To examine whether xenobiotics impair teleost reproduction by altering reproductive endocrine function, steroid hormone secretion and ovarian growth were investigated in female Atlantic croaker (Micropogonias undulatus) after chronic oral administration with sublethal doses of three classes of reproductive toxins (lead, benzo[a]pyrene and Aroclor 1254). All the sublethal treatments significantly impaired ovarian growth as assessed by the gonadosomatic index. Reduced ovarian growth was accompanied by a significant decline in circulating 17β-estradiol levels in fish exposed to lead and benzo[a]pyrene. All three toxicants significantly decreased plasma testosterone levels. However, the steroidogenic capacity of ovarian tissue in vitro was not altered by xenobiotic exposure. The data suggest that the decreased ovarian growth in croaker after pollutant exposure may be a consequence of the decline in plasma 17β-estradiol levels. Further, this decrease in circulating levels of gonadal steroids does not appear to be caused by a direct effect of the chemicals on ovarian steroid synthesis.  相似文献

Embryos of the estuarine teleost, Fundulus heteroclitus, were exposed during development to different concentrations of the water-soluble fractions (WSF) of a No. 2 fuel oil under various temperature-salinity combinations. Temperatures and salinities were, respectively: 20, 25 or 30°C and 10, 20 or 30%S. The WSF were extracted from No. 2 fuel oil daily and diluted to concentrations of 15, 20 or 25% of the original mixture. Controls were exposed to seawater without WSF at each temperature-salinity combination.The length of eleutheroembryos was markedly decreased by exposure to WSF. Temperature exerted an effect on growth at low WSF concentrations (0 and 15% WSF) but not at higher ones. The two highest WSF concentrations had a predominant effect on growth that was not further modified by physical factor influences. Conversely, as the WSF exposure increased, the yolk diameter increased, indicating a decreased utilisation of yolk material during development. The mean number of vertebrae in the eleutheroembryos decreased at high WSF concentrations and at high temperatures. Morphological abnormalities consisted predominantly of spinal deformations (scoliosis and lordosis). The data indicate that the survivors of chemical toxicity during embryo development exhibit sublethal responses which would reduce their survival potential in later life.  相似文献

The chronic effects of polycyclic aromatic hydrocarbons (PAHs) on ovary development, total hepatic lipids and plasma sex- and corticosteroid levels in female flounder (Platichthys flesus) were examined. Sexually mature feral female flounder were exposed via the diet to phenanthrene (0.5, 2.5 or 12.5 nmol/g food) or chrysene (0.4 nmol/g food) for 12 weeks, during the previtellogenic phase of the annual reproductive cycle. PAH exposure did not directly affect germ cell development since no structural and/or developmental differences were observed between control and exposed fish. On the contrary, all treatments resulted in altered plasma steroid levels. The most pronounced effect was the significant decrease in plasma 17 beta-estradiol to 19 +/- 11%, 27 +/- 7%, 63 +/- 20% and 61 +/- 12% in relation to control fish, respectively, in flounders exposed to 12.5, 2.5 or 0.5 nmol phenanthrene/g food and 0.4 nmol chrysene/g food. Impaired ovarian growth was not observed, most likely because experiments were ended before the period of vitellogenesis, even though a non-significant general decline in total hepatic lipids could be observed. Moreover, all exposed flounders, except fish fed with the highest amount of phenanthrene, showed a negative correlation between plasma 17 beta-estradiol and 17 alpha-hydroxyprogesterone levels (r = -0.46). One possible explanation is that PAH action may be mediated by a specific inhibition of steroidogenic enzymes. These findings provide evidence that selected PAHs are antiestrogenic xenobiotics with the capability to impair female teleost reproductive function.  相似文献

Pacific herring (Clupea harengus pallasi) larvae were exposed either directly to the water-soluble fraction (WSF) of crude oil or indirectly via oil-contaminated prey (OCP) (Artemia salina) for <- 28 days, to determine the relative effects of diet and water as routes of contamination. Larvae were affected rapidly by 0·9 ppm WSF, which caused high larval mortality, reduced swimming ability and rapid reductions in feeding rates. Larval length was significantly reduced by 0·7 ppm WSF in 7 days and by 0·3 ppm WSF within 14 days. The WSF exposure also caused similar reductions in larval weights. Highly contaminated prey (6 ppm prey exposure) caused significant mortality, but surviving larvae appeared robust. The OCP did not affect swimming, feeding, or growth. Exposure of larvae was not significantly extended by OCP, which rapidly depurated WSF in clean water (98% in 1 day). Therefore, OCP is probably not an important source of low molecular weight petroleum contamination to larval fish in the marine environment.  相似文献

研究了实验室条件下原油水溶性组分(WSF)暴露对黑鲷、黄鳍鲷和褐菖鲉肝微粒体EROD活性的剂量-效应,时间-效应和恢复过程。实验结果表明,在剂量诱导实验中,褐菖鲉肝EROD活性在原油WSF浓度为50μg/dm3时呈现生物统计学上的显著差异,而黑鲷和黄鳍鲷肝EROD活性在75μg/dm3时才呈现生物统计学上的显著差异;褐菖鲉肝EROD活性诱导倍数最高,但黑鲷的诱导浓度范围较广。在时间诱导实验中,在40μg/dm3原油WSF暴露下黄鳍鲷肝EROD活性在2 d时首先呈现显著差异;三种鱼肝EROD活性均在第4天达到最高,并呈现显著性变化,此后随着暴露时间的延长而逐渐下降并接近对照组水平。在恢复实验中三种鱼肝EROD活性下降并恢复到对照组水平。研究结果表明:对于石油污染物,黑鲷、黄鳍鲷和褐菖鲉肝EROD活性都可以作为污染生化效应监测指标,然而就三种鱼类比较而言,褐菖鲉最敏感,更适合于作为石油类污染及其生化效应,尤其是低剂量效应的监测生物。  相似文献

The oxygen consumption reflects the aerobic energy production of an animal. For fish eggs and larvae the normal oxygen consumption represents the optimal conditions for growth and development. Under the stress of environmental pollution gross deviation in oxygen uptake rate of the developing fish embryo may reflect metabolic disturbances and impede the development. Since the eggs and larvae of cod (Gadus morhua L.) are likely to be exposed to oil pollution from offshore installations in the North Sea we decided to investigate the effects of the water-soluble fraction (WSF) of North Sea crude oil on the oxygen consumption of cod eggs and larvae. The results showed that oxygen consumption of the larvae at the time of final yolk absorption (5–7 days post hatching at 5°C) is strongly suppressed by oil exposure at concentrations down to 50 ppb. No effect on the oxygen uptake, however, was found during the egg stage.  相似文献

The arctic amphipod Boeckosimus (= Onisimus) affinis was exposed to the water soluble fraction (WSF) of Prudhoe Bay crude oil. Animals were constantly exposed or one-time exposed for 3 or 10 days and then removed to clean water. In constant exposure experiments mortality was correlated to the strength of the solution for 6 weeks; afterwards animals in all solutions experienced similar mortality. Food search success lessened for 2 weeks, then gradually increased. Distance moved and time spent moving decreased as the concentration of the WSF increased.Animals one-time exposed to the WSF showed initial mortality rates of 15–20%, with little further mortality. Food search success was lessened relative to the strength of the WSF and duration of exposure, with general recovery within two weeks. Distance moved was decreased, but time spent moving was not. Recent studies have shown that several environmental chemicals which disrupt classical genomic steroid actions can also interfere with nongenomic steroid actions initiated at the cell surface. The effects of bisphenol-A and atrazine on the nongenomic actions of a progestin, 17,20 beta,21-trihydroxy-4-pregnen-3-one (20 beta-S), on oocyte maturation (OM) were investigated an in vitro Atlantic croaker (Micropogonias undulatus) oocyte bioassay. Treatment of croaker follicle-enclosed oocytes with either bisphenol-A or atrazine blocked OM in response to 20 beta-S in a concentration-dependent manner at 10-25 microM (2.2-5.7 ppm) and higher concentrations. These compounds were also effective competitors at concentrations of 10(-6)-10(-5)M of [3H]-20 beta-S binding to the ovarian progestin membrane receptor that mediates the OM response to 20 beta-S. The results suggest that diverse classes of environmental chemicals can disrupt nongenomic progestin actions via receptor-mediated mechanisms.  相似文献

Maximum oocyte size was used to assess seasonal ovarian development in sole. Fish age, especially the adolescent period, appeared to affect the start of vitellogenin-dependent oocyte development in the annual reproductive cycle and the subsequent oocyte growth rate. The majority of oocyte growth occurred between September and March. Several other aspects of ovarian development were also age-dependent, including the increase in ovary condition factor (ovary weight/fish length3) and the size of oocytes commencing nuclear migration. Evidence is presented that in the recruiting year class of sole abortive maturation occurs where oocytes develop yolk but spawning does not take place. The implications of this study on the estimation of female spawning stock biomass are discussed.  相似文献

Experimental exposure tanks were set up containing 'clean' marine sediment spiked with set doses of polyaromatic hydrocarbons (PAH), or polychlorinated biphenyls (PCB), or both. Reference tanks contained only 'clean' sediment. Female dab Limanda limanda L., exposed to these sediments for 7 days showed no differences in serum total protein concentration, serum lysozyme activity or body growth. Their kidney leucocytes were assayed for extracellular production of reactive oxygen species of the phagocytic respiratory burst. No significant differences in superoxide anion (O2-) production were found between fish exposed to the 'spiked' sediments and those on the 'clean' reference sediment. The production of hydrogen peroxide (H2O2) was, however, reduced in the fish held on the PAH and PAH/PCB spiked sediments. H2O2 levels for fish exposed to PCBs alone, did not differ significantly from those of control fish. Our results suggest that the decrease in H2O2 production was due to exposure to PAHs, rather than to PCBs.  相似文献

Understanding the ecological significance of oestrogenic biomarker responses, seen in field surveys of some UK estuarine fish, requires a suitable model to link biochemical and reproductive effects. The sand goby (Pomatoschistus minutus) is a common inshore fish that has been successfully bred in laboratory aquaria. Juvenile sand goby were continuously exposed to 17beta-oestradiol (E2) during gonadal maturation, and their subsequent reproductive success investigated. Exposure endpoints included hepatic vitellogenin (VTG) mRNA expression and maturation (male nuptial colouration, sperm duct gland- and gonado-somatic indices (SDGSI, GSI)). Reproductive endpoints included spawning behaviour, brood size, fertility and population-level fertile egg production. Measured E2 exposure concentrations were <5 (controls), 17, 71 and 530 ngl(-1). Male maturation decreased with increasing E2 concentration. VTG mRNA expression increased with E2 concentration, but not with duration of exposure. Reproductive endpoints showed progressive impairment with increasing E2 concentration. For most endpoints, the lowest observed effect concentration (LOEC) was 71 ngl(-1), and EC50s were in the range 87-165 ngl(-1). Pre-breeding male colouration, VTG expression and SDGSI were good predictors of subsequent population-level fertile egg production, but GSI was not. The sensitivity of oestrogenic markers in sand goby are comparable to those used in other species, and feral fish populations exhibiting such oestrogenic responses at molecular and organ levels are likely to have reduced fertile egg production.  相似文献

为了解和探讨3~5环PAHs对海水鱼类胚胎发育的毒性及作用方式,比较研究了菲(phenanthrene,Phe)、芘(pyrene,Py)、苯并(a)芘(benzo(a)pyrene,BaP)单一暴露和三者各自与α-萘黄酮(α-naphthoflavone,ANF)联合暴露对海水青鳉(marine medaka, Oryzias melastigma)胚胎发育的毒性效应。胚胎体内EROD活性、发育畸形、孵化率和心律等毒性指标被测定,结果显示:Phe,Py和BaP对海水青鳉胚胎体内EROD活性的诱导能力大小为BaP>Py>Phe,各化合物对EROD诱导与发育畸形之间的关系较为复杂,除Phe所引起的EROD诱导与畸形指数之间呈显著相关(r=0.95,p=0.015)外,Py和BaP均无相关性;在100 μg/dm3 ANF影响下,CYP1A活性诱导被抑制,但胚胎发育的畸形指数被显著提高,ANF分别与Phe,Py和BaP的联合暴露对胚胎发育呈潜在的协同作用。本文研究初步表明,3~5环PAHs化合物对海水青鳉胚胎发育的毒性作用方式可能不同;CYP1A活性抑制在PAHs混合物对海水青鳉胚胎发育的毒性作用过程中未起到缓解毒性的作用,CYP1A抑制剂与PAH型CYP1A诱导剂的混合物对鱼类胚胎发育具有潜在的协同毒性作用,现有的PAHs混合物毒性风险评价方法可能低估了实际环境中PAHs的风险;海水青鳉早期生活阶段的心脏发育对PAHs混合物暴露较为敏感,可推荐其作为生物标志物指示PAHs或溢油污染。  相似文献

以原油水溶性成分(water soluble fraction of crude oil,WSF)为污染物,采用暴露实验法,研究了栉孔扇贝(Chlamys farreri)鳃和消化腺组织中超氧化物歧化酶(Superoxide Dismutase,SOD)和过氧化氢酶(Catalase,CAT)活性的变化.结果表明,WSF污染下,鳃和消化腺组织SOD和CAT酶活性随暴露时间增加一般表现为降低-升高-降低的趋势,酶活性达到最高的时间随浓度不同而变化.第1天时消化腺SOD在0.08 mg/L浓度下被诱导,而后随时间增加表现为抑制效应;浓度为0.21和0.88 mg/L时消化腺SOD酶活性被抑制,随暴露时间延长而活性增加.暴露时间为4d时,石油烃浓度在0.08和0.88 mg/L时鳃组织SOD酶活性均被抑制,而浓度为0.21 mg/L时被诱导.消化腺和鳃组织SOD可以作为扇贝被污染胁迫的指标.  相似文献

Recently evidence has been obtained for reproductive impairment in estuarine populations of Atlantic croaker exposed to seasonal hypoxia. However, it is not known whether a similar disruption of reproductive function occurs in croaker inhabiting a much larger hypoxic area, the extensive dead zone in coastal regions of the northern Gulf of Mexico extending from the Mississippi Delta to East Texas. Gonadal development in male Atlantic croaker collected in September 2008 at six sites in the dead zone was compared to that in male fish sampled from three reference sites east of the Mississippi Delta which do not experience persistent hypoxia. Croaker testes collected from the dead zone were at an earlier stage of spermatogenesis than those from the reference sites. Histological examination of the testes collected from the dead zone showed that their tubules had small lumens that contained very little sperm compared to the lumens of the reference fish. Overall, sperm production was 26.2% that of the control fish at the reference sites. This decrease in spermatogenesis at the dead zone sites was accompanied by an approximately 50% decrease in testicular growth compared to that in the reference fish. The results suggest that reproductive impairment can occur over regional scales in marine fish populations exposed to extensive seasonal hypoxia in dead zones with potential long-term impacts on population abundance.  相似文献

The asteroid Patiria miniata was used to develop a static bioassay for various crude oil water-soluble fractions (WSF). The criterion used is the size of 48-h exposed embryos. The reasons for choosing this species and the growth criterion are discussed. Experiments show that the body length of 48-h exposed embryos is inversely proportional to hydrocarbon concentration of the WSF and the time of exposure. It is also shown that the toxicity of the WSF decreases with the age of the preparation and that the first 12 h following fertilisation are most sensitive to the effects of WSF.  相似文献

The role of vitellogenin in the transport of organochlorines was investigated in Atlantic croaker (Micropogonias undulatus) by exposing them through the diet to o,p′-DDT at a concentration of 10.8 μg/100 g fish/day for 14 days or Aroclor 1254 (PCB) at a concentration of 0.5 mg/100 g fish/day for 30 days during gonadal recrudescence. Tissue samples were taken from the fish at various times after initial exposure, and o,p′-DDT and PCBs were extracted in acetonitrile and analyzed by gas chromatography. Analysis of the ovarian tissue collected 2 and 3 weeks from the start of exposure revealed that the o,p′-DDT concentration increases as the gonadosomatic index (GSI) increases (r2 = 0.63), with accumulation ranging from less than 1% to as much as 8% of the total dosage. Interestingly, o,p′-DDT did not accumulate in the testes during the same exposure period. Accumulation of PCBs was found to be 40 times higher in the ovaries than in the testes. Gel filtration of plasma from exposed females showed that o,p′-DDT elutes in the low density lipoprotein and vitellogenin fractions. Control plasma incubated with o,p′-DDT at 4 °C for 16 h followed by chromatography on Sepharose 6B gave similar results with an o,p′-DDT concentration of 0.6μg/mg protein in the vitellogenin fraction. Furthermore, both o,p′-DDT and PCBs were found to bind to purified croaker vitellogenin. These results suggest that lipoproteins, including vitellogenin, are involved in the transport and accumulation of organochlorines in the ovaries of exposed fish.  相似文献

We exposed the bat starfish, Patiria miniata, to the water-soluble fraction (WSF) of oil from a natural seep near Santa Barbara, California, during the first 48 hours of embryological growth. There were no consistent differences among populations whose parents were from areas with differing levels of hydrocarbon exposure. In a second experiment, the order of decreasing toxicity of crude oils to the embryos was: Monterey Zone > Rincon Zone > Prudhoe Bay > Isla Vista Seep. The results are discussed in relation to the effects of natural seeps and oil characteristics.  相似文献

To investigate the effects of a polychlorinated biphenyl (PCB) mixture (Aroclor 1254) on reproductive neuroendocrine function, male Atlantic croaker (Micropogonias undulatus) were exposed to the PCB mixture in the diet (0.1 mg/100. g body wt/day) for 30 days during gonadal recrudescence. Concentrations of the monoamines (5-hydroxytryptamine (5-HT), dopamine (DA) and norepinephrine (NE)) and their major metabolites (5-hydroxyindolacetic acid, dihydroxyphenylacetic acid and 3-methoxytyramine) were measured in the preoptic-anterior hypothalamus (POAH) and medial and posterior hypothalamus (MPH) using HPLC with electrochemical detection. There was a significant decline in 5-HT and DA concentrations and an increase in their metabolite to parent amine ratios in both the POAH and MPH of Aroclor 1254-exposed fish. In addition, Aroclor 1254 exposure resulted in the loss of the in vitro gonadotropin (GtH) response to stimulation by a luteinizing hormone-releasing hormone analog (LHRHa). The reduced availability of 5-HT in the nerve terminals may be responsible for the loss of pituitary GtH response to stimulation by LHRHa. These results support the hypothesis that Aroclor 1254-induced alterations in pituitary gonadotropin release may be mediated, at least partially, by altered hypothalamic serotonergic activity.  相似文献

Naphthalene (NAP) and phenanthrene (PHE) effects on Salminus brasiliensis, a carnivorous freshwater fish, were investigated using behavioral tests. Larval stages of S. brasiliensis were exposed to water concentrations of 0, 0.04 mg/l, 0.20mg/l and 0.50mg/l for naphthalene and 0, 0.01 mg/l, 0.05 mg/l and 0.1mg/l for phenanthrene during two developmental phases. The prey fish Prochilodus lineatus were not exposed. Visual acuity was measured at the end of phase 2 in individual S. brasiliensis, which were also tested at the end of each phase for number of attacks on prey, number of prey captured, prey capture efficiency, and distance swam. Vision was impaired by PHE exposure, as acuity angles increased in exposed fish. At Stage I 2.3+/-1.2 prey were captured with 46% efficiency in controls compared to 0.4+/-0.3 prey captured with 13.4% efficiency in fish exposed to 0.05 mg PHEl(-1), the LOEL for both endpoints. At Stage II 4.0+/-1.1 preys were captured in controls compared to 2.5+/-0.9 preys captured in fish exposed to 0.01 mg PHEl(-1), the LOEL. Stage II control fish captured prey with 70% efficiency compared to 30% efficiency at Stage II fish exposed to 0.05 mg PHEl(-1), the LOEL. Distance swam was not affected by either NAP or PHE exposure. The exposure of larval stages of S. brasiliensis to realistic water concentrations of PHE impairs foraging skills and could affect recruitment of the species.  相似文献
