富营养化湖泊水体中藻蓝蛋白提取方法的对比   总被引:1,自引:1,他引:0  
藻蓝蛋白是蓝藻的指示型色素,目前还没有标准的提取方法,特别是萃取剂的选择品种较多,对测量结果影响较大.分别以室内培养的铜绿微囊藻和巢湖夏季野生蓝藻为提取对象,运用液氮反复冻融法破碎蓝藻细胞,分别以Asolctin-CHAPS缓冲液(AC)、磷酸盐缓冲液(PBS)和Tris-HCl缓冲液来提取藻蓝蛋白,通过分光光度法分析提取液的吸收光谱、藻蓝蛋白浓度以及藻蓝蛋白浓度与叶绿素a浓度的相关性,比较3种缓冲液的提取效果.结果显示:3种缓冲液提取的藻蓝蛋白提取液在620 nm处都出现了藻蓝蛋白的特征吸收峰,但AC缓冲液和PBS缓冲液提取效率明显高于Tris-HCl缓冲液.另外,虽然AC缓冲液提取效率最高,但其成本昂贵、配制过程复杂且不易控制、难保存,不适宜大规模地推广应用;而PBS缓冲液的提取效率略低于AC缓冲液,但其配制方便快捷,更适合于大规模湖泊水质监测的要求,因此推荐PBS缓冲液作为常用的藻蓝蛋白提取液.  相似文献

Sb, As, Cu, and Zn toxicity and contamination have become a growing concern in recent years. Phytoremediation, a plant based and cost effective technology, may be an effective approach in the cleanup of water contaminated by these metals. In this study, the aquatic macrophyte Eleocharis acicularis was used in laboratory and field experiments to assess its capability to accumulate Sb, As, Cu, and Zn, and thereby investigate its potential application in phytoremediation. The results showed that E. acicularis adapted well to water contaminated by these metals. The removal rates of Sb, As, Cu, and Zn in the laboratory experiment were 3.04, 2.75, 0.417, and 1.49 μg/L/day, respectively. The highest concentrations of these metals accumulated in E. acicularis after 10 days of the laboratory experiment were 6.29, 6.44, 20.5, and 73.5 mg/kg dry weight, respectively. Only 8% of As, 12% of Sb, 87% of Cu and 93% of Zn removed from the water were used by E. acicularis. The highest concentrations of Sb, As, Cu, and Zn accumulated in E. acicularis after 10 wk of the field experiment were 76.0, 22.4, 33.9, and 266 mg/kg dry weight, respectively. The results indicate that E. acicularis has the ability to accumulate Sb, As, Cu, and Zn from contaminated water.  相似文献

The objective of this study was to determine the concentrations and possible sources of heavy metals and persistent organic pollutants (POPs) in water and estuarine sediments from Gao-ping River in order to evaluate the environmental quality of aquatic system in southern Taiwan. High concentrations of heavy metals including Cr, Zn, Ni, Cu and As, ranging from 10.7 to 180 mg/kg-dry weight (dw), were detected in sediments from Gao-ping River. When normalized to the principal component analysis (PCA), swinery and electroplating wastewaters were found to be the most important pollution sources for heavy metals. Of various organochlorine pesticide (OCP) residues detected, aldrin and total-hexachlorocyclohexane (HCH) were frequently found in sediments. The total concentrations of OCPs were in the range 0.47-47.4 ng/g-dw. Also, the total-HCH, total-cyclodiene, and total-dichlorodiphenyltrichloroethane (DDT) were in the range 0.37-36.3, 0.21-19.0, and 0.44-1.88 ng/g-dw, respectively. The polychlorinated biphenyl (PCB) concentrations in sediments from Gao-ping River ranged between 0.37 and 5.89 ng/g-dw. The PCB concentrations are positively correlated to the organic contents of the sediment particles. alpha-HCH was found to be the dominant compound of HCH in the sediments, showing that long-range transport may be the possible source for the contamination of HCH in sediments from Gao-ping River. In summary, trace amounts of POPs in estuarine sediments from Gao-ping River were detected, showing that there still exist a wide variety of POP residues in the river sediments in Taiwan. These POP residues may be mainly from long-range transport and weathered agricultural soils, while heavy metal contamination is primarily from the swinery and industrial wastewaters.  相似文献

Green-lipped mussels, Perna viridis, and Manila clams, Ruditapes philippinarum were sourced from “clean” sites in the Hong Kong region, depurated in a laboratory using uncontaminated filtered seawater for 8 days, and transplanted to a suspected gradient of chemically polluted sites in Hong Kong. After 14- and 28-days of field exposure, several antioxidant parameters including glutathione S transferase (GST), catalase (CAT), glutathione peroxidase (GPx), and glutathione (GSH) were quantified in gill and hepatopancreas tissues. Whole body tissue concentrations of polycyclic aromatic hydrocarbons (PAHs), petroleum hydrocarbons (PHCs), polychlorinated biphenyls (PCBs) and organochlorine pesticides (OCs) were determined in pooled site samples. Chemical analyses indicated that: (a) clams had higher levels of PAHs, PHCs, DDTs and PCBs, whereas mussels had higher hexachlorocyclohexane (HCHs) and there was no difference between species for dieldrin and remaining OCs; (b) Kat O should not be continued as a “clean” reference site for Hong Kong, because of the levels of contaminants measured and (c) PAH concentrations in the current survey were similar to those previously measured. Toxicological conclusions were: (a) antioxidant responses were different between species; (b) CAT and GST have highest utility in clams for field use in Hong Kong, whereas CAT in both gill and hepatopancreas tissue showed most potential in mussels; (c) significant induction of antioxidant responses over day 0 (excluding GPx in both tissues, and GST in mussel hepatic tissue); (d) groups of contaminants do not consistently induce antioxidant responses and (e) organochlorines and PCBs correlated significantly with CAT and GST in clam hepatopancreas and with CAT in mussel gill and hepatic tissue. Multivariate statistical techniques indicated little relationship between the site patterns for antioxidant responses and the contaminant gradients identified in body burden analysis.  相似文献

Total Hg (∑Hg), organic-Hg (O-Hg) and Se bioaccumulations in small cetaceans distributed in Taiwanese waters of the Taiwan Strait and the southwestern Pacific have been investigated for the first time. The results could represent the baseline metal concentrations of marine mammals in the southwestern Pacific, where volcanic activities are possibly the major source of mercury to the environments. Muscle samples of four species of small cetaceans were collected from animals accidentally caught by tuna-longline fisheries from 1994 through 1995. In total, 53 pantropical spotted dolphins, Stenella attenuata, nine spinner dolphins, S. longirostris, five bottlenose dolphins, Turiops truncatus and four Risso's dolphins, Grampus giseus were analyzed. In addition, two stranded pantropical spotted dolphins were investigated. Cold vapour AAS and ICP-MS were used in the analysis of Hg and Se, respectively. Significant species difference was found in the four species of small cetaceans. Among them, the pantropical spotted dolphin showed the highest mean concentration (mg/kg wet wt.) of both ∑Hg (3.64±2.19) and O-Hg (2.79±1.23), whereas the Risso's dolphin had the highest mean concentrations of Se (1.77±1.29). There was no significant sex difference with respect to metal bioaccumulation in the samples of S. attenuata. Significant correlations between body length (BL) and ∑Hg, as well as O-Hg concentrations were observed in pantropical spotted (Sa) and spinner dolphins (Sl). The linear relationships were Sa: ∑Hg=−8.290+0.066BL, r=0.421; Sl: ∑Hg=−2.735+0.025BL, r=0.875; Sa: O-Hg=−3.723+0.036BL, r=0.408; and Sl: O-Hg=−3.017+0.025BL, r=0.870. However, a demethylation phenomenon that decreasing the percentage of O-Hg coupled with increasing levels of Se was observed when the ∑Hg concentrations in the muscle tissues of dolphins reached 4 mg/kg wet wt.  相似文献

Data-based models, namely artificial neural network (ANN), support vector machine (SVM), genetic programming (GP) and extreme learning machine (ELM), were developed to approximate three-dimensional, density-dependent flow and transport processes in a coastal aquifer. A simulation model, SEAWAT, was used to generate data required for the training and testing of the data-based models. Statistical analysis of the simulation results obtained by the four models show that the data-based models could simulate the complex salt water intrusion process successfully. The selected models were also compared based on their computational ability, and the results show that the ELM is the fastest technique, taking just 0.5 s to simulate the dataset; however, the SVM is the most accurate, with a Nash-Sutcliffe efficiency (NSE) ≥ 0.95 and correlation coefficient R ≥ 0.92 for all the wells. The root mean square error (RMSE) for the SVM is also significantly less, ranging from 12.28 to 77.61 mg/L.  相似文献

There is a lack of information on monitoring neurotoxicity in offshore commercial species. To help fill this gap, we sampled hake (Merluccius merluccius) and Norway lobster (Nephrops norvegicus) in fishing grounds off the coast of l’Ametlla de Mar (NW Mediterranean) in June 2005 at a depth of 100 m and 400 m. Additionally, at 400 m depth, two other fish species, Micromesistius poutassou and Phycis blennoides were included. Neurotoxicity markers such as Colinesterases (ChEs), namely acethyl- (AChE), butyryl- (BChE), propionyl- (PrChE) and carboxilesterase (CbE) were measured in muscle. Lipid peroxidation (LP), a marker of oxidative damage, was also included. The results are discussed in relation to the animal’s sex, size and fishing depth. A comparison of esterases and LP levels between muscle and liver of hake and between muscle and hepatopancreas of Norway Lobster was made.

AChE was dominant in muscle and CbE in hepatopancreas. No differences between fish species were seen for AChE. However, N. norvegicus, presented lower levels of ChEs and LP. A size-dependence in ChEs was seen for M. merluccius, with larger animals showing significantly lower activities (p < 0.05). Sex-dependence was seen in N. norvegicus for most esterases, except AChE, with males displaying higher activities (p < 0.05). A sampling-depth effect was also seen in the crustacea, with animals from 100 m generally presenting lower esterase activities and higher LP levels.  相似文献

Riparian vegetation can trap sediment and nutrients sourced from hillslopes and reduce stream bank erosion. This study presents results from a 10-year stream monitoring program (1991–2000), in a 6 km2 agricultural catchment near Albany, Western Australia. After 6 years, a 1.7 km stream reach was fenced, planted with eucalyptus species and managed independently from the adjacent paddocks. Streamflow, nutrient and sediment concentration data were collected at the downstream end of the fenced riparian area, so there are data for before and after improved riparian management. Suspended sediment (SS) concentrations fell dramatically following improved riparian management; the median event mean concentration (EMC) dropped from 147 to 9.9 mg l−1. Maximum SS concentrations dropped by an order of magnitude. As a result, sediment exports from the catchment decreased following improved riparian management, from over 100 to less than 10 kg ha−1 yr−1. Observations suggest that this was the result of reduced bank erosion and increased channel stability. Riparian management had limited impact on total phosphorus (TP) concentrations or loads, but contributed to a change in phosphorus (P) form. Before improved riparian management, around half of the P was transported attached to sediment, but after, the median filterable reactive P (FRP) to TP ratio increased to 0.75. In addition, the median FRP EMC increased by 60% and the raw median FRP concentration increased from 0.18 to 0.35 mg l−1. These results suggest that there was a change in the dominant P form, from TP to FRP. Changes in total nitrogen (TN) following improved riparian management were less clear. There were reductions in TN concentrations at high flows, but little change in the loads or EMC. This study demonstrates the benefits of riparian management in reducing stream bank erosion, but suggests that in catchments with sandy, low P sorption soils, there may be limitations on the effectiveness of riparian buffers for reducing P and N exports.  相似文献

Declining water quality on the south coast of Western Australia has been linked to current agricultural practices. Riparian buffers were identified as a tool available to farmers and catchment managers to achieve water quality improvements. This study compares 10 m wide regenerating grass and Eucalyptus globulus buffer performance. Surface and subsurface water quality were monitored over a 3‐year period. Nutrient and sediment transport were both dominated by subsurface flow, in particular through the B‐horizon, and this may seriously limit the surface‐runoff‐related functions of the riparian buffers. Riparian buffer trapping efficiencies were variable on an event basis and annual basis. The grass buffer reduced total phosphorus, filterable reactive phosphorus, total nitrogen and suspended sediment loads from surface runoff by 50 to 60%. The E. globulus buffer was not as effective, and total load reductions in surface runoff ranged between 10 and 40%. A key difference between the grass and E. globulus buffers was the seasonality of sediment and nutrient transport. Surface runoff, and therefore sediment and nutrient transport, occurred throughout the year in the E. globulus buffer, but only during the winter in the grass buffer. As a consequence of high summer nutrient and sediment concentrations, half the annual loads moving via surface runoff pathways through the E. globulus buffer were transported during intense summer storms. This study demonstrates that grass and E. globulus riparian buffers receiving runoff from pasture under natural rainfall can reduce sediment and nutrient loads from surface runoff. However, in this environment the B‐horizon subsurface flow is the dominant flowpath for nutrient transport through the riparian buffers, and this subsurface flow pathway carries contaminant loads at least three times greater than surface runoff. 本文利用一个三层剪切型结构模型在各种激励下的振动台试验,研究经验模分解(empirical mode decomposition,EMD)和小波分析(wavelet analysis,WA)在结构损伤识别中的应用。研究针对结构刚度突然损失的损伤类型,并在试验中通过连接在模型两侧的弹簧的断裂来模拟。利用EMD和WA分析试验记录到的结构加速度信号来识别结构损伤发生的时刻和位置。试验结果表明,EMD和WA方法均可利用分解信号中的尖峰准确识别结构损伤发生的时刻,并利用信号尖峰在结构上的空间分布来确定损伤位置。研究表明,EMD和WA都是进行结构在线检测的良好方法。  相似文献

A comprehensive study aimed at evaluating the occurrence, significance of concentrations and spatial distribution of priority pollutants (PPs) along the Comunidad Valenciana coastal waters (Spain) was carried out in order to fulfil the European Water Framework Directive (WFD). Additionally, PP concentrations were also analysed in the effluent of 28 WWTPs distributed along the studied area. In coastal waters 36 organic pollutants of the 71 analysed, including 26 PPs were detected although many of them with low frequency of occurrence. Only 13 compounds, which belong to four different classes (VOCs, organochlorinated pesticides, phthalates and tributyltin compounds (TBT)) showed a frequency of occurrence above 20% in coastal waters. In the results obtained until now, octylphenol, pentachlorobenzene, DEHP and TBT exceeded the annual average concentration (EQS-AAC), and only TBT surpassed the maximum allowable concentration (EQS-MAC). The most frequent contaminants determined in coastal waters were also present in WWTP effluents.  相似文献

In this study, the distribution and partition of polybrominateddiphenyl ethers (PBDEs) and polychlorinated biphenyls (PCBs) in water from the Pearl River Estuary were investigated. A seasonal dependence for dissolved and particle phase PCB and PBDE levels was observed. A high dissolved phase level occurred in the dry season. The positive correlation between POC content in the particles and phase distribution coefficients (Kp) suggested that POC may play an important role in determining the partition of PBDEs and PCBs. A linear correlation between the observed log KOC and log KOW for PBDEs and PCBs was found with slopes less than one. The observed log KOC values were lower than the predicted log KOC values for PBDEs (except for BDE28). The log KOC values were higher than the predicted values for PCBs. A combination of sorption to colloids (10-98% for PBDEs and less than 25% for PCBs) and adsorption to BC was responsible for this observation.  相似文献

Seventeen organic phosphorus pesticides (OPs) and 18 organic chlorine pesticides (OCs) at water from Jiulong River Estuary (JLRE) are determined by SPE–GC–(FPD and μECD). The contents of the total OPs (17) ranged from 134.8 to 354.6 ng/l (the mean is 227.2 ng/l). Total OCs concentrations varied from 115.4 to 414.7 ng/l in water (mean 237.7 ng/l). The levels of total hexachlorocyclohexane in water varied from 31.95 to 129.8 ng/l (mean 62.51 ng/l), and those of total DDTs were in the range 19.24–96.64 ng/l (mean 48.69 ng/l). The distribution and behavior characteristics of OPs indicate that five main OPs (methamidophos, dichlorvos, malathion, omethoate and dimethoate) occupied the most part of the total OPs, in addition, the sources of the OPs (mainly from current usage) are not stable. While the OCs mainly came from early application and were more stable. The OPs did not have obvious correlation with the dissoved organic carbon and suspended particulate matter at water from JLRE. It maybe due to the complicated hydrological kinetics in the estuary, and the higher water solubility and weak absorption (with organic matter) action of OPs. When compared with other areas, the OPs and OCs level in JLRE are some moderate.  相似文献

Incoherent scatter radars (ISR) are versatile instruments for continuous monitoring of ionisation processes in the Earths atmosphere. EISCAT, The European Incoherent Scatter facility has proven effective also in meteor studies. The time resolution of the radar can be reduced to a few milliseconds, sufficient to resolve the passage of individual meteors through the narrow ISR beam. Methods for group and phase velocity determination of the meteoroids and the discrepancy between the results related to the target behaviour are presented. The radar cross sections of echoes associated with moving meteoroids (–meteor head echoes) are very small and increase with decreasing wavelength. The parent meteoroids are found to have visual magnitudes far below the detection limit of most optical observations. The equivalent visual magnitude limit of the smallest objects observed by EISCAT in the current experiments has been estimated by two different methods, both from the cross-section measurements and from the measured event rates. Both methods give a limit value of +10 for the smallest objects while the upper limit is +4. The lower limit of the visual magnitude for the collocated optical measurement system is +4. Thus the two detection systems observe two different meteor size ranges, with the radar almost reaching micrometeorite population. Meteor fluxes estimated from the event rates and the radar system parameters agree well with previous extrapolated values for this size range.  相似文献

Daily values of the ionospheric characteristics foF2 and M(3000)F2 for a given hour and month are correlated with the corresponding daily values of sunspot number using measured data collected at seven European locations. The significance of applying different-order polynomials is considered and the times are confirmed when the higher-order terms are important. Mean correlation coefficients for combined data sets over all hours, months and stations are determined, together with the standard errors of estimates. Comparisons are made with corresponding figures for monthly median values derived from the same data sets.  相似文献

基于背景噪声的地震方法发展迅速,已广泛应用于全球和区域地球内部结构研究、浅地表地质调查及油气田勘探开发.本文简要介绍了背景噪声的来源,回顾了噪声地震学的发展历程.给出了基于背景噪声的全波场和面波格林函数恢复的公式,较为详细综述了噪声源的分布和记录台站间距对格林函数恢复的影响.讨论了两台站互相关法和空间自相关法获取面波频散特性的区别与理论连接.对基于噪声的面波层析成像法、程函方程层析成像法、空间自相关法的原理进行了总结.介绍了噪声地震学方法在各领域特别是浅地表方面的应用现状.最后简要展望了噪声地震学的发展前景.  相似文献

The levels and distribution of tris-(2,3-dibromopropyl) isocyanurate (TBC) and hexabromocyclododecanes (HBCDs) of surface sediments in the Yellow River Delta wetland had been investigated. The concentrations of TBC and ∑ HBCDs ranged from 0.20 to 29.03 ng·g? 1 dw and below limits of detections (LODs) to 20.25 ng·g? 1 dw. The average composition profile of three HBCDs isomers were 10.1%, 6.1% and 83.8% for α-, β- and γ-HBCD, respectively. Moreover, correlation analysis indicated there are similar sources among three isomers and positive correlations between total organic carbon (TOC) content and concentrations of TBC and HBCDs. The mass inventory of TBC,α-, β-, γ-HBCD, ΣHBCDs in surface sediments of Yellow River Delta wetland were estimated about 725.50, 72.76, 44.29, 548.34, 665.39 kg. Therefore, further investigations on potential human health and environmental risk assessments of TBC and HBCDs were needed.  相似文献

This paper is the first one of a series of three papers on the fluid evolution of the crust-upper mantle and the causes of earthquakes. Their relationship between the deep-seated fluids and the seismic activities are discussed from aspects of their macoscopic scale, microscopic mechanism and dynamic behaviors in the three papers respectively. Based on magnetotelluric sounding (MT) measurements conducted by Chinese geophysicists in more than 20 years, the maps of the upper mantle conductive layer (MCL) with a buried depth of >50 km and the crustal conductive layer (CCL) with a buried depth of >15 km in the Chinese mainland are presented in this paper. The resistivity structure, the causes of conductive layers in crust-mantle and the relationships between earthquake distribution and conductive layers are discussed.  相似文献

Grass buffer strips impact the hydrology of flow and consequently the fate of sediment. A complex process‐based model is developed to predict flow characteristics as well as sediment deposition and transport upstream, and within grass strips. The model is capable of estimating the proportion and amount of different sediment particle size classes in the outflow. The modified Green–Ampt equation was used to simulate infiltration. Gradually varied flow and kinematic wave approximation were used to model flow characteristics upstream and within grass strips. The GUEST model approach has been modified in order to use its basic approaches in sediment transport module in grass strips. Model predictions agree well with the results of two sets of controlled experiments. The bias, coefficient of model efficiency and the root mean squared error of the modelled efficiency of grass strips in reducing sediment concentration were 0.93–0.99, 0.58–0.99 and 8.9–12.7, respectively. The sensitivity analysis showed that the initial soil moisture and flow rate are the most sensitive parameters in predicting runoff loss. Expected annual loss (EAL), which can be expressed in dollars, is an effective way of communicating the seismic vulnerability of constructed facilities to owners and insurers. A simplified method for estimating EAL without conducting time‐consuming non‐linear dynamic analyses is presented. Relationships between intensity measures and engineering demand parameters resulting from a pushover analysis and a modified capacity‐spectrum method are combined with epistemic and aleatory uncertainties to arrive at a probabilistic demand model. Damage measures are established to determine thresholds for damage states from which loss ratios can be defined. Financial implications due to damage can then be quantified in the form of EAL by integrating total losses for all likely earthquake scenarios. This rapid loss estimation method is verified through the computationally intensive incremental dynamic analysis, with the results processed using a distribution‐free methodology. To illustrate the application of the proposed method, the seismic vulnerability of two highway bridge piers is compared; one bridge is traditionally designed for ductility while the other is based on an emerging damage avoidance design (DAD) philosophy. The DAD pier is found to have a clear advantage over the conventional pier; the EAL of the DAD pier is less than 20% of its ductile counterpart. This is shown to be primarily due to its inherent damage‐free behaviour for small to medium earthquake intensities, whose contribution to EAL is significantly more than that of very rare events.
