Mussels are used as sentinel organisms and bioindicators to evaluate the toxic effects of chemical pollutants in marine organisms, especially heavy metals, representing an important tool for biomonitoring environmental pollution in coastal areas. Antioxidant defence enzymes play an important role in cellular antioxidant defence systems and protect from oxidative damage by reactive oxygen species (ROS). Indigenous mussels Mytilus galloprovincialis of the Saronikos Gulf of Greece were used for monitoring heavy metal pollution in three polluted sites in the area and in one unpolluted site. Seasonal variations of the activity of antioxidant defence enzymes, superoxide dismutase (SOD) and catalase (CAT), as well as lipid peroxidation (LP) were measured as biomarkers in a period of three years in relation to concentrations of trace metals in their gills and mantle and compared to mussels from an unpolluted sampling site. SOD activity increased at least 2 fold at the polluted sites when compared to the control site (the high activity was recorded in the spring time). CAT activity was increased 2-3 times at the polluted sites, with high activity in the winter and spring time, compared to the control site. LP concentration was twice higher at the polluted sites, following the same seasonal pattern. Trace metals contents in mussels collected at polluted sites were 3-4 fold higher compared to the control site and showed moderate variations along the months, with a winter maximum followed by a summer pre-spawning minimum matching the seasonal trends of temperature and salinity. Our results showed that metal pollution in the Elefsis Bay (the most polluted coastal area) causes relatively medium levels of oxidative stress in tissues of mussels due to cellular oxy-radical generation. This study, which is the first in the area, showed that seasonal variations of the activity of antioxidant defence enzymes and LP concentrations in mussels can be used as potential biomarkers of toxicity for long-term monitoring in marine coastal ecosystems.  相似文献

In the period 1997-2000, approximately 1,800,000 m3 of material dredged from the Port of Leghorn was discharged into a sea dumping site located 14 miles from the coast. The red mullet (Mullus barbatus) was used as a bioindicator species for monitoring the biological impact of these discharges on a geographical and temporal scale. Organisms were sampled over three years (1998-2000) at different stations and several biomarkers, both of exposure and effect, were analyzed. Bioavailability of specific classes of pollutants was evaluated by analyzing levels of metallothioneins, the activity of cytochrome P450 1A (CYPIA) and of glutathione S-transferases. Among biomarkers of effect, special attention was paid to the balance between prooxidant challenge and antioxidant defenses, and to the appearance of damage caused by oxidative stress. The analyses of the main components of the antioxidant system included superoxide dismutase, catalase, glutathione peroxidases, glutathione reductase, glyoxalase I and II, and total glutathione. These data were integrated with the measurement of total oxyradical scavenging capacity (TOSC) as an indication of the overall biological resistance to toxicity of different forms of oxyradicals (peroxyl radicals, hydroxyl radicals and peroxynitrite). Results indicated a biological impact in organisms sampled near the disposal site; the impact was particularly evident during 1999 and mainly related to organic chemicals such as PAH. Exposure to these pollutants also caused variations in the levels and activity of several antioxidants. The analysis of TOSC, however, revealed that the overall capacity of specific tissues of organisms to absorb various oxidants was not seriously compromised when challenged with increased prooxidant pressures. Variations of single antioxidants were useful in revealing early warning "biological responses", while integration with TOSC analyses indicated if such changes also reflect a more integrated and functional "biological effect" with possible consequences at the organisms level. The red mullet appears to be a useful sentinel species for a biomarker approach to monitoring impact caused by dredged materials.  相似文献

Local marine environments in China's Pearl River Delta (PRD), the most rapidly developing region in one of the world's fastest growing economies, have been experiencing significant environmental stress during the past decades. This investigation was conducted to determine the status and trends of persistence organic pollutants (POPs) such as polycyclic aromatic hydrocarbons (PAHs), petroleum hydrocarbons (PHCs), polychlorinated biphenyls (PCBs), organochlorine (OC) pesticides and dioxin-related compounds in marine sediments collected from sixteen coastal stations in the Pearl River Delta (PRD) in March 2003. Elevated concentrations of PAHs (94-4300 ng/g), PCBs (6.0-290 ng/g), PHCs (14-150 microg/g), and DDTs (1.4-600 ng/g) were detected in sediment samples. In addition, 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD)-like activities in the sediment samples were estimated to range from 0.3 to 440 pg TCDD-EQ/g. Sediments collected from Xiashan contained the greatest concentrations of trace organic contaminations amongst all the sampling stations in the present study. The degree of trace organic contamination was, in general, more severe at stations situated along the west shores of the PRD than their counterparts in the east. A preliminary assessment was performed to examine the probable risks to the marine ecosystem due to POPs. The results showed that OC pesticide contamination in the PRD was particularly serious and might pose a threat to the health of the marine inhabitants.  相似文献

Aliphatic hydrocarbons and PAHs in the whole soft tissues of Bathymodiolus azoricus from three Mid-Atlantic Ridge hydrothermal vents (Menez-Gwen, Lucky Strike and Rainbow), and Mytilus galloprovincialis from three contaminated coastal sites in South Portugal were analysed, and its effects on the digestive gland microsomes mixed-function oxygenase system (MFO) were assessed. Aliphatic hydrocarbons levels were present in the same magnitude in both coastal and hydrothermal environments, while the UCM (unresolved complex mixture) for coastal mussels were higher than in vent mussels. In general, significantly higher PAHs concentrations were found in coastal mussels, compared to B. azoricus where low molecular weight PAHs (2-3 rings) represented the majority of PAHs contrarily to what was observed in M. galloprovincialis. The MFO components were present in both mussel species, and were detected in vent mussels for the first time. However this system seems to have different roles in species from these contrasting environments. In coastal mussels MFO responded to hydrocarbon contamination while response in hydrothermal organisms appeared to be related mainly to endogenous factors.  相似文献

Polycyclic aromatic hydrocarbons (PAHs) and organochlorine pesticides (OCs) are generally present in the marine environment in complex mixtures. The ecotoxicological nature of contaminant interactions, however, is poorly understood, with most scientific observations derived from single contaminant exposure experiments. The objective of this experiment was to examine dose-response relationships between antioxidant parameters and body contaminant levels in mussels exposed to different exposure regimes under laboratory conditions. Accordingly, the green-lipped mussel, Perna viridis, was challenged with a mixture of PAHs (anthracene, fluoranthene, pyrene, benzo[a]pyrene) and OC pesticides (α-HCH, aldrin, dieldrin, p,p′-DDT) over a 4 week period. Contaminants were delivered under four different dosing regimes, with all treatments receiving the same total contaminant load by the end of the exposure period. Antioxidant biomarkers were measured after 1, 2, 3 and 4 weeks, including glutathione (GSH), gluathione-S-transferase (GST), superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), glutathione reductase (GR) and lipid peroxidase (LPO). GST and CAT were induced in hepatic tissues in most of the exposure regimes, with the majority of significant induction occurring in a constant exposure regime and a two-step alternate exposure regime. Significant differences among exposure regimes were detected in the body burden of contaminants after 28 days. Hepatic CAT and GSH are proposed as potentially useful biomarkers as they showed good correlation with target contaminants and were not readily affected by different dosing patterns.  相似文献

The concentrations of BHCs, DDTs and PCBs have been determined in samples of Engraulis encrasicholus, Mytilus galloprovincialis, Nephrops norvegicus, Mullus barbatus, Euthymnus alletteratus and Sarda sarda, collected in different stations of the Ligurian Sea. Increasing levels of contamination were found in the order: Nephrops, Mytilus and Engraulis, Mullus and Euthymnus, Sarda.A comparison with the data available in the literature relevant to Mytilus samples collected in Genoa in 1973–1974 showed a notable decrease in the concentration of chlorinated residues.  相似文献

Polycyclic aromatic hydrocarbons (PAHs) are important environmental pollutants due to their persistence and bioaccumulation potential both in organisms and in sediments. In this study, bioaccumulation and depuration experiments were performed employing local Mediterranean mussel species, Mytilus galloprovincialis, with two biomarkers: filtration rate and lysosomal stability (neutral red retention) assay. Benzo(a)anthracene (BaA) was chosen as the model PAH compound due to its common presence in several matrices in the marine environment. Bioconcentration Factors (BCFs) for the mussels exposed to different BaA concentrations were calculated from both kinetic rate constants and from the experimental data and found between 27-3184 and 16-2745, respectively. Experimental QSAR values were found comparable to the previous QSAR models created for BaA. The effect of BaA exposure on the mussels showed a more clear dose-response relationship according to the results of lysosomal stability compared to the filtration rate.  相似文献

Zooplankton is an essential component of the food chains of marine and brackish environments. Ecotoxicological risk to zooplankton, evaluated by the biomarker approach, can be used as an early warning signal of risk to the health of marine ecosystems. The aim of this project is to evaluate the application of certain biomarkers in zooplankton. In the present paper, laboratory and field studies were performed with the copepods Acartia margalefi, Acartia latisetosa and the mysid Siriella clausi. Acetylcholinesterase activity (AChE) was determined in homogenates of whole organisms. Mean AChE activity was 10.05 micromol min g(-1) for A. margalefi, 3.30 micromol min g(-1) for A. latisetosa and 79.70 micromol min g(-1) for S. clausi. A. margalefi and S. clausi also showed a linear increase in enzyme activity with increasing concentrations of samples. Preliminary laboratory work was also carried out with specimens of A. latisetosa, which were exposed to an organophosphorus insecticide (parathion). Moderate inhibition of AChE (19%) with respect to controls was observed. AChE activities were two orders of magnitude higher in the zooplankton than in hemolymph samples of the decapod Carcinus aestuarii (Fossi et al., 1996) indicating that these species have a high metabolic rate, which makes them suitable for biomarker studies.  相似文献

However, in more recent years it has been used to characterize sublethal actions of pollutants in aquatic organisms. For instance, in vivo NMR involves live, intact organisms or cell cultures and the application of chemical stressors to reveal toxic mechanisms in real time. Alternatively, NMR-based metabolomics involves rapid cessation of metabolic activity following chemical exposure (via liquid N(2)) to provide an assessment of metabolic actions via more traditional NMR analysis. Two examples are briefly presented to exemplify the power of NMR for assessing toxic actions in marine and freshwater organisms.  相似文献

In Hong Kong, discharges of domestic and industrial wastewaters and disposal of contaminated muds into coastal waters have resulted in high levels of persistent organic contaminants in the water column, biota and bottom sediments. Although the acute toxicity of effluents has been tested by government regulators using mortality-based bioassays, the sublethal effects of many of the pollutants, including environmental carcinogens, have not been well studied. The objective of the present work was to examine the efficiency and efficacy of using: (1) DNA strand breaks (comet assay and alkaline agarose gel electrophoresis assay); (2) oxidative DNA damage (levels of 8-hydroxy-2-deoxyguanosine in cellular DNA) and (3) micronuclei (MN) formation as early warning toxicological indicators of potential environmental carcinogen exposure. Green-lipped mussels (Perna viridis) were transplanted to study sites and retrieved after 4, 8, 12, 16 and 30 days. The bioaccumulation of organic pollutants, including the polycyclic aromatic hydrocarbons (PAHs), organochlorine pesticides (OC pesticides) and polychlorinated biphenyls (PCBs) was observed, and compared with biomarker assays. Some significant dose- and time-dependent inductions of the genotoxic biomarkers, including 8-OHdG, DNA strand breaks and micronuclei were observed. In particular, several very strong (r>0.9) and significantly positive relationships were found at all sites during days 0-12. In addition, significant strong relationships between certain organic contaminants (carcinogenic PAHs and 4 common OC pesticides) and micronuclei were detected at the polluted sites, Tsim Sha Tsui and Sai Wan Ho. The results suggest that the use of micronuclei as a biomarker is potentially effective, especially at the more polluted sites. Nonetheless, further research is required to validate their effectiveness under varying field conditions, especially to elucidate responses under lower contaminant loads, interactions between the environmental and physiological factors affecting organisms in differing coastal environments, and changes in biotransformation and DNA repair enzymes in test organisms.  相似文献

The presence of alkylphenols (APs) in coastal and marine ecosystems is not as well-documented as it is in freshwater ecosystems. This paper reviews reported concentrations of alkylphenol ethoxylates (APEOs) and APs in seawater, sediments and organisms of marine environments such as estuaries, coastal lagoons, bights, harbours or deep sea in order to study their distribution. Overall contamination of marine aquatic compartments by APs and APEOs has been observed, while coastal areas in the vicinity of wastewater discharges are more impacted than deep sea environments, but to a lesser extent than freshwater sites. Sediments act as sinks for APs and APEOs, especially around wastewater discharge sites. Reported AP concentrations in marine organisms are higher in bivalves and gastropods than in fishes. As nonylphenols and octylphenols are estrogenomimetic, biological responses induced in marine organisms are discussed. Finally, we describe the cell bioassay approach for the biodetection of APs.  相似文献

Sediment toxicity assessments using caged organisms present advantages over using laboratory and native community studies. The use of caged Arenicola marina in sediment toxicity assessments was evaluated. Lugworms were exposed in situ to sediments from coastal and port areas in Spain for seven days, and the activities of the biotransformation enzymes ethoxyresorufin O-deethylase, dibenzylfluorescein dealkylase and glutathione S-transferase, the activities of the antioxidant enzymes glutathione reductase and glutathione peroxidase and lipid peroxidation were then analyzed as biomarkers. Biomarker results and sediment physicochemical data were integrated. Cádiz Bay (SW Spain) sediments presented metal contamination that was not linked to a biochemical response. In LPGC Port (SW Spain), Pb contamination exhibited a moderate toxic potential, while PAHs, and presumably pharmaceuticals, provoked biochemical responses that efficiently prevented lipid peroxidation. In Santander Bay (N Spain), exposure to PAHs and, presumably, pharmaceuticals induced biomarker responses, but lipid peroxidation occurred nevertheless. These results indicated that caged A. marina were effective for the assessment of sediment quality and that the selected biomarkers were sufficiently sensitive to identify chemical exposure and toxicity.  相似文献

Baseline information is presented on embryo malformation rate and biomarkers in fish as indicators of sub-lethal stress caused by pollution in coastal waters of Xiamen, PR China. Fish and eggs were sampled from several areas in Xiamen coastal waters (Xiamen Harbour, Maluan and Tongan Bays and East Channel), where varying levels of pollutant input have been documented. Comparative sampling was done at a "cleaner" reference site at Dongshan Island. Embryonic malformation rates, which indicate general water quality, varied with location and species of fish, and exceeded background levels for unpolluted waters (assumed approximately 5%) by up to eightfold at some sites. Generally, sites around Xiamen Harbour show signs of poor water quality having highest mean levels of embryo deformity (20-30%) and these decreased towards open waters (Tongan Bay, Eastern Channel) where abnormalities approached background levels. An indication that toxic contaminants may be having a localised effect in the region, particularly in the harbour was reinforced by the biomarker assays. However, activities of the biomarkers ethoxyresorufin O-deethylase (EROD) and glutathione S-transferase in fish livers indicate no clear pattern, and there is no evidence that fish from the four sampling areas have been more or less exposed to PAHs and other compounds that induce these biomarkers. Antioxidant biomarkers (glutathione peroxidase, catalase, superoxide dismutase, and reduced glutathione) suggest that exposure to xenobiotics appears to be lowest in Dongshan and Maluan and highest in the harbour and Tongan. Inhibition of acetylcholinesterase in fish muscle indicated possible effects by organophosphate and carbamate pesticides in Xiamen waters and these effects may be greatest in the area of the harbour.  相似文献

The influence of three pesticides, chlorphenvinphos, carbaryl and propoxur on phosphatase, dehydrogenase and catalase activities of different bacterial species isolated from water were investigated. The pesticides tested were found to inhibit the phosphatase and dehydrogenase of the respiration cycle of bacteria and disturbed seriously the catalytic function of catalase by inhibition or excessive stimulation. The pesticides affected most strongly the phosphatase activity of bacteria causing a potential hazard to the phosphorus regime of water reservoirs. The degree of impairment exerted by the pesticides tested on the enzymatic activity of bacteria depended definitively upon the characteristics of bacteria studied, the chemical structure of the pesticides and the doses applied.  相似文献

Summary Two methods of the evaluation of sensible heat flux are briefly presented from the view point of their application to micrometeorology. The comparison of sensible fluxes for coastal and inland stations is presented, in order to explain the influence of the marine atmosphere on these fluxes. They reach a maximum value in summer at the coastal stations, due to maximum wind speed and high convective activity developed by the influence of the marine atmosphere.  相似文献

Microplastics have been recently identified as marine pollutants of significant concern due to their persistence, ubiquity and potential to act as vectors for the transfer and exposure of persistent organic pollutants to marine organisms. This study documents, for the first time, the presence and abundance of microplastics (>1.6 microm) in Singapore's coastal environment. An optimized sampling protocol for the collection and analysis of microplastics was developed, and beach sediments and seawater (surface microlayer and subsurface layer) samples were collected from nine different locations around the coastline. Low density microplastics were separated from sediments by flotation and polymer types were identified using Fourier transform infrared (FTIR) spectrometry. Synthetic polymer microplastics identified in beach sediments included polyethylene, polypropylene, polystyrene, nylon, polyvinyl alcohol and acrylonitrile butadiene styrene. Microplastics were detected in samples from four out of seven beach environments, with the greatest quantity found in sediments from two popular beaches in the eastern part of Singapore. Polyethylene, polypropylene and polystyrene microplastics were also found in the surface microlayer (50-60 microm) and subsurface layer (1m) of coastal waters. The presence of microplastics in sediments and seawater is likely due to on-going waste disposal practices from industries and recreational activities, and discharge from shipping.  相似文献

The common mussel Mytilus galloprovincialis was selected as unique biomonitor species to implement a regional monitoring programme, the CIESM Mediterranean Mussel Watch (MMW), in the Mediterranean and Black Seas. As of today, and upon standardization of the methodological approach, the MMW Network has been able to quantify (137)Cs levels in mussels from 60 coastal stations and to produce the first distribution map of this artificial radionuclide at the scale of the entire Mediterranean and Black Seas. While measured (137)Cs levels were found to be very low (usually < 1 Bq kg(-1) wet wt) (137)Cs activity concentrations in the Black Sea and North Aegean Sea were up to two orders of magnitude higher than those in the western Mediterranean Basin. Such effects, far from representing a threat to human populations or the environment, reflect a persistent signature of the Chernobyl fallout in this area.  相似文献

Sea-surface microlayer (SML) and seawater samples collected from Singapore's coastal marine environment were analyzed for selected chlorinated pesticides and polychlorinated biphenyls (PCBs). The SML is a potential site of enrichment of persistent organic pollutants (POPs) compared to the underlying water column. The concentration ranges of SigmaHCH, SigmaDDT and SigmaPCB in subsurface (1 m depth) seawater were 0.4-27.2 ng/l (mean 4.0 ng/l), 0.01-0.6 ng/l (mean 0.1 ng/l) and 0.05-1.8 ng/l (mean 0.5 ng/l) respectively. In the SML, the concentration ranges of SigmaHCH, SigmaDDT and SigmaPCB were 0.6-64.6 ng/l (mean 9.9 ng/l), 0.01-0.7 ng/l (mean 0.2 ng/l) and 0.07-12.4 ng/l (mean 1.3 ng/l) respectively. High spatial and temporal distribution was observed for all POPs measured. However, overall levels measured in the SML were lower than levels reported in the literature for SML samples from temperate coastal regions-possibly due to loss of semi-volatile compounds in the tropical climate of Singapore. Atmospheric wet deposition during the monsoon season may be an important source of POPs to the SML. This study provides the first scientific data on POP concentrations and enrichment factors in the SML for Southeast Asia.  相似文献

Sediments and marine organisms such as mussels (Mytilus galloprovincialis), fish (Mullus barbatus, Serranus scriba), crabs (Polybius henslowi) and shrimp-like crustaceans (mysids, euphausiids) were sampled on European coasts, France, Spain) and analysed for their PAH content by gas chromatography/mass spectrometry (GC-MS). The PAH concentrations determined in the organisms were significantly negatively correlated with logKow values, but to different extents according to the species. This was explained by the greater exposure of the organisms to the lower molecular weight compounds with differences resulting from different feeding habits, habitat, and biotransformation capacities of the organisms in relation to trophic levels. The influence of these observations on the use of different sentinel species in biomonitoring programmes is discussed.  相似文献
