Thirty-six strains of marine actinomycetes were isolated from a sample of marine sediment collected from the Yellow Sea and evaluated in terms of their inhibitory activity on the growth of Aspergillus parasiticus and the production of norsolorinic acid using dual culture plate assay and agar diffusion methods. Among them, three strains showed strong antifungal activity and were subsequently identified as Streptomyces sp. by 16S rRNA gene sequencing analysis. The supernatant from the fermentation of the MA01 strain was extracted sequentially with chloroform and ethyl acetate, and the activities of the extracts were determined by tip culture assay. The assay results show that both extracts inhibited mycelium growth and toxin production, and the inhibitory activities of the extracts increased as their concentrations increased. The results of this study suggest that marine actinomycetes are biologically important for the control of mycotoxins, and that these bacteria could be used as novel biopesticides against mycotoxins.  相似文献

Compositional data on the sterol and alcohol fractions isolated from deep-sea marine sediments from the Okinawa Trough were obtained to determine the relative contribution from. marine and terrestrial inputs. Following extraction, the sterol plus alcohol fraction was isolated by layer chromatography, derivatized with BSTFA and then analysed by capillary GC and GC-MS. A suite of C26-C29 stenols and stanols and C30-C32 keto - alcohols were identified in the sediments. The thermal stability of the compounds in these sediments was studied by heating portions of the surface sediment in glass tubes fpr 16 hours at temperatures from 50℃ to 200℃.The C27 stanol/stenol ratio increased when temperatures went up to 175℃ , but the distribution of C30-C32 Keto - alcohols remained unaffected. At 200℃ most of the sterols and Keto - alcohols were destroyed.  相似文献

In order to determine and assess the concentrations of trace elements in coral reefs sediments from Red Sea of Yemen, sediment samples were collected, treated and analyzed for cadmium, chromium, cobalt, copper, manganese, nickel, lead, iron, zinc and vanadium by the atomic absorption spectrometric analysis. The result is that cadmium, cobalt and lead concentrations were high and other elements are low or the same as natural background. It is concluded that the high cadmium, cobalt and lead levels in coral reefs sediments will have negative effects on marine life of the sites, so further researches are needed to characterize the sources fate, biogeochemical processes and impacts of these trace elements on coral reefs and marine of the region.  相似文献

As one of the main groups of marine planktonic protozoa, oligotrich ciliates have shown a high biodiversity. Recently, a new aloricate oligotrich ciliate, Strombidium pseudorapulum sp. n. was isolated from coastal waters of southeast China. The living morphology and infraciliature of this species were studied using live cell observation and protargol impregnation. Strombidium pseudorapulum sp. n. differs from its congeners by having a radish-like body with a bumped anterior end and an elongated tail-like posterior end, possessing a red ‘eyespot' in apical protrusion of cell, adoral zone of membranelles composed of 12–16 collar and 9–12 buccal membranelles, one spindle-shaped macronucleus, girdle kinety in anterior 2/5 of cell with 45–60 densely spaced dikinetids, and ventral kinety occupying about the posterior 1/3 of the cell with 16–19 dikinetids. Strombidium pseudorapulum sp. n. clusters together with those species having ‘eyespot' in apical protrusion in the phylogenetic tree based on the small subunit rRNA gene sequence. It is a sister species of S. cuneiforme with 31 unmatched nucleotides between their SSU rRNA gene sequences.  相似文献

A poorly-described marine planktonic ciliate, Tintinnopsis brasiliensis Kofoid & Campbell, 1929, collected from the Taiping Cape of Qingdao, China, was morphologically investigated based on permanent preparation after protargol impregnation and was compared with other related congeners. According to the infraciliature, three ciliary groups can be recognized, which reveals a very stable structure among specimens and denotes that the pattern of infraciliature is, apart from the features of the lorica, a highly reliable criterion for species identification.  相似文献

We investigated the effects of lipopolysaccharide(LPS) and dopamine(DA) on the activation of the prophenoloxidase(proPO) system of Litopenaeus vannamei.LPS and DA were shown with a negative dose-dependent effect on hyalne cells(HC),semi-granular cells(SGC),large granular cells(LGC),and total haemocyte count(THC).When haemocytes were treated with LPS or DA,serine proteinase activity and intracellular phenoloxidase(PO) activity were significantly reduced,but extracellular PO activity increased significantly.These findings indicated that the reduction in haemocyte counts was mainly because of the degranulation and activation of the proPO system from semi-granule and large granule cells.The PKC inhibitor,chelerythrine,and the TPK inhibitor,genistein,had an inhibitory effect on extracellular PO activity,while serine proteinase and intracellular PO activity increased.This suggests that the LPS and DA induce the activation of proPO in haemocytes via PKC and TPK-related signaling pathways,but serine proteinase may be activated only by PKC,as the genistein effects were not statistically significant.Electrophoresis analysis revealed that POs induced by LPS or DA have the same molecular mass and high diphenolase activity.Two PO bands at 526 kDa and 272 kDa were observed in PAGE,while in the haemocyte lysate supernatant(HLS),only a 272-kDa band was observed.This band was resolved after SDS-PAGE under non-reducing and reducing conditions into two groups of POs,166 kDa and 126 kDa,and 78.1 kDa and 73.6 kDa,respectively,suggesting that PO in L.vannamei is an oligomer,which may have different compositions intra-and extracellularly.  相似文献

Temporal and spatial heterogeneity identifications of marine environment and its changes have great significances in marine spatial planning and offshore pollution control.In this study,considering the integration of marine ecological environment and sea-land interaction,we built a spatialized evaluation model named Marine Environment Change Spatial Assessment(MECSA)to quantify the marine environment status and changes.In MECSA,we applied the geospatial model and the pressure-state-response(PSR)model for processing and integrating evaluation indicators.A case study in the Laizhou Bay showed that the marine environment quality was generally in a declining state from 2009 to 2015.In 2015,the Marine Environment Index(MEI)had decreased by 0.1 compared with 2009,although the two years all reached a‘Good'level.The spatial MEI layers of the two years showed a same distribution:the southwestern part was in poorer status,with a fan ring shape gradually getting better to the northeast.The Marine Organisms Response Index(MORI)contributed the most to the MEI.Therefore,future marine environmental assessment and spatial planning should focus on the identification the marine environment with its changes from the perspective of spatial heterogeneity and systemicity.Based on single indicators and comprehensive evaluation results,we can propose spatially targeted policies and recommendations scientifically.  相似文献

The rapid economic and social developments in the Luoyuan and Lianjiang counties of Fujian Province, China, raise certain environment and ecosystem issues. The unusual phytoplankton bloom and eutrophication, for example, have increased in severity in Luoyuan Bay(LB). The constant increase of nutrient loads has largely caused the environmental degradation in LB. Several countermeasures have been implemented to solve these environmental problems. The most effective of these strategies is the reduction of pollutant loadings into the sea in accordance with total pollutant load control(TPLC) plans. A combined three-dimensional hydrodynamic transport-transformation model was constructed to estimate the marine environmental capacity of chemical oxygen demand(COD). The allowed maximum loadings for each discharge unit in LB were calculated with applicable simulation results. The simulation results indicated that the environmental capacity of COD is approximately 11×104 t year-1 when the water quality complies with the marine functional zoning standards for LB. A pollutant reduction scheme to diminish the present levels of mariculture- and domestic-based COD loadings is based on the estimated marine COD environmental capacity. The obtained values imply that the LB waters could comply with the targeted water quality criteria. To meet the revised marine functional zoning standards, discharge loadings from discharge units 1 and 11 should be reduced to 996 and 3236 t year-1, respectively.  相似文献

Karenia is one of the most harmful and representative red tide genus in a temperate zone. Blooms caused by this genus have resulted in massive fish death in the South China Sea and the East China Sea. However, the potential effects of this dinoflagellate on human health through the transfer of toxins via marine food webs, and the mechanisms of toxicity, are still unknown. Therefore, we examined the toxic effects of a strain of K. mikimotoi (isolated from the South China Sea) on the proliferation and morphology of four mammalian cell lines (two normal cell lines and two cancer cell lines). In addition, we carried out a preliminary investigation on the mechanism of toxicity of the alga. The results show that the polar lipid-soluble component of K. mikimotoi significantly inhibited proliferation of the four cell lines, and resulted in the cells becoming spherical, swollen and damaged. The result of Annexin V and PI double-staining confirmed that cell membranes were disrupted. The malonaldehyde (MDA) contents in the medium of the four cell lines treated with the polar-lipid extracts all increased significantly, which indicates that the polar-lipid toxins produced by K. mikimotoi could adversely affect mammalian cells by inducing lipid peroxidation. We conclude that K. mikimotoi is a potential threat to human health, and the comprehensive effect of this dinoflagellate and its mechanisms should be investigated further.  相似文献

Dinoflagellates in the genus Symbiodinium, including nine clades(A–I), mainly form mutualistic symbioses with corals. More than 100 Symbiodinium molecular types have been identified by the ITS2-based genotype method within any given clade, and specifically within Symbiodinium clade C. However, the genotype identification method using the ITS2 sequence is likely to lead to high diversity estimates due to the intra-genomic variations in the ITS2 space; thus, further validation is essential for a correct identification. In this study, the molecular diversity of Symbiodinium ITS2 sequences cloned from two stone corals, Acropora sp. SY-01 and Pocillopora sp. SY-05, and one soft coral, Sarcophyton sp. SY-07, living in the northern part of South China Sea(SCS), were analyzed and compared using the ITS2-based genotype identification method, coupled with ITS2-based secondary structural and phylogenetic analyses. As the result, 12 Symbiodinium ITS2 genotypes were identified, while only six and three Symbiodinium ITS2 genotypes were supported by ITS2-based secondary structural and phylogenetic analyses, respectively. In addition, no shared Symbiodinium ITS2 genotypes were observed among the three coral species, suggesting coral species-dependent Symbiodinium genotypes were within clade C. In summary, the present study provides a theoretical basis for validating the molecular diversity of Symbiodinium ITS2 genotypes in corals.  相似文献

The present study aimed at assessing the antifouling activity of bacteria associated with marine sponges. A total of eight bacterial strains were isolated from the surface of sponge Sigmadocia sp., of them, SS02, SS05 and SS06 showed inhibitory activity against biofilm-forming bacteria. The extracts of these 3 strains considerably affected the extracellular polymeric substance producing ability and adhesion of biofilm-forming bacterial strains. In addition to disc diffusion assay, microalgal settlement assay was carried out with the extracts mixed with polyurethane wood polish and coated onto stainless steel coupons. The extract of strain SS05 showed strong microalgal settlement inhibitory activity. Strain SS05 was identified as Bacillus cereus based on its 16S rRNA gene. Metabolites of the bacterial strains associated with marine invertebrates promise to be developed into environment-friendly antifouling agents.  相似文献

In China, promoting the development of coastal areas has been included in a series of national strategic development plans.At the same time, many marine environmental problems have been associated with the rapid development of coastal sea use. In order to quantify the impact of human activities on the coast, the characteristics of coastlines and near-shore sea use of Liaodong Bay, Northeast China, were first classified using multi-source, remotely sensed imagery using automatic or semi-automatic extraction methods for five periods between 1993 and 2015. Sea use dynamics and coastline dynamics resulting from human activates were analyzed. Results showed a significant trend of continuous growth in sea use and a progressive increase in the total length of artificial coastline, but a noticeable loss of natural coastline during the five periods. Reclaimed land and enclosed areas were the main types of sea use. Most coastal human activities were distributed in the northern part of the bay. In recent years, rapid industrialization and urbanization in China's coastal areas have promoted large-scale land reclamation. Accordingly, the observed coastline changes during each period had a close relationship with coastal development and sea area utilization. Based on marine functional zoning(MFZ), the sea use carrying capacity was evaluated by means of indexes to describe human exploitation of the marine and coastal environments in the bay. This showed that the intensity of coastal utilization in Liaodong Bay has increased year-on-year. Sea use carrying capacity reached a ‘critically loaded'state by 2008 and was ‘overloaded' by 2015.  相似文献

A total of 78 Yarrowia lipolytica yeast strains from seawater, sediments, mud of salterns, the guts of marine fish, and marine algae were obtained. After the crude protein of the yeasts was estimated by the method of Kjehldahl, we found that seven strains of the marine yeasts grown in soy bean cake hydrolysate with 20 g L-1 of glucose for 48 h at 28℃ contained more than 41.0 g protein per 100 g of cell dry weight and the cell dry weight was more than 4.4 g per L of the culture. Among them, strain SWJ-1b contained the highest crude protein. The results of Biolog identification and molecular methods further confirmed that they indeed belonged to Y. lipolytica.  相似文献

Gelatin from the sea cucumber(Paracaudina chinens var.) was hydrolyzed by bromelain and the hydrolysate was found to have a high free radical scavenging activity. The hydrolysate was fractionated through an ultrafiltration membrane with 5 kDa molecular weight cutoff(MWCO). The portion(less than 5 kDa) was further separated by Sephadex G-25. The active peak was col-lected and assayed for free radical scavenging activity. The scavenging rates for superoxide anion radicals(O2·-) and hydroxyl radi-cals(·OH) of the fraction with the highest activity were 29.02% and 75.41%,respectively. A rabbit liver mitochondrial free radical damage model was adopted to study the free radical scavenging activity of the fraction. The results showed that the sea cucumber gelatin hydrolysate can prevent the damage of rabbit liver and mitochondria.  相似文献

One new marine oligotrich ciliate, Omegastrombidium hongkongense n. sp., was isolated from a bloom of Noctiluca scuntillans near Port Shelter, Hong Kong. The morphology and infraciliature of this new species were studied on both living and protargol-stained specimens. Its phylogenetic position was discussed based on the sequence of the small subunit r RNA gene. O. hongkongense is different from its congeners with special characters. The cells are usually heart-shaped, and the cell size usually is(20–35) ×(20–30) μm in vivo. Its deep buccal cavity extends obliquely to about 1/2 of cell length. It shows prominent apical protrusion. The adoral zone of membranelles is divided into 17–19 collar membranelles and four buccal membranelles. It has one ball-like macronucleus. The girdle kinety forms a closed loop which obliquely surrounds the body. The ventral kinety and thigmotactic membranelles are not observed. The SSU r RNA sequence of O. hongkongense was close to those of Strombidium paracalkinsi and Varistrombidium kelum with approximately 99% similarity. In the phylogenetic trees, O. hongkongense can be grouped with O. elegans and V. kielum species with very low support(16% ML).  相似文献

The effects of salinity on hemolymph osmotic pressure, Na^+ concentration and Na^+-K^+-ATPase activity of gill of Chinese crab Eriocheir sinensis were studied. The results showed that hemolymph osmotic pressure and Na^+ concentration increased significantly (P〈0.05), and the Na^+-K^+-ATPase activity of gills decreased significantly (P〈0.05) when salinity increased from 0 to 16. The hemolymph osmotic pressure and Na^+ concentration in each treatment group rose remarkably at 0.125 d or 0.25 d, while the Na^+-K^+-ATPase activity of gill reduced gradually with increased experiment time in 3 d. Then the three parameters remained at a constant level after 0.25 d, 0.125 d and 3 d, respectively, and higher hemolymph osmotic pressure, higher Na^+ concentration and lower Na^+-K^+-ATPase activity of gill occurred at higher salinity. The effect of salinity change on protein concentration of hemolymph was indistinct (P〉0.05); However, the protein concentration decreased gradually with the increase of salinity from 0.25 d to 1d, and then tended to be stable from day 1 to day 15.  相似文献

We present results on the effect of modified clay on cyst formation of Scrippsiella trochoidea in harmful algal bloom (HAB). Modified clay (in concentration of 0, 0.1, 0.5, and 1.0 g/L) were added to cultures, and observations were made on cysts of S. trochoidea under controlled laboratory conditions. Results indicate that the removal rate of algal cells reached 97.7% at the clay concentration of 1.0 g/L. The cyst formation rate increased from 4.6% to 24.6% when the concentration of clay was increased from 0 to 1.0 g/L. Two cyst metamorphs were observed: spinal calcareous cysts and smooth noncalcareous ones. The proportion of the spinal cysts decreased from 76.9% to 24.1% when clay concentration increased from 0 to 1.0 g/L. In addition, modified clay affected cyst germination. The germination rate decreased with the increases in the clay concentrations. Non-calcareous cysts had a lower germination rate with a longer germination time. We conclude that modified clay could depress algal cell multiplication and promote formation of temporal cysts of S. trochoidea, which may help in controlling HAB outbreaks.  相似文献

Two species of jellyfish, Cyanea nozakii Kishinouye and Nemopilema nomurai Kishinouye, have occurred offcoastal areas of the northeastern China Sea, Yellow Sea, and Bohai Sea in recent years. They influence marine ecosystem safety and fishery production, and also pose a risk to human health. The current study examined the hemolytic and cytotoxic activities of crude venoms extracted from the nematocysts of C. nozakii and N. nomurai. The results showed that there were more nematocysts on tentacles from C. nozakii than on tentacles of the same length from N. nomurai. The protein concentration per nematocyst extracted from N. nomurai was higher than that from C. nozakii. Both nematocyst venoms showed dose-and timedependent hemolytic activity on erythrocytes from chicken, pigeon, and sheep, with sheep erythrocytes being the most sensitive, with EC 50 values of 69.69 and 63.62 μg/m L over a 30-min exposure with N. nomurai and C. nozakii nematocyst venoms, respectively. A cytotoxic assay of both jellyfish venoms on A431 human epidermal carcinoma cells resulted in IC 50 values of 68.6 and 40.9 μg/mL after 24-h incubation, respectively, with venom from C. nozakii showing stronger cytotoxic activity than that from N. nomurai. The results of current study indicate that nematocyst venom from C. nozakii had stronger hemolytic and cytotoxic activities than that from N. nomurai and, thus, C. nozakii might be more harmful to the health of humans and other species than are N. nomurai when they appear in coastal waters.  相似文献
