以哈维弧菌(Vibrio harveyi)、鲍类疱疹病毒(Abalone herpesvirus,Ab HV)悬液对杂色鲍(Haliotis diversicolor)注射刺激,研究其血淋巴中可溶性总蛋白浓度,免疫相关酶超氧化物歧化酶(Superoxide Dismutase,SOD)、酸性磷酸酶(Acid Phosphatase,ACP)、碱性磷酸酶(Alkaline Phosphatase,AKP)活性的变化,杂色鲍外套膜、鳃、肝胰腺、腹足中血蓝蛋白(Hemocyanin,Hc)基因Hc1、Hc2相对表达量的变化以及感染后杂色鲍无细胞血浆的抑菌性。结果显示:(1)杂色鲍血淋巴中可溶性总蛋白浓度变化显著,呈现出先升高后降低的趋势。(2)无细胞血浆中的SOD、ACP、AKP活性与对照组相比变化显著,哈维弧菌注射组的SOD活性在感染后第6小时显著升高;Ab HV注射组SOD活性与对照组比较在注射后的各个时相均呈下降趋势,从注射后6 h各个时相的SOD活性与对照组相比均差异显著(P0.05)。哈维弧菌注射组ACP活性在注射后的第48小时显著升高;Ab HV注射组ACP活性在注射后的第12小时显著升高。哈维弧菌注射组AKP活性在注射后48 h显著升高;AbHV注射组AKP活性在注射后第12小时显著升高。(3)杂色鲍外套膜、鳃、肝胰腺以及腹足中血蓝蛋白两个基因Hc1、Hc2的相对表达量均在注射后的不同时间点出现升高。(4)杂色鲍无细胞血淋巴对哈维弧菌、创伤弧菌和溶珊瑚弧菌这三种贝类病原均表现出了不同程度的抑菌性增强的作用。这些结果表明细菌和病毒的刺激引起了杂色鲍免疫相关因子的变化,使得血淋巴的抑菌性增强。本研究为今后进一步研究杂色鲍的非特异性免疫以及在生产实践中应用免疫技术防治病害提供参考。  相似文献

杭州湾内水温在不同时间尺度下均存在变化,这对河口生态等会产生影响。由于昼夜气温变化大,空气和海水的热量交换较为频繁。另外,随着气温的逐日下降,水温也随之降低。为研究日气温变化下杭州湾内的水温变化,建立了三维数学模型,通过考虑水面和空气的热交换通量,复演了日气温变化下湾内水温特征。经过和实测潮流数据比较,本文模型很好地模拟出了杭州湾内大、中和小潮期间水流的变化。在综合考虑太阳辐射、长波辐射和水气感热交换下,模型成功再现了日气温变化下水温的变化过程。研究结果一方面有助于认识强潮河口湾内水温变化特征,另一方面将为研究温度对泥沙运动的影响提供前期基础。  相似文献

Extracellular enzyme activities were compared among surface water, bottom water, and sediments of the Delaware Estuary using six fluorescently labeled, structurally distinct polysaccharides to determine the effects of suspended sediment transport on water column hydrolytic activities. Potential hydrolysis rates in surface waters were also measured for the nearby shelf. Samples were taken in December 2006, 6 months after a major flood event in the Delaware Basin that was followed by high freshwater run-off throughout the fall of 2006. All substrates were hydrolyzed in sediments and in the water column, including two (pullulan and fucoidan) that previously were not hydrolyzed in surface waters of the Delaware estuary. At the time of sampling, total particulate matter (TPM) in surface waters at the lower bay, bay mouth, and shelf ranged between 31 mg l−1 and 48 mg l−1 and were 2 to 20 times higher than previously reported. The presence of easily resuspended sediments at the lower bay and bay mouth indicated enhanced suspended sediment transport in the estuary prior to our sampling. Bottom water hydrolysis rates at the two sites affected by sediment resuspension were generally higher than those in surface waters from the same site. Most notably, fucoidan and pullulan hydrolysis rates in bay mouth bottom waters were 22.6 and 6.2 nM monomer h−1, respectively, and thus three and five times higher than surface water rates. Our data suggest that enhanced mixing processes between the sediment and the overlying water broadened the spectrum of water column hydrolases activity, improving the efficiency of enzymatic degradation of high molecular weight organic matter in the water with consequences for organic matter cycling in the Delaware estuary.  相似文献

Few phosphorus-depleted coastal ecosystems have been examined for their ability to hydrolyze phosphomonoesters. We examined seasonal (August 2006–April 2007) alkaline phosphatase activity in Florida Bay, a phosphorus-limited shallow estuary, using fluorescent substrate at low concentrations (≤2.0 μM). In situ dissolved inorganic and organic phosphorus levels and phosphomonoester concentrations were also determined. Water column alkaline phosphatase activity was partitioned into two particulate size fractions (>1.2 and 0.2–1.2 μm) and freely dissolved enzymes (<0.2 μm). Water column alkaline phosphatase activity was also compared to leaf and epiphyte activity of the dominant tropical seagrass Thalassia testudinum. Our results indicate: (1) potential alkaline phosphatase activity in Florida Bay is high compared to other marine ecosystems, resulting in rapid phosphomonoester turnover times (2 h). (2) Water column alkaline phosphatase activity dominates, and is split equally between particulate and dissolved fractions. (3) Alkaline phosphatase activity was highest during cyanobacterial blooms, but not when normalized to chl a. These results suggest that dissolved, heterotrophic and autotrophic alkaline phosphatase activity is stimulated by phytoplankton blooms. (4) The dissolved alkaline phosphatase activity is relatively constant, while the particulate activity is seasonally and spatially dynamic, typically associated with phytoplankton blooms. (5) Phosphomonoester concentrations throughout the bay are low, even though potential hydrolysis rates are high. We propose that bioavailable dissolved organic P is hydrolyzed by dissolved and microbial alkaline phosphatase enzymes in Florida Bay. High alkaline phosphatase activity in the bay is also promoted by long hydraulic residence times. This background activity is primarily driven by carbon and phosphorus limitation of microorganisms, and regeneration of enzymes associated with cell lysis. Pulses of inorganic phosphorus and labile organic phosphorus and nitrogen may stimulate autotrophs, particularly cyanobacteria, which in turn promote biological activity that increase alkaline phosphatase activity of both autotrophs and heterotrophs in the bay.  相似文献

以大菱鲆为实验材料,研究其繁殖周期内卵子和卵巢液中磷酸酶活性变化规律及其与受精率相关性。结果显示:在大菱鲆繁殖周期内,卵子碱性磷酸酶和酸性磷酸酶活性呈相反的变化趋势,且排卵中期卵子碱性磷酸酶和酸性磷酸酶活性分别显著低于和高于早期和晚期(P<0.05),而卵巢液中碱性磷酸酶和酸性磷酸酶活性呈同卵子中相反的变化趋势;此外卵子碱性磷酸酶和酸性磷酸酶及卵巢液酸性磷酸酶活性同受精率显著相关(P<0.05)。以上结果表明,大菱鲆卵子和卵巢液中碱性磷酸酶和酸性磷酸酶活性同受精率存在着显著相关性,其活性变化在一定程度上反映卵子质量优劣,可作为评判卵子质量辅助指标。  相似文献

Rates of short-term (up to 2 years) bed elevation change and sedimentation from mudflats to salt marshes were measured in a rapidly infilling macrotidal estuary using an original combination of three high-resolution techniques: an ultrasonic altimeter, the Rod Surface-Elevation Table (RSET) method, and filter traps. The Authie estuary is located on a straight, sand-rich coast and is undergoing rapid infill under the influence of flood-dominant tides reinforced by wave action. The estuarine sediment suite consists of both mud and sand derived from the sea, of sand derived from storm wave erosion of dunes lining the north bank of the estuary, and, to a much smaller extent, of mud from the river catchment. Bed elevation change and sedimentation rates show an expected increase with the duration of tidal flooding (hydroperiod) in both space and time. The estuarine bed sediment suite changes from sandy at the mouth to muddy within the low-energy inner estuary, where mudflats are rapidly accreting, paving the way for the formation of increasingly denser and mature salt marshes from the high-sedimentation pioneer zone to the upper marsh where annual sedimentation is very low. Recorded variability in rates of bed elevation change and sedimentation reflect the influence of estuarine macro-scale and local sediment transport and depositional processes in a macrotidal context dominated by high inputs of allochthonous sediments.  相似文献

The active lifetime of extracellular enzymes is a critical determinant of the effectiveness of enzyme production as a means for heterotrophic marine microbes to obtain organic substrates. Here, we report lifetimes of three classes of extracellular enzyme in Arctic seawater. We also investigated the relative importance of photochemical processes and particle-associated processes in inactivating extracellular enzymes. Enzyme inactivation in filtered seawater was slow, with apparent half-lives of enzyme activities on the order of hundreds of hours. The presence of particles (including cells) did not significantly change inactivation rates, suggesting that the long half-lives observed in filtered seawater were realistic for enzymes in unfiltered seawater. Phosphatase and leucine aminopeptidase were susceptible to photoinactivation, but only under high intensity UV-B and UV-C illumination; there was no evidence for increased inactivation rates under natural illumination at our study site in Ny Ålesund, Svalbard. Comparison of inactivation rates of commercially-obtained enzymes from non-marine sources with the extracellular enzymes naturally present in Arctic seawater suggests that the natural enzymes contain structural features that confer longer lifetimes, consistent with observations reported by others from a range of field sites that cell-free enzymes can contribute a substantial fraction of total hydrolytic activity in the water column.  相似文献

The paper presents information on joint research of the short-term (tidal and synoptic) variability of the hydrological and hydrochemical parameters during the summer 2016 low-water period in mesotidal estuary of the Kyanda River, which flows into Onega Bay, the White Sea. It is demonstrated that semidiurnal, diurnal, and synoptic variations of almost all observed parameters are significant and differ notably along the estuary.  相似文献

In Toulon Bay (France), very high phosphatase activities have been found in the zooplankton fraction>90 microm. This work was intended to specify their origin. For that purpose, larvae, juvenile and adult Crustacea (Copepods: Calanoids, Cyclopoids, Branchiopods: Cladocera, and Cirripeds) were isolated. Their activities were measured using paranitrophenyl phosphate dissolved in sea water in order to calculate Km (the enzyme half saturation concentration) and Vmax (the reaction rate when the enzyme is saturated with substrate). Vmax were referred to protein contents of the isolated organisms to calculate specific activities. For all zooplankton groups high and low affinity phosphatase activities were found. The low affinity enzyme was responsible for at least 70% of the total phosphatase activity. Its specific activity was higher for larvae than for copepodites and adults. In Cirriped nauplii this activity was particularly high with values which were several hundred times higher than that in other Crustacea. These enzymes had optimum pH close to 8.4, magnesium requirement and were competitively inhibited by orthophosphate. Experiments with intact and lysed Cirriped nauplii confirmed that living organisms had only a weak external activity and showed that most of the activity of these larvae was primarily intracellular.  相似文献

Extracellular enzymatic activity (EEA) of five enzymes (peroxidase, phenol oxidase, beta-glucosidase, beta-N-acetylglucosaminidase and acid phosphatase) was analysed in sediments colonised by Spartina maritima in two salt marshes (Rosário and Pancas) of the Tagus estuary (Portugal) with different characteristics such as sediment parameters and metal contaminant levels. Our aim was a better understanding of the influence of the halophyte on microbial activity in the rhizosphere under different site conditions, and its potential consequences for metal cycling and phytoremediation in salt marshes. Acid phosphatase and beta-N-acetylglucosaminidase presented significantly higher EEA in Rosário than in Pancas, whereas the opposite occurred for peroxidase. This was mainly attributed to differences in organic matter between the two sites. A positive correlation between root biomass and EEA of hydrolases (beta-glucosidase, beta-N-acetylglucosaminidase and acid phosphatase) was found, indicating a possible influence of the halophyte in sediment microbial function. This would potentially affect metal cycling in the rhizosphere through microbial reactions.  相似文献

强人类活动(如航道疏浚)和自然气候变化(如海平面上升)对近岸河口环境不同影响的辨识是目前河口海岸学研究的热点和难点问题。在地形概化和动力简化条件下, 解析模型能够快速辨识强人类活动和自然气候变化对河口环境的影响, 它是探讨河口动力过程对外界干扰的响应机制的重要工具。本文基于前人对葡萄牙Guadiana河口不同分潮之间非线性相互作用的研究, 采用一维水动力解析模型探讨河口不同分潮潮波传播过程对水深变化(模拟航道疏浚和河道淤积过程)的响应机制。研究结果表明: 平均水深$\overline{h}$的变化影响无量纲河口地形参数γ和摩擦参数χ, 进一步影响河口动力参数包括潮波振幅参数ζ、流速振幅参数μ、波速参数λ、潮波振幅增大/衰减率参数δ以及流速与水位之间的相位差?等; 平均水深变化对河口中下游段(x=0~60km)的潮汐动力影响较大, 而对河口上游段(x=60~78km)影响较弱; 主要半日分潮(M2、S2、N2)对水深变化的响应略大于全日分潮(K1、O1); 航道疏浚幅度小于2m时, 对河口潮汐动力格局影响不大, 而当疏浚幅度大于2m时, 将对河口潮汐动力格局及水环境(如盐水入侵等)产生较大影响; 河道淤积将导致潮汐动力减弱, 流速振幅、潮波振幅及传播速度减小, 流速和水位之间的相位差也减小。  相似文献

为模拟夏季水分蒸发水体盐度快速升高对凡纳滨对虾(Litopenaeusvannamei)生长性能及理化调节的影响,试验设置盐度从30突变至35、40、45、50、55及60,以盐度30为对照,突变盐度下养殖28d,每7d检测凡纳滨对虾的存活率、相对增重率、体长增长率,试验结束时检测血清Na+/K+-ATP酶、总ATP酶、碱性磷酸酶(AKP)、酸性磷酸酶(ACP)和超氧化物歧化酶(SOD)活力。结果表明,高盐突变显著抑制凡纳滨对虾的存活率和相对增重率(P0.05),随着突变盐度的升高,凡纳滨对虾的相对增重率逐渐降低,60盐度组仅为对照组的15.53%。盐度突变至50后,凡纳滨对虾存活率显著下降。随着高盐突变幅度的增加,Na+/K+-ATP酶和ACP酶活力受到显著影响(P0.05),其中Na+/K+-ATP酶活力逐渐上升,在突变60盐度时表现为最高;ACP酶表现为先上升再下降的单峰变化趋势。总ATP酶、SOD酶、AKP酶受影响不显著(P0.05)。结果表明,高盐突变幅度越大,凡纳滨对虾存活率越低、生长越缓慢,Na+/K+-ATP酶活力升高,渗透调节能力增强,ACP酶活力升高,说明高盐突变激发凡纳滨对虾机体代谢活力。  相似文献

Light attenuation (Kd) of photosynthetically active radiation (PAR) by chromophoric dissolved organic matter (CDOM), total suspended solids (TSS) and chlorophyll a (Chl a) were measured at nine stations along an estuarine gradient in the Swan River, Western Australia, over 15 months. There were strong spatial gradients associated with the marine-freshwater transition along the 32 km of estuary sampled, as well as seasonal gradients mainly associated with rainfall, 80% of which occurs between May and September. CDOM absorbances at 440 nm reached a maximum of 10.9 m−1 with the freshwater inflow but concentrations of suspended matter remained low throughout the sampling period (1.0–21.0 mg l−1) under the diurnal tides of the estuary. CDOM was the dominant constituent of Kd and a stepwise multiple regression showed that 66% (p < 0.0001) of the variation in Kd can be explained by CDOM and an additional 8% (p < 0.0001) by TSS. As a consequence of this result, analysis into the influence of river discharge rates on CDOM absorbance levels was examined for 2002 using data collected during this study, and for 2000 and 2001 using historical dissolved organic carbon (DOC) and river discharge data. The outcome of this analysis infers that greater river discharge rates result in increased CDOM absorbances in the Swan River estuary.  相似文献

通过对比鸭绿江河口不同地区柱状样的粒度参数及敏感组分变化规律,分析了河口不同地区沉积动力环境和沉积特征的差异;结合代表性柱状样的210Pb测年,分析了柱状样在不同时期自然和人类活动影响下的沉积特征变化。结果表明,西水道主要受潮流作用而含有泥沙互层,粒度参数变化小;西岸潮滩受潮流和波浪作用含有贝壳碎屑,并在顶部出现粗化层。近几十年来,鸭绿江河口地区沉积特征的变化主要由于人类活动对水沙入海通量的影响:1941年以前柱状样的波动沉积特征反映了河流山溪性特征;1941—1970(1980)年随水沙入海通量减少而呈现砂含量降低、粉砂增加的特征;1970(1980)—1990年由于水沙入海通量变化较小,而呈现沉积物组成和敏感组分保持稳定的特征;但1990年后随水沙入海通量的进一步降低,邻近河口区域的沉积物出现明显的粗化。  相似文献

大菱鲆仔稚鱼期消化酶及碱性磷酸酶活性的变化   总被引:11,自引:2,他引:11  
针对海水鱼类仔稚鱼期高死亡率的现状,本实验采集不同发育阶段的大菱鲆,分析测定了酸、碱性蛋白酶、淀粉酶和脂肪酶以及消化道碱性磷酸酶的活性变化,以期为提高大菱鲆仔稚鱼期的成活率和生长率以及优化饵料配方提供理论依据。结果表明,大菱鲆初孵仔鱼体内就已具有碱性蛋白酶活性,而酸性蛋白酶活性在15日龄左右胃腺出现时才测得。蛋白酶活性的3个极低值分别出现于仔鱼开口期,卵黄囊完全消失期和仔鱼向稚鱼的转变期。早期仔鱼体内表现出较强的淀粉酶活性。5日龄时其活性急剧下降,并一直保持较低的比活力水平。脂肪酶活性在仔稚鱼期一直很弱,进入幼鱼期后比活力迅速升高。碱性磷酸酶活性从初孵到50日龄左右始终保持较小幅度的上升,直到60日龄变态完成时才出现大幅度的升高。大菱鲆仔稚鱼发育过程中,消化道中各种酶活性变化显著,仔稚鱼变态完成后,蛋白酶、脂肪酶和碱性磷酸酶活性显著增强,这标志着大菱鲆由稚鱼期向幼鱼期的转变及消化机能的完善。  相似文献

Modelling sedimentation rates within an estuary over the time scale of years to decades is a difficult undertaking. The complex nature of sediment transport and the compounding errors associated with making predictions over longer time-scales introduce a high degree of uncertainty when predicting the fate of catchment-derived sediments. In this paper a methodology is presented that links catchment and estuary models that simulate the runoff of sediment from catchments and its subsequent dispersal within the estuary to provide estimates of annual sedimentation rates within the estuary. The models are calibrated against short-term field data and the methodology is validated against sedimentation rates obtained from sediment cores.The catchment of the Mahurangi Estuary delivers between 3800 and 39,000 tonnes/yr with an average load of just over 15,000 tonnes/yr being delivered to the estuary. Data from sediment cores show that over 80% of this load is deposited within the estuary resulting in sedimentation rates of 25 mm/yr in the upper estuary and less than 5 mm/yr in the lower sections of the estuary. The methodology predicts these rates of sedimentation across a range of sub environments within the estuary.  相似文献

河口近岸海域潮流不对称对于物质输运、地形地貌变化有重要作用,因此研究河口近岸海域潮流不对称具有重要的意义。从统计学角度出发推导了一种潮流不对称的计算方法,并将该方法应用于全日潮类型的北仑河口。研究认为:从三阶原点矩出发推导的计算方法能够合理的量化潮流不对称;潮流不对称的方向由分潮的相对相位决定,不同分潮对潮流不对称的影响大小由其相对振幅和相对相位共同决定;在正规全日潮海区,三阶原点矩即可满足对潮流不对称的计算。北仑河口潮流表现为落潮流速占主导,潮流不对称空间上表现为口门强于外海,浅滩强于深槽。  相似文献

以哈维氏弧菌(Vibrio harveyi)对条纹斑竹鲨(Chiloscyllium plagiosum)进行病原接种试验,分别在4、8、12、24、48、72、96h后测定肝脏、脾脏、鳃及血清的酸性磷酸酶(acid phosphatase,ACP)、碱性磷酸酶(alkaline phosphatase,AKP)、超氧化物歧化酶(superoxide dismutase,SOD)和血清中溶菌酶(lysozyme,LSZ)的活力.实验结果表明:对照组条纹斑竹鲨的这4种酶的活力呈现出明显的组织差异性,血清中SOD活力为124.32 U/cm3,显著高于其他3种酶的活力;其他组织中3种酶的活力由高到低依次为SOD、ACP、AKP.在实验组条纹斑竹鲨被感染4~72h期间,血清中SOD活力明显下降,ACP活力持续下降,感染96h后除LSZ外,其他3种酶的活力都呈回升趋势;其他组织中ACP与AKP活力变化均为被感染4h后下降,12h后明显回升,SOD则在被感染4h后活力上升,而后下降,96h后回升.这4种酶的活力的变化主要是应激作用与非特异性免疫防御作用共同作用导致的结果.  相似文献

采用生理生化及分子生物学方法,对刚植片的三角帆蚌进行了为期57天的研究,探讨三角帆蚌珍珠形成初期钙的代谢特征。实验每周取样一次,分别测定了珍珠囊和珍珠的直径,珍珠囊和珍珠的重量及水温,外套膜、鳃、围心腔三种组织的钙含量,酸、碱性磷酸酶的比活力,以及外套膜中钙调素基因的表达量。结果表明,植片后三角帆蚌的珍珠囊及珍珠在第22天和43天各出现一个快速生长期。其外套膜和鳃中的酸性磷酸酶比活力第8天升到最高,随后逐渐下降;外套膜中的碱性磷酸酶比活力第15天达到最高,第29天后开始快速下降;鳃和围心腔中的碱性磷酸酶比活力第15天后才开始上升,第29天达到最高。三种组织的钙含量总体呈现上升趋势,其中鳃的钙含量最高并在第43天达到最高值。植片后三角帆蚌外套膜中的钙调素RNA表达量明显增强,第8天和43天的表达量尤其显著。三角帆蚌珍珠的形成主要依赖于鳃和外套膜对钙的代谢,在此过程中酸、碱性磷酸酶以及钙调素起着重要的作用。可能依照钙调素基因上调表达、酸性和碱性磷酸酶激活、鳃和外套膜钙含量升高、珍珠生长的顺序来进行,植片后的第8天、15天和43天是珍珠形成的关键期。  相似文献
