南极产低温几丁质酶菌株的筛选及分子鉴定   总被引:2,自引:0,他引:2  
从南极普里兹湾深海底泥样品中分离得到一批嗜冷细菌,对这些嗜冷细菌进行了产几丁质酶菌株的筛选,对其中一株具有较高产酶活性的菌株AC167所分泌的几丁质酶进行了性质分析,并通过16S rDNA序列分析进行了菌株的分子鉴定。该菌株最适生长温度为10℃,最高生长温度为25℃,是典型的嗜冷菌,经16S rDNA序列比较及系统发育分析鉴定为假单胞菌属(Pseudomonas)。该菌株只在低于25℃的条件下分泌几丁质酶,酶的最适反应温度为30°C,属于低温酶。  相似文献

从日晒盐场结晶池中筛选到1株产C_(50)类胡萝卜素的红色极端嗜盐古菌。该菌株为革兰氏阴性菌,短棒状,最适生长盐度为250,最适生长pH为7。表型鉴定方法结合16S rDNA序列分析判定,该菌属于极端嗜盐古菌盐红菌属Halorubrum,命名为Halorubrum sp.Sp1(16S rRNA Genbank登录号为KF697239)。根据紫外-可见光扫描特征光谱,确定该菌株主要色素为C_(50)类胡萝卜素。pH 8时单位细胞色素积累量最大,在盐度150~300范围内随盐度升高,单位细胞色素积累量逐渐降低。  相似文献

从同安盐田土壤中分离到一株中度嗜盐菌TA-4,经过16S rDNA序列分析、形态学和生理生化特征分析,该茵株初步鉴定为盐单胞菌(Halomonas salina).TA-4能在2%-25%NaC1的培养基中生长,最适生长NaC1浓度为6%,最适生长温度为35℃,最适pH为7.0~8.0.在高盐条件下,TA-4能够在细胞内合成四氢嘧啶以维持渗透压的平衡,在15%培养基中其含量达到60.3mg/g细胞干重.渗透冲击试验表明TA-4细胞内的四氢嘧啶含量受培养基中的盐浓度调控,并且在低渗冲击时能够快速分泌到细胞外,在高渗冲击冲剂后的6h内又可以恢复到原来的90%以上.  相似文献

嗜酸氧化亚铁硫杆菌(Acidithiobacillus ferrooxidans,简称A.f)是一种能够以亚铁、单质硫、以及硫化物作为能量来源的化能自养菌,因为此特性,其是生物湿法冶金中主要浸矿菌种之一.本试验中,通过从4个不同来源样品中分离到了4株A.f菌,然后通过改变影响其生长的因素来比较其氧化亚铁以及硫的活性.试验中使用了6种不同条件,其中每个条件下仅改变一个影响因子,这些影响因子包括温度、离子强度、pH、重金属离子浓度、外加离子浓度、硫培养、浸矿培养.通过比较结果,发现不同条件下,4株菌表现均不一样,而活力最强的菌株也不一样.例如,A.f.3菌株在以亚铁为能量来源的时候,一般是活性最强的菌株,而A.f.2一般是活性最差的菌株;但是在用硫和黄铁矿为能量来源的时候,A.f.2的活性不再是最差的,硫培养时,A.f.2是活性最强的菌株.通过比较,人们可以筛选得到一个高活性以及工业应用价值更高的菌株.  相似文献

以萘为唯一碳源和能源从南,中绳海槽深海沉积物中分离得到一株能降解萘的海洋嗜低温细菌Nah-1,测定了该茵的最适生长条件及生长曲线。16S rRNA基因(16SrDNA)序列同源性分析表明该菌属于解环菌属(Cyczocznsticus)。PCR扩增萘降解基因得到目标片段,比对结果表明,相似度最高的基因phnAl来自Cycloclasticus sp.A5,为99%,该基因编码的蛋白是萘双加氧酶大亚基。  相似文献

以从海南三亚近海海水中采集的样品为研究对象,利用平板筛选法筛到一株产酯酶的中度嗜盐菌菌株LYG1-1。通过形态学、生理生化及16S rRNA序列分析,发现该菌株为革兰氏阴性菌,其最适生长温度为30℃,最适生长pH为7.5,最适NaCl浓度为10%。16S rRNA基因序列分析显示,菌株LYG1-1与Halomonas salina亲缘性最近,16S rRNA基因序列相似性为99.72%。在含10%NaCl的0.05mol·L-1Tris-HCl(pH 8.0)缓冲液中,45℃条件下作用,酶活性最高,40℃处理24h仍保持83%的活性。金属离子浓度为5mmol·L-1时,Ca2+、Mg2+对菌株LYG1-1酶活具有明显的激活作用,Hg2+表现出强烈的抑制作用。  相似文献

微生物聚合物聚-?-羟基丁酸酯(PHB)具有良好的生物降解性、生物相容性和热塑性,在环保、医药和农业领域具有广阔的应用前景。高PHB积累菌株的获得及其发酵条件的研究具有很好的实用价值。作者从渤海湾日晒盐场盐田卤水中分离出一株杆状细菌,经16S r DNA序列测序比对,确定为盐杆菌属(Halomonas),命名为Halomonas 100-16-2,其最适生长盐度为50~100,最适生长p H为7~8,属中度嗜盐菌。在酸水解酪蛋白(7.5 g/L)的基础培养基中分别添加0、10、20和30 g/L葡萄糖,研究盐度100条件下不同葡萄糖添加量对Halomonas 100-16-2生长和PHB含量的影响。结果表明,随着葡萄糖浓度的增加,该菌株的生长和PHB积累显著提高。葡萄糖质量浓度为30 g/L时,细胞干质量达8.60 g/L,PHB含量达到5.57 g/L(即达到细胞干质量的65%)。本研究为渤海湾盐田微生物资源的开发和利用提供了有价值的基础数据。  相似文献

印度洋深海热液区可培养细菌的分子鉴定与系统发育分析   总被引:1,自引:0,他引:1  
采用MMJHS寡营养培养基从印度洋深海热液区沉积物和热液硫化物中分离获得16株细菌,通过16S rDNA序列比对和生理生化分析,对它们进行了鉴定并构建了系统发育树.结果表明,12株细菌属于γ-变型菌(γ-Gammaproteobacteria),其中6株属于盐单胞菌属(Halomonas),4株属于嗜冷杆菌属(Psychrobacter),2株属于食碱菌属(Alcanivorax);其余4株属于芽孢杆菌(Bacillus),其中1株为地衣芽孢杆菌(Bacillus licheniformis).分离获得的16株细菌中,4株为革兰氏阳性细菌,12株为革兰氏阴性细菌;H2S反应、吲哚测定反应、M.R和V-P均呈阴性.本研究为深入认识和开发利用深海热液区微生物资源奠定了基础.  相似文献

从东太平洋热液区E53站位的深海沉积物样品中分离出1株能在65℃生长的嗜热菌(DYth03).该菌的16S rDNA序列与地芽孢杆菌属(Geobacillus)内各种之间的同源性为98%以上.克隆得到DYth03的DNA聚合酶基因(DYth-pol),序列分析表明该基因全长为2 631 bp,G+C含量为55.5%,推测编码为876个氨基酸,与BstDNApolI的同源性最高(达98%).将该聚合酶基因克隆到pTTQ-h表达载体上,并在大肠杆菌DH1中进行表达.对纯化到的表达产物进行酶活性测定,结果表明该酶具有聚合酶活性和5’-3’外切酶活性.  相似文献

Diversity of bacteria was studied in deep-sea sediments from the Shikoku Basin in the Northwest Pacific Ocean by PCR, RFLP and sequence analysis of 16S rDNA and comparing with Genbank database. Based on the RFLP profile generated, 77 clones from the 16S rDNA library were divided into 27 types. Phylogenetic analysis showed that the 27 independent clones fell into four groups: Proteobacteria (62.96%), Chloroflexi (14.81%), Planctomycetes (14.81%) and Acidobacteria (7.41%). Among all sequenced clones, 6 were related to the sulfur or sulfate metabolism bacteria and the results also demonstrated that some bacteria in deep-sea sediments had relation to matter-energy circulation.  相似文献

台湾地处西太平洋构造活动带,近海发育了多处热液活动,其中最典型的为龟山岛和绿岛热液体系。本文对海峡两岸在龟山岛和绿岛热液的地球化学特征以及周边生物体的响应的研究进展进行了综述。龟山岛热液喷出流体具有全球最低的pH(1.52),富含重金属元素和CO2等酸性气体,是周围海水中有色溶解有机质的来源;绿岛具有全球热液中最低的溶解有机碳浓度(14 μmol /L),且具有特殊的动力学特性。喷口周边分布了较为罕见的自然硫烟囱体和硫磺球。喷口的高毒性、高酸性热液改变了热液区生物体如螃蟹的生活习性和解毒机制。热液区的主要活跃菌群为参与碳、硫和氮代谢途径的γ-和ε-变形菌。主要生物质合成以硫还原和硫氧化的化能无机自养型生物为主,微生物硫代谢促进了热液系统中的微生物能量流动和元素循环作用。某些热液生物采用繁殖期迁徙的机制应对高毒性、高酸性热液环境。在热液活动的胁迫下,这些微生物产生了新颖独特的代谢产物。此外,龟山岛和绿岛热液体系还受到了潮汐、台风和地震等灾害性事件的影响。台湾近海热液体系的研究对认识热液地球化学循环、探讨热液的生态环境效应等具有重要的意义。  相似文献

Spatial distribution, diversity, and composition of bacterial communities within the shallow sub-seafloor at the deep-sea hydrothermal field of the Suiyo Seamount, Izu-Bonin Arc, Western Pacific Ocean, were investigated. Fluids were sampled from four boreholes in this area. Each borehole was located near or away from active vents, the distance ranging 2–40 m from active vents. In addition, fluids discharging from a natural vent and ambient seawater were sampled in this area. We extracted DNA from each sample, amplified bacterial 16S rRNA genes by PCR, cloned the PCR products and sequenced. The total number of clones analyzed was 348. Most of the detected phylotypes were affiliated with the phylum Proteobacteria, of which the detection frequency in each clone library ranged from 84.6% to 100%. The bacterial community diversity and composition were different between hydrothermal fluids and seawater, between fluids from the boreholes and the vent, and even among fluids from each borehole. The relative abundances of the phylotypes related to Thiomicrospira, Methylobacterium and Sphingomonas were significantly different among fluids from each borehole. The phylotypes related to Thiomicrospira and Alcanivorax were detected in all of the boreholes and vent samples. Our findings provide insights into bacterial communities in the shallow sub-seafloor environments at active deep-sea hydrothermal vent fields.  相似文献

本文记录了西太平洋深海围线海绵科1新种——扭形白须海绵。该海绵共含2个标本,其中1个采集于雅浦海山932米处,另1个采集于卡罗琳海山775.3米处。该新种含有椭圆形大双盘骨针、3种类型的双盘骨针、棒状骨针和节杖骨针,使其很容易与该属下的现有8种区分开。该种为西北太平洋报道的第4种白须海绵属的种。  相似文献

现代海底热液沉积物的硫同位素组成及其地质意义   总被引:17,自引:0,他引:17  
共收集到现代海底热液沉积物的1264个硫同位素数据,结合我们对冲绳海槽Jade热液区和大西洋中脊TAG热液区中表层热液沉积物的硫同位素研究成果,对比分析了不同地质-构造环境中海底热液沉积物的硫同位素组成特征及其硫源问题.结果表明:(1)现代海底热液沉积物中硫化物的硫同位素组成集中分布在1‰~9‰之间,均值为4.5‰(n=1042),而硫酸盐矿物的硫同位素组成主要分布在19‰~24‰之间,均值为21.3‰(n=217);(2)无论在洋中脊还是在弧后盆地扩张中心,无沉积物覆盖热液活动区中热液沉积物与有沉积物覆盖热液活动区相比,其硫同位素组成的分布范围相对狭窄;(3)各热液活动区中硫化物硫同位素组成的不同,反映出各自硫源的差异性.无沉积物覆盖洋中脊中热液成因硫化物的硫主要来自玄武岩,部分来自海水,是玄武岩和海水硫酸盐中硫不同比例混合的结果,而在弧后盆地和有沉积物覆盖的洋中脊,除了火山岩以外,沉积物和有机质均可能为热液硫化物的形成提供硫;(4)现代海底热液沉积物硫同位素组成的变化和硫源的不同可能归因于海底热液体系中流体物理化学性质的变化、岩浆演化和构造-地质背景的不同.  相似文献

以东太平洋海隆附近深海柱状沉积物为材料,通过化学裂解和酶消化相结合的方法提取了沉积物微生物的总基因组DNA,并进行了纯化。结果表明所得到的DNA分子片段大小在21kb左右,纯化后的DNA可直接用于PCR等分子生物学操作。细菌16SrDNAV3可变区的PCR—DGGE图谱展示出15条以上条带,表明深海沉积物中细菌多样性较高,群落结构比较复杂。对其中9条主要条带进行回收、测序和系统发育分析,结果表明所获得的序列分属放线菌门(Actinobacteria),绿弯菌门(Chloroflexi),γ-变形细菌亚门(Gamma—proteobacteria),α-变形细菌亚门(Alpha—proteobacteria)和嗜酸菌门(Acidobacteria)5个大类群。  相似文献

Earth’s fastest present seafloor spreading occurs along the East Pacific Rise near 31°–32° S. Two of the major hydrothermal plume areas discovered during a 1998 multidisciplinary geophysical/hydrothermal investigation of these mid-ocean ridge axes were explored during a 1999 Alvin expedition. Both occur in recently eruptive areas where shallow collapse structures mark the neovolcanic axis. The 31° S vent area occurs in a broad linear zone of collapses and fractures coalescing into an axial summit trough. The 32° S vent area has been volcanically repaved by a more recent eruption, with non-linear collapses that have not yet coalesced. Both sites occur in highly inflated areas, near local inflation peaks, which is the best segment-scale predictor of hydrothermal activity at these superfast spreading rates (150 mm/yr).  相似文献

Abstract

Hydrothermal iron‐rich crusts have been recovered from a number of hot spot volcanos including Crough Seamount, Pitcairn Volcanos 2 and 1, Cyana Seamount, Teahitia, Moua Pihaa, and Macdonald Seamount in the S. W. Pacific. Mineralogically, the crusts consists of ferrihydrite with traces of the weathering products of volcanic ash (feldspar, nontronite, pyroxene, and serpentinite). The iron oxyhydroxide phase has a mean particle size of 3–4 nm indicating rapid deposition. Electron microprobe studies have revealed the presence of filamentous iron‐silica deposits within the crusts reflecting the possible bacterial oxidation of iron from the hydrothermal fluids. The crusts display wide variability in composition both between individual sampling stations and between seamounts. Endmember analysis shows that the compositional data can be resolved into three endmembers: a Fe‐rich endmember, a light and heavy rare earth element endmember, and a Ba (barite)‐rich endmember. The Fe‐rich endmember appears to contain very low concentrations of most trace elements. For bulk samples, the composition of the iron‐rich crusts reflects dilution of the iron oxyhydroxide phase by volcanic ash and, to a lesser extent, a hydrogenous component. This is illustrated by the wide variability in SiO2 (11.1–71.3%) and Mn (0.01–1.21%) contents of the crusts. For iron‐rich crusts containing greater than 40% Fe, the Pitcairn crusts display lower contents of Pb, Ba, Mo, U, Th, As, and rare earth elements (REE) and lower cerium anomalies than those from Teahitia. REE profiles of crusts from each of the hot spot volcanoes are characterized by small negative cerium anomalies but pronounced positive europium anomalies. The low average La/Fe ratios of the crusts from the various seamounts (47–572 X 10‐6) and positive Eu anomalies of the crusts suggest rapid deposition of the iron oxyhydroxide near the hydrothermal vent. The high Fe/Mn ratio of vent fluids at hot spot volcanoes (8.5–5.6) may account for the formation of these iron‐rich crusts. The present data indicate that there may be differences in the nature of the iron‐rich crusts based on the depth of occurrence. This influences the temperature of the venting hydrothermal fluids and the possibility of occurrence of submarine phreatomagmatic eruptions.  相似文献

Since the discovery of hydrothermal vents in the late 1970s, deep-sea hydrothermal vent fields have attracted great attention as biological hotspots. However, compared with other ocean ridges, the structure and function of microbial communities inhabiting vent fields in the Central Indian ridge (CIR) remain understudied. Here, we provide for the first time 16S rRNA gene-based comparative metagenomic analysis of the sediment-associated microbial communities from three newly discovered vent fields in the CIR. Sediment samples collected in the Invent B, Invent E and Onnuri vent fields varied in geochemical properties, elemental concentrations and associated microbial communities. Proteobacteria (Gammaproteobacteria) was the dominant phylum in Invent B and Onnuri vent fields. In contrast, Invent E mainly consisted of Chloroflexi and Euryarchaeota. Predicted functional profiling revealed that the microbial communities in the three vents are dominated by chemoheterotrophic functions. In addition, microbial communities capable of respiration of sulfur compounds, nitrification, nitrite oxidation, methylotrophy, and methanotropy were found to be the main chemolithoautotrophs. Compared to other vent fields, Invent E showed a predominance of archaeal methanogens suggesting it exhibits slightly different geochemistry. Multivariate analysis indicated that the biogeochemical and trace metal differences are reflected in the sediment microbial compositions of the three vent fields. This study expands our current understanding of the microbial community structure and potential ecological functions of the newly discovered hydrothermal vent fields in the CIR.
