Here, a novel one-dimensional composite of poly(m-phenylenediamine)s coating on filamentous Streptomyces was successfully constructed via a controllable polymerization reaction. The synthesized composites were characterized by scanning electron microscopy, Fourier transform infrared spectroscopy, and X-ray photoelectron spectroscopy. Their adsorption isotherm and kinetics for aqueous hexavalent chromium were also systematically examined. The results of scanning electron microscopy analysis indicated that the obtained composites based on Streptomyces were showed a uniform and stable one-dimensional morphology with distinct core–shell configuration. Moreover, the Langmuir isotherm model (R 2 > 0.96) and pseudo-second-order equation (R 2 = 0.9996) described well the equilibrium adsorption behavior and kinetics of hexavalent chromium adsorption by the composites. In addition, bath adsorption experiments demonstrated the highest adsorption capacity of hexavalent chromium by the composites reached 320.03 mg g?1 in an acid solution, which was 5.6 times as that of the pure Streptomyces filaments. The results of Fourier transform infrared spectroscopy and X-ray photoelectron spectroscopy analyses suggested that the adsorption of hexavalent chromium by the composites possibly involved the protonation, redox, and chelation reactions. Therefore, a promising application of these composites in treating acid hexavalent chromium-contaminated wastewater is expectable.  相似文献

New bio-adsorbent carbon materials were synthesized from the leaves and veins of Mucuna pruriens and Manihot esculenta plants, which are locally available in abundance. The synthesized carbons were activated using 0.01N HNO3. Surface area of the activated carbons from M. pruriens and M. esculenta plants was found to be quite high, i.e., 918 and 865 m2/g, respectively. Scanning electron microscopy analysis of the carbons reflects complex disorganized surface structures of different open pore sizes, shapes and dimensions. These properties of the newly synthesized activated carbons led to the development of a sand-supported carbon column, for its possible use in the removal of coliform bacteria and Escherichia coli (E. Coli) from raw water samples. The removal percentage of E. coli was found to be 100% with both the types of carbon adsorbents, as confirmed from the McCardy most probable number table. Similarly, the removal percentage of coliform bacteria was found to be 99 and 98.7% by M. pruriens and M. esculenta carbon columns, respectively. These activated carbons synthesized from locally available plants possess the characteristics of good low-cost adsorbents which can be easily used for the removal of bacteria from water by adsorption method.  相似文献

Novel bionanocomposites, S. cerevisiae–AgNPs, were synthesized by in situ formation of AgNPs on S. cerevisiae surface using fulvic acids as reductants under simulated sunlight. S. cerevisiae–AgNPs were characterized using UV–Vis spectroscopy, scanning electron microscope, transmission electron microscope and Fourier transform infrared spectroscopy. These analyses showed that AgNPs were distributed on the surface of S. cerevisiae. The application of S. cerevisiae–AgNPs in bacteria killing and heavy metal removal was studied. S. cerevisiae–AgNPs effectively inhibited the growth of E. coli with increasing concentrations of S. cerevisiae–AgNPs. E. coli was killed completely at high concentration S. cerevisiae–AgNPs (e.g., 100 or 200 µg mL?1). S. cerevisiae–AgNPs as excellent heavy metal absorbents also have been studied. Using Cd2+ as model heavy metal, batch experiments confirmed that the adsorption behavior fitted the Langmuir adsorption isotherms and the Cd2+ adsorption capacity of S. cerevisiae–AgNPs was 15.01 mg g?1. According to adsorption data, the kinetics of Cd2+ uptake by S. cerevisiae–AgNPs followed pseudo second-order kinetic model. Moreover, S. cerevisiae–AgNPs possessed ability of different heavy metals’ removal (e.g., Cr5+, As5+, Pb2+, Cu2+, Mn2+, Zn2+, Hg2+, Ni2+). The simulated contaminated water containing E. coli, Cd2+ and Pb2+ was treated using S. cerevisiae–AgNPs. The results indicated that the bionanocomposites can be used to develop antibacterial agents and bioremediation agents for water treatment.  相似文献

The main objective of this study was to investigate the efficiency of biological treatment of azo dye-containing wastewater with a sequencing batch reactor system, followed by ultrafiltration. The performance of the system was quantified by measuring the chemical oxygen demand and azo dye concentration. The biodegradation was carried out under combined alternating anaerobic and aerobic conditions with Nylosan Yellow E2RL SGR as a model azo dye contaminant. The bioprocess revealed a maximal reduction in chemical oxygen demand and dye removal efficiency of 91 and 85%, respectively. After ultrafiltration of effluent from the biological treatment, the efficiency increased to 94% for chemical oxygen demand and to 97% for the azo dye decolourisation. Samples of activated sludge from the bioprocess were collected for microbial characterisation. Bacteria and fungi were isolated and identified by 16S rRNA gene and ITS1-5.8S rDNA-ITS2 sequence analysis, respectively. Serratia marcescens and Klebsiella oxytoca were the most common bacteria with the highest number present during the aerobic and anaerobic phases of the bioprocess. In addition, a high number of Elizabethkingia miricola, Morganella morganii, Comamonas testosteroni, Trichosporon sp. and Galactomyces sp. were detected. Taken together, our results demonstrated that the sequencing batch reactor system combined with ultrafiltration is an efficient technique for treatment of wastewater containing azo dye. Moreover, the ultrafiltration effectively removes the microbiota from the final effluent resulting in stable product water.  相似文献

Cadmium and other heavy metals lead to environmental danger, and these heavy metals are a great threat to human and other animal’s health. Investigation of the relationship between survival of E. coli and metallothionein smtA gene expression against cadmium ion is the goal of this research. Survival of recombinant bacteria containing smtA gene was analyzed against various concentrations of cadmium chloride salt using optical density (OD). At the resistive range, recombinant bacteria were subjected to different treatments. At the logarithmic phase of bacterial growth, sampling, RNA extraction and cDNA synthesis were performed and smtA gene expression was then analyzed by real-time PCR using designed primers for smtA gene and Amp resistance (as the calibrator gene). Relative gene expression was calculated using the ??Ct method. The resistive range against cadmium chloride was 0.5–0.7 mM (minimum inhibitory concentration (MIC = 0.5 mM)). Survival and gene expression analysis showed that in induced bacteria, smtA expression was increased significantly that in turn conferred resistance to cadmium chloride prominently. There was a direct relationship between increased smtA gene expression and survival of the recombinant bacteria. Therefore, our result may help to confront to cadmium metal environmental pollution using overexpression of smtA gene expression in recombinant bacteria.  相似文献

This paper reports the results of the experimental determination of the coefficient of sorption of rare-earth elements and yttrium (REY) under conditions of acidic pH using several strains of heterotrophic bacteria (Microbacterium sp., Curtobacterium sp., Bacillus subtilis, Pseudomonas putida, and Bacillus pumilis) that are widespread in natural and technogenic waters of the Far East (Russia). Insignificant fractionation between heavy and light rare-earth elements, negative cerium and dysprosium anomalies, and a positive europium anomaly were revealed. The selectivity of REY biosorption by gram-positive and gram-negative bacteria, as well as inertia of the biosorption process under more acidic conditions of the medium, were shown.  相似文献

The accumulation efficiency of selected trace elements in the leaves of Melandrium album and Robinia pseudoacacia grown on heavy metal contaminated sites in comparison with a non-contaminated one was evaluated. The study was undertaken to calculate air pollution tolerance index and to determine the contents of selected metabolites: glutathione, non-protein thiols, ascorbic acid, chlorophyll and the activity of antioxidant enzymes: guaiacol peroxidase and superoxide dismutase. Such estimations can be useful in better understanding of plants defense strategies and potential to grow in contaminated environments. The results in the most contaminated site revealed higher contents of metals in M. album leaves, especially Zn, Cd and Pb (3.4, 6 and 2.3 times higher, respectively) in comparison with the R. pseudoacacia. Better accumulation capacity found in M. album was shown by metal accumulation index values. The plants could be used as indicators of Zn, Cd (both species) and Pb (M. album) in the soil. Glutathione content (in both species) and peroxidase activity (in M. album), general markers of heavy metals contamination, were increased in contaminated sites. In most cases in contaminated areas R. pseudoacacia had decreased ascorbic acid and chlorophyll levels. Opposite tendency was recorded in M. album leaves, where similar or higher contents of the above-mentioned metabolites were found. In our study, M. album and R. pseudoacacia proved to be sensitive species with the air pollution tolerance index lower than 11 and can be recommended as bioindicators.  相似文献

Four bacterial strains, capable of degrading diesel oil, n-alkanes or hexadecane, were isolated from soils contaminated with petroleum oil and identified. Strains of Pseudomonas sp., Pseudomonas putida TPHK-1 and Pseudomonas aeruginosa TPHK-4, were more efficient in degrading high concentrations of the hydrocarbons than the other two strains, Stenotrophomonas maltophilia TPHK-2 and Acenitobacter sp. TPHK-3. P. putida TPHK-1 exhibited tolerance to very high concentrations of heavy metals such as cadmium, lead, zinc and copper. The innate ability of P. putida TPHK-1, as evidenced by the amplified genes alkB1 and alkB2 that encode alkane hydroxylases, and cat12o and cat23o coding for catechol dioxygenase, in degrading diesel oil in the presence of heavy metals is far greater than that of the strains reported in the literature. Heavy metal tolerance coupled with rapid degradation of hydrocarbons, even at high concentrations, suggests that P. putida TPHK-1 has a great potential in remediating soils contaminated with mixtures of hydrocarbons and heavy metals.  相似文献

The study of low-cost techniques for the tertiary treatment of wastewater is of global interest; above all low-energy techniques that do not require the use of chemicals. In this study, a wastewater treatment technology based on the filtration by a zooplanktonic population (Daphnia magna) is studied in controlled laboratory and mesocosm experiments for different hydraulic retention times (HRT). The efficiency of the treatment is evaluated in terms of particle removal efficiency. From laboratory experiments, HRT over 12 h and Daphnia concentrations above 50 individuals l?1 guarantee a particle removal efficiency greater than 30 %. However, low HRT of 6 h would require Daphnia concentrations above 70 individuals l?1 in order to obtain a particle removal efficiency of 20 %. The minimum removal efficiency of 2 % was for HRT = 3 h, independent of the Daphnia concentration. In the mesocosm, the growth of Daphnia individuals enhanced Daphnia magna filtering rates and higher removal efficiencies than those in the laboratory for the same HRT range. In the mesocosm experiments E. coli concentrations were reduced to a maximum of 2 logarithmic units. A balance equation model is proposed to predict particle removal efficiencies for varying HRT.  相似文献

The development of the bioaugmentation during the phytoremediation of contaminated water with diesel in pilot horizontal subsurface flow constructed wetlands was investigated for 63 days. The objective of this study was to examine the enrichment of rhizobacteria in a pilot-scale system for efficient treatment of total petroleum hydrocarbon (TPH) effluent. A consortium of three rhizobacteria strains (Bacillus aquimaris, Bacillus anthracis and Bacillus cereus), which were able to utilize hydrocarbon compounds as sole carbon sources, was injected into the constructed wetlands (batchwise operation) planted with Scirpus grossus. The TPH removals from water, without or with the addition of rhizobacteria, were found to be 72 and 84%, while from sand was found to be 59 and 77%, for each treatment, respectively. These results showed that the rhizobacteria strains could enhance S. grossus growth by decreasing diesel stress and protecting S. grossus against diesel, with 12 and 18% additional TPH removal from water and sand, respectively. Our results demonstrate that S. grossus is potential to improve the phytoremediation of hydrocarbon contaminants through inoculation with effective rhizobacterial strains.  相似文献

This research is focused on evaluating heavy metals (Cd, Cu, Fe, Mn, Pb, and Zn) uptake and removal by Eleocharis ovata, Cyperus manimae, Typha dominguensis, and Pteridium aquilinum in a natural wetland impacted by mining activities. We analyzed heavy metals content and distribution in native plants, soils, and water of a semipermanent natural wetland in Taxco de Alarcón, Guerrero, and we also determined the physicochemical characteristics of the water. Translocation factor (TF) and bioconcentration factor (BCF) were evaluated. Results showed that physical and chemical conditions are favorable for plants development. Correlation analysis showed a good and positive relation (0.95) between Cu and Pb in soils and plants. In the analyzed matrices: Zn (0.62–2.20 mg/L) exceeded the permissible limits in water, high concentrations of Pb and Zn (26.57–525.67 and 266.67–983.33 mg/kg, respectively) were detected in the studied soils, and Pb exceeded the normal range for E. ovata and P. aquilinum in the analyzed plants. Uptake of heavy metals in the tissues of different species was found in the following order: root > leaf. Data of TF and BCF showed that E. ovata is a tolerant plant with respect to heavy metals exposure since TF value was greater than 1. This study showed that E. ovata could be considered as a bioaccumulator of heavy metals in contaminated soils.  相似文献

This study investigated the relationship between wastewater environments and the nitrifiers at a full-scale plant using principal component analysis. Ammonia-oxidizing bacteria and nitrite-oxidizing bacteria were detected by florescent in situ hybridization, polymerase chain reaction, phylogenetic analysis, real-time quantitative polymerase chain reaction. Pyrosequencing was also used in profiling the ammonia monooxygenase locus of ammonia-oxidizing bacteria community. It was found that the dominant ammonia-oxidizing bacteria sequences were related to uncultured ammonia-oxidizing bacterium, uncultured Nitrosomonadaceae bacterium, Nitrosomonas sp., and uncultured bacterium. In addition, Nitrobacter clones were related to uncultured alpha proteobacterium, uncultured bacterium, uncultured Nitrobacter sp., and uncultured Bradyrhizobium sp., whereas Nitrospira clones were similar to uncultured bacterium, Candidatus Nitrospira defluvii, uncultured Nitrospira sp., and uncultured Nitrospirae bacterium. The ammonia-oxidizing bacteria and nitrite-oxidizing bacteria ranged 2.83 × 108–1.33 × 1010 and 1.25 × 1010–1.13 × 1011 copies L?1, respectively, equivalent to nitrite-oxidizing bacteria: ammonia-oxidizing bacteria ratio of 10:1. The first three parts of the principal components analysis accounted for 76.8% of the explained variance. The first principal component (44.4%) designated that ammonia-oxidizing bacteria and Nitrospira were mainly influenced by seasonal variations, followed by chemical oxygen demand concentration and nitrogen species (i.e., ammonia, nitrite, and nitrate). The second principal component (19.1%) showed no information about the nitrifiers’ interaction with environmental factors, whereas Nitrobacter demonstrated a high correlation with ammonia on the third principal component (13.3%). These results revealed that the species of Nitrobacter were less influenced by environmental conditions than ammonia-oxidizing bacteria and Nitrospira spp.  相似文献

In this study, Illumina sequencing was used for the identification of bulking and foaming bacteria in industrial wastewater treatment plants. The reliable identification of bulking and foaming bacteria represents the first step in developing effective and specific control strategies to avoid disturbances in activated sludge systems. Illumina sequencing revealed 432 16S rRNA operational taxonomic units, representing phylotypes and including 21 bulking and foaming bacteria in the two investigated industrial wastewater treatment plants. Foaming represents the most severe problem in the cascade biology system. Up to 22.5% of all sequencing reads are bulking and foaming bacteria, including Chryseobacterium, Candidatus Microthrix parvicella and Gordonia sp. as the dominant bulking and foaming bacteria which are known for foam formation. Moreover, Illumina sequencing revealed an increase in Candidatus Microthrix parvicella and Gordonia sp. reads from activated sludge to foam and scum samples, indicating a preferred flotation and/or growth advantages in the foam and scum layers. Analyses of the taxonomic assignment and distribution showed that the phylum Actinobacteria is the most dominant phylum, underlining the key role of Actinobacteria in bulking and foaming. Multivariate data analysis was applied, revealing that the dominant bulking and foaming bacteria are positively correlated with the sludge age and influent flow and negatively correlated with the dissolved oxygen level and the temperature. In terms of developing a specific control strategy, the positive linear relationships to the fatty acid and surfactant sludge loadings are highlighted and the removal of lipid compounds from the wastewater influent could avoid an overgrowth of bulking and foaming bacteria.  相似文献

Phenol and its methylated derivatives, cresol isomers, are hazardous pollutants that are commonly present in various industrial effluents and known to have detrimental effect on aquatic life as well as human health, due to their toxic and carcinogenic nature. It is essential, therefore, to reduce the concentration of these contaminants in industrial effluent to acceptable levels prior to being discharged into the environment. Bacterial cells of the strain Pseudomonas putida, with excellent biodegradation capabilities and high tolerance of cresols, were extracted and immobilized in polyvinyl alcohol (PVA) gel for cresols biodegradation. The biodegradation was carried out at different operating conditions, in both batch and continuous modes, using a cylindrical spouted bed bioreactor. Factors affecting o-cresol and m-cresol degradation were studied in batch experiments, and the results showed that the immobilized bacteria could tolerate cresols concentration up to 200 mg/l. Moreover, the experiments indicated that the biodegradation rate was highly affected by the operating parameters such as pH and temperature, with optimum ranges of 6–8 for pH and 30–35 °C for temperature. However, the optimum conditions were different for each cresol isomer. The potential of P. putida in degrading binary and ternary mixtures of cresols was also examined in the continuous process and compared with single component biodegradation. The experimental results revealed that the biodegradation of o-cresol was highly inhibited by the presence of p-cresol and m-cresol.  相似文献

The aim of this work was to evaluate the usefulness of Lemna minuta Kunth for the simultaneous removal of Cr(VI) and phenol. The impact of these contaminants on plant growth and some biochemical processes have also been discussed for a better understanding and utilization of this species in the field of phytoremediation. The optimal growth conditions and plant tolerance to Cr(VI) and/or phenol as well as removal were determined. Plants exposed to Cr(VI) and phenol were able to efficiently grow and remove both contaminants at high concentrations (up to 2.5 and 250 mg/L, respectively) after 21 days, indicating that they were resistant to mixed contamination. There were no significant differences between chlorophyll, carotene and malondialdehyde content of treated plants with respect to the controls, which would be due to an efficient antioxidant response. L. minuta showed a higher biomass than control without contaminant when was exposed to low concentrations of Cr(VI), suggesting an hormesis effect. The main removal process involved in chromium phytoremediation would be sorption or accumulation in the biomass. Moreover, our results suggest that phenol could be used as a donor of carbon and energy by these plants. These findings demonstrated that Lemna minuta Kunth might be suitable for treatment of different solutions contaminated with Cr(VI) and phenol, showing a high potential to be used in the treatment of effluents containing mixed contamination.  相似文献

Lack of nutrients is a common challenge for enteric pathogens, like Shigella, when discharged in waters and soils by infected persons. These bacteria must evolve protective mechanisms to survive nutrient changing conditions. Many studies have demonstrated that bacterial starvation can affect cell morphology and surface properties such as lipopolysaccharide content. This study focuses on the morphological and physiological alterations of Shigella flexneri, a human pathogen, under nutrient starvation during 30 days. Our results showed that S. flexneri can survive under nutrient deficiency during 4 weeks. However, starved cells revealed several morphological changes indicating decrease of the size and change of the cell shape as observed by transmission electron microscopy. Furthermore, the outer membrane proteins and lipopolysaccharide profiles of starved cells revealed the disappearance, the appearance, and changes of the protein band levels. The survival and physiological adaptation of S. flexneri under nutrient limitation during 30 days may increase the risk of human and animal infections.  相似文献

Axenic culture of microalgae Chlorella vulgaris ATCC® 13482 and Scenedesmus obliquus FACHB 417 was used for phycoremediation of primary municipal wastewater. The main aim of this study was to measure the effects of normal air and CO2-augmented air on the removal efficacy of nutrients (ammonia N and phosphate P) from municipal wastewater by the two microalgae. Batch experiments were carried out in cylindrical glass bottles of 1 L working volume at 25 °C and cool fluorescent light of 6500 lux maintaining 14/10 h of light/dark cycle with normal air supplied at 0.2 L min?1 per liter of the liquid for both algal strains for the experimental period. In the next set of experiments, the treatment process was enhanced by using 1, 2 and 5% CO2/air (vol./vol.) supply into microalgal cultures. The enrichment of inlet air with CO2 was found to be beneficial. The maximum removal of 76.3 and 76% COD, 94.2 and 92.6% ammonia, and 94.8 and 93.1% phosphate after a period of 10 days was reported for C. vulgaris and S. obliquus, respectively, with 5% CO2/air supply. Comparing the two microalgae, maximum removal rates of ammonia and phosphate by C. vulgaris were 4.12 and 1.75 mg L?1 day?1, respectively, at 5% CO2/air supply. From kinetic study data, it was found that the specific rates of phosphate utilization (q phsophate) by C. vulgaris and S. obliquus at 5% CO2/air supply were 1.98 and 2.11 day?1, respectively. Scale-up estimation of a reactor removing phosphate (the criteria pollutant) from 50 MLD wastewater influent was also done.  相似文献

Waste engine oil causes a vital environmental pollution when it spill during change and transportation and products of waste engine oil causes lethal effects to the living systems. Thus, abiotic and biotic approaches are being extensively used for removal of waste engine oil pollution. Therefore in present study, waste engine oil degradation was accomplished by a new bacterial culture, isolated from the soil by an enrichment technique. Morphological, biochemical and gene sequence analysis revealed that isolate was Bacillus cereus. Subsequently, biodegradation potential of B. cereus for waste engine oil was studied. Experimental variables, such as pH, substrate concentration, inoculum size, temperature and time on the biodegradation, were checked in mineral salt medium. The biodegradation efficiency of B. cereus was determined by gravimetry, UV–visible spectrophotometry and gas chromatography. In addition, waste engine oil was also characterized by GC–MS and FTIR for its major constituents, which showed total 38 components in waste engine oil, including hopanes, benzopyrene, long-chain aliphatic hydrocarbons, dibenzothiophenes, biphenyl and their derivatives. Results of successive biodegradation indicated that B. cereus was capable to degrade 1% of waste engine oil with 98.6% degradation potential at pH 7 within 20 days. Hence, B. cereus presents an innovative tool for removing the engine oil from the contaminated area.  相似文献

Moringa oleifera seed was used as a biosorbent for the removal of water treatment disinfection by-products, haloethers, trihalomethanes and haloketones from water samples. M. oleifera seed has polar functional groups such as O–H, C=O, C–N and others which facilitate the extraction of these disinfection by-products. Experiments were conducted in batch and fixed bed column modes. Using batch mode, different parameters were optimized such as M. oleifera seed dosage, sample pH, contact time and agitation speed. The effect of the thickness layer and flow rate were also studied using the fixed bed mode. Maximum adsorption occurs using the batch mode with removal efficiencies of 120.5, 114 and 111.5 mg/g for haloethers, trihalomethanes and haloketones, respectively. Adsorption equilibrium followed Langmuir model, and the kinetic data obeyed the pseudo-second-order model, revealing that the M. oleifera seed had higher adsorption capacity than other reported sorbents. Shorter removal time with higher adsorption capacity of disinfection by-products by M. oleifera seed suggests that this material was effective for water treatment, in dealing with the removal of the disinfection by-products considered.  相似文献

Contamination with heavy metals in surface and groundwater is a threat to human health and ecosystems. Due to this, the need arises to remediate water polluted through ecological and profitable technologies, such as phytoremediation. The objective of the work was to evaluate the concentration of lead (Pb) and zinc (Zn) in the floating macrophytes Limnobium laevigatum and Ludwigia peploides, after being exposed to contaminated water experimentally. In this way to be able to determine if these plants have mechanisms that allow them to accumulate the metals in the roots and to perform the translocation of these to different vegetative organs, L. laevigatum and L. peploides were placed in solutions contaminated with Pb ([Pb]?=?5 mg/l) and Zn ([Zn]?=?20 mg/l). The concentrations of metals in water, root and leaf samples were evaluated as a function of time (0, 1, 2 and 4 days). The determination of the metals was performed by the atomic absorption spectrophotometry technique. After 4 days of exposure to Pb and Zn, the plants showed high metal removal efficiencies of water, more to 70% in all cases. Pb was accumulated fundamentally by roots, while Zn was accumulated more in the leaves. In addition, the bioconcentration and translocation factors for each metal were calculated.  相似文献
