In this study, we are reporting bacterial diversity in crude oil-contaminated soil of Assam, India. Integration of physiological community profiling, culture-dependent and culture-independent (metagenome) approaches, was employed to obtain a complete picture of the total bacterial diversity. Samples collected from 10 sites contaminated with crude oil ranging from 0.22 to 89.36% were analysed, and altogether 160 culturable bacteria were isolated (117 Gram-positive and 43 Gram-negative bacteria). Molecular identification showed the predominance of genera Lysinibacillus, Alcaligenes, Bacillus, Clostridium, Enterobacter and Pseudomonas. Conversely, denaturing gradient gel electrophoresis (DGGE) profiles of 16S rDNA phylotypes showed the predominance of Sphingomonas, Ralstonia, Sphingobium, Massilia, Acinetobacter and Pseudomonas. Both culture-dependent and culture-independent approaches resulted in 11 genera of which Bacillus and Pseudomonas were the key inhabitants creating most favourable bacterial milieu in the presence of crude oil contaminants. Metabolic fingerprints data depicted in PCA plot demonstrated that sites CTF-D-1 and Core-10 were most diverged. It was further confirmed that variations of bacterial species dominance in different sites were due to origin of hydrocarbon contamination. We here claim that the present findings is a first-hand report on combined physiological community profiling, culture-based and culture-independent approaches in assessing total bacterial diversity in crude oil-contaminated soil of Assam.  相似文献

The lipA gene, encoding a solvent-tolerant extracellular lipase from Proteus sp. SW1, was displayed on the cell surface of Escherichia coli by fusing it to an antigen 43 anchoring motif. The display of LipA on the Escherichia coli cell surface was directly confirmed by immunofluorescence microscopy and flow cytometry. After 6 days of incubation in media containing 1 % used cooking oil, an Escherichia coli strain expressing surface displayed lipase was able to degrade 27 % of the oil. The biosurfactant, pseudopyronine B, was purified from culture supernatants of Pseudomonas sp. SL31. Its critical micelle concentration was determined to be 1400 mg/l, and the surfactant was stable within a temperature range from 0 to 120 °C and a pH range of 3–11. Pseudopyronine B-containing crude media extracts efficiently removed up to 51 % of the cadmium from contaminated water. We demonstrated the oil degradation ability of the mixed culture of four bacterial strains, namely the recombinant Escherichia coli expressing cell surface displayed lipase (pKKJlipA), His-tagged lipase (pETlipA), extracellular lipase-producing Proteus sp. SW1, and pseudopyronine B-producing Pseudomonas sp. SL31 by culturing in LB media containing 1 % oil. The consortium degraded 29 % of oil in one day and reached 84 % after 7 days.  相似文献

The aim of this research was to evaluate the potential of six legumes: Medicago sativa L., Glycine max, Arachis hypogea, Lablab purpureus, Pheseolus vulgaris and Cajanus cajan to restore within a short period of time soil contaminated with 3% crude oil. The legumes in five replications were grown in crude oil-contaminated and crude oil-uncontaminated soil in a completely randomized design. Plants were assessed for seedling emergence, plant height and leaf number. GC–MS was used to analyze the residual crude oil from the rhizosphere of the legumes. Plant growth parameters were reduced significantly (P < 0.05) for legumes in contaminated soil compared to their controls. In the 4th week after planting (WAP), shoot height increased across the species up to the 8th WAP. However, in the 12 WAP no significant increase in the shoot of all species was observed. Two WAP legumes planted in contaminated soil had significantly (P < 0.05) higher leaf number than these planted in uncontaminated soil with the exception of M. sativa. In the 4th WAP, only A. hypogea and P. vulgaris had increased leaf number, while in the 6th WAP, only L. purpureus had increased leaf number and survived up to the 12th WAP while most of the legumes species died. Chromatographic profiles indicated 100% degradation of the oil fractions in C. cajan and L. purpureus after 90 days. For other legumes however, greater losses of crude oil fractions C1–C10 and C10–C20 were indicated in rhizosphere soil of P. vulgaris and G. max, respectively. The most effective removal (93.66%) of C21–C30 components was observed in G. max-planted soil even though vegetation was not established. The legumes especially C. cajan, L. purpureus and A. hypogea are promising candidates for phytoremediation of petroleum hydrocarbon-impacted soil.  相似文献

Four bacterial strains, capable of degrading diesel oil, n-alkanes or hexadecane, were isolated from soils contaminated with petroleum oil and identified. Strains of Pseudomonas sp., Pseudomonas putida TPHK-1 and Pseudomonas aeruginosa TPHK-4, were more efficient in degrading high concentrations of the hydrocarbons than the other two strains, Stenotrophomonas maltophilia TPHK-2 and Acenitobacter sp. TPHK-3. P. putida TPHK-1 exhibited tolerance to very high concentrations of heavy metals such as cadmium, lead, zinc and copper. The innate ability of P. putida TPHK-1, as evidenced by the amplified genes alkB1 and alkB2 that encode alkane hydroxylases, and cat12o and cat23o coding for catechol dioxygenase, in degrading diesel oil in the presence of heavy metals is far greater than that of the strains reported in the literature. Heavy metal tolerance coupled with rapid degradation of hydrocarbons, even at high concentrations, suggests that P. putida TPHK-1 has a great potential in remediating soils contaminated with mixtures of hydrocarbons and heavy metals.  相似文献

Waste engine oil causes a vital environmental pollution when it spill during change and transportation and products of waste engine oil causes lethal effects to the living systems. Thus, abiotic and biotic approaches are being extensively used for removal of waste engine oil pollution. Therefore in present study, waste engine oil degradation was accomplished by a new bacterial culture, isolated from the soil by an enrichment technique. Morphological, biochemical and gene sequence analysis revealed that isolate was Bacillus cereus. Subsequently, biodegradation potential of B. cereus for waste engine oil was studied. Experimental variables, such as pH, substrate concentration, inoculum size, temperature and time on the biodegradation, were checked in mineral salt medium. The biodegradation efficiency of B. cereus was determined by gravimetry, UV–visible spectrophotometry and gas chromatography. In addition, waste engine oil was also characterized by GC–MS and FTIR for its major constituents, which showed total 38 components in waste engine oil, including hopanes, benzopyrene, long-chain aliphatic hydrocarbons, dibenzothiophenes, biphenyl and their derivatives. Results of successive biodegradation indicated that B. cereus was capable to degrade 1% of waste engine oil with 98.6% degradation potential at pH 7 within 20 days. Hence, B. cereus presents an innovative tool for removing the engine oil from the contaminated area.  相似文献

Maryland Coastal Bays differ in hydrography from river-dominated estuaries because of limited freshwater inflow from tributary creeks and more marine influence. Consequently, the copepod community structure may be different from that of the coastal ocean and river-dominated estuaries in the mid-Atlantic region. A 2-year study was conducted to describe copepod species composition and seasonal patterns in abundance and factors influencing the community structure. Seven copepod genera, Acartia, Centropages, Pseudodiaptomus, Parvocalanus, Eurytemora, Oithona, and Temora, in addition to harpacticoids were found. The copepod community was dominated by Acartia spp. (64%), followed by Centropages spp. (30%), unlike in river-dominated estuaries in the region where the copepod community is usually dominated by Acartia spp. followed by Eurytemora affinis. Acartia tonsa was the most abundant in summer and fall whereas Centropages spp., Temora sp., Oithona similis, E. affinis, and harpacticoids were most abundant in winter and early spring. Parvocalanus crassirostris and Pseudodiaptomus pelagicus were present in fall and winter but at relatively low densities. The highest mean density of copepods occurred in winter 2012 (36,437 m?3) and the lowest in spring 2013 (347 m?3). Low densities occurred through early summer (614 m?3) coinciding with peak spawning by bay anchovy (Anchoa mitchilli). Bottom-up control via low phytoplankton biomass coupled with top-down control by ctenophores (Mnemiopsis sp.), mysids (Neomysis americana), and bay anchovy was probably responsible for the low copepod densities in spring and early summer. Temperature and salinity were also important factors that influenced the seasonal patterns of copepod species occurrence. The observed seasonal differences in the abundance of copepods have important implications for planktivorous fishes as they may experience lower growth rates and survival due to food limitation in spring/early summer when copepod densities are relatively low than in late summer/fall when copepod abundance is higher.  相似文献

Low saline water flooding (LSWF) had proved to be an efficient method for enhanced oil recovery in clay-bearing hydrocarbon reservoirs, but the interaction mechanisms among in-situ rocks – fluids and injection fluids within the reservoir – are not yet known properly. Understanding the molecular level interaction among these components is critical for designing and field scale implementation of LSWF in clay-bearing crystalline reservoir rocks, which is very limited in the existing literature. A weathered amphibolite rock and one dead crude oil from the Bakrol field (Cambay basin, India) have been used in this study. The presence of clay minerals in the weathered amphibolite rock was observed using a polarising microscope and characterised by the X-ray diffraction (XRD) and Fourier transform infrared (FTIR) techniques. The crude oil and its fractionated SARA components have been extensively studied by spectroscopic techniques for their characterisation. The interaction study among the rock powder, hydrocarbon crude oil and saline water has been performed in the present work for gaining better insight for designing the injection fluid for LSWF. The weathered amphibolite rock powder was mixed with the dead crude oil and kept for 30 days in room temperature (T) and pressure (P) for proper interaction. The XRD, FTIR and cation exchange capacity results clearly demonstrated the incorporation of crude oil components in the interlayer surfaces of clay minerals. The oil removal efficiency, from the oil-treated rock powder of three saline water samples having NaCl concentration of 3000, 5000 and 8000 ppm, was investigated using the UV–Vis and fluorescence spectroscopies. The low saline NaCl water is capable of removing the maximum amount of polar components from the oil-treated rock powder. These molecular level insights are valuable for designing effective injection fluid for enhancing the oil recovery from the clay-rich crystalline reservoir rock.  相似文献

The revised representatives of ammonite genera Malbosiceras and Pomeliceras from the Berriasian of the Crimean Mountains are classed with seven species, four of the first genus [M. malbosi (Pictet), M. chaperi (Pictet), M. broussei (Mazenot), M. pictetiforme Tavera] and three of the second one [P. aff. boisseti Nikolov, P. breveti (Pomel), P. (?) funduklense Lysenko et Arkadiev sp. nov.]. The identified species are described. The genus Mazenoticeras is considered as synonym of Malbosiceras. The above species prove that all the Berriasian zones (jacobi, occitanica and boissieri) are characteristic of corresponding deposits in the Crimean Mountains.  相似文献

We analyze three-phase flow of immiscible fluids taking place within an elementary capillary tube with circular cross-section under water- and oil-wet conditions. We account explicitly for momentum transfer between the moving phases, which leads to the phenomenon of viscous coupling, by imposing continuity of velocity and shear stress at fluid-fluid interfaces. The macroscopic flow model which describes the system at the Darcy scale includes three-phase effective relative permeabilities, K i j,r , accounting for the flux of the ith phase due to the presence of the jth phase. These effective parameters strongly depend on phase saturations, fluid viscosities, and wettability of the solid matrix. In the considered flow setting, K i j,r reduce to a set of nine scalar quantities, K i j,r . Our results show that K i j,r of the wetting phase is a function only of the fluid phase own saturation. Otherwise, K i j,r of the non-wetting phase depends on the saturation of all fluids in the system and on oil and water viscosities. Viscous coupling effects (encapsulated in K i j,r with ij) can be significantly relevant in both water- and oil-wet systems. Wettability conditions influence oil flow at a rate that increases linearly with viscosity ratio between oil and water phases.  相似文献

The development of the bioaugmentation during the phytoremediation of contaminated water with diesel in pilot horizontal subsurface flow constructed wetlands was investigated for 63 days. The objective of this study was to examine the enrichment of rhizobacteria in a pilot-scale system for efficient treatment of total petroleum hydrocarbon (TPH) effluent. A consortium of three rhizobacteria strains (Bacillus aquimaris, Bacillus anthracis and Bacillus cereus), which were able to utilize hydrocarbon compounds as sole carbon sources, was injected into the constructed wetlands (batchwise operation) planted with Scirpus grossus. The TPH removals from water, without or with the addition of rhizobacteria, were found to be 72 and 84%, while from sand was found to be 59 and 77%, for each treatment, respectively. These results showed that the rhizobacteria strains could enhance S. grossus growth by decreasing diesel stress and protecting S. grossus against diesel, with 12 and 18% additional TPH removal from water and sand, respectively. Our results demonstrate that S. grossus is potential to improve the phytoremediation of hydrocarbon contaminants through inoculation with effective rhizobacterial strains.  相似文献

The Jurassic successions represent a wide distribution in North of the Kerman province. These successions include Ab-Haji, Badamu, and Hojedk formations. The Hojedk Formation contains the plant fossils. The Lenjan section is one of the suitable areas for paleontological studies on the Hojedk Formation. The study section is mostly composed of green sandstone and shale with several interbedded coal veins with different thicknesses. The thickness of the Hojedk Formation is about 200 m in the Lenjan section. In this study, seven genera and 13 species of macro plant fossils were identified and described, including Nilssonia undulata, Nilssonia bozorga, Nilssonia berriesi, Nilssonia sp., Klukia cf. exilis, Klukia exilis, Cladophlebis antarctica, Coniopteris lobata, Coniopteris murrayana, Elatocladus confertus, Podozamites sp., Equisetites sp., and Coniopteris sp. The Bajocian–Bathonian can be attributed to the Lenjan section based on the recognized flora.  相似文献

Fine sediment inputs can alter estuarine ecosystem structure and function. However, natural variations in the processes that regulate sediment transport make it difficult to predict their fate. In this study, sediments were sampled at different times (2011–2012) from 45 points across intertidal sandflat transects in three New Zealand estuaries (Whitford, Whangamata, and Kawhia) encompassing a wide range in mud (≤63 μm) content (0–56 %) and macrofaunal community structure. Using a core-based erosion measurement device (EROMES), we calculated three distinct measures of sediment erosion potential: erosion threshold (? c ; N m?2), erosion rate (ER; g m?2 s?1), and change in erosion rate with increasing bed shear stress (m e ; g N?1 s?1). Collectively, these measures characterized surface (? c and ER) and sub-surface (m e ) erosion. Benthic macrofauna were grouped by functional traits (size and motility) and data pooled across estuaries to determine relationships between abiotic (mud content, mean grain size) and biotic (benthic macrofauna, microbial biomass) variables and erosion measures. Results indicated that small bioturbating macrofauna (predominantly freely motile species <5 mm in size) destabilized surface sediments, explaining 23 % of the variation in ? c (p ≤ 0.01) and 59 % of the variation in ER (p ≤ 0.01). Alternatively, mud content and mean grain size cumulatively explained 61 % of the variation in m e (p ≤ 0.01), where increasing mud and grain size stabilized sub-surface sediments. These results highlight that the importance of biotic and abiotic predictors vary with erosion stage and that functional group classifications are a useful way to determine the impact of benthic macrofauna on sediment erodibility across communities with different species composition.  相似文献

Coal contains abundant microbial genera which include archaebacteria. The study of archaea kingdom in coal mines is a significant tool for knowing the relationship between coal and archaebacteria, the major role in geochemical cycle and application for further coal bio–beneficiation. The present study related to exploration of archaebacteria and their habitat in coal mining area of Dhanbad with reference to their ecology and nutrient availability that have evolve to grow under extreme conditions. Total six different sites such as two underground coal mines (Sudamdih shaft and Chasnalla underground mine), two opencast coal mines (Chandan project and Bhowra abandoned mine), Jharia mine fire and Sudamdih coal washery of Dhanbad was selected. Seven gram negative obligate anaerobic bacteria were isolated from the selected sites. The isolated species were rod and cocci shaped and the colony was round, smooth, off white in colour and with entire margin and little are cluster of cocci in shape. The isolated species were identified as Methanococcus spp, Methanobacterium spp and Methanosarcina spp. Apart from that two thermoacidophilic sulfur oxidizing bacteria Sulfolobus spp was also isolated from Jharia Coal Mine Fire. The physicochemical and biological characterization of the habitat was also studied for the entire selected area.  相似文献

Spectroscopic and photometric data for the two rapidly rotating members of the α Persei cluster He 373 and AP 225 are analyzed. Improved estimates have been obtained for the projected equatorial rotation velocities: v sin i = 164 km/s for He 323 and v sin i = 129 km/s for AP 225. Multi-band photometric mapping is used to map the spot distributions on the surfaces of the two stars. The fractional spotted areas S and mean temperature difference ΔT between the unspotted photosphere and the spots are estimated (S = 7% and ΔT = 1000 K for He 373; S = 9% and ΔT = 800 K for AP 225). The H α line profiles of both stars have variable emission components whose widths are used to deduce the presence of extended regions of emission reaching the corotation radius.  相似文献

The removal of petroleum and petroleum-based products from the environment is of great importance. The objectives of this study were to investigate the most suitable physiological conditions and the effects of additional carbon, nitrogen and surfactant sources on petroleum biodegradation by Klebsiella pneumoniae ATCC13883 isolated from drilling fluid and to evaluate petroleum biodegradation with detailed hydrocarbon analysis by GC–MS. The results indicated that the highest biodegradation rate of 66.5% for K. pneumoniae was obtained under the conditions of pH 7, petroleum concentration 1% (v/v) and 7-day incubation at 150 rpm and 25 °C, proving to be the most effective physical conditions for petroleum biodegradation in this present study. Additional sources such as Triton X: 100, glucose and yeast extract significantly enhanced the petroleum biodegradation of K. pneumoniae to 68, 71 and 72.5%, respectively. In the last stage of this study, biodegradation rates were above 90% for hydrocarbons ranging from C10 and C20, above 70% for hydrocarbons ranging from C21 and C22 and above 40% for hydrocarbons ranging from C31 and C32. In conclusion, oil field adapted K. pneumoniae could efficiently degrade short-, medium- and long-chain alkanes in petroleum and thus is a potential source for advanced petroleum treatment.  相似文献

We obtained speckle interferometric and spectroscopic observations of the system 41 Dra during its periastron passage in 2001. The components’ lines are resolved in the spectral interval 3700–9200 Å. The observed wavelength dependence of the brightness difference between the components is used to estimate the B-V indices separately for each of the components: B-V = 0.511 for component a and B-V = 0.502 for component b. We derived improved effective temperatures of the components from their B-V values and hydrogen-line profiles. The observations can be described with the parameters for the components T eff a = 6370 K, log ga = 4.05 and T eff b = 6410 K, log gb = 4.20. The iron, carbon, nitrogen, and oxygen abundances in the atmospheres of the components are log N(Fe)a = 7.55, log N(Fe)b = 7.60, log N(C)a = 8.52, log N(C)b = 8.58, log N(N)a = 8.05, log N(N)b = 7.99, log N(O)a = 8.73, log N(O)b = 8.76.  相似文献

Three commercial bio-aid samples have shown good troubleshooting capabilities earlier during several biological wastewaters processing, but their microbial contents were still unclear. This study was aimed to assess eubacteria contents for the three bio-aids based on sequence comparisons of 16S ribosomal deoxyribonucleic acid (rDNA). Majority of target 16S rDNA sequences of bacteria within or cultured from bio-aids were obtained via pyrosequencing-based approach, and few sequences were from isolation- or cloning-sequencing approaches. Results showed that a total of 8114, 2327, and 5402 sequences were retrieved from pyrosequencing-based methods for the three bio-aids, respectively, which were affiliated with 100, 80, and 59 bacterial species based on sequence comparison using Ribosome Data Project (RDP) Classifier and Basic Local Alignment Search Tool (BLAST). Target sequences matched Bacillus sp., Enterococcus sp., and Clostridium sp. of Firmicutes phylum were the most abundant for the bio-aids, while some sequences identical to bacteria of phyla Actinobacteria, Bacteroidetes, Chloroflexi, Cyanobacteria, and Proteobacteria were also found. Additionally, most of sequences from the isolation- or cloning-sequencing-based methods were not retrieved by the pyrosequencing-based methods. Culture media appeared to potentially select several bacteria species within phyla Firmicutes and/or Proteobacteria, but sequences from the culture-based methods did fulfill some absences for the uncultured method. Based on principal coordinate analysis, bacteria community structures of the three bio-aids were different. This study provided insights into bacteria contents for the three bio-aids as fundamental information in attempts for future selection or formulation of suitable bio-aids for solving various troubles during wastewater biological processing.  相似文献

A rich assemblage of palynomorphs including miospores and megaspores has been recorded from the bore core no. IBKAN-2 drilled at a depth of 17.80m in Kuraloi Block-A, south-west part of the Ib-River Coalfield, Jharsuguda district, Odisha. The miofloral assemblage recovered shows prominence of cingulate, zonate, taeniate, non-striate and non saccate palynotaxa which indicate an early Triassic age and is comparable with the palynoassemblages of the same age known from the Damodar basin, India. The megaspore assemblage includes eight genera and fifteen species namely, Banksisporites karanpuraensis, B. indicus, B. utkalensis, Banksisporites sp., Barakarella pantii, Barakarella sp., Biharisporites spinosus, Biharisporites sp., Erlansonisporites triassicus, Erlansonisporites cf. erlansonii, Erlansonisporites sp. Jhariatriletes sp., Singhisporites sp., Ramispinatispora sp. and Talchirella trivedii. Records of megaspores are sparse in the early Triassic of Gondwana basins of India. The present study records the first occurrence of miospores and megaspores from the early Triassic of Mahanadi basin besides substantiating the earlier data.  相似文献

Plug-shaped ichnofossils Conichnus conicus, Conostichus broadheadi and C. stouti are found in the intercalated micritic sandstone and sandy allochemic limestone shale sequence of Bagh Group, Narmada district, Gujarat. These ichnospecies occur at two stratigraphic levels and shows distinct morphological features interpreted as resting/dwelling structures of sea anemone. The occurrence of these ichnospecies along with oyster fossils genera like Bosostrea and Indostrea indicate shallow marine environment.  相似文献

The correlation between oxides of nitrogen emission and in-cylinder temperature of diesel engine fueled with various alternative fuels has been investigated in this research paper. Experimentations were performed in engine without any modifications using pure high-speed diesel fuel, used cooking oil biodiesel (UCO20), animal fat residue biodiesel (AFR20) and camphor oil (CMR20) at 20% volume concentration of biodiesel each. From combustion analysis, the heat release rate and peak cylinder pressure of biodiesel blends were about 13.487% lower and 4.819% higher than those of diesel fuel on an average, respectively. Longer combustion duration has been observed for all biodiesel blends at all load conditions. Oxides of nitrogen emission level show 16.405, 10.352 and 7.524% increment for UCO20, AFR20 and CMR20, respectively. Noteworthy NO x reduction of about 43.8% was recorded for diesel blended with camphor oil when compared to other biodiesel blends. The relationship between in-cylinder temperature and NO x emission concentration was premeditated through thermal imager. The result depicted that the increase in NO x concentration depends on augmented in-cylinder temperature for all test fuels.  相似文献
