In this research, the bioremediation of dispersed crude oil, based on the amount of nitrogen and phosphorus supplementation in the closed system, was optimized by the application of response surface methodology and central composite design. Correlation analysis of the mathematical‐regression model demonstrated that a quadratic polynomial model could be used to optimize the hydrocarbon bioremediation (R2 = 0.9256). Statistical significance was checked by analysis of variance and residual analysis. Natural attenuation was removed by 22.1% of crude oil in 28 days. The highest removal on un‐optimized condition of 68.1% were observed by using nitrogen of 20.00 mg/L and phosphorus of 2.00 mg/L in 28 days while optimization process exhibited a crude oil removal of 69.5% via nitrogen of 16.05 mg/L and phosphorus 1.34 mg/L in 27 days therefore optimization can improve biodegradation in shorter time with less nutrient consumption.  相似文献

Effects of nutrients on crude oil biodegradation in the upper intertidal zone   总被引:10,自引:0,他引:10

To enhance biodegradation, nutrients in the form of slow-release fertilizer (SRF) were applied to oil-contaminated microcosms (3%, v/v) which simulated intertidal environmental systems. Although nutrient concentrations in the interstitial water were not proportional to those in amended SRF, microbial activity, growth of oil-degrading microorganisms, and oil-degradation rate were closely related to the concentration of nutrients in the interstitial water. Adding nutrients at higher dose (microcosm I, 144.4 mg C/kg sand/day, versus microcosm II, 8.5 mg C/kg sand/day) had a positive effect on oil degradation rate, which was especially obvious during the early phase of treatment. Use of pristane, phytane, and nor-hopane as biomarkers enabled the detection of significant treatment differences in hydrocarbon biodegradation, which were not reliable enough when the data were normalized to sand mass.  相似文献

Nitrogen and phosphorous fertilizers were used to determine their short-term summer effects on algal colonization, abundance, and species composition in moderate herbivory treatments. Secondary succession of algae on coral skeletons was examined in four treatments: an untreated control, a pure phosphate fertilizer, a pure nitrogen fertilizer, and an equal mix of the two fertilizers. Turf algae cover was the only measure of algae abundance to respond significantly to fertilization. Turf cover was three times higher in treatments with added nitrogen when compared with the pure phosphorus treatment. These turfs were dominated by green and cyanobacteria taxa, namely Enteromorpha prolifera, Lyngbya confervoides, and two species of Cladophora. The phosphate treatment was dominated by encrusting corallines and the cyanobacteria L. confervoides, while the controls had the highest cover of frondose brown algae, namely Padina sanctae-crucis and two species of Dictyota. Results indicate that turf algae were co-limited by nitrogen and phosphorus but enrichment appeared to inhibit brown frondose algae that currently dominate these reefs. Number of species was lowest on the pure phosphorus and nitrogen treatments, highest in the controls and intermediate in the mixed treatments, which suggests that diversity is reduced most by an imbalanced nutrient ratio.  相似文献

Petroleum biodegradation and oil spill bioremediation   总被引:27,自引:0,他引:27
Hydrocarbon-utilizing microorganisms are ubiquitously distributed in the marine environment following oil spills. These microorganisms naturally biodegrade numerous contaminating petroleum hydrocarbons, thereby cleansing the oceans of oil pollutants. Bioremediation, which is accomplished by adding exogenous microbial populations or stimulating indigenous ones, attempts to raise the rates of degradation found naturally to significantly higher rates. Seeding with oil degraders has not been demonstrated to be effective, but addition of nitrogenous fertilizers has been shown to increase rates of petroleum biodegradation. In the case of the Exxon Valdez spill, the largest and most thoroughly studied application of bioremediation, the application of fertilizer (slow release or oleophilic) increased rates of biodegradation 3–5 times. Because of the patchiness of oil, an internally conserved compound, hopane, was critical for demonstrating the efficacy of bioremediation. Multiple regression models showed that the effectiveness of bioremediation depended upon the amount of nitrogen delivered, the concentration of oil, and time.  相似文献

Many research studies have demonstrated the feasibility and efficacy of fertilization with nitrogen and phosphorus to combat oil spills in marine environments. Rapid dilution of water-soluble nutrients can be overcome by oleophilic formulations that retain optimal nutrient concentrations at the oil-water interface where biodegradation occurs. Previous work has demonstrated that biodegradation processes are enhanced by the addition of lipophilic fertilizers of natural origin (uric acid and lecithin). In this work, we examined the effectiveness of these nutrients in combination with biosurfactants (rhamnolipids) and molasses (source of C and vitamins) to enhance the biodegradation by naturally occurring microorganisms. It was found that the use of biosurfactants resulted in an increased removal of petroleum hydrocarbons (96% removal of C19-C34 n-alkanes within a period of 18 days) as well as in a reduction of the lag phase (almost 80% removal was achieved within the first week of biosurfactant application).  相似文献

This field study was a combined chemical and biological investigation of the relative rates of weathering and biodegradation of oil spilled in sediments and testing the influence of a bioremediation protocol. The aim of the chemistry work presented here was to determine whether the bioremediation protocol affected the rate of penetration, dissipation or long-term retention of a medium range crude oil (Gippsland) and a Bunker C oil stranded in tropical Rhizophora sp. mangrove and Halosarcia sp. salt marsh environments. Permission for the planned oil spills was granted in the Port Authority area of Gladstone, Queensland (Australia). Sediment cores from three replicate plots of each treatment for mangroves and four replicate plots for the salt marsh (oil only and oil plus bioremediation) were analysed for total hydrocarbons (THC) and for individual alkane markers using gas chromatography with flame ionization detection (GC–FID). Sediments were collected at day 2, then 1, 2, 5 or 6 and 12 or 13 months post-spill for mangroves and day 2, 1, 3 and 9 months post-spill for salt marshes. Over this time, hydrocarbons in all of the oil treated plots decreased exponentially. There was no statistical difference in initial oil concentrations, penetration of oil to depth, or in the rates of oil dissipation between untreated oil and bioremediated oil in the mangrove plots. The salt marsh plots treated with the waxy Gippsland oil showed a faster rate of biodegradation of the oil in the bioremediated plots. In this case only, the degradation rate significantly impacted the mass balance of remaining oil. The Bunker C oil contained only minor amounts of highly degradable n-alkanes and bioremediation did not significantly impact its rate of loss in the salt marsh sediments. At the end of each experiment, there were still n-alkanes visible in the gas chromatograms of residual oils. Thus it was concluded that there was unlikely to be any change in the stable internal biomarkers of the oils over this time period. The predominant removal processes in both habitats were evaporation and dissolution, with a lag-phase of 1–2 months before the start of microbial degradation.  相似文献

Fertilizers and liming agents are generally used to achieve optimal economic yields. However, several negative effects of long‐term annual fertilization of nitrogen (N) in particular have been observed, such as reduced cation exchange capacity and decreased base saturation, which may stimulate accelerated leaching loss of other nutrients. Equilibrium‐tension lysimeters installed at a depth of 1.4 m were used to evaluate leaching of soil‐solution ionic constituents from a tallgrass prairie restoration and optimally and deficiently N‐fertilized, no‐tillage (NT) and chisel‐plowed (CP) maize (Zea mays L.) agroecosystems on Plano silt loam soil (fine‐silty, mixed, superactive, mesic Typic Argiudoll). This study was conducted in south‐central Wisconsin, USA during a 1‐year period of above‐normal precipitation between January 2000 and January 2001. The loss of soluble constituents added to agroecosystems to maintain adequate soil fertility and pH, such as N, phosphorus, potassium, calcium, and magnesium, was generally numerically smaller from the natural prairie 25 years after conversion from cultivated agriculture than from optimally and deficiently N‐fertilized, NT and CP maize agroecosystems, though the differences were not significant. Tillage and fertilizer N‐rate generally did not significantly affect drainage, ionic concentrations, or their leaching losses in the maize agroecosystems, though all parameters evaluated tended to be numerically smaller in the deficiently than optimally N‐fertilized maize agroecosystems. Nitrate‐N leaching losses were generally significantly positively correlated with leaching losses of K, Ca, Mg, and Na in the maize agroecosystems, but not for the prairie, indicating that nitrate‐N leaching plays a significant role in the concomitant loss of cations to maintain soil‐solution charge balance in N‐fertilized maize agroecosystems in a temperate environment.  相似文献

Crude oil hydrocarbon composition characteristics and oil viscosity prediction are important bases in petroleum exploration.A total of 54 oil/heavy-oil samples and 17 oil sands were analyzed and quantified using both comprehensive 2D gas chromatography(GC×GC)and comprehensive 2D gas chromatography/time-of-flight mass spectrometry(GC×GC/TOFMS).The results show that crude oil in the West slope is mainly heavy oil and its hydrocarbon composition is characterized overall by paraffinsmono-aromaticsnaphthenesnon-hydrocarbonsdi-aromaticstri-aromaticstetra-aromatics.Aromatics are most abundant and non-hydrocarbons are least abundant, whilst content differences among paraffins, naphthenes, aromatics, and non-hydrocarbons are less than 15%.There are two types of heavy oil, secondary type and mixing type.Biodegradation is the main formation mechanism of heavy oil.Biodegradation levels cover light biodegradation, moderate biodegradation, and severe biodegradation. With increasing biodegradation, paraffin content decreases while contents of aromatics and nonhydrocarbons increase. In contrast, naphthene content increases first and then decreases with increasing biodegradation. In severe biodegradation stage, naphthenes decrease more quickly than aromatics and non-hydrocarbons.This provides a new method for studying oil/heavy-oil biodegradation mechanism and biodegradation resistance of different hydrocarbons at different biodegradation stages. In the Longhupao-Daan terrace and Qijia-Gulong depression, most crude oil is conventional oil.Its composition is dominated by paraffins with the lowest content of aromatics. In some casual oil wells from the Longhupao-Daan terrace, crude oil from Saertu oil reservoirs is moderately biodegraded whereas crude oil from Putaohua oil reservoir is lightly biodegraded.Chemical parameters using saturate hydrocarbons and aromatics are usually not suitable for determining organic type and thermal maturity of biodegraded oil, especially of moderately or severely biodegraded oil, whilst Ts/(Ts+Tm)ratio can be used to determine thermal maturity of both conventional crude oil and heavy oil.  相似文献

A preliminary study was undertaken to determine the optimal conditions for the biodegradation of a crude oil. Among 57 oil‐degrading bacterial cultures isolated from oil‐contaminated soil samples, Bacillus sp. IOS1‐7, Corynebacterium sp. BPS2‐6, Pseudomonas sp. HPS2‐5, and Pseudomonas sp. BPS1‐8 were selected for the study based on the efficiency of crude oil utilization. Along with the selected individual strains, a mixed bacterial consortium prepared using the above strains was also used for degradation studies. The mixed bacterial consortium showed more growth and degradation than did individual strains. At 1% crude oil concentration, the mixed bacterial consortium degraded a maximum of 77% of the crude oil. This was followed by 69% by Pseudomonas sp. BPS1‐8, 64% by Bacillus sp. IOS1‐7, 45% by Pseudomonas sp. HPS2‐5, and 41% by Corynebacterium sp. BPS2‐6. The percentage of degradation by the mixed bacterial consortium decreased from 77 to 45% as the concentration of crude oil was increased from 1 to 12%. Temperature of 35°C and pH 7 were found to be optimum for maximum degradation.  相似文献

Nutrient concentration and hydrocarbon bioavailability are key factors affecting biodegradation rates of oil in contaminated beach sediments. The effect of a slow-release fertilizer, Osmocote, as well as two biopolymers, chitin and chitosan, on the bioremediation of oil-spiked beach sediments was investigated using an open irrigation system over a 56-day period under laboratory conditions. Osmocote was effective in sustaining a high level of nutrients in leached sediments, as well as elevated levels of microbial activity and rates of hydrocarbon biodegradation. Chitin was more biodegradable than chitosan and gradually released nitrogen into the sediment. The addition of chitin or chitosan to the Osmocote amended sediments enhanced biodegradation rates of the alkanes relative to the presence of Osmocote alone, where chitosan was more effective than chitin due to its greater oil sorption capacity. Furthermore, chitosan significantly enhanced the biodegradation rates of all target polycyclic aromatic hydrocarbons.  相似文献

Among six crude oil-degrading yeasts that were isolated from an oil-polluted area in the Persian Gulf, two yeast strains showed high degradation activity of aliphatic hydrocarbons. From an analysis of 18S rRNA sequences and biochemical characteristics, these strains were identified as Yarrowia lipolytica strains PG-20 and PG-32. Gas Chromatography (GC) analysis of the crude oil remaining in the culture medium after 1 week at 30°C showed that the strains PG-20 and PG-32 degraded 68% and 58% of crude oil, respectively. The optimal growth condition and biodegradation of hydrocarbons was in ONR medium with an acidic pH (pH5). These two strains may degrade aliphatic hydrocarbons more efficiently than aromatic hydrocarbons, although strain PG-20 had better degradation than strain PG-32. The two Y. lipolytica strains reduce surface tension when cultured on hydrocarbon substrates (1% v/v). These strains showed a cell surface hydrophobicity higher than 70%. These results suggested that Y. lipolytica strains PG-20 and PG-32 have high crude oil degrading activity due to their high emulsifying activity and cell hydrophobicity. In conclusion, these yeast strains can be useful for the bioremediation process in the Persian Gulf and decreasing oil pollution in this marine ecosystem.  相似文献

Lipophilic phosphorus and nitrogen-containing compounds were screened to attain a fertilizing composition to enhance microbial degradation of oil pollutants in the aquatic environment.Soya-bean lecithin and ethyl allophanate have been proved to be good sources of phosphorus and nitrogen, respectively, for oil degrading microorganisms. A laboratory study was conducted to examine oil biodegradation in the presence of a fouling organism community. It is shown that the majority of fouling species exhibit relatively weak tolerance to oil. Oil degradation in seawater is shown to proceed quicker and more completely in the presence of the fouling organism community than it does without such organisms. The population of specific bacterial groups is shown first to grow and next decrease as a result of accumulation in water of toxic products of their vital activity. Oil biodegradation is found to cause substantial changes in the water hydrochemical characteristics in experimental vessels.  相似文献

Coral reef degradation resulting from nutrient enrichment of coastal waters is of increasing global concern. Although effects of nutrients on coral reef organisms have been demonstrated in the laboratory, there is little direct evidence of nutrient effects on coral reef biota in situ. The ENCORE experiment investigated responses of coral reef organisms and processes to controlled additions of dissolved inorganic nitrogen (N) and/or phosphorus (P) on an offshore reef (One Tree Island) at the southern end of the Great Barrier Reef, Australia. A multi-disciplinary team assessed a variety of factors focusing on nutrient dynamics and biotic responses. A controlled and replicated experiment was conducted over two years using twelve small patch reefs ponded at low tide by a coral rim. Treatments included three control reefs (no nutrient addition) and three + N reefs (NH4Cl added), three + P reefs (KH2PO4 added), and three + N + P reefs. Nutrients were added as pulses at each low tide (ca twice per day) by remotely operated units. There were two phases of nutrient additions. During the initial, low-loading phase of the experiment nutrient pulses (mean dose = 11.5 microM NH4+; 2.3 microM PO4(-3)) rapidly declined, reaching near-background levels (mean = 0.9 microM NH4+; 0.5 microM PO4(-3)) within 2-3 h. A variety of biotic processes, assessed over a year during this initial nutrient loading phase, were not significantly affected, with the exception of coral reproduction, which was affected in all nutrient treatments. In Acropora longicyathus and A. aspera, fewer successfully developed embryos were formed, and in A. longicyathus fertilization rates and lipid levels decreased. In the second, high-loading, phase of ENCORE an increased nutrient dosage (mean dose = 36.2 microM NH4+; 5.1 microM PO4(-3)) declining to means of 11.3 microM NH4+ and 2.4 microM PO4(-3) at the end of low tide) was used for a further year, and a variety of significant biotic responses occurred. Encrusting algae incorporated virtually none of the added nutrients. Organisms containing endosymbiotic zooxanthellae (corals and giant clams) assimilated dissolved nutrients rapidly and were responsive to added nutrients. Coral mortality, not detected during the initial low-loading phase, became evident with increased nutrient dosage, particularly in Pocillopora damicornis. Nitrogen additions stunted coral growth, and phosphorus additions had a variable effect. Coral calcification rate and linear extension increased in the presence of added phosphorus but skeletal density was reduced, making corals more susceptible to breakage. Settlement of all coral larvae was reduced in nitrogen treatments, yet settlement of larvae from brooded species was enhanced in phosphorus treatments. Recruitment of stomatopods, benthic crustaceans living in coral rubble, was reduced in nitrogen and nitrogen plus phosphorus treatments. Grazing rates and reproductive effort of various fish species were not affected by the nutrient treatments. Microbial nitrogen transformations in sediments were responsive to nutrient loading with nitrogen fixation significantly increased in phosphorus treatments and denitrification increased in all treatments to which nitrogen had been added. Rates of bioerosion and grazing showed no significant effects of added nutrients. ENCORE has shown that reef organisms and processes investigated in situ were impacted by elevated nutrients. Impacts were dependent on dose level, whether nitrogen and/or phosphorus were elevated and were often species-specific. The impacts were generally sub-lethal and subtle and the treated reefs at the end of the experiment were visually similar to control reefs. Rapid nutrient uptake indicates that nutrient concentrations alone are not adequate to assess nutrient condition of reefs. Sensitive and quantifiable biological indicators need to be developed for coral reef ecosystems. The potential bioindicators identified in ENCORE should be tested in future research on coral reef/nutrient interactions. Synergistic and cumulative effects of elevated nutrients and other environmental parameters, comparative studies of intact vs. disturbed reefs, offshore vs. inshore reefs, or the ability of a nutrient-stressed reef to respond to natural disturbances require elucidation. An expanded understanding of coral reef responses to anthropogenic impacts is necessary, particularly regarding the subtle, sub-lethal effects detected in the ENCORE studies.  相似文献

Bioremediation was conducted in the field on a mature Rhizophora stylosa mangrove stand on land to be reclaimed near Fisherman’s Landing Wharf, Gladstone Australia. Gippsland crude oil was added to six large plots (>40 m2) and three plots were left untreated as controls. Bioremediation was used to treat three oiled plots and the remaining three were maintained as oiled only plots. The bioremediation strategy consisted of actively aerating the sediment and adding a slow-release fertilizer in order to promote oil biodegradation by indigenous micro-organisms. Oil addition stimulated the numbers of alkane-degrading bacteria slightly to levels of 104–105/g sediment. Bioremediation of the oiled sediment had a marked effect on the alkane-degrading population, increasing the population size by three orders of magnitude from 105 to 108 cells/g of sediment. An effect of bioremediation on the growth of aromatic-degraders was detected with numbers of aromatic-degraders increasing from 104 to 106 cells/g of sediment. Active aeration and nutrient addition significantly stimulated the growth of hydrocarbon-degraders in oiled mangrove sediment in the field.  相似文献

Erosion effect on the productivity of black soil in Northeast China   总被引:9,自引:0,他引:9
Continuous soil erosion has caused serious land degradation in the black soil area of Northeast China. The primary objective of this study was to determine the effects of accelerated erosion on soil productivity, as measured by soybean (Glycine max L. Merr.) yields. Eight erosion levels, 0, 10, 20, 30, 40, 50, 60, and 70 cm, were simulated by imitating the integrated process of natural erosion and tillage activity. Each erosion level had two sub-treatments: conventional fertilization and no fertilization. Soil erosion was found to affect survival probability and to cause remarkable reductions in the Leaf Area Index (LAI), plant height, pod number, biomass, and yield. Soybean yield was exponentially decreased with the increase of soil erosion depth. Compared to erosion depth of 0 cm, erosion levels of 10, 20, 30, 40, 50, 60, and 70 cm experienced reductions in soybean yield by 28.8%, 37.8%, 43.5%, 52.6%, 53.1%, 52.9%, and 64.1% respectively when fertilized whereas the reductions at those levels were 32.6%, 42.2%, 53.0%, 54.0%, 65.8%, 69.7%, and 72.6%, respectively, when unfertilized. At the erosion depths of 10, 20, 30, 40, 50, 60, and 70 cm, the yield reductions per 10 cm of soil eroded when fertilized were 28.8%, 18.9%, 14.5%, 13.2%, 10.6%, 8.8%, and 9.2%, averaged 14.9%, but when unfertilized they were 32.6%, 21.1%, 17.7%, 13.5%, 13.2%, 11.6%, and 10.4%, averaged 17.1%. The results also showed that chemical fertilizers could enhance the yields of eroded soil, but could not recover the yields to the pre-erosion level. Additionally, the results indicated that the primary reason for the decrease in soybean yield with increasing erosion depth was the loss of soil organic matter, soil N and P. These results may aid in selecting effective soil erosion control strategy, forecasting land degradation, establishing soil erosion tolerance, and evaluating the economic cost of soil erosion in the black soil region in Northeast China Supported by National Natural Science Foundation of China (Grant No. 40471082) and National Basic Research Program of China (Grant No. 2007CB407204)  相似文献

We studied the responses of algae, corals, and small fish to elevated inorganic fertilizer, organic matter, and their combination over a 49-day summer period in cages that simulated the coral reef in the remote Glovers reef atoll, Belize. The addition of organic matter reduced while fertilization had no effect on the numbers of herbivorous damsel and parrotfishes. All measures of algal biomass were influenced by fertilization. The combined inorganic and organic enrichment produced the highest algal biomass, which is most likely due to the combined effect of higher nutrients and lower herbivory. The cover of turf and total algae were influenced by all treatments and their interactions and most strongly and positively influenced by fertilization followed by organic matter and the combination of organic matter and inorganic fertilizer. The inorganic and combined treatments were both dominated by two turf algae, Enteromorpha prolifera and Digenia simplex, while the nonfertilized treatments were dominated by brown frondose algae Lobophora variegata, Padina sanctae, and Dictyota cervicornis. The organic matter treatment had greater cover of P. sanctae and D. cervicornis than the untreated control, which was dominated by Lobophora variegata, also the dominant algae on the nearby patch reefs. Crustose corallines grew slowly ( approximately 2.5 mm/49 days) and were not influenced by the treatments when grown on vertical surfaces but decreased on horizontal coral plates in the combined organic matter and fertilization treatment. No mortality occurred for the two coral species that were added to the cages. Porites furcata darkened in the fertilized cages while there was a mix of paling and darkening for a small amount of the coral tissue of Diploria labyrinthiformes. Inorganic fertilization stimulates small filamentous turf algae and Symbiodinium living in coral but inhibits brown frondose algae. Organic matter inhibits small herbivorous fish, L. variegata, and encrusting coralline algae when growing on horizontal surfaces.  相似文献

Both laboratory experiments and numerical modelling were conducted to study the biodegradation and transport of benzene–toluene–xylenes (BTX) in a simulated semi‐confined aquifer. The factors incorporated into the numerical model include advection, hydrodynamic dispersion, adsorption, and biodegradation. The various physico‐chemical parameters required by the numerical model were measured experimentally. In the experimental portion of the study, BTX compounds were introduced into the aquifer sand. After the contaminants had been transported through the system, BTX concentrations were measured at 12 equally spaced wells. Subsequently, microorganisms obtained from the activated sludge of a sewage treatment plant and cultured in BTX mixtures were introduced into the aquifer through the 12 sampling wells. The distribution data for BTX adsorption by the aquifer sand form a nonlinear isotherm. The degree of adsorption by the sand varies, depending on the composition of the solute. The degradation time, measured from the time since the bacteria were added to the aquifer until a specific contaminant was no longer detectable, was 35–42 h for BTX. The dissolved oxygen, after degradation by BTX compounds and bacteria, was consumed by about 40–60% in the entire simulated aquifer; thus the aerobic conditions were maintained. This study provides insights for the biodegradation and transport of BTX in aquifers by numerical modelling and laboratory experiments. Experimental and numerical comparisons indicate that the results by Monod degradation kinetics are more accurate than those by the first‐order degradation kinetics. Hydrocarbon degraders from tropical marine environments   总被引:1,自引:0,他引:1
Analysis of 20 samples of marine mud and water around Mumbai resulted in the isolation of 17 bacteria and yeasts all of which were able to degrade more than 10% of the supplied crude oil. The yeasts strains were important degraders of the aliphatic fraction of crude. All the isolated yeasts belonged to the genus Candida. Using biochemical tests these were identified as Candida parapsilosis, C. albicans, C. guilliermondii, Yarrowia lipolytica, C. tropicalis and C. intermedia. Y. lipolytica was the best degrader utilizing 78% of the aliphatic fraction of Bombay High crude oil. None of these isolates degraded the aromatic or ashphaltene fractions. All the isolates required aeration, nitrogen and phosphate supplementation for optimal degradation. Four out of the six yeasts are human pathogens.  相似文献
