We monitored seasonal changes of the abundance and composition of microorganisms in the fish-farm sediment in Kusuura Bay, Amakusa, Japan, using the quinone profiling technique, during bioremediation by introducing cultured colonies of polychaete, Capitella sp. I. In November 2004, approximately 9.2 million cultured worms were transferred to the fish-farm sediment, which increased rapidly, and reached 458.5 gWW/m(2) (528,000 indiv./m(2)) in March 2005. During this fast-increasing period of Capitella, the microbial quinone content of the surface sediment (0-2 cm) also increased markedly, and reached 237 micromol/m(2) in January 2005, although the water temperature decreased to the lowest levels in the year. Particularly, the mole fraction of ubiquinone-10 in total quinones in the sediment, indicating the presence of alpha subclass of Proteobacteria, increased by 9.3%. These facts suggest that the bacterial growth was enhanced markedly by the biological activities of worms in the sediment, and the bacteria played an important role in the decomposition of the organic matter in the sediment.  相似文献

We assessed the occurrence of pollution indicators and antibiotic resistant bacterial isolates from water and sediment samples of three different eco-regions of the Chennai coast between March - May of 2010. Total of 960 bacterial strains belonging to four genera were isolated which show the highest frequencies of resistance to vancomycin (53.6%) and penicillin (52.6%) (except Enterococcus sp., which is highly resistant to erythromycin) and lowest frequencies of resistance to chloramphenicol (3.43%), ciprofloxacin (3.95%), gentamicin (4.68%), and tetracycline (6.97%). The E. coli, Vibrio sp., Salmonella sp. and Enterococcus sp. show high frequency of resistance to 2-5 antibacterials of 60.4%, 45.83%, 69.16% and 46.6%, respectively. High pollution indices (PI - 6.66-14.06) and antibiotic resistance indices (ARI - 0.29-0.343) indicate that the coastal environment is highly exposed to antibiotic sources that suggesting to avoid direct contact.  相似文献

In this study, we determined pH, phosphorus content and the number of P-dis- solving/decomposing bacteria in relation to the depth in the sediment of Guanting reservoir in Beijing. The pH value was slightly increased from 8.0 in the surface to 8.5 in the bottle (69 cm in depth) of sediment. The highest P content, 1269 mg/kg, was detected in the 35 cm layer, followed by the 5 cm (993 mg/kg) and 69 cm (580 mg/kg) layers. The number of inorganic P-dissolving bacteria varied from 6(102 to 8(104 and the organic P-discomposing bacteria were from 1.9(103 to 6.3(104 per gram sediment in different depths, which were counted under 28℃ and 20℃ ( pH7.5 and 8.5. The number of P-dissolving/discomposing bacteria was directly correlated to the P content in each layer of sediments. The analysis of P-dissolving/decomposing ability of bacteria showed that some of these bacteria were also capable of accumulating P intracellularly. The intracellular P-accumulation was more efficient at lower temperature; in contrast, the activity of P-dissolving/decomposing was stronger at higher temperature. So the content of dissolved P in water body, or quality of water, could be affected by the change of temperature via the regulation of bacterial activity. On the basis of 16S rDNA sequencing, the 13 efficient P-dis- solving/decomposing bacteria were identified as Bacillus spp., Bacterium sp., Microbacterium sp., Paenibacillus sp. and Pseudomonas sp.  相似文献

附生细菌在微囊藻水华暴发过程中可能发挥了重要作用,但是,缺少关于微囊藻附生细菌特性的研究报道.本文从4种群体微囊藻(铜绿微囊藻、惠氏微囊藻、水华微囊藻和坚实微囊藻)中分离得到了18种附生细菌,分别隶属于六大类群:α-变形菌纲、β-变形菌纲、γ-变形菌纲、放线菌纲、纤维粘网菌纲和黄杆菌纲.BIOLOG板分析显示除了α-变形菌纲中的2种细菌以及放线菌纲中的1种细菌以外,其余细菌都能够利用10种以上的碳源.18种附生细菌中有12种具有趋化性,12种具有疏水性,14种具有自聚能力.除了黄杆菌纲菌株都具有亲水性外,其余5大类群菌株都含有疏水性种类.六大类群中都含有具有趋化性和自聚能力的种类.每种微囊藻中的大多数附生细菌种类都能利用10种以上的碳源,都具有疏水性.每种微囊藻中至少有一组细菌表现出共聚能力.除了水华微囊藻外,其余3种微囊藻中附生细菌的大多数种类都具有趋化性.除了惠氏微囊藻外,其余3种微囊藻的附生细菌中大多数种类都具有自聚能力.结果显示细菌旺盛的代谢潜力、趋化性、疏水性、自聚能力和共聚能力可能有利于附生细菌定殖于微囊藻群体.  相似文献

Effects of spilled oil on microbial communities in tidal flats were examined by use of a simulator for a tidal flat ecosystem. The simulator is composed of a wave generator, a tide control device, and a tidal flat. Sediment for the tidal flat was obtained at a natural tidal flat in Hiroshima Bay, Japan. After stabilizing the benthic organisms, fuel oil C was added to the surface of the flat at 1 lm(-2). Although the total number of micro-organisms remained at 1.5-3.5 x 10(9) cells g(-1) dry sediment irrespective of the addition of oil, bacterial communities which were analyzed based on the 16S rDNA showed clear changes after the addition of fuel oil C and after a subsequent recovery period. Bacterial colonies were randomly isolated from the oil-supplemented sediment during the experiments, and the isolates were examined for susceptibility to hydrocarbons in order to screen the oil-susceptible bacteria. The proportion of oil-susceptible bacteria in the isolates decreased with the addition of the oil. Oil-susceptible bacteria showed an inability to assimilate petroleum compounds as well as an inhibition of growth. The possibility of using oil-susceptible bacteria as an indicator of bioremediation in tidal flats was discussed.  相似文献

Biodegradation of polycyclic aromatic hydrocarbons (PAHs) in contaminated sediment is an attractive remediation technique and its success depends on biodegradation kinetics, and the optimal condition for the PAH-degrading isolates; however, information on this aspect is still scarce. The effects of multi-factors on biodegradation of phenanthrene, a 3-ring model PAH, in contaminated sediment slurry by Sphingomonas sp. a bacterial strain isolated from surface mangrove sediment, were investigated using the orthogonal experimental design (form L(16)(4(5))). The most significant factors were salinity and inoculum size, while the effects of phenanthrene concentrations, nutrient addition and temperatures were insignificant. The optimal biodegradation condition in contaminated mangrove sediment slurry was 30 degrees C, 15 ppt salinity, a carbon/nitrogen ratio of 100:1 (the background ratio in sediment) and an inoculum size of 10(6) most probable number g(-1) sediment. The phenanthrene biodegradation could be best described by the first order rate model, C=C(0)e(-kt), where k (the rate constant) is equaled to 0.1185, under the optimal condition. The kinetic model was verified and its validity in predicting biodegradation by Sphingomonas sp. at various phenanthrene concentrations was proved by experimental data.  相似文献

The spatial distribution of polychaeta along pollution gradients often reflects different degrees of disturbance. In order to evaluate polychaeta fauna of an organically polluted tropical bay, 20 sandy beaches distributed in five areas were sampled. The relationship between community structure, slope, beach index, exposure, sediment and water quality parameters were analysed. Multivariate analysis of variance (PERMANOVA) showed differences among areas and beaches. Scolelepis chilensis dominated at mouth of bay beaches whereas Streblospio gynobranchiata and Capitella capitata complex, at inner beaches. Highest polychaete density was recorded at areas 3 and 5 with the dominance of Saccocirrus sp. and the organic indicator species C. capitata complex and Polydora sp. The most important factors obtained from canonical analysis were sorting, slope, mud and organic matter percentage. Marine biotic index (AMBI) showed that areas 3 and 5 were highly affected by anthropogenic factors, given that a poor polychaeta fauna, dominated by opportunistic species, were found. Polychaete assemblages were affected by eutrophication along an estuarine gradient as well as by morphodynamic condition of the beaches.  相似文献

Passive multilevel samplers (MLS) containing a solid matrix for microbial colonization were used as in situ microcosms in conjunction with a push-pull biostimulation experiment designed to promote biological U(VI) and Tc(VII) reduction. MLS were deployed at 24 elevations in the injection well and two downgradient wells to investigate the spatial variability in microbial community composition and growth prior to and following biostimulation. The microbial community was characterized by real-time quantitative polymerase chain reaction (Q-PCR) quantification of bacteria, NO(3)(-)-reducing bacteria (nirS and nirK), delta-proteobacteria, Geobacter sp., and methanogens (mcrA). Pretest cell densities were low overall but varied substantially with significantly greater bacterial populations detected at circumneutral pH (t-test, alpha= 0.05), suggesting carbon substrate and low pH limitations of microbial activity. Although pretest cell densities were low, denitrifying bacteria were dominant members of the microbial community. Biostimulation with an ethanol-amended ground water resulted in concurrent NO(3)(-) and Tc(VII) reduction, followed by U(VI) reduction. Q-PCR analysis of MLS revealed significant (1 to 2 orders of magnitude, Mann-Whitney U-test, alpha= 0.05) increases in cell densities of bacteria, denitrifiers, delta-proteobacteria, Geobacter sp., and methanogens in response to biostimulation. Traditionally, characterization of sediment samples has been used to investigate the microbial community response to biostimulation; however, collection of sediment samples is expensive and not conducive to deep aquifers or temporal studies. The results presented demonstrate that push-pull tests with passive MLS provide an inexpensive approach to determine the effect of biostimulation on contaminant concentrations, geochemical conditions, and the microbial community composition and function.  相似文献

太湖水体中不同形态磷含量与磷转化细菌的关系研究结果显示:太湖水体中总磷和活性磷的含量分别为0.113mg,L和O.O11mg/L;无机磷和有机磷分解菌在底泥中达6.73x103cells/g,而在水体中仅为71cells/m1,且存在明显的时间和空间差异;根据菌落形态特征,分离筛选了3株有代表性的无机磷转化菌和7株占优势的有机磷分解菌.3tt无机磷转化菌经鉴定分别与巨大芽孢杆菌(Bacillus megaterium)、假单孢菌(Pseudomonas sp.)和类芽孢杆菌(Paenibacillus sp.)比较接近.而7株有机磷分解菌则分别与为苏云金芽孢杆菌(Bacillus thurigiensts)、短小芽孢杆菌(Bacillus pumilus)、芽孢杆菌(Bacillus SP.)、无芽孢杆菌(Bacterium sp.)、氧化微杆菌(Microbacterium oxydans)腊状芽孢杆菌(Bacillus cereus)、简单芽孢杆菌(Bacillus simples)接近;太湖水体中磷分解细菌主要归属于芽孢杆菌属(Bacillus)和假单孢菌属(Pseudomonas),对细菌降解性能进行研究的结果显示:无机磷分解细菌对太湖水体活性磷的贡献显著大于有机磷分解细菌的贡献率.  相似文献

Understanding the mechanisms of bacterial transport in aquifers is important in developing bioremediation strategies. Collision efficiency (alpha) is one important parameter used in modeling bacterial transport. This study was undertaken to measure change in alpha with distance by performing a bacterial transport experiment in Oyster, Virginia. Following injection of a bacterium, Comamonas sp., into a well, water samples were collected at various distances along the flowpath and injected into columns packed with homogenized South Oyster focus area sediment. Zeta potentials of the bacteria in the samples were measured. Values of alpha were determined at various locations in the field in two ways: based on field breakthrough concentrations at the sampling points and based on column breakthrough concentrations. The alpha values estimated from field breakthrough decreased with distance, whereas those estimated from column breakthrough increased with distance. Bacterial cell surface charge became progressively more negative with distance in the field. We hypothesize that the apparent contradiction between field and column alpha values was caused by differences in the flow of the two systems. Flow in the columns was forced to occur through fine-grained zones of iron and aluminum hydroxide coatings that selectively removed the most negatively charged bacteria. In contrast, in the field, the injected cells did not come into contact with the positively charged coatings because the bulk solution bypassed them due to heterogeneous hydraulic properties. These results suggest that laboratory-based models may underestimate bacterial transport distance in the field. A more realistic approach may be necessary to capture the degree of heterogeneity.  相似文献

A two-year study was conducted in the vicinity of a harbour in sub-tropical Hong Kong, to examine the progress of improvement in sediment quality and recovery of macrobenthic community upon cessation of organic pollution caused by sewage discharge. Median sediment particle diameter (Mdphi) and levels of total organic carbon (TOC), total Kjeldahl nitrogen (TKN), ammonia-nitrogen (NH(3)-N) and total phosphorus (TP), as well as macrobenthic species composition and abundance were determined bi-monthly at three inside-harbour and two outside-harbour locations. At the two inside-harbour locations, significantly higher levels of TOC, TKN, NH(3)-N and TP in sediments were observed than the outside-harbour locations. However, no significant temporal change of nutrient levels was detected at all sampling locations during the two-year study, except a significant decrease in TKN and NH(3)-N levels at one outside-harbour location and a decline in NH(3)-N content at another outside-harbour location. Spatially, the highest in mean total species number, individual number, species diversity H' and lowest evenness J was found at one outside-harbour location, whereas the other four locations had relatively similar values. H' was negatively correlated with TOC, TKN, NH(3)-N and TP content in sediments while J was positively correlated with MDphi. Across the study period, the patterns of macrobenthic community were significantly different among all samplings and that the spatial difference of benthic community was best correlated with MDphi, TOC, TKN and water depth. A weak sign of recovery in macrobenthic community structure was detected at inside-harbour locations, with replacement of opportunistic by ubiquitous species over the two-year study. The slow recovery of macrobenthic community was related to sediment characteristics. Results of a larval settlement bioassay using the polychaete Capitella sp. I also demonstrated that the inside-harbour sediments were still unfavourable for colonization and larval settlement of species sensitive to pollution. The slow biodegradation of the organic pollutants and continuous presence of heavy metals in the sediment may hinder settlement and colonization of benthic animals. However, increases of H' and J were observed in a longer time scale when comparing the present data with those obtained four years ago. This suggested that a detectable recovery of benthic community in the harbour may take at least three years and a complete recovery may even take longer duration.  相似文献

In this study, DNAs were extracted from sediment samples at depths of 5, 35, and 69 cm from eutrophic Guanting reservoir, China. 16S rDNAs were amplified by PCR and clone libraries were constructed. The depth-related distribution of bacterial community in the sediment was characterized by using amplified 16S rDNA restriction analysis (ARDRA) and sequencing of the dominant clones. The results indicated that species diversity in the sediment of Guanting reservoir was rather high with the Shannon-Wiener index about 5.8. Bacterial richness varied in different depths: the highest in the sample of 35 cm in depth; followed by the sample of 5 cm in depth; and the lowest bacterial richness in the sample of 69 cm. Dominant species from the three samples were different although there were some common clones. PhyIogenetic analysis showed that all of the dominant clones in the three layers were uncultured bacteria and distantly related to the previously reported species in beta or gamma subclass of proteobacteria, including bacterial groups that have the ability to degrade aromatic hydrocarbons, n-all:anes, chlorinated organic compounds, or to accumulate polyphosphate, etc. Changes of deptt-related bacterial community in the Guanting reservoir sediment might reflect the pollution history and the water quality of the reservoir. In addition, the cloned sequences from the Guanting reservoir sediment were all different from the presently reported ones, indicating that there were some particular bacteria in that environment.  相似文献

The polychaetes of the tidal flats near the town of Iqaluit, Baffin Island were analysed along gradients of environmental disturbance resulting from human activity. Sources of environmental disturbance include a sewage lagoon, garbage sites; and an area of the tidal flat that is cleared by bulldozer. Sampling of the tidal flats included 300 biological sediment cores taken from 75 sites along seven transects. Environmental disturbance has resulted in four zones of polychaete communities with increasing distance. The heavily disturbed zone is closest to the disturbances and is devoid of polychaetes. The disturbed zone follows and is characterized by low diversity the result of increased densities of a few opportunistic species such as, Capitella 'capitata' sp. The moderately disturbed zone is characterized by increased species diversity due to organic enrichment from the disturbances. The undisturbed zone, located the furthest from the sources of disturbance, is characterized by moderate levels of diversity compared to the other three zones.  相似文献

Heterotrophic and hydrocarbonoclastic bacteria were enumerated at the water surface and in sediment at 14 stations located in the Ixtoc I area. The distribution of heterotrophic bacteria in water and in sediment seems to be influenced by the river waters and the characteristics of sediments. The concentrations of hydrocarbonoclastic bacteria in sediments are low, and their relative percentage in regard to the heterotrophic bacteria, which are in general less than 1%, lead us to believe that the area studied was not affected by the Ixtoc I blowout.  相似文献

水体和沉积物是湖泊生态系统中迥异但又紧密相连的两类生境,栖息在这两类生境中的细菌在维持生态系统平衡和驱动元素循环中起着关键性作用。为了探究湖泊水体和沉积物细菌群落的分布格局,本文对太湖四个湖区水体和沉积物中细菌群落进行调查,基于高通量测序技术和统计分析手段,分析这两类生境中的细菌群落组成和多样性水平、分布特征及其驱动因素。结果表明:放线菌门(Actinobacteria)、变形菌门(Proteobacteria)和蓝细菌门(Cyanobacteria)是水体细菌群落中最主要的细菌门,而沉积物中Proteobacteria占据优势地位。在两类生境中,太湖西部区域细菌群落丰富度和独特性(LCBD)相对较高,各区域群落结构表现出显著性差异。对水体而言,电导率、pH值、PC1(重金属组成)和沉积物孔隙度是驱动细菌丰富度的重要因子,PC1、水温及pH值是影响细菌群落LCBD的重要因子,而细菌网络复杂性随pH值的增加而增加,且在高pH环境中占主导地位;对沉积物而言,其丰富度和LCBD的重要影响因子均是沉积物中总磷和锂,细菌网络复杂性随金属元素施加的环境压力增大而降低,但随总磷、磷酸盐和铵态氮浓度的...  相似文献

为探讨水丝蚓(Tubificid worms)扰动对磷在湖泊沉积物-水界面间迁移的影响,选取太湖梅梁湾与大浦口两富营养化湖区为研究对象,通过室内培养实验,利用Rhizon间隙水采样器等技术,研究了水丝蚓扰动对太湖沉积物-水界面理化性质及溶解活性磷(SRP)在界面通量的影响.结果表明水丝蚓扰动能够增大表层沉积物含水率、氧化还原电位,减小间隙水中Fe2+浓度.水丝蚓没有显著改变梅梁湾间隙水中SRP浓度,同时促进了梅梁湾沉积物中SRP向上覆水的释放;但水丝蚓显著减小了大浦口间隙水中SRP浓度,并抑制了大浦口沉积物中SRP向上覆水的释放.水丝蚓扰动对磷在沉积物-水界面间迁移的不同影响可能是由沉积物中Fe2+含量差异较大造成的.  相似文献

A bacterial copper-resistant strain was isolated from a hatchery-conditioned adult of the scallop Argopecten purpuratus and was identified as Vibrio sp. according to its physiological characteristics. The lowest concentration of Cu2+ required for its complete inhibition in VNSS medium was 50 microg ml(-1). The Vibrio strain was found to accumulate copper, exhibiting cellular and loosely bound copper levels of 201.14 and 493.21 microg g(-1) dry weight, respectively, after 24 h of incubation in VNSS medium supplemented with 15 microg ml(-1) of Cu2+, with cellular concentration factors of 10.17 and 14.35 after 12 and 24 h of exposure. When a scallop larvae culture was exposed to a concentration of 6.46 x 10(2) CFU ml(-1) of Cu-enriched Vibrio, they accumulated 20.42 +/- 1.12 and 30.96 +/- 1.85 microg Cu2+ g(-1) dry weight after 12 and 24 h, respectively. This study suggests that bacterial copper accumulation could be very active in marine environments increasing the occurrence of copper transfer to marine food chains.  相似文献

To achieve a better contaminant removal efficiency in a low-temperature and high-salt environment, two novel strains of cold- and salt-tolerant ammonia-oxidizing bacteria (AOB), i.e., Ochrobactrum sp. (HXN-1) and Aquamicrobium sp. (HXN-2), were isolated from the surface sediment of Liaohe Estuarine Wetland (LEW), China. The optimization of initial ammonia nitrogen concentration, pH, carbon–nitrogen ratio, and petroleum hydrocarbons (PHCs) to improve the ammonia-oxidation capacity of the two bacterial strains was studied. Both bacterial strains showed a high ammonia nitrogen removal rate of over 80% under a high salinity of 10‰. Even at a temperature as low as 15 °C, HXN-1 and HXN-2 could achieve an ammonia nitrogen removal rate of 53% and 62%, respectively. The cold- and salt-tolerant AOB in this study demonstrated a high potential for ammonia nitrogen removal from LEW.  相似文献
