利用辽宁大连的紫贻贝(Mytilus galloprovincialis)与福建宁德的厚壳贻贝(M.coruscus)为亲本,采用2×2双列杂交设计,构建了紫贻贝自交组(M.galloprovincialis♀×M.galloprovincialis♂)、正交组(M.galloprovincialis♀×M.coruscus♂)、反交组(M.coruscus♀×M.galloprovincialis♂)与厚壳贻贝自交组(M.coruscus♀×M.coruscus♂)4个遗传组合,比较分析了受精率、孵化率、子代的成活率、生长数据等,探讨了紫贻贝与厚壳贻贝种间杂交的可行性,同时为贻贝良种选育提供参考。结果表明,自交组与反交组的受精强度具有不对称性,正交组与自交组受精率无差异,反交组的受精率及孵化率显著低于正交组与自交组(P0.05),且浮游幼虫不能正常存活;正交组子代具有生长优势,第9、11、13天的浮游幼虫及第78天的稚贝壳长均显著大于自交组(P0.05);正交组与紫贻贝自交组在整个幼虫阶段存活率无显著差异,在壳顶幼虫期显著高于厚壳贻贝自交组(P0.05)。  相似文献

为了研究紫贻贝与厚壳贻贝种间杂交的核相变化,并评估其杂交优势,本实验以两种贻贝亲本为材料建立了4个交配组合,分别为:紫贻贝自交组EE(Mytilus edulis♀×M.edulis♂)、厚壳贻贝自交组CC(Mytilus coruscus♀×M.coruscus♂)、正交组EC(M.edulis♀×M.coruscus♂)和反交组CE(M.coruscus♀×M.edulis♂)。通过Hoechst 33258染色、荧光显微观察的方法,对4个交配组从受精、减数分裂到卵裂的核相变化过程进行了观察,并对孵化后幼虫到稚贝期的生长情况进行了记录。结果表明:正反交组的受精细胞学过程与自交组较为相似,但第二极体排出时间延长。对各实验组子一代早期的生长和存活研究表明:浮游阶段,杂交组幼虫生长优势不明显;附着变态后,正交组EC在壳长及壳高等生长指标的杂交优势表现明显,且正交组(EC)大于反交组(CE)。自交组的受精率、孵化率、成活率均高于杂交组。  相似文献

为研究东海区厚壳贻贝(Mytilus coruscus)遗传多样性,以厚壳贻贝F mtDNA D-loop为标记,对浙江省舟山嵊山岛、宁波渔山岛、温州南麂岛、福建省宁德湾和莆田南日岛五个海区的厚壳贻贝群体进行了遗传分析。结果表明,厚壳贻贝各群体的遗传多样性差异不明显,在5个群体中,宁德群体的遗传多样性相对最丰富;将5个群体作为一个整体时,呈现出较高的单倍型多样性和较低的核苷酸多样性。对厚壳贻贝5个群体间的遗传分化系数(Fst)和基因流(Nm)进行检测,结果显示群体间Fst值都很低,但Nm值都很高(Nm绝对值1),表明5个群体间存在丰富的基因交流。但宁德群体与浙江沿海的3个群体(嵊山、渔山、温州)的Fst值相对较高,且差异显著(P0.5),表明宁德群体与这3个群体间出现遗传分化。本研究旨在为海洋经济贝类资源的保护管理提供理论依据。  相似文献

以筛选的2个可鉴别的地中海贻贝(Mytilus galloprovincialis)、厚壳贻贝(M.coruscus)及其杂交后代的PCR标记(扩增核基因Glu-5’特异片段PCR标记Me14/Me17和延伸因子1α第一个内含子区段PCR标记引物EFbisF/EFbisR),对舟山海域3个贻贝主产区(马鞍列岛海域、浪岗山列岛海域、中街山列岛海域)采集的贻贝样本的种类进行鉴别.研究结果显示:100个马鞍列岛海域贻贝个体和30个中街山列岛海域贻贝个体均为厚壳贻贝;而54个浪岗山列岛海域贻贝样本中,16个个体(29.6%)为厚壳贻贝,38个个体(70.4%)为地中海贻贝;3个群体中均未检测到2种贻贝的杂交个体.因此,推测舟山海域地中海贻贝自然分布仅局限于局部海区,但在适宜的海区地中海贻贝可取代厚壳贻贝成为贻贝床的优势物种,舟山海域贻贝床的动态变化值得进一步关注;贻贝样本中没有检测到地中海贻贝与厚壳贻贝的杂交个体,表明调查海域可能没有杂交贻贝分布或数量极少,地中海贻贝通过杂交、基因渐渗污染厚壳贻贝基因组的风险较小.  相似文献

基于线粒体DNA COI基因序列分析了浙江和福建沿海的厚壳贻贝Mytilus coruscus群体遗传结构及遗传多样性现状。研究结果显示:在长度为564bp的COI基因片段上,厚壳贻贝的5个群体均呈现较高的单倍型多样度(0.867±0.129~1.000士0.177)和较低的核苷酸多样度(0.007±0.005~0.011±0.008);邻接关系树显示,5个群体的单倍型相互混杂,仅有1个小的分支存在;厚壳贻贝单倍型网络关系图呈星状结构,存在1个与系统树相对应的分支;不同群体间的F_(ST)值较小,且统计检验的结果均不显著,表明浙江、福建沿岸的厚壳贻贝群体问在COI基因上存在同质性;Tajima's D和Fu's F_s中性检验的结果(Tajima's D=-1.828,P=0.017;Fs=-22.954,P=0.000)都显著偏离中性,提示厚壳贻贝经历了群体扩张。  相似文献

以壳长SL、壳宽SW、壳高SH(BD)、OA(壳顶至韧带末端的直线距离)、OB(壳顶至壳背面最高点的直线距离)、OC(壳顶至壳后端最远点的直线距离)、OD(壳顶至壳高性状在腹缘的落点的直线距离)、AB(韧带末端至壳背缘最高点的直线距离)、BC(壳背缘最高点至壳后端最远点的直线距离)、CD(壳后端最远点至壳高性状在腹缘的落点的直线距离)为贝体框架变量,采用多元分析方法系统比较了嵊泗列岛海域厚壳贻贝、紫贻贝和"杂交贻贝"贝体框架特征的差异,结果表明:(1)在所涉9项贝体框架特征指标中,紫贻贝与厚壳贻贝间无显著差异的指标仅为L5(OC/SL)和L7(AB/SL)(P0.05),而"杂交贻贝"各项指标则均与厚壳贻贝和紫贻贝具显著差异(P0.05),厚壳贻贝和紫贻贝变异系数大于10%的指标均仅为L7(AB/SL),而"杂交贻贝"则仅为L3(OA/SL);(2)厚壳贻贝与紫贻贝间的欧氏距离最短(P0.05),仅为0.160;厚壳贻贝与"杂交贻贝"间和紫贻贝与"杂交贻贝"间的欧氏距离相近(P0.05),分别为0.452和0.418;(3)经主成分分析,提取到的3个特征值均大于1的主成分,累计贡献率达82.928%,其中第一主成分、第二主成分、第三主成分可依次归为与滤食功能区水平剖面占比相关的贝体框架因子,与消化功能区水平剖面占比相关的贝体框架因子,和与消化功能区垂直剖面占比相关的贝体框架因子,通过第一主成分仅能较清晰地区分厚壳贻贝和"杂交贻贝";(4)采用逐步判别法,以判别贡献率较大的L1(SW/SL)、L3(OA/SL)、L4(OB/SL)、L5(OC/SL)、L6(OD/SL)和L7(AB/SL)为自变量,所建Fisher分类函数方程组可较清晰区分厚壳贻贝、紫贻贝和"杂交贻贝",三者的判别准确率依次为94.6%、94.6%和100%,综合判别准确率为96.4%。  相似文献

采用16S rRNA基因测序技术,对我国东南沿海4个地理群体的厚壳贻贝遗传结构及遗传变异进行研究。通过对4个厚壳贻贝群体共83个个体的线粒体16S rRNA基因进行测序,获得1个长度为305bp的同源序列,共检测到150个多态位点,多态位点比例达49.18%。83个个体中共检测到28个单倍型,单倍型多样性指数(Hd)为0.810,核苷酸多样性指数(Pi)为0.09602,平均核苷酸差异数(K)达27.846。结果表明,我国东南沿海厚壳贻贝群体具有较高的遗传多样性水平。遗传结构检测结果表明,舟山群体、温州群体、宁德群体间的遗传距离小,遗传分化系数(Fst)为-0.0141—0.0059之间,群体内部无显著分化(P>0.05),而福州群体与其它群体间遗传距离较大,为0.215—0.217之间,遗传分化系数(Fst)也较大,为0.6217—0.6319之间,存在极显著的遗传分化(P<0.001)。  相似文献

以嵊泗列岛海域2+龄厚壳贻贝同生群养殖个体、同域生长的1+龄紫贻贝同生群养殖个体以及混于厚壳贻贝和紫贻贝养殖筏架中的"杂交贻贝"为实验对象,以壳宽SW、壳长SL、壳高SH(BD)、OA(壳顶至韧带末端的直线距离)、OB(壳顶至壳背面最高点的直线距离)、OC(壳顶至壳后端最远点的直线距离)、OD(壳顶至壳高性状在腹缘的落点的直线距离)、AB(韧带末端至壳背缘最高点的直线距离)、BC(壳背缘最高点至壳后端最远点的直线距离)、CD(壳后端最远点至壳高性状在腹缘的落点的直线距离)为贝体框架变量,采用多元分析方法系统比较了三者贝体框架性状对壳重的影响效应。结果表明:(1)3种实验贝壳重的变异系数均大于各贝体框架性状,"杂交贻贝"各测定性状的变异系数均为最大,而厚壳贻贝和紫贻贝间则均接近;(2)3种实验贝各贝体框架性状与壳重的相关系数均达到极显著水平(P0.01),三者中仅"杂交贻贝"各贝体框架性状间的相关系数也均达到极显著水平(P0.01);(3)经通径分析,厚壳贻贝被保留的3个贝体框架性状与壳重的复相关指数为0.868,它们对壳重的直接作用呈OCSHSW,紫贻贝被保留的2个贝体框架性状与壳重的复相关指数为0.700,它们对壳重的直接作用呈OCSW,"杂交贻贝"被保留的3个贝体框架性状与壳重的复相关指数为0.931,它们对壳重的直接作用呈SLSHOA;(4)经多元回归分析,得到了用于估算厚壳贻贝、紫贻贝和"杂交贻贝"壳重的回归方程。  相似文献

肠道细菌对厚壳贻贝稚贝附着的作用研究   总被引:1,自引:0,他引:1  
为探讨肠道细菌形成的微生物被膜对贝类附着的作用,从厚壳贻贝(Mytilus coruscus)成体肠道中分离10株肠道细菌,调查厚壳贻贝肠道细菌形成微生物被膜特性,分析微生物被膜密度、细菌种属与厚壳贻贝稚贝附着之间的相互关系。所有肠道细菌均可有效促进稚贝附着。其中,Paracoccus sp.1微生物被膜诱导的稚贝附着率最高(61%),且微生物被膜膜厚较厚,细菌分布较密集;仅5株肠道细菌Roseovarius sp.1、Winogradskyella sp.1、Tenacibaculum sp.4、Bacillus sp.5和Nonlabens sp.1的被膜密度与附着显著相关(P0.05)。系统发育分析发现肠道细菌诱导附着率与其种属无关。本研究表明源于厚壳贻贝成贝肠道细菌可以促进其稚贝的附着,说明肠道细菌对厚壳贻贝的生长发育具有一定的作用。本研究结果为深入探讨肠道细菌与厚壳贻贝相互作用和推动该种规模化养殖提供理论依据。  相似文献

A biochemical genetic study of the mussels Perna perna and Choromytilus meridionalis on the west coast of South Africa revealed the presence of an unreported mytilid mussel that had previously been mistaken, because of its shell colour polymorphism, for either P. perna or a hybrid between P. perna and C. meridionalis. The gene products of 19 protein-coding loci in P. perna. C. meridionalis and in the newly recognized mussel were examined by means of horizontal starch-gel electrophoresis. The results showed that there was no allele-frequency overlap between any of these mussel taxa, thus implying little or no genetic relatedness between them. A morphological examination showed that the previously unreported mytilid has a pitted resilial ridge (similar to that of Perna), an anterior adductor muscle (unlike either Perna or Choromytilus), and an undivided posterior foot retractor-muscle scar (unlike Perna). Such features are diagnostic of the genus Mytilus. The South African Mytilus has morphological traits that are more characteristic of M. galloprovincialis of the Mediterranean Sea than of the more cosmopolitan M. edulis. which occurs in the northern and southern Pacific and Atlantic oceans. A large heterozygosity in Mytilus sp. argues against a recent dispersal from the Mediterranean Sea via a small founder population. Rather, the presence of Mytilus sp. in South Africa may represent a relict population of a wider geographic distribution of M. galloprovincialis resulting from Pleistocene cooling. However, the warmer water at lower latitudes could have still prevented dispersal of M. edulis, a species adapted to colder waters.  相似文献

The genetic constitution of mussels (Mytilus spp.) was studied by means of three nuclear (Me 15/16, EF-bis, ITS) and one mtDNA (ND2-COIII) marker on a large European scale. In addition to a sharp cline between Atlantic and Mediterranean M. galloprovincialis, we observed a clear genetic distinction between the Black Sea and Mediterranean populations and a higher incidence of M. trossulus than reported so far in northern European populations. The frequency of M. galloprovincialis nuclear alleles was high along the Iberian Peninsula and decreased abruptly along the French coasts with a high frequency of M. edulis alleles in the Bay of Biscay, The Netherlands, Germany, Iceland, Barents and White Seas, and with little evidence of introgression between the two taxa. M. trossulus alleles were observed in the Baltic Sea and Danish Straits as expected. In addition, occurrence of M. trossulus alleles in cold waters of Iceland, Barents Sea and White Sea is reported for the first time.  相似文献

The blue mussels Mytilus edulis and Mytilus trossulus occur sympatrically and are able to hybridize in populations on the eastern coast of Newfoundland, Canada, presenting an opportunity to study their aggregational behavior. Aggregation behavior may therefore provide insight into post‐settlement interactions and pre‐zygotic reproductive isolation between the species. Three treatments were designed using M. edulis and M. trossulus to investigate their intraspecific and interspecific spatial distribution patterns. With Ripley’s K‐function and Monte Carlo simulation analysis, we found that in the single‐species treatment, M. edulis aggregated significantly but not M. trossulus. Based on results of two‐way ANOVAs, both the number of aggregations and the moving distance were significantly affected by the treatments (single‐species or mixed‐species treatment) and times (24, 48, 72 and 96 h). In further pairwise comparisons using Tukey’s test, M. edulis aggregated differently with or without M. trossulus occupying the same tank, suggesting that the aggregational behavior of M. edulis could be driven by species‐specific chemical cues. The result that M. edulis aggregates intraspecifically may increase the probability of intraspecific fertilization of the spawned gametes and thus function as a pre‐zygotic reproductive isolation mechanism maintaining the blue mussel hybrid zone.  相似文献

拟相手蟹属因其形态极其相似成为相手蟹科分类中最有疑问的一个属。通过对中国沿海近亲拟相手蟹Parasesarma affine、斑点拟相手蟹P. pictum、三栉拟相手蟹P. tripectinis、P. ungulatum及褶痕拟相手蟹P. plicatum 5种拟相手蟹的形态对比发现,可从它们的螯足形状(包括螯足掌节背面的梳状栉,可动指背面突起数目)及雄性第一腹肢形状对其进行区分。对其线粒体16S rRNA基因序列进行分子系统发育分析,结果表明5种拟相手蟹之间的遗传距离为1.9%~9.3%,达到了种间差异水平;构建的系统发育树显示近亲拟相手蟹与褶痕拟相手蟹汇聚成一支,随后与P. ungulatum聚在一起,斑点拟相手蟹与三栉拟相手蟹汇聚成独立支系。形态和分子证据均支持5种拟相手蟹分别为独立有效物种。  相似文献

Ecosystem engineers such as mussels may affect strongly both the structure of benthic assemblages and the ecosystem functioning. The black-pygmy mussel Limnoperna securis is an invasive species that is spreading along the Galician coast (NW Spain). Its current distribution overlaps with the distribution of the commercial native mussel species Mytilus galloprovincialis, but only in the inner part of two southern Galician rias. Here, we analysed the assemblages associated with clumps of the two mussel species and evaluated if the invasive species increased complexity of habitat. To measure complexity of clumps we used a new method modified from the “chain and tape” method. Results showed that the identity of the mussel influenced macrofaunal assemblages, but not meiofauna. L. securis increased the complexity of clumps, and such complexity explained a high percentage of variability of macrofauna. The shift in dominance from M. galloprovincialis to L. securis may alter habitat structure and complexity, affecting the macrofaunal assemblages with unpredictable consequences on trophic web relations.  相似文献

Up to 12 marine mollusc and three crab species are thought to have been deliberately or accidentally introduced to South Africa. Of these, only the Mediterranean mussel Mytilus galloprovincialis and the European shore crab Carcinus meanas have become invasive. M. galloprovincialis, probably introduced in the late 1970s, is already the dominant mussel throughout the Cape west coast. As a result, indigenous intertidal mussels Aulacomya ater have been largely displaced, mussel standing stock has increased and the upshore limit of mussel beds has become considerably elevated. Implications include competitive exclusion of large limpets but enhanced recruitment of juvenile limpets, increased habitat availability for mussel infauna, reduction of habitat for algal infauna and enhanced food availability for predators, particularly terrestrial species. C. maenas was first recorded from Table Bay Docks in 1983 and currently ranges from Camps Bay to Saldanha Bay. Although it is a voracious predator, it appears unable to colonize wave-swept shores, so is unlikely to displace indigenous crabs, or to severely impact prey species on the open coast. Valuable conservation areas and mariculture sites in sheltered lagoonal areas are, however, threatened by it.  相似文献

贻贝隶属于软体动物门(Mollusca)、双壳纲(Bivalvia)、翼形亚纲(Pteriomorphia)、贻贝目(Mytilida)、贻贝超科(Mytiloidea),大约有400种贻贝分布在世界各地,可适应淡水、潮间带至深海多种生境。本实验以贻贝科6亚科12属28种中国沿海常见贻贝的28SrDNA为目的片段,构建系统发育进化树,运用最大似然法和贝叶斯推演法分析了贻贝科的系统发生,并追踪贻贝科物种系统演化历史。结果显示:贻贝亚科Mytilinae、偏顶蛤亚科Modiolinae、石蛏亚科Lithophaginae均非单系群。在属阶元,深海偏顶蛤属Bathymodiolus、贻贝属Mytilus和股贻贝属Perna为单系群。本研究发现应接受将原隔贻贝属Septifer分为Septifer属和Mytilisepta属的分类提议;应接受将原石蛏属Lithophaga中的膜石蛏亚属Leiosolenus提升至属的地位的分类提议。此外,短齿蛤属Brachidontes的单系性不被支持,刻缘短齿蛤Brachidontessetiger并未与短齿蛤属其他物种在系统发育树上聚拢,亲缘关系较远,为不同属物种,建议恢复刻缘短齿蛤原属名Volsella (Dunker, 1857)。  相似文献

CYP1A-immunopositive protein can be elevated in response to planar PAHs and PCBs in Mytilus sp. digestive gland whilst CYP3A-immunopositive protein has been associated with testosterone 6β-hydroxylation in fish. Levels of CYP1A- and CYP3A-immunopositive protein were determined in Mytilus galloprovincialis digestive gland microsomes collected from 12 sites in the Mediterranean Sea during May and September 2001. CYP1A-immunopositive protein was significantly highest at contaminated sites whilst CYP3A-immunopositive protein was significantly lowest. A weak negative correlation (r2=0.21) was seen between CYP1A- and CYP3A-immunopositive protein. Little evidence of differences at the different sampling times was observed. These results confirm previous work indicating elevation of CYP1A-immunopositive protein in Mytilus sp. digestive gland at contaminated sites. Further study is required to characterise CYP3A-like expression in Mytilus and to elucidate the consequences of possible CYP3A-like down-regulation at contaminated sites.  相似文献

From June 1984 to May 1985, monthly and seasonal panel tests of fouling organisms were carried out on rafts at Tuoji Island. Seventy - three species were obtained. The occurring frequencies of the following species are all at least 30%: Ectocarpus confervoides, Polysiphonia urceolata, Tubularia mesembryanthmum, Cfytia edwardsi, Tubulipora pulchra, Celleporella hyalina, Mytilus edulis, Hiatella arctica, Caprella acanthogaster, Corophium crassicome, Jassa cf.falcata and Podocerus cf . inconspicuus. Large or calcareous species are not dominant ones. The wet weight is comparatively light, the yearly wet weight being 9.12 kg m-2. Attaching seasons of major species are from June to October and no bio - attachment occurs between January and April. Fouling organisms at Tuoji Island differ rather greatly from those in the harbours along the continental coast, showing the charcteristics of fouling organisms in the northern island harbours of China.  相似文献

A second marine mussel is shown to have invaded South Africa's west coast. Molecular techniques, based on intraspecific gene sequence divergences, prove its identity as Semimytilus algosus, a member of the family Mytilidae, native to Chile. The identity of an older introduced population found in Namibia is also confirmed. The present geographic range of S. algosus in South Africa extends some 500 km, from Groenriviersmond in the north to Bloubergstrand in the south. Together with Mytilus galloprovincialis, another previously established invasive mussel, S. algosus has become a dominant intertidal organism on wave-exposed rocky shores across this region. Both species are now much more abundant intertidally than either of the indigenous mussels Aulacomya ater and Choromytilus meridionalis, which have become largely confined to sublittoral and sand-inundated habitats respectively. The two invasive mussels display strong spatial segregation, with M. galloprovincialis dominating the midshore and S. algosus blanketing the lower shore. Through a combination of its small size and high abundance, S. algosus is likely to greatly increase food availability for a range of intertidal predators, many of which cannot consume mussels above a threshold size.  相似文献
