benthic communities.  相似文献

Rainbow vent field is one of the most metal‐contaminated hydrothermal sites on the Mid‐Atlantic Ridge near the Azores region. Two hydrothermal shrimp species dominate the fauna at the Rainbow site along with the mussel Bathymodiolus azoricus. Although the levels of essential and non‐essential metals in these shrimps have been studied, the biological consequences of a metal‐rich environment are still largely unknown. Therefore, the aim of this study was to determine the levels of metal‐binding proteins – metallothioneins (MT) and the activities of antioxidant enzymes – superoxide dismutase, catalase, total glutathione peroxidase and selenium‐dependent glutathione peroxidase in two hydrothermal vent shrimps (Mirocaris fortunata and Rimicaris exoculata) collected from the Rainbow site and to compare them with two coastal shrimps (Palaemon elegans and Palaemonetes varians) from a south Portugal lagoon (Ria Formosa) to evaluate their different adaptation strategies towards metals in their environment. Results show significant differences in MT levels and antioxidant enzymatic activities between vent and coastal shrimps and also between shrimp species collected from the same site. This suggests that biochemical responses in both vent and coastal shrimps are affected not only by the environmental characteristics but also by inter‐specific differences. Nevertheless, these responses apparently confer successful adaptation for survival in a metal‐extreme environment.  相似文献

This work investigated the natural variability of several biomarkers in Tapes philippinarum and Mytilus galloprovincialis, sampled from Northern Adriatic where these organisms are important sentinel species for future environmental impact assessment. Levels of metallothioneins, peroxisomal enzymes and acetylcholinesterase, showed a significant seasonality and marked differences between clams and mussels. Among antioxidant enzymes, catalase and GST decreased during the warmer period, the latter enzyme activity resulting particularly high in clams. The total oxyradical scavenging capacity toward peroxyl radicals decreased in mussels from winter to summer, indicating a prooxidant challenge due to higher seawater temperature and intensity of light irradiance. Lysosomal membrane stability did not exhibit significant seasonal variations, while some variations were observed for DNA damages. Overall results indicated a significant influence of seasonal variability on several biomarkers and species-specific differences which should be considered to discriminate the appearance of anthropogenic disturbance.  相似文献

Oil spills can result in the deposition of large quantities of petroleum hydrocarbons into intertidal and shallow waters seriously impacting bivalve populations. Petroleum hydrocarbons are enriched in polycyclic aromatic hydrocarbons (PAH) and PAH analogs many of which may have potential to damage DNA. The Comet assay is useful for assessing DNA damage and has been used to a limited degree with aquatic organisms, but mostly with studies in vitro. We have carried out studies with the Comet assay to assess the DNA damaging potential of complex mixtures of petroleum hydrocarbons for bivalves. Experiments were carried out with mussels (Mytilus edulis) and clams (Mya arenaria) with dispersions and water soluble fractions of an Arabian crude oil which was also chemically characterized in detail by GC-MS. Pilot studies were first conducted to evaluate test performance and reproducibility. An interindividual coefficient of variation ranging from 17 to 30% was established for the assay with hemocytes and digestive gland cells of both species. Exposure to hydrocarbon fractions had no significant impact on clams. However, an increase in DNA damage was observed at P < 0.1 with digestive gland cells of mussels exposed to aqueous fractions of a light crude oil. These studies have demonstrated a potential for DNA damage in bivalves exposed to oil spills in inshore waters as well as potential for interspecies sensitivity.  相似文献

Connectivity between estuarine fish nurseries and coastal adult habitats can be affected by variations in juvenile growth and survival. Condition indices are renowned proxies of juvenile nutritional status and growth rates and are valuable tools to assess habitat quality. Biochemical (RNA:DNA ratio) and morphometric (Fulton's condition factor K) condition indices were determined in juveniles of Solea solea, Solea senegalensis, Platichthys flesus, Diplodus vulgaris and Dicentrarchus labrax collected in putative nursery areas of nine estuaries along the Portuguese coast (Minho, Douro, Ria de Aveiro, Mondego, Tejo, Sado, Mira, Ria Formosa and Guadiana) in the Spring and Summer of two consecutive years (2005 and 2006) with distinct climatic characteristics. Individual condition showed significant variation amongst species. The combined use of both condition indices highlighted the low correlation between them and that RNA:DNA had a higher sensitivity. RNA:DNA varied between years but overall the site relative patterns in condition were maintained from one year to the other. Higher RNA:DNA values were found in Spring than in Summer in most species. Intra-estuarine variation also occurred in several cases. Species specific trends in the variability of condition amongst estuaries were highlighted. Some estuaries had higher juvenile condition for more than one species but results did not reveal an identical trend for all species and sites, hindering the hypotheses of one estuarine nursery promoting superior growth for all present species. Significant correlations were found between condition indices, juvenile densities and environmental variables (water temperature, salinity and depth) in the estuarine nurseries. These influenced juvenile nutritional condition and growth, contributing to the variability in estuarine nursery habitat quality. Management and conservation wise, interest in multi-species approaches is reinforced as assessments based on a single species may not reflect the overall nursery habitat quality.  相似文献

Cellular and humoral immune parameters are often used as biomarkers to trace environmental and physiological stresses in marine bivalves. In this study, we compared various immune parameters of Manila clams (Ruditapes philippinarum) under normal conditions and under a high level of desiccation, using flow cytometry. The immune parameters analyzed included, total hemocyte count, hemocyte mortality, hemocyte DNA damage, reactive oxygen species (ROS) production, and phagocytosis activity. Total hemocyte count, hemocyte DNA damage, and hemocyte mortality were significantly elevated among clams under high desiccation stress, while phagocytosis activity and spontaneous ROS production were significantly lower compared to those parameters of the control clams (p<0.05). These data suggest that the immune parameters analyzed in this study well reflect the physiological status of clams.  相似文献

The seasonal plasticity of individual Zostera noltii architectural, reproductive and elemental content features, of plant epiphyte load and of meadow biomass–density relationships was investigated along a vertical intertidal gradient at Ria Formosa lagoon, southern Portugal. The vertical variability of the seagrass environment was evident in the sediment characteristics, which showed coarser grain size, less organic matter, lower N content and higher ammonium concentration in the low intertidal than in medium and high intertidal. A clear vertical differentiation in Z. noltii morphology was observed from longer and wider leaves, longer and wider internodes and shorter roots at low intertidal, to shorter and narrow leaves, shorter and narrower internodes and longer roots at high intertidal. The leaf size was negatively related to light availability and positively related to nutrient availability whereas the root size was negatively related to nutrient availability. The lower leaf N and P content found in low intertidal plants may reflect a dilution effect of the nutrients due to higher growth rates. Lower N content of low intertidal leaves supports previous findings that the nitrate reductase activity is lower in plants from this level. The higher epiphyte load observed in Z. noltii leaves of the low intertidal may be a consequence of the lower exposure period, but also of higher hydrodynamics that increase the availability of nutrients. No evidence of the influence of the intertidal level on the flowering shoot density was found. The cyclic temporal pattern of the biomass–density relationship was much wider at low and medium intertidal than at high intertidal. At low intertidal, the decline in shoot density during fall and winter was coincident with a biomass decrease and its increase in spring and summer coincided with the biomass increase. In medium and high intertidal, the biomass and density seasonal variations were decoupled. As a result, only at low intertidal there was a significant positive relationship between biomass and density. This suggests that Z. noltii population structure along the intertidal is regulated by different factors. Light is probably the most important factor regulating the population structure in the low intertidal, whereas desiccation is probably the main factor regulating the populations in upper intertidal. Zostera noltii showed a considerable plasticity at a physiological-, plant- and population-level along the intertidal zone, indicative of the species acclimation to the steep environmental gradient of this particular ecosystem.  相似文献

The assessment of DNA damage by the Comet assay has been described as a useful non-specific general biomarker of stress in many marine organisms. In field situations it has successfully been employed to distinguish between reference and polluted sites and in the laboratory it has been widely used as a mechanistic tool to determine pollutant effects and mechanisms of DNA damage. To date a wide range of marine vertebrates and invertebrates have been used, however, the usefulness of this assay as a biomarker in cnidarians has not yet been assessed. The aims of this study were to optimize the Comet assay for cnidarian cells and to assess its utility for detecting genotoxic damage in these cells. Cells were isolated from the North American pacific coast temperate sea anemone Anthopleura elegantissima using a non-enzymatic dissociation procedure and viability was determined to be in excess of 90%. Cells were incubated either with (1 h acute exposures) hydrogen peroxide (H(2)O(2)), ethylmethanesulphonate (EMS) or benzo(a)pyrene (B[a]P). In comparison to other marine species, anemone cells exhibited high control or background levels of DNA strand breaks. Despite this, however, we observed dose responses for each of the study chemicals with no reduction in cell viability. This study demonstrates that anemone cells respond to known DNA damaging agents, including B[a]P which requires metabolism to exert its genotoxic effect, and that the Comet assay may prove to be a useful biomarker of stress in cnidarian species.  相似文献
