Tilapia is a common fish species inhabiting inland waters and estuarine regions in Hong Kong and Southeast Asia, and useful for bio-monitoring of metal pollution. Metallothionein (MT) gene expression in fish tissues has been useful to sub-lethal risk assessment as biomarker of exposure to metal ions in fishes inhabiting metal contaminated area. To investigate metal inductions of Tilapia MT gene expression in vivo, Tilapias were injected with different concentrations of heavy metals and tissues were then removed for quantitative PCR assay using mimic PCR methods. All of the metal ions tested (Cu2+, Cd2+, Hg2+, Ni2+, Pb2+ and Zn2+) were able to induce hepatic MT mRNA levels. Renal MT mRNA levels of Cd2+ and Zn2+ treated fish was not induced with significant fold induction, however MT mRNA levels in gills were sensitive to the administrations of these metal ions. These data indicated that Tilapia MT mRNA levels in gills and liver are sensitive biomarker of exposure to various metal ions.  相似文献

Most indigenous New Zealand freshwater fish are benthic, and use substrate elements for shelter, cover, and nesting sites. Sedimentation can reduce the amount of interstitial space provided by the substrate, thus reducing the amount of available cover and potentially influencing fish abundance. To test the effects of sedimentation on fish density, upland bullies (Gobiomorphus breviceps Stokell) were introduced into experimental channels with different substrate treatments. After an acclimatisation period, they were allowed to emigrate. The maximum fish density and relative habitat suitability was then determined by the number of fish remaining after 6 days. Bully numbers declined by over 50% after 6 days in the trials with no cover, whereas with 20 cobbles providing cover for 40 bullies, numbers reduced by only 10%. When sufficient fine sediment was added to fill the spaces under the cobbles, bully numbers reduced by more than 60%, regardless of the number of cobbles. However, when cobbles were raised above the fine sediment, the number of bullies remaining in the channels was similar to the number remaining with cobbles and no sediment. The overall effect of high levels of sedimentation on bully density was similar to that of providing no cover, with the effect diminishing as the level of sedimentation decreased.  相似文献

The objective of this study was conducted to identify the possibility of using Chironomus metallothionein (MT) and vitellogenin (VTG) as biomarkers of stress caused by endocrinedisrupting chemicals (EDCs), heavy metals, herbicides and veterinary antibiotics. We characterized the MT and VTG cDNA in Chironomus riparius and evaluated their mRNA expression profiles following exposure to different environmental pollutants. The gene expression analysis showed that the MT mRNA levels increased significantly after long-term exposure to cadmium (Cd), copper (Cu), Lead (Pb), di(2-ethylhexyl) phthalate (DEHP), and 2,4-dichlorophenoxyacetic acid (2,4-D). Moreover, the VTG mRNA expression increased significantly in C. riparius larvae exposed to BPA, NP, DEHP, Cd, 2,4-D and fenbendazole. Evaluation of the long-term effects of environmental pollutants revealed up regulation of Chironomus MT mRNA in response to DEHP exposure among EDCs, and the level of the VTG mRNA was increased significantly following treatment with Cd and herbicide 2,4-D at all concentrations in a dose-dependent manner. These results indicate that VTG could be used as a potential biomarker of herbicide and Cd as well as EDCs, while MT was a potential biomarker of heavy metals such as Cd, Cu, and Pb in aquatic environments.  相似文献

This study compared the longitudinal size distribution of bluegill bullies (Gobiomorphus hubbsi) from Hutt River with existing data on the size distributions of bluegill bullies and torrentfish (Cheimarrichthys fosteri) from Rakaia River, New Zealand. All populations had longitudinal trajectories that showed some increase in size with distance upstream. This increase in size appeared to primarily reflect the influence of amphidromous life styles, where juveniles diffuse upstream from the sea. However, a quantile regression analysis revealed differences in growth and migration rate between the two bluegill bully populations; bluegill bullies from Rakaia River grew at a slower rate and showed variation in migration rate within their population that was not evident in the Hutt River population. Null models were also generated for each population to test for the presence of all size classes of fish in the lower reaches of each river. Both bluegill bully populations showed a significant absence of the largest size classes in the lower reaches and the size trajectories differed significantly from the null models. This difference suggests that all individual bluegill bullies continuously move upstream throughout their lives. In comparison, the size distribution of torrentfish closely resembled the null model, indicating that some individuals did not migrate upstream to the same extent as others.  相似文献

Several water quality parameters, marginal vegetation, and the fish community in Lake Ellesmere, a large, shallow, turbid lake situated on the east coast of the South Island, New Zealand, were sampled along selected transects in late summer to determine species spatial patterns. Thirteen species offish were caught, with seven of them comprising major populations, although only four of these were adequately sampled by the method used—they included inanga (Galaxias maculatus), common smelt {Retropinna retropinna), shortfinned eel (Anguilla australis), and common bully (Gobiomorphus cotidianus). Inanga were almost exclusively confined to areas on the western side of the lake which has a convoluted shoreline with a variety of small embayments with scattered vegetation along the margins. Common smelt occupied both marginal and offshore areas, but they too were more abundant along the margins particularly on the western side. Both the common bully and shortfinned eel were widespread and abundant in the lake, although their abundance and biomass were higher within the vicinity of the lake outlet, major inflowing tributaries, and other marginal habitats. All three species of flounders of the genus Rhombosolea were poorly represented in our samples. However, in commercial catches they were reasonably abundant, with the sand flounder (R. plebeia) and yellowbelly (R. leporina) being more common in offshore than inshore catches, whereas the black flounder (R. retiaria) showed no difference in the catches between areas. Our overall findings are consistent with the hypothesis that bottom‐dwelling species (eels, bullies, flounders) are more widely distributed than free‐swimming forms (inanga, common smelt), which were largely confined to areas sheltered from prevailing winds. The highly turbid nature of the lake, primarily because of re‐suspension of fine bottom sediments by frequent winds, is considered an important factor in the distribution of the fish fauna.  相似文献

The acute toxicity of seawater solutions of copper, zinc, lead, cadmium and mercury to two species of free-living marine nematodes was studied. Enoplus brevis (Bastian) is a euryhaline species inhabitating intertidal estuarinemud. The closely related Enoplus communis (Bastian) is stenohaline and may be found as a commensal in the byssus threads of seed mussels. Specimens of both species were collected from two sites on the Northeast coast of England showing different levels of pollution by heavy metals. Both species were more susceptible to copper, lead and mercury than to zinc and cadmium, but the order of toxicity of the metals changed with exposure concentration. The euryhaline species survived better than the strictly marine species and there is a suggestion that animals from a polluted site were more tolerant of elevated of heavy metals.  相似文献

The blue crab, Callinectes sapidus, is distributed along the east coast of the United States from Cape Cod, Mass., through the Gulf of Mexico, including both relatively unpolluted coastal areas and estuaries contaminated with trace metals. Cadmium is of particular concern because it is concentrated in the digestive glands of blue crabs and can be passed on to consumer organisms. Tissue concentrations and partitioning of trace metals from crabs exposed in the laboratory to 10 ppb dissolved cadmium for 40 days were compared with blue crabs collected from two locations on the Hudson River, NY, Foundry Cove and Haverstraw Bay, Foundry Cove and Haverstraw Bay, both of which have elevated trace metal levels relative to estuarine areas near Beaufort, NC. Crab digestive glands, gills and muscle were removed and analyzed for total cadmium, copper, zinc and nickel concentrations using acid digestion and atomic absorption spectrophotometry, and metal-binding (metallothionein-like) proteins were determined by gel-filtration chromatography. In crabs exposed to cadmium in the laboratory, the cytosolic partitioning was similar to previous investigations at our laboratory where higher levels of cadmium (100 ppb) and shorter exposure times (14 days) were used. The similarity in cadmium partitioning from these two separate experiments indicates dose independence. In crabs from polluted environments the digestive glands contained the highest concentrations of trace metals. Chromatograms of the cytosol from the digestive glands and gills from both field and laboratory exposed crabs showed similar distributions of cadmium, copper and zinc. The gills of both field and laboratory-exposed crabs had metal-binding proteins that contained mostly cadmium, and the digestive glands had metallothionein-like proteins that contained cadmium, copper and zinc. Estimated molecular weights for these proteins were similar to the metallothioneins found in other crustaceans and mammals.  相似文献

To establish a base line against which future pollution may be measured, eight common commercial species of New Zealand sea fish were analysed for cadmium, chromium, copper, iron, lead, manganese, nickel, and zinc. One sample of edible muscle tissue was analysed for each of 70 samples of each species. The internal organs of up to five specimens of each species were also analysed. The fish studied were: kahawai, Arripis trutta, trevally, Caranx lutescens, tarakihi, Cheilodactylis macropterus, snapper, Chrysophrys auratus, moki, Latridopsis ciliaris, hapuku, Polyprion oxygeneios, kingfish, Seriola grandis and gurnard, Trigla kumu.

Although, none of the edible parts of the fish appeared to have trace element levels likely to be a public health problem (cadmium 0.002–0.02 ppm, chromium 0.01–0.04 ppm, copper 0.04–0.95 ppm, iron 0.9–13.5 ppm, lead ‘ 0.14–0.95 ppm, manganese 0.04–2.00 ppm, nickel 0.01–0.08 ppm, zinc 2.0–36.0 ppm), some of the organs (particularly the liver) had relatively high concentrations of elements such as cadmium (up to 54 ppm). Only if some industry were to seek to exploit internal organs of fish for human consumption would such levels become important. There was some evidence for a relationship between trace element concentrations and fish size for copper in kingfish and snapper, iron in hapuku, manganese in gurnard, and for zinc in kingfish and tarakihi. Some element pairs such as copper and zinc, iron and manganese, appeared to have concentrations which were mutually related. It was assumed that the elemental concentrations reported represent natural levels and are not the result of significant man‐made pollution because there are no major industries or large urban centres adjacent to the fishing grounds.  相似文献

Rainbow vent field is one of the most metal‐contaminated hydrothermal sites on the Mid‐Atlantic Ridge near the Azores region. Two hydrothermal shrimp species dominate the fauna at the Rainbow site along with the mussel Bathymodiolus azoricus. Although the levels of essential and non‐essential metals in these shrimps have been studied, the biological consequences of a metal‐rich environment are still largely unknown. Therefore, the aim of this study was to determine the levels of metal‐binding proteins – metallothioneins (MT) and the activities of antioxidant enzymes – superoxide dismutase, catalase, total glutathione peroxidase and selenium‐dependent glutathione peroxidase in two hydrothermal vent shrimps (Mirocaris fortunata and Rimicaris exoculata) collected from the Rainbow site and to compare them with two coastal shrimps (Palaemon elegans and Palaemonetes varians) from a south Portugal lagoon (Ria Formosa) to evaluate their different adaptation strategies towards metals in their environment. Results show significant differences in MT levels and antioxidant enzymatic activities between vent and coastal shrimps and also between shrimp species collected from the same site. This suggests that biochemical responses in both vent and coastal shrimps are affected not only by the environmental characteristics but also by inter‐specific differences. Nevertheless, these responses apparently confer successful adaptation for survival in a metal‐extreme environment.  相似文献

The diet of brown trout (Salmo trutta) and rainbow trout (S. gairdnerii) was studied in specimens from Lake Benmore, a deep, oligo‐trophic lake in South Island, New Zealand. Between November and July, both species fed mainly on small molluscs (Potamopyrgus antipodarum, Physa sp., and Gyraulus corinna) gleaned from the littoral weed beds. Energy values for the three species of mollusc were determined: P. antipodarum, 6000 J g‐1; G. corinna, 5500 J g‐1; Physa sp., 9800 J g‐1. Potamopyrgus antipodarum yielded little energy to the fish, unless its shell broke during passage through the gut. Physa sp. was the most profitable mollusc, irrespective of shell breakage and Potamopyrgus antipodanim the least profitable. The mean energy value per snail for G. corinna and Physa sp. eaten by rainbow trout was 25–30% less than for snails eaten by brown trout, possibly because rainbow trout ingested empty shells from the sediment surface. Rainbow trout extracted about 20% more energy than brown trout from unbroken shells. In July, 84% of the brown trout switched to predation of common bullies, Gobio‐morphus cotidianus, probably as a result of bully reproductive behaviour. Rainbow trout did not show the same change, apparently because they were feeding in deeper water where few bullies were available. The low‐energy diet and its possible connection with growth rate are discussed.  相似文献

Metallothionein has been assayed in a range of aquatic animal tissues as an indicator of metal exposure. We sequenced chub (Leuciscus cephalus) metallothionein cDNA which showed over 90% homology to common carp, goldfish and stone loach and 77% homology to rainbow trout sequences for metallothionein. We then used the extended primer method to develop an accurate quantitative competitive RT-PCR assay for metallothionein mRNA. RT-PCR was used to measure metallothionein mRNA in feral chub from a range of field sites, with different levels of heavy metal pollution, in the West Midlands, UK. Measurements were complemented by analysis of liver and gill metallothionein protein by capillary electrophoresis. There was no significant difference in the metallothionein protein levels between fish of different rivers and there was no evidence of elevation of mRNA at the sites of highest metal exposure. The level of metal exposure (e.g. zinc, nickel and cadmium each ranging between 15 and 28 microg/l ) at the pH (7.5-8.5) of these rivers appears insufficient to elevate hepatic or gill metallothionein in chub. A lack of elevation of hepatic metallothionein mRNA in chub exposed to zinc, copper and manganese for 24 h and 10 days in the laboratory also suggests a non-responsiveness of this species.  相似文献

Organisms have evolved a cellular response called stress protein response that increases their tolerance in adverse environmental conditions. Well known stress proteins that bind essential and toxic metals are metallothionein (MT). The scallop Mizuhopecten yessoensis is the most interesting organism because it is able to accumulate toxic cadmium in its digestive gland. However, in the tissue of the digestive gland of Mizuhopecten yessoensis MT (metallothioneins) have not been found. Eastern scallops, Mizuhopecten yessoensis, were collected from two locations ?? one clean and one polluted site. The concentrations of cadmium (Cd), copper (Cu) and zinc (Zn) were measured in the digestive gland. There was a significant increase in Cd concentrations in this studied tissue. We found that in the presence of cadmium Mizuhopecten yessoensis can induce high molecular proteins. The results of experiments have shown that Cd-binding ligands have a number of properties similar to MT: acetone and temperature stability; the ability to bind some metals, including Cd, Cu and Zn. Protein chromatography (FPLC, Superosa 12) from the digestive gland of scallop M. yessoensis has shown that cadmium is associated with high molecular weight Cd-binding proteins (72 kDa and 43 kDa). The major cadmium-binding protein 72 kDa is glycoprotein. In experiments we have demonstrated that Cd-binding proteins can be induced when there is cadmium exposure. The results of this study strongly suggest that the far eastern scallop Mizuhopecten yessoensis has a unique and well-developed system for the detoxification of heavy metals and it allows for biochemical systems to be maintained in a relatively stable manner in the presence of heavy metals.  相似文献

Laeonereis acuta (Polychaeta, Nereididae) was collected in an unpolluted (UP) and an polluted (P) site at the Patos Lagoon estuary (Southern Brazil) and maintained under control conditions (UPC and PC, respectively) or exposed to waterborne copper (UPCu and PCu; 500 μg Cu/l), for 48 h. Four groups (aaUPC, aaPC, aaUPCu, and aaPCu) were also pre-exposed for 48 h to ascorbic acid (aa; 0.1 mM) before copper exposure. Histological and morphological alterations, as well as oxygen consumption changes were evaluated. Independently of the sampling site and the pre-exposure to the ascorbic acid, morphological abnormalities were evident in more than 80% of worms exposed to copper. Conspicuous histological changes (coeloma obliteration, cuticle separation from the epidermis, and absence of dorsal vessel) were also observed. In addition, PCu worms showed loss of the digestive epithelium and coiling behavior. Similar oxygen consumption values were observed in control and copper exposed worms.  相似文献

Accumulation of cadmium, copper, lead and zinc in the tissues of Mytilus edulis planulatus was examined under cyclic conditions of exposure in order to establish whether the rate of accumulation of the metal is proportional to the time exposed to the elevated concentrations. When exposed to a single metal, the accumulation of lead and zinc was directly proportional to the exposure time, but that of copper was not. Under simultaneous exposure to all four metals both lead and cadmium were accumulated in direct proportion to the exposure time, while copper and zinc were not. Accumulation of the three metals cadmium, copper and zinc was influenced by the presence of other metals.  相似文献
