为探讨菌株Salinivibrio sp. YH4分泌的丝氨酸蛋白酶EYHS的耐盐性及结构特征,采用明胶底物酶谱法分析EYHS的耐盐性,并应用生物信息学手段对EYHS及6种耐盐的S8家族丝氨酸蛋白酶结构特征进行分析。结果表明EYHS在4 mol/L的NaCl溶液中仍具有活性,属于耐盐蛋白酶。EYHS及6种S8家族丝氨酸蛋白酶分子表面的loop区等无规则卷曲所占比例较高,α-螺旋与β-片层则主要位于酶分子内部。EYHS分子表面酸性氨基酸含量较高,且具有弱疏水内核。多序列比对发现蛋白酶的催化三联体两侧存在高度保守的基序和保守的极性氨基酸及芳香族氨基酸,并存在多个保守的Gly与Ala。同源模建和表面电荷分布显示,α螺旋和β片层围成了蛋白酶的催化腔,EYHS活性中心包含由Asp32、His65与Ser215组成的催化三联体,且催化位点区域表面静电势为负。上述结构特征可能有助于耐盐丝氨酸蛋白酶EYHS在高盐环境下维持其稳定性和适度柔性,并有助于其催化功能的发挥,为深入研究耐盐丝氨酸蛋白酶的高盐环境适应性提供了一定的理论依据。 相似文献

从北极楚科奇海水中分离到一株产蛋白酶耐冷河豚毒素假交替单胞菌BSw20353。该菌可合成多种胞外蛋白酶,而培养温度影响蛋白酶的合成。分离纯化出其中一种在15℃、25℃培养时均有生成的酶:该酶最适作用温度55℃;最适pH8;金属离子Hg2+、Sr2+抑制酶活,Fe2+、Mn2+则具有激活作用;该酶属于金属蛋白酶,受螯合剂邻二氮杂菲、EDTA及EGTA抑制,基本不受丝氨酸蛋白酶抑制剂PMSF、TPCK及TLCK抑制,半胱氨酸蛋白酶抑制剂E-64对该酶无抑制作用,天冬氨酸蛋白酶抑制剂pepstatin则具有一定抑制作用;SDS、二硫苏糖醇、碘乙酸可抑制酶活;而在变性剂尿素、盐酸胍作用下,酶活反而有所增高。 相似文献

土壤盐分过高抑制植物生长，已成为盐碱地区域农业发展的主要限制因素。植物根际促生细菌（Plant growth-promoting rhizobacteria，PGPR）促进盐碱地作物生长，增强作物耐盐能力，进而诱导作物盐耐受性。对能诱导植物盐耐受性的PGPR种类作了总结，并从PGPR自身产生物质及植物受到PGPR作用后生理变化两个方面评述PGPR提高植物耐盐性的机制。了解PGPR提高植物耐盐性的机理，为大规模利用微生物提高作物在盐碱等条件下的稳产高产提供理论依据。 相似文献

骆驼蓬属叶中氨基酸组成与抗逆性的关系 总被引：9，自引：0，他引：9
实验结果证明：西北地区骆驼蓬属叶中总氨基酸和游离氨基酸组成中均以天冬氨酸、苏氨酸、丝氨酸、谷氨酸、丙氨酸、缬氨酸、苯丙氨酸、组氨酸、和脯氨酸为主要组分。其中，天冬氨酸、苏氨酸、谷氨酸、苯丙氨酸、和脯氨酸表现出种间的量值差异。不同生境下的骆驼蓬和多裂骆驼蓬均以天冬氨酸等９种氨基酸为主要组分，但因生境不同而在部分氨基酸含量上有明显差异。生于盐碱荒地的骆驼蓬，其脯氨酸含量为不同生境下同种的１１．３倍和８．８５倍。不同生境下的多裂骆驼蓬其脯氨酸含量依干旱胁迫减弱而降低。表明细胞中脯氨酸积累与植物的耐盐、抗旱性有关 相似文献

黄河三角洲湿地典型盐生植物群落土壤酶活性研究 总被引：1，自引：0，他引：1
选择黄河三角洲湿地盐角草、盐地碱蓬、二色补血草、中亚滨藜、獐毛、星星草6种典型盐生植物群落,研究土壤蔗糖酶、纤维素酶、脲酶、蛋白酶、磷酸酶的活性特征。结果显示:土壤蔗糖酶活性和蛋白酶活性逐渐升高,9月以上各植物群落蔗糖酶活性依次分别为279.05 U/g、291.62 U/g、304.82 U/g、334.60 U/g、333.03 U/g、357.30 U/g,均显著高于6月;蛋白酶活性分别为21.20 U/g、24.01 U/g、25.40 U/g、30.09 U/g、30.89 U/g、33.12 U/g,除盐角草群落外均显著高于6月。9月獐毛和星星草群落纤维素酶活性显著高于6月。所有植物群落脲酶活性先升高后降低,8月达最高,分别为151.65 U/g、165.36 U/g、177.85 U/g、197.30 U/g、189.71 U/g、203.00 U/g,除盐角草群落外,分别显著高于6月。磷酸酶活性7月在除盐角草群落外的其他植物群落中比6月显著提高,后期则变化不大。相同生长期不同植物群落之间,几种酶活性均表现出在星星草和獐毛群落高于盐角草和碱蓬群落。本研究结果可为深入理解滨海湿地不同盐生植物群落土壤特性、盐碱地改良及群落演替等提供相关理论参考和依据。 相似文献

为了研究盐、旱及其交互胁迫下植物生长和生理特性的变化，了解沙漠腹地极端生境下互 叶醉鱼草（Buddleja alternifolia）幼苗的适应性，确定合理的灌溉方式，为沙漠腹地醉鱼草的引种提 供理论依据。通过大田控制实验，设计了 3 个水分梯度和 4 个盐分梯度随机结合实验，定位测量生 长量和生化指标，最后基于隶属函数法综合评价醉鱼草的抗旱耐盐性。结果表明：（1）在干旱胁迫 或低浓度盐胁迫下，醉鱼草的地径、新枝和冠幅生长量均呈上升趋势，而在高盐浓度胁迫下均降 低；水盐交互胁迫下,不同的盐浓度与干旱交互胁迫效果不同,低盐浓度与干旱交互胁迫时（W1S1、 W2S1），植物生长量均上升，高盐浓度与干旱交互胁迫时（W1S3、W2S3），植物生长量均降低。（2）无论 是盐胁迫、干旱胁迫还是盐、旱交互胁迫，醉鱼草游离脯氨酸（Pro）含量随着胁迫程度的增加均呈显 著上升的趋势。（3）醉鱼草的超氧化物歧化酶（SOD）活性和过氧化物酶（POD）活性变化相对复杂， 除了在干旱胁迫时，SOD 活性和 POD 活性都显著下降外,在盐胁迫和交互胁迫时它们各自的变化并 不一致。醉鱼草在盐胁迫、交叉胁迫和干旱胁迫时，渗透调节物质和酶活性各自对应的发生了变 化，没有明显的同步性，存在相互协调的可能，同时植物细胞的敏感性远远强于表型生长的变化， 高盐浓度的伤害并没有表现在植物外部形态上，但酶活性及渗透调节物质却有显著变化。通过隶 属函数分析显示，醉鱼草最佳的生长水盐条件是盐浓度为 8 g·L-1、灌水量为 25 L·次-1，对水盐胁迫 的抗旱耐盐性为：干旱胁迫＞交互胁迫＞盐胁迫。 相似文献

3种典型荒漠灌木内生固氮菌及固氮酶基因nifH多样性分析 总被引：1，自引：0，他引：1
具有特殊生境的内生固氮菌,对改善植物营养、增强植物抗逆性及群落稳定性具有重要作用,是一类潜力巨大、尚待开发的微生物资源。以新疆3种典型荒漠灌木多枝柽柳（Tamarix ramosissima）、黑果枸杞（Lycium ruthenicum）、盐穗木（Halostachys caspica）为材料,从根和枝条组织中分离筛选获得137个内生固氮菌菌株,采用16S rDNA PCR-RFLP、16S rDNA序列测定、BOXAIR-PCR指纹图谱、nifH PCR-RFLP等方法分析其遗传多样性及系统发育关系。结果表明：分离菌株经16SrDNA PCR-RFLP酶切图谱聚类划分为9个遗传类型。序列测定和系统发育分析显示,代表菌株分属于拉恩氏菌属（Rahnella）、假单胞菌属（Pseudomonas）、泛菌属（Pantoea）、芽孢杆菌属（Bacillus）、寡养单胞菌属（Stenotrophomonas）和申氏杆菌属（Shinella）,其中Rahnella为优势种群。一个测定菌株的16SrDNA序列同源性低于97%,提示可能为一个潜在的新种。BOXAIR-PCR分析优势种群的基因组结构特征,获得12种指纹图谱类型,显示出该种群具有丰富的基因组多样性。nifH PCR-RFLP分析分离内生固氮菌菌株固氮酶基因nifH的分子多态性,将其划分为3种基因型,体现了nifH具有高度保守性,同时在不同固氮微生物种群和菌株间也存在一定的多样性。研究结果丰富了固氮微生物物种资源库和基因库,对于内生固氮菌资源的保护和利用奠定了科学基础。 相似文献

从北极海洋环境中筛选分离出一株产胞外碱性蛋白酶的革兰氏阴性杆菌 Ar/ W/ b/ 75°2 5′ N/ 1。该菌能在 7～ 30℃范围内生长 ,并且在 30℃条件下生长良好 ,但不能在 40℃条件下生长。该菌在 0 .5%～ 1 0 % Na Cl的盐浓度范围内能良好生长 ,表现出一定的嗜盐性。葡萄糖、蔗糖及可溶性淀粉等碳源物质可被该菌株利用以供生长之需 ,其中以葡萄糖最佳。氮源物质中 ,蛋白胨是最有利于菌株生长、产酶的有机氮源 ,而 NH4 NO3则是效果最好的无机氮源。对该菌蛋白酶的酶学特性研究表明 :所产胞外蛋白酶占其蛋白酶总量的 75.7%左右 ;蛋白酶反应的最适温度为40℃ ,酶活力在温度不高于 40℃条件下保持稳定 ;该菌蛋白酶具有较宽的 p H活性范围 ,在 p H7～ 1 1均表现出较高酶活力 ,其最适 p H在 8左右 相似文献
