利用二乙胺基乙基琼脂糖凝胶柱层析纯化自养小球藻(Chlorella autotropica)粗多糖,得到3个硫酸多糖组分(A,B,C);利用高效液相色谱仪和熔点仪测定了3个多糖的分子质量和熔点;利用BaCl2浊度法测定了硫酸多糖中硫酸根的质量分数;利用傅立叶变换红外光谱分析了海水自养小球藻3个硫酸多糖组分的环状结构、半缩醛羟基构型及取代基类型。结果显示,3个硫酸多糖组分的硫酸根的质量分数分别为0.77%,8.46%,2.69%,3个组分均含有硫酸基、酰氨基,葡萄糖是3个硫酸多糖组分的主要组成单糖;3个硫酸多糖组分组成单糖的环构型均是吡喃环,成苷的半缩醛羟基构型有一定差异,A是α构型,B、C是α、β两种半缩醛羟基构型并存。为深入研究自养小球藻硫酸多糖的结构及抗癌活性提供基础。  相似文献

采用多种方法提取了龙须菜(Gracilaria lemaneiformis)中的琼胶,并研究了碱处理、微波处理、超声波处理等方法对龙须菜的琼胶提取率、硫酸基质量分数和多糖质量分数的影响。结果表明:当碱质量分数由2.5%增加到10%时,琼胶产率由15%降低到9.8%,硫酸基质量分数由2.66%降低到2.06%,同时,多糖质量分数由98.3%增加到102.6%;微波和超声波处理在不同程度上增加了琼胶的提取率和多糖质量分数,同时降低了硫酸基质量分数。另外,还从宏观和微观两个角度考察了龙须菜碱处理前后藻体的变化。  相似文献

以分子量为考查指标,采用正交实验法研究了海洋硫酸多糖916的氧化降解最佳条件。考察了H2O2浓度、原料用量和反应时间对降解后分子量的影响。结果表明,最佳降解条件为体系中H2O2浓度为3%,原料用量为2%,反应时间为3h。该法简单易行,操作简便,所用试剂廉价易得,是一种简单有效的降解方法。  相似文献

慢性肾衰的进程中伴随着肾脏炎症的发生,研究表明褐藻多糖硫酸酯具有抗炎作用,且对于慢性肾衰的早期治疗具有一定的作用。本文通过脂多糖(lipopolysaccharide,LPS)诱导,建立大鼠肾小球系膜细胞的炎症模型,用来源于海带的高分子质量褐藻多糖硫酸酯F1和低分子质量褐藻多糖硫酸酯F2对细胞进行处理,24 h后MTT法检测各组细胞活力,Griess法检测24 h及48 h后细胞培养液中NO的含量。结果表明,各组间细胞活力无明显差异;脂多糖可以诱导大鼠肾小球系膜细胞NO产量增加,F1和F2均可在一定程度上抑制脂多糖诱导的NO产量的增加,从而减轻细胞炎症反应。剂量为25μg/m L的F1和F2处理细胞48 h后,与模型组相比,F1和F2组细胞培养液中NO含量分别下降22.52%和38.65%,低分子质量褐藻多糖硫酸酯抗炎效果明显高于高分子质量褐藻多糖硫酸酯。  相似文献

以海萝藻(Gloiopeltis furcata)为原料,依次经冷水、热水和热碱溶液提取,从中获得了3种多糖(GFW,GFH和GFA),其得率分别为57.9%,2.5%和2.6%。运用气相色谱法(GC)、高效凝胶渗透色谱法(HPGPC)、红外光谱(IR)及核磁共振碳谱(13C-NMR)等方法对其单糖组成、分子量以及结构进行了分析。结果表明,GFW和GFH都只含有D-半乳糖(Gal)和3,6-内醚-L-半乳糖(AnG),而GFA除了含有Gal和AnG外,还有木糖;3种多糖的分子量分别为22.6 kD,26.5 kD和49.8 kD;硫酸基含量分别为31.2%,25.1%,22.7%。IR和13C-NMR数据均表明其硫酸基在半乳糖残基C6位。结论:海萝藻中多糖均为硫酸多糖,其中2种为硫琼胶,另一种为含有木糖的硫酸半乳聚糖。  相似文献

以κ-卡拉胶(硫酸基含量25.1%,分子质量83.1 kD)为原料,研究了二甲基亚砜-甲醇法(DMSO法)、三甲基氯硅烷法(CTMS法)以及苯并四甲酸-三氧化二锑法(PMA法)对其脱硫后多糖样品的得率、硫酸基含量及分子质量的影响.结果表明,κ-卡拉胶经三种方法脱硫后,样品得率分别为45.3%、50.6%和62.0%,脱硫率分别为42.2%、48.6%和82.8%,分子质量分别为15.7、22.3和4.75 kD.通过红外光谱和硫酸基含量分析表明,PMA法脱硫效果优于DMSO和CTMS法.PMA法不仅对κ-卡拉胶有效,而且适合于l-卡拉胶、λ-卡拉胶和岩藻聚糖硫酸酯的脱硫.  相似文献

本研究对一株经太空搭载后选育的海水钝顶螺旋藻诱变株H_(11)的室外产多糖能力进行了评估。结果显示,在广州秋季室外平均温度为27°C下,诱变株H_(11)的生物量产率、总糖含量、总多糖含量和水溶性多糖含量分别为19.09±0.03 mg/(L·d)、27.89±1.28%DW、20.95±0.87%DW和19.74±0.93%DW。与野生株WT相比,H_(11)的生物量、总糖、总多糖和水溶性多糖的产率分别提高了15.39%,148.93%,176.50%和187.70%。在冬季室外平均培养温度为17°C条件下,诱变株和野生株生长速率及多糖产率都有所降低,但诱变株在产多糖能力上仍显著高于野生株(P0.05)。研究表明海水钝顶螺旋藻诱变株H_(11)产多糖能力极高,具有可观的产业应用前景。  相似文献

主要采用傅里叶红外光谱(fourier transform infrared spectroscopy, FTIR)、气相色谱(gas chromatography, GC)和凝胶渗透色谱(gel permeation chromatography, GPC)方法对5种海藻粗多糖的单糖组成、官能团、分子量进行了初步分析。结果显示5种海藻粗多糖均含有木糖和岩藻糖, 其中南方团扇藻多糖含有较为丰富的山梨糖, 属于大分子量均一性多糖, 而孔石莼多糖分子量分布较宽, 均一性较差。红外光谱分析结果表明, 5种粗多糖均为酸性多糖, 棒叶蕨藻、琼枝麒麟菜和匍枝马尾藻多糖具有明显的硫酸基团吸收峰, 琼枝麒麟菜多糖存在呋喃糖和吡喃糖, 并且吡喃糖苷键可能存在β和α两种类型。明胶氯化钡实验结果指出南方团扇藻多糖的硫酸基团含量为38.43%(±2.38%), 明显高于其他几种海藻多糖, 而棒叶蕨藻多糖的硫酸基团含量最低, 仅为8.92%(±1.04%)。  相似文献

采用人肠道微生物体外厌氧发酵技术,研究人肠道微生物对不同分子量岩藻聚糖硫酸酯的降解利用。分别将五个志愿者的肠道微生物接种到以高分子量和低分子量岩藻聚糖硫酸酯为唯一碳源的培养基中,酵解48h后,采用TLC和PAGE分析分子量变化,PMP-HPLC分析单糖组成变化,GC分析短链脂肪酸的生成情况。结果发现:在人肠道微生物体外降解岩藻聚糖硫酸酯的体系中,岩藻聚糖硫酸酯寡糖和低分子量组分(10—20k Da)含量显著降低,而且酵解产物的单糖组成没有变化,说明被人肠道微生物彻底降解利用;而人肠道微生物对高分子量岩藻聚糖硫酸酯(20k Da)的降解和利用度很低,产物的半乳糖和甘露糖的比例明显下降。低分子量和高分子量岩藻聚糖硫酸酯在酵解后均生成短链脂肪酸乙酸、丙酸和丁酸,但在高分子量岩藻聚糖硫酸酯发酵液中还生成了支链脂肪酸(异丁酸、异戊酸)和戊酸。研究结果表明,人肠道微生物能够彻底降解利用复杂的海带岩藻聚糖硫酸酯寡糖和低分子量组分,但是对高分子量岩藻聚糖硫酸酯的降解率很低。单糖分析说明人肠道微生物能够降解和利用多糖中的所有单糖,产生有利于肠道健康的短链脂肪酸,但是只有低分子量岩藻聚糖硫酸酯能够抑制支链脂肪酸生成。  相似文献

褐藻多糖硫酸酯(fucoidan,FPS)是所有褐藻中所固有的细胞间多糖,存在于细胞壁基质中。生长于潮间带、长时间与空气接触的褐藻种类中,如多年生的墨角藻(Fucus vesiculosus)类,其褐藻多糖硫酸酯的含量可高达20%;生长在较深处的海带(Saccharina japonica)类中含量较低,约为1%~2%。褐藻多糖硫酸酯是一类独特的结合有硫酸基的水溶性杂聚糖,其化学组成和结构非常复杂,以岩藻糖和硫酸基为主,随着褐藻的种类不同还含有半乳糖、木糖、糖醛酸等其他成分。褐藻多糖硫酸酯具有多种生物活性,目前针对墨角藻、泡叶藻(Ascophyllum nodosum)、鼠尾藻(Sargassum thunbergii)等褐藻中的褐藻多糖硫酸酯研究较多,发现其具有免疫调节、抗病毒、抗肿瘤、抗凝血、抗氧化等生物活性。本文主要就褐藻多糖硫酸酯的提取、纯化、结构及生物活性等方面研究进行综述。  相似文献

Stocking density is widely recognized as a critical factor in aquaculture and a potential source of long-term stress.The influence of stocking density on growth and stress response of juvenile turbot(Scophthalmus maximus, ~3–75g, initial to final weight) was examined in fish held under low(LD, ~0.21–5.31 kg/m~2, initial to final density),medium(MD, ~0.42–10.81 kg/m~2) and high stocking density(HD, ~0.63–14.27 kg/m~2) for 120 days in a recirculating aquaculture system(RAS). In this trial, the growth curve for weight of juvenile turbot in RAS, all fitted by the Schnute model. No significant difference was found in growth performance among the three densities until at the final sampling(Day 120). The final weight and body weight increase(BWI) in the HD group were significantly lower than in other groups(P0.05, weight:(75.83±2.49) g,(75.39±2.08) g,(65.72±2.86) g and BWI:(2 436.12±28.10)%,(2 421.29±4.64)%,(2 097.88±20.99)% in LD, MD and HD groups, respectively). Similarly, the specific growth rate(SGR), feed conversion ratio(FCR) and coefficient of variation for weight(CV_w) were adversely affected by high stocking density(P0.05). However, there was no difference in survival and Fulton's condition factor(K) of turbot among the different groups. Physiological analyses demonstrated a clear increase in the plasma cortisol level and an obvious decrease in growth hormone(GH) concentration in the HD group on Day120(P0.05). There was no significant effect of stocking density on plasma glucose, Cl– and protein levels. All these findings would provide a reference for selecting the optimal stocking density of juvenile turbot in RAS.  相似文献

South African abalone Haliotis midae farms utilise large volumes of seawater (c. 500–1 500 l s–1) and produce relatively dilute effluents that are potentially suitable for the integrated culture of other species. To test this hypothesis, a marine finfish, silver kob Argyrosomus inodorus, and a detritivorous polychaete, bloodworm Arenicola loveni loveni, were cultured in abalone farm effluent and the results compared to controls reared in unused seawater. The silver kob were fed a nutritionally complete pelleted diet whereas the bloodworm were placed in shallow tanks with a low water velocity that allowed suspended organic solids to settle for the detritivorous worms to feed on. Silver kob growth rate (0.48% body weight d–1; SE = 0.01%), mortality (1.8 ± 0.5%), feed conversion ratio (3.0 ± 0.2) and protein efficiency ratio (1.0 ± 0.1) did not differ significantly between the effluent and control treatments. Bloodworm reared in abalone effluent grew well on the particulate organic waste matter in the effluent (0.39% body weight gain d–1; SE = 0.07%), whereas those in the seawater control lost weight at 0.19 ± 0.04% body weight d–1 over the experimental period. Bloodworm mortality did not differ significantly between effluent (6 ± 3%) and unused seawater (11 ± 8%) treatments. The faster growth of bloodworm in the abalone farm effluent was ascribed to the higher deposition rate of enriched organic solids (182 ± 56 g m–2 d–1) compared with those grown in the seawater control (46 ± 13 g m–2 d–1). It was concluded that abalone farm effluent is potentially suitable for the culture of both bloodworm and silver kob.  相似文献

Patterns of ventilation, oxygen consumption (VO2), blood gas tensions (PaO2 and PaCO2) and pH were determined in shortfin eels (Anguilla australis schmidtii (Phillips)). At rest, shortfin eels (average weight 646.5 ± 64.5 g) had a VO2 of 20.4 ± 1.2 ml. kg‐1.h‐1 (n = 13) at 17.5 ± 0.2°c, with smaller fish having the highest metabolic rates. The frequency of ventilation was inversely proportional to body weight in both A. australis schmidtii and A. dieffenbachii. In air‐saturated water 10 eels exhibited periodic apnoea (mean duration 3.59 min); periods of ventilation were more variable in duration (mean 4.92 min). After 2.62 min of apnoea, the PaO2 of dorsal aortic blood had fallen from 9.12 to 1.91 kPa. Thus, although the blood has a high affinity for oxygen and the haemoglobins are 30% oxygen‐saturated at this low PaO2, the eel allows its blood to be significantly depleted in oxygen during apnoeic pauses at rest. When ventilating its gills at rest, PaO2 does not approach the PO2 of the inspired water. It is suggested that these features of respiration in eels result in a saving of metabolic costs involved in ventilation. The results are discussed in terms of the eel's ability to withstand hypoxic conditions.  相似文献

在工厂化循环水养殖系统中,将初始体质量为186 g±2.0 g的大菱鲆(Scophthalmus maximus)放养于低(9.4 kg/m~2±0.2 kg/m~2)、中(13.6 kg/m~2±0.8 kg/m~2)、高(19.1 kg/m~2±1.3 kg/m~2)3个养殖密度,以研究不同养殖密度对大菱鲆生长、消化酶和蛋白质代谢的影响。养殖120 d后,低、中、高试验组养殖密度分别增长至26.1 kg/m~2±1.2 kg/m~2、38.2 kg/m~2±2.5 kg/m~2、52.3 kg/m~2±3.6 kg/m~2。结果表明:低密度组和中密度组中大菱鲆增质量率、特定生长率、肥满度和蛋白质效率均显著(P0.05)高于高密度组;而饲料系数显著低于高密度组(P0.05)。低密度组和中密度组大菱鲆总蛋白酶和淀粉酶活力显著低于高密度组(P0.05);但脂肪酶活力在3个密度组之间无显著性差异。与低密度组相比,高密度组显著提高了谷氨酸脱氢酶活力,同时降低了谷草转氨酶和亮氨酸氨肽酶活力(P0.05),而对谷丙转氨酶活力无任何影响。综上所述,在工厂化循环水系统中,增加养殖密度能提高养殖的产量,但过高的养殖密度会对大菱鲆生长、消化酶活力以及蛋白质代谢产生不利的影响。  相似文献

The effect of dietary protein to energy ratio (P:E) on the growth of dusky kob Argyrosomus japoni-cus was investigated as a first step towards formulating a practical diet for this potential mariculture species in South Africa. The effects of dietary protein and lipid on growth, feed conversion ratio (FCR) and nitrogenous waste production were determined in cultured juveniles (initial weight 113.8 ± 2.0 g fish?1). Fish were fed diets that contained either 42 or 46% protein with 6, 12 or 18% lipid, within a P:E range of 20.5–26.7 g protein MJ?1. Weight gain and specific growth rate (SGR) were significantly influenced by an interaction between dietary protein and energy (p < 0.001). An increase in dietary lipid produced increasing growth rates (ranging from 0.95 ± 0.04% to 1.10 ± 0.01% day?1) among fish fed the 46% protein diets, with no significant differences in FCR (1.05 ± 0.02 to 1.11 ± 0.02) or protein efficiency ratio (PER; 1.95 ± 0.06 to 1.66 ± 0.02). By contrast, there was a trend of decreasing growth (0.96 ± 0.04% to 0.64 ± 0.03% day?1) and a deterioration in FCR (1.3 ± 0.10 to 1.93 ± 0.19) and PER (1.12 ± 0.11 to 1.68 ± 0.11) with increasing dietary lipid among fish fed 42% protein diets, indicating a protein– energy imbalance. The protein and lipid levels tested here had no significant effect on total ammonia production, with a combined mean production of 2.54 ± 0.55 mg kg?1 I?1 h?1, 10 hours postprandial (p = 0.68). The dietary lipid level appeared to directly determine the carcass lipid content, with the mean lipid content ranging from 4.2% to 17.8% for fish fed diets ranging from 6% to 18% respectively after 12 weeks (r2 = 0.98). A 46% protein and 18% lipid (P:E ratio of 22.0 g protein MJ?1) diet is recommended for rearing juvenile A. japonicus of a mass range of 100–300 g.  相似文献

The brown sea cucumber Isostichopus fuscus (Ludwig, 1875) has been subjected to strong fishing pressure and is currently considered an endangered species by the IUCN Red List of Threatened Species. Spatial and temporal variations in density were analysed and population parameters of I. fuscus were estimated in three localities of the Southern Mexican Pacific. Density was estimated using circular transects covering a 300 m2 area in each locality. Body length and weight were also measured. The mean density was 0.018 ± 0.013 individuals·m?2, and was statistically significantly lower during summer. The mean length and weight were 22.4 ± 3.8 cm and 396 ± 130 g, respectively, and statistically significant differences were found among localities. The weight–length relationship was W = 0.28·L1.32 and the mean relative condition index (Kn) = 1.04 ± 0.31. The von Bertalanffy growth equation was Lt = 38.7·(1–exp(?0.4·t)). The lowest values of length, weight and age were found at Dos Hermanas, suggesting that this locality could be favorable for the settlement of larvae and recruitment and growth of juveniles. The present study is the second to estimate densities of I. fuscus and the first one to publish data on size structure and growth for the population at Bahias de Huatulco. The densities were so low in the three sampled localities that none of them would be able to support a fishery, even under a sustainable management scheme. The information collected in the present study could be helpful for management programs of the Huatulco National Park, as all the data used for these currently come mainly from the populations of I. fuscus in the Gulf of California.  相似文献

Size trends and population characteristics of striped marlin, Tetrapturus audax, caught in the New Zealand recreational fishery were evaluated using club records from the Bay of Islands Sword‐fish Club (BOISC) from 1925 to 2003 (n = 15 114) and biological samples from northern New Zealand collected from 1985 to 1994 (n = 684). Average weight (kg) ± SE of striped marlin declined 6–9 kg every 20 years between 1925 (124.2 ± 1.0 kg) and 2003 (100.1 kg ± 0.4). Weight (H14843 = 444.58, P < 0.001) and condition (H443 = 37.54, P < 0.001) increased significantly throughout the recreational fishing season (December‐May). No differences were observed in the lower jaw‐fork length (LJFL)‐weight (W) relationship between sexes or years but average weight of females (106.2 ±1.1 kg) was significantly greater than males (90.2 ±1.2 kg) from 1985 to 1994 (F684 = 88.37, P < 0.001). Negative allometric growth (W = 2E ‐08 LJFL2.88) was recorded for all striped marlin. Individual growth was modelled by fitting backcalculated LJFL to eight age classes derived from dorsal spine measurements (n = 94) using the von Bertalanffy growth equation (r2 = 0.96). The parameters estimated in the von Bertalanffy growth equation were asymptotic length (L ) = 3010 mm, annual growth rate (K) = 0.22, age (yr) at hypothetical length 0 (t0) = ‐0.04.  相似文献

Pueruli and post‐pueruli, early juveniles and sub‐adults of the spiny lobster, Panulirus homarus and juveniles of P. ornatus were grown in different floating sea cages along the southeast coast of India from May 2003 to May 2007. The first type of cage had a galvanised iron pipe frame (2.0 m × 2.0 m × 1.2 m) with steel woven mesh and four inner detachable compartments (0.75 m × 0.75 m × 1.10 m). Fibre‐reinforced plastic was used subsequently to fabricate cages (1 m × 1 m × 1 m). Pueruli and post‐pueruli of P. homarus (1.58 ± 0.62 g SD), stocked at 60 individuals/m2, grew to an average weight of 123.10 ± 26.22 g in 266 days with a survival rate of 70%. Sub‐adults of P. homarus with an average weight (± SD) of 123.61 ± 29.26 g reached 341.25 ± 46.22 g in 225 days at a stocking density of 21 individuals/m2 with a survival of 73 ± 6%. The post‐pueruli grew by 0.46 ± 0.10 g per day with a specific growth rate (SGR) of 1.64, whereas sub‐adults had a growth rate of 0.97 ± 0.20 g per day with a SGR of 0.43. At a higher stocking density of 80 individuals/m2, juveniles (51.83 ± 10.32 g and 58.20 ± 28.22 g) of P. homarus recorded growth rates of 0.86 ± 0.25 (SGR 0.82) and 0.97 ± 0.34 g (SGR 0.96) per day. This study indicates that post‐pueruli of P. homarus can be grown to over 200 g in 12 months and up to 350 g in 16 to 17 months in sea cages. Juveniles (average weight 76.35 ± 34.50 g) of P. ornatus, reared with P. homarus at a stocking density of 80 individuals/m2, recorded a weight gain of 139 g in 155 days at arate of 0.89 ± 0.32 g per day with an SGR of 0.67. Marine live clam, Donax spp., was the main feed supplemented with the gastropod, Xancus pyrum, the green mussel, Perna viridis, marine crab (Charibdis sp.), squid (Loligo sp.), and fish such as clupeids and Leognathus sp. Pueruli and post‐pueruli settled in large numbers (up to 35 individuals/month in one cage) both inside and outside the cages.  相似文献

Ecological experiments were conducted to examine the effects of seawater containing elevated par- tial pressure of carbon dioxide (p CO2 800 × 10 -6 , 2 000 × 10 -6 , 5 000 × 10 -6 and 10 000 × 10 -6 ) on the survival and reproduction of female Acartia pacifica, Acartia spinicauda, Calanus sinicus and Centropages tenuiremis, which are the dominant copepods in the southern coastal waters of China. The results show that the effects of elevated p CO2 on the survival rates of copepods were species-specific. C. sinicus, which was a macro-copepod, had a higher survival rate (62.01%-71.96%) than the other three species (5.00%-26.67%) during the eight day exposure. The egg production rates of C. sinicus, A. spinicauda and C. tenuiremis were significantly inhibited by the increased p CO2 and the exposure time duration. There were significantly negative impacts on the egg hatching success of A. spinicauda and C. tenuiremis in the p CO2 2 000 × 10 -6 and 10 000 × 10 -6 groups, and, in addition, the exposure time had noticeably impacts on these rates too. This study indicates that the reproductive performances of copepods were sensitive to elevated p CO2 , and that the response of different copepod species to acidified seawater was different. Furthermore, the synergistic effects of seawater acidification and climate change or other pollutant stresses on organisms should be given more attention.  相似文献

Holothuria arguinensis is a potential species for sea cucumber aquaculture in Europe. In a first experiment, feeding rate (FR), growth, absorption efficiency (AE) and survival of adults (135.01?±?10.87?g eviscerated weight) were assessed under sediment tank conditions. In a second experiment, the feasibility to use sea bream (Sparus aurata) biodeposits as a food source was studied for adults (168.69?±?14.59?g eviscerated weight). Individuals in the first experiment showed an increase of weight (specific growth rate (SGR)?=?0.2% d?1), positive FRs and mean value of AE close to 80% with a mean organic matter content of 90.07?±?11.5?mg/g in the offered sediment. Sea cucumbers fed with fish biodeposits showed a reduction of weight (SGR??1), low FRs and negative values of AE. The influence of low salinity and temperature during experiments should be considered to explain these results. Indeed, sea cucumbers can be affected by a decrease of these parameters during winter months resulting in a period of inactivity.  相似文献
