A 1-year cycle of observations was performed in four Sicilian transitional water systems (Oliveri-Tindari, Cape Peloro, Vendicari and Marsala) to characterise their ecological status. A panel of variables among which trophic and microbial (enzyme activities, abundance of hetetrophic bacteria and of bacterial pollution indicators) parameters, were selected. Particulate organic carbon (POC) and nitrogen (PON) and chlorophyll-a (Chl-a) contents defined the trophic state, while microbial hydrolysis rates and abundance gave insights on microbial community efficiency in organic matter transformation and on allochthonous inputs. To classify the trophic state of examined waters, the synthetic trophic state index (TRIX) was calculated.Microbial hydrolysis rates correlated positively with POC and Chl-a, which increased along the eutrophication gradient. The significant relationships among TRIX, trophic and microbial parameters suggested the use of leucine aminopeptidase, alkaline phosphatase and POC as suitable parameters to implement the Water Framework Directive when assessing the ecological status of transitional water systems.  相似文献

The bacteriological indices of water quality, including coliforms, sulfite-reducing clostridia, total microbial count, coliphages, and fecal streptococci were analyzed. A model describing fluctuations in microorganism population in the aquatic environment is proposed. A stochastic differential equation was obtained and used to derive a lognormal distribution of organism population. The model was applied to describe time series of bacteriological indices of the Moskva River Water Source. Satisfactory agreement was obtained between theoretical distributions and empirical data in a wide range of index values embracing one to three orders of variations in microorganism population. A forecast procedure was developed and applied to calculate the exceedance probabilities of different levels of bacterial pollution of the water source. Time periods with higher bacterial pollution were identified.  相似文献

Sponges can filter large amounts of water, which exerts an important grazing impact on free bacteria, an important component of the diet of sponges. We examined the accumulation of bacteria in the Demospongiae (Hymeniacidon perlevis). Analyses were performed on homogenates from unstarved and starved sponges in seawater from their sampling site (the Ionian Sea). Culturable heterotrophic bacteria (22 °C), total culturable bacteria (37 °C) and vibrios densities were measured on marine agar 2216, plate count agar and TCBS agar, respectively. Total and fecal coliforms, as well as fecal streptococci, were determined by the most probable number method (MPN). H. perlevis was able to accumulate all of the six microbiological groups. Bacterial groups differed in their resistance to digestion by H. perlevis. Our data suggest that H. perlevis may accumulate, remediate and metabolize bacteria and that they may be employed as a useful bioindicator and bioremediator.  相似文献

The genus Vibrio, belonging to Gammaproteobacteria of the phylum Proteobacteria, is a genetically and ecologically diverse group of heterotrophic bacteria, that are ubiquitous in marine environments, especially in coastal areas. In particular, vibrios dominate, i.e. up to 10% of the readily culturable marine bacteria in these habitats. The distribution of Vibrio spp. is shaped by various environmental parameters, notably temperature, salinity and dissolved organic carbon. Vibriospp. may utilize a wide range of organic carbon compounds, including chitin (this may be metabolized by most Vibrio spp.), alginic acid and agar. Many Vibrio spp. have very short replication times (as short as ~10 min), which could facilitate them developing into high biomass content albeit for relatively short durations. Although Vibriospp. usually comprise a minor portion (typically ~1% of the total bacterioplankton in coastal waters) of the total microbial population, they have been shown to proliferate explosively in response to various nutrient pulses, e.g., organic nutrients from algae blooms and iron (Fe+) from Saharan dust. Thus, Vibrio spp. may exert large impacts on marine organic carbon cycling especially in marginal seas. Genomics and related areas of investigation will reveal more about the molecular components and mechanisms involved in Vibrio-mediated biotransformation and remineralization processes.  相似文献

The deltaic estuarine system of the Matang Mangrove Forest Reserve of Malaysia is a site where several human settlements and brackish water aquaculture have been established. Here, we evaluated the level of fecal indicator bacteria (FIB) and the presence of potentially pathogenic bacteria in the surface water and sediments. Higher levels of FIB were detected at downstream sampling sites from the fishing village, indicating it as a possible source of anthropogenic pollution to the estuary. Enterococci levels in the estuarine sediments were higher than in the surface water, while total coliforms and E. coli in the estuarine sediments were not detected in all samples. Also, various types of potentially pathogenic bacteria, including Klebsiella pneumoniae, Serratia marcescens and Enterobacter cloacae were isolated. The results indicate that the Matang estuarine system is contaminated with various types of potential human bacterial pathogens which might pose a health risk to the public.  相似文献

Human fecal contamination of coral reefs is a major cause of concern. Conventional methods used to monitor microbial water quality cannot be used to discriminate between different fecal pollution sources. Fecal coliforms, enterococci, and human-specific Bacteroides (HF183, HF134), general Bacteroides-Prevotella (GB32), and Clostridium coccoides group (CP) 16S rDNA PCR assays were used to test for the presence of non-point source fecal contamination across the southwestern Puerto Rico shelf. Inshore waters were highly turbid, consistently receiving fecal pollution from variable sources, and showing the highest frequency of positive molecular marker signals. Signals were also detected at offshore waters in compliance with existing microbiological quality regulations. Phylogenetic analysis showed that most isolates were of human fecal origin. The geographic extent of non-point source fecal pollution was large and impacted extensive coral reef systems. This could have deleterious long-term impacts on public health, local fisheries and in tourism potential if not adequately addressed.  相似文献

Limited by sampling frequency and detection methods, current recreational water monitoring programs often fail to protect public health, especially after heavy rain or flooding, when microbial water quality can change rapidly. In order to assess the variations in the microbial indicators and to develop a scientific warning system, we conducted an intensive sampling project at the No. 1 Bathing Beach. The results show that, during dry weather, the detection rate of Enterococcus was significantly lower than that of Faecal coliform bacteria. On these days, water quality was mainly impacted by pollutants brought in by swimmers than by stormwater outfall. During wet weather, rainfall and microbial bacterial concentrations showed a positive correlation. Trends in the two microbial bacteria were approximately the same. With increasing distance from the shore, the detection rate of microbial bacteria gradually decreased. Microbial bacteria concentrations increased markedly during high tide and under a south wind.  相似文献

The goals of this study were to assess watersheds impacted by high densities of OSDS (onsite sewage disposal systems) for evidence of fecal contamination and evaluate the occurrence of human pathogens in coastal waters off west Florida. Eleven stations (representing six watersheds) were intensively sampled for microbial indicators of fecal pollution (fecal coliform bacteria, enterococci, Clostridium perfringens and coliphage) and the human enteric pathogens, Cryptosporidium, Giardia, and enteroviruses during the summer rainy season (May-September 1996). Levels of all indicators ranged between < 5 and > 4000 CFU/100 ml. Cryptosporidium and Giardia were detected infrequently (6.8% and 2.3% of samples tested positive, respectively). Conversely, infectious enteroviruses were detected at low levels in 5 of the 6 watersheds sampled. Using cluster analysis, sites were grouped into two categories, high and low risks, based on combined levels of indicators. These results suggest that stations of highest pollution risk were located within areas of high OSDS densities. Furthermore, data indicate a subsurface transport of contaminated water to surface waters. The high prevalence of enteroviruses throughout the study area suggests a chronic pollution problem and potential risk to recreational swimmers in and around Sarasota Bay.  相似文献

Analysis of the physical, chemical and biological parameters assessing water quality in Harris Neck estuary indicated that the average dissolved oxygen level was 8.6 mg/L, it maintained moderate levels of total dissolved nitrogen (2.7-4.6 mg/L) and total dissolved phosphorous (<0.05 mg/L), chlorophyll a was above 5.0 μg/L and it is contaminated with low levels of fecal bacteria. Bifidobacterium adolescentis, a putative marker of human fecal pollution, was detected once at stations 3 and 5. Overall the Harris Neck water quality analyses indicated a relatively pristine and a healthy functioning marine environment.  相似文献

In light of water quality monitoring paradigms shifting to a more holistic approach, it is essential that environmental microbiologists embrace new methodological developments in clinical virology to create rapid, laboratory‐free methods for the identification of wastewater pollution. It is widely accepted that routine monitoring of fecal indicator bacteria (FIB) does not adequately reflect human health risks associated with fecal pollution, especially risks posed by viruses. Enteric viruses are typically more resistant to wastewater treatment and persist longer in the environment than FIB. Furthermore, enteric viruses often have extremely low infectious doses. Currently, the incorporation of sanitary surveys, short‐term monitoring of reference pathogens, exploratory quantitative microbial risk assessments, and predictive ecological models is being championed as the preferred approach to water management. In addition to improved virus concentration methods, simple, point‐of‐use tests for enteric viruses and/or improved viral indicators are needed to complement this emerging paradigm and ensure microbial safety worldwide.

The effects from two winter rain storms on the coastal ocean of the Southern California Bight were examined as part of the Bight ‘03 program during February 2004 and February–March 2005. The impacts of stormwater from fecal indicator bacteria, water column toxicity, and nutrients were evaluated for five major river discharges: the Santa Clara River, Ballona Creek, the San Pedro Shelf (including the Los Angeles, San Gabriel, and Santa Ana Rivers), the San Diego River, and the Tijuana River. Exceedances of bacterial standards were observed in most of the systems. However, the areas of impact were generally spatially limited, and contaminant concentrations decreased below California Ocean Plan standards typically within 2–3 days. The largest bacterial concentrations occurred in the Tijuana River system where exceedances of fecal indicator bacteria were noted well away from the river mouth. Maximum nitrate concentrations (~40 μM) occurred in the San Pedro Shelf region near the mouth of the Los Angeles River. Based on the results of general linear models, individual sources of stormwater differ in both nutrient concentrations and the concentration and composition of fecal indicator bacteria. While nutrients appeared to decrease in plume waters due to simple mixing and dilution, the concentration of fecal indicator bacteria in plumes depends on more than loading and dilution rates. The relationships between contaminants (nutrients and fecal indicator bacteria) and plume indicators (salinity and total suspended solids) were not strong indicating the presence of other potentially important sources and/or sinks of both nutrients and fecal indicator bacteria. California Ocean Plan standards were often exceeded in waters containing greater than 10% stormwater (<28–30 salinity range). The median concentration dropped below the standard in the 32–33 salinity range (1–4% stormwater) for total coliforms and Enterococcus spp. and in the 28–30 salinity range (10–16% stormwater) for fecal coliforms. Nutrients showed a similar pattern with the highest median concentrations in water with greater than 10% stormwater. Relationships between colored dissolved organic matter (CDOM) and salinity and between total suspended solids and beam attenuation indicate that readily measurable, optically active variables can be used as proxies to provide at least a qualitative, if not quantitative, evaluation of the distribution of the dissolved, as well as the particulate, components of stormwater plumes. In this context, both CDOM absorption and the beam attenuation coefficient can be derived from satellite ocean color measurements of inherent optical properties suggesting that remote sensing of ocean color should be useful in mapping the spatial areas and durations of impacts from these contaminants.  相似文献

The effects of precipitation on the hygienic quality of water and blue mussels collected from five different localities in the urban areas in the Inner Oslofjord were investigated, with samples analysed for Escherichia coli, Salmonella spp., pathogenic Vibrio spp., Norovirus, Sapovirus, Cryptosporidium spp. and Giardia duodenalis. The sampling sites were located at varying distances from the outlet of combined sewer overflows (CSO)-impacted rivers/streams. In general, 1–3 log10 increases in fecal indicator bacteria and human pathogens were observed after heavy rainfalls. Blue mussels appeared to be a useful indicator of the impact of sewage at these sites, and generally a good correlation was identified between concentrations of E. coli and other human pathogens in the mussels. Provision of general advice to the public of avoiding areas near the outlets of CSO-impacted rivers after heavy rainfall may reduce the risk of gastroenteritis by bathers and others that may swallow water during recreational activities.  相似文献

Riparian invasion by non-native trees may lead to changes in the quality of leaf litter inputs into freshwater ecosystems. Different plant species may affect the community of decomposers and the rate of litter decay in different ways. We studied the microbial colonization and decomposition of leaf litter of the invasive to Lithuania Acer negundo and native Alnus glutinosa during 64-day litterbag experiments in the littoral zones of mesotrophic and eutrophic lakes. The decomposition of A. negundo leaf litter proceeded faster than that of A. glutinosa irrespective of differences in the trophic conditions of the lakes. The amount of terrestrial and cellulose-degrading fungi (during the initial period) and bacterial numbers (during the experiment) were higher on A. negundo leaves than on A. glutinosa in both lakes. Differences in the assemblages of aquatic fungi colonizing the leaves of both types might be one of the reasons causing variation in their decay. The trophic conditions of the lakes did not significantly determine the extent of differences in decomposition rates between the two leaf species, but affected the microbial decomposers. The sporulation rate and diversity of aquatic fungi, especially on A. glutinosa leaves, was higher in the mesotrophic lake than in the eutrophic lake, while heterotrophic bacteria were more numerous on the leaves in the eutrophic lake. Generally, differences in the colonization dynamics of heterotrophs and the faster decay of A. negundo litter than of A. glutinosa suggest that the replacement of native riparian species such as the dominating A. glutinosa by invasive A. negundo may cause changes of organic matter processing in the littoral zones of lakes.  相似文献

We evaluated microbiological water quality in the Pettaquamscutt River (Rhode Island, USA), an estuarine river. Fecal coliform (FC) and enterococci (FE) bacteria, presence of Bifidobacterium adolescentis DNA (indicating human fecal contamination), and optical brightener (OB) fluorescence (associated with laundry detergents) were determined for 14 stations from May to September 2010. Six stations had high counts of FE and FC, and the presence of B. adolescentis DNA and high OB fluorescence indicated human fecal contamination - four had septic systems as likely sources of contamination; the others were in sewered areas. The ability of FC and FE to indicate human fecal contamination was assessed against a positive B. adolescentis test. FC and FE had false positive rates of 25% and 17%, respectively, and false negatives of 44% for FC and 63% for FE. Inclusion of molecular and chemical indicators should improve tracking of human fecal contamination sources in the river.  相似文献

Mineral constituents of clay materials may promote interaction, adsorption and attachment of microorganisms, often resulting in biofilms’ formation. In this study investigation is made to determine how littoral clayey materials on the shores of a river promote accumulation of bacteria and increase contamination of river water. Clayey samples were collected at various points along the shore of a river around Mondeor in Johannesburg and the mineralogical composition was determined using XRD and XRF. Microorganisms in clay-biofilm and river water were identified by DNA sequencing and plate count. Results showed that total coliforms, Escherichia coli, Pseudomonas sp. and presumptive indigenous microorganisms attached to littoral clayey materials containing the mineral muscovite (characterising argillaceous soils). Bacteria number on clayey materials was significantly higher than on overlying water especially before rainy season. However a decrease of the number of bacteria in clayey materials concurrent with an increase in the number of suspended bacteria after rain events, was the result of the action of high and fast flows in the basin, eroding the biofilms. Attachment of microorganisms in clayey material as observed in this study could be ascribed to the glue-like aspect of soil (due to muscovite) that facilitates adhesion. It therefore demonstrates the potential of clayey materials to encourage biofilm formation and enhance microbial contamination of river water as shown here.  相似文献

The bacterial communities in the anoxic layer of a heavily polluted microbial mat and their growth on hydrocarbons under sulfate-reducing conditions were investigated. Microbial communities were dominated by members of Alphaproteobacteria (27% of the total rRNA), Planctomycetes (21.1%) and sulfate-reducing bacteria (SRB: 17.5%). 16S rRNA cloning revealed sequences beloning to the same bacterial groups with SRB affiliated to the genera Desulfobulbus, Desulfocapsa, Desulfomicrobium, Desulfobacterium and Desulfosarcina/Desulfococcus. The derived enrichment cultures on crude oil, hexadecane and toluene were dominated by SRB. While most SRB sequences of the toluene and hexadecane cultures were related to the sequence of Desulfotignum toluolica, the crude oil enrichment showed a more diverse bacterial community with sequences from the genera Desulfotignum, Desulfobacter, Desulfatibacillus, Desulfosalina, and Desulfococcus. We conclude that the anoxic layer of the studied mats contains a diverse community of anaerobic bacteria, dominated by SRB, some of which are able to grow on hydrocarbons.  相似文献

Corals and reef environments are under increased stress from anthropogenic activities, particularly those in the vicinity of heavily populated areas such as the Florida Keys. The potential adverse impacts of wastewater can affect both the environment and human health; however, because of the high decay rate of bacterial indicators in coral reef waters it has been difficult to document the presence of microbial contaminants and to assign risks in these environments. Here we show initial evidence that microorganisms associated with human feces are concentrated along the surface of coral heads relative to the overlying water column in the Florida Keys. Bacterial indicators (fecal coliform bacteria, enterococci or Clostridium perfringens) were detected in 66.7% of the coral surface microlayer (CSM) samples at levels between five and 1000 CFU/100 ml, but were found infrequently and at low numbers in the overlying water column ( < or = 2.5 CFU/100 ml). Similarly, enterovirus nucleic acid sequences, an indicator of human-specific waste, were detected in 93.3% of the CSM samples and only once in the water column by cell culture. Results show that coral mucus may accumulate enteric microorganisms in reef environments, and may indicate a risk to public and environmental health despite low indicator levels in the surrounding water.  相似文献

Although urban wet weather discharges may have elevated concentrations of fecal indicator bacteria impacting water quality at swimming beaches, not all of these bacteria may arise from human sources. In this study, the contribution of non-human fecal indicator bacteria was quantified by sampling coastal reference beaches in southern California. Samples were collected at beaches near stormwater discharges from undeveloped watersheds and analyzed for total coliform, Escherichia coli, and enterococci. Surfzone samples exceeded water quality thresholds >10 times more frequently during wet weather than dry weather. Exceedences were greatest <24 h following rainfall, then steadily declined on successive days. Early season storms exceeded thresholds more frequently, and by greater magnitude, compared to late season storms. Large storms exceeded thresholds more frequently than smaller-sized storms, partly due to the breaching of sand berms. When discharges did reach the surf zone, bacterial concentrations in the wave wash were correlated with watershed bacterial flux.  相似文献

Bathing water quality is an important public health issue, mainly because of fecal contamination. In 2006, the European Commission (EC) adopted a new directive with respect to recreational bathing waters that calls for stricter standards and reduces the number of laboratory tests done in routine beach monitoring from nineteen to two bacterial indicators, namely, Escherichia coli and intestinal enterococci, replacing policies of the EC Bathing Water Directive that have existed since 1976. Our practice in Portugal is in line with this international development, and this study demonstrates the equivalency of the new bacteriological parameters with the old. The water quality of 25 coastal beaches was surveyed using both new and old microbiological indicators of fecal contamination. Statistical analysis demonstrated equivalency of the results obtained for fecal coliforms with Escherichia coli and for fecal streptococci with intestinal enterococci.  相似文献
