红球菌(Rhodococcus sp.)B7740分离自北冰洋海水样品,前期实验表明其能产生大量黄色素,但是该菌生物量很低。本实验通过碳源、氮源、培养温度、发酵液起始pH值、菌种接种量等单因素和正交实验优化了该菌株的培养条件,在此基础上,通过对提取剂种类及pH、提取时间、料液比等的研究,优化了色素的提取条件。实验结果表明,1L陈海水中,添加5g酵母粉,5g葡萄糖,3g乳糖,7g牛肉膏;起始pH7,培养温度25℃,菌种接种量1.0%,160r/min摇床培养7d时,生物量从6.1mg/mL上升到23.08mg/mL,提高了278.4%。以上述培养条件下得到的干菌体为材料,采用pH7的无水乙醇,当料液比1:30,浸提温度75℃,浸提1h重复提取2次时,色素含量为1160.625μg/g干菌体。对色素的分析表明,该色素为类胡萝卜素,实验结果为进一步研究极地微生物红球菌(Rhodococcus sp.)B7740产类胡萝卜素的提取工艺提供了基础。 相似文献

凤眼莲净化水中重金属的研究 总被引：3，自引：0，他引：3
通过静态水培养实验探讨了凤眼莲对水中重金属离子铅、锌、铬的短期去除能力.实验表明:凤眼莲对这3种重金属的耐受能力和去除能力都是Pb(II)＞Cr(III)＞Zn(II);pH值、凤眼莲生物量以及重金属离子的初始浓度对净化效果均有明显的影响;其中凤眼莲对含铅水溶液有很好的净化效果,经过4天的培养,300 g凤眼莲对4 L浓度为5～30 mg/L的铅离子溶液的去除率均可达到99%.因此,凤眼莲在植物修复重金属污染水体方面具有很大的潜能. 相似文献

主要研究了反硝化聚磷微生物对湿地受氮、磷污染水体的净化功能及在实践应用中的技术可行性.研究基于从大庆龙凤湿地富营养化水体中分离并鉴定到的一株假单胞菌(Pseudomonas putida),将其命名为DQ1.结果表明,菌株DQ1在以硝酸盐为唯一氮源,以柠檬酸为唯一碳源,在30℃的条件下静止培养30h,能将培养液中91.64％的总氮去除,去除速率达到11.33 mg/(L·h);在厌氧条件下培养2h,上清液总磷质量浓度上升27 mg/L,菌株DQ1表现出明显的有氧反硝化聚磷的特性;静止培养28h,聚磷量则可达到94.72 mg/L,磷去除率为99.6％.在高碳磷比和低碳磷比的污染体系中,菌株DQ1都对磷有较好的去除效果.DQ1固定化技术在湿地水体修复方面有很强的应用潜力,经过6h,水体中的氮和磷都能被有效去除,氨氮质量浓度由5.38 mg/L降至检测限以下,而硝态氮去除率可达98.48％,磷去除率达到97.46％. 相似文献

以柴油为唯一碳源和能源,从南极海水海冰微生物资源库中筛选到一株石油烃低温降解菌希瓦氏菌NJ49,并对影响其生长和降解率的环境因素(pH、温度、盐度、营养盐和表面活性剂)进行了初步研究。结果表明:希瓦氏菌NJ49可作为低温海域石油烃污染生物修复的菌源,其生长和降解的最适条件为:初始pH7.5,温度15℃,盐度6%,摇瓶装量80ml,最佳氮源硝酸铵,最佳磷源为磷酸二氢钾和磷酸氢二钾的混合物,添加表面活性剂可促进希瓦氏菌NJ49的生长和生物降解率。 相似文献

基础原料铬酸钠的质量直接影响后续系列铬盐产品的品质。电化学氧化技术获得的铬酸钠电解液中可溶性硅在后期系列铬酸盐的制备过程中无法从自体系中去除,会残留在最终产品中进而影响后续铬盐产品的质量。为提高以铬酸钠电解液为原料制备的三氧化二铬产品品质,研究了以铬酸为酸化剂,调节铬酸钠电解液pH去除硅的方法。结果表明,优化反应条件:铬酸钠电解液pH=8,反应温度30℃,反应时间16 h,搅拌速度300 rpm。在此条件下,铬酸钠电解液中二氧化硅浓度降至200 mg/L以下。以除硅后的铬酸钠电解液为原料制备的三氧化二铬纯度达99.43%,满足颜料级三氧化二铬纯度要求(≥99.00%)。 相似文献

从北极海洋环境中筛选分离出一株产胞外碱性蛋白酶的革兰氏阴性杆菌 Ar/ W/ b/ 75°2 5′ N/ 1。该菌能在 7～ 30℃范围内生长 ,并且在 30℃条件下生长良好 ,但不能在 40℃条件下生长。该菌在 0 .5%～ 1 0 % Na Cl的盐浓度范围内能良好生长 ,表现出一定的嗜盐性。葡萄糖、蔗糖及可溶性淀粉等碳源物质可被该菌株利用以供生长之需 ,其中以葡萄糖最佳。氮源物质中 ,蛋白胨是最有利于菌株生长、产酶的有机氮源 ,而 NH4 NO3则是效果最好的无机氮源。对该菌蛋白酶的酶学特性研究表明 :所产胞外蛋白酶占其蛋白酶总量的 75.7%左右 ;蛋白酶反应的最适温度为40℃ ,酶活力在温度不高于 40℃条件下保持稳定 ;该菌蛋白酶具有较宽的 p H活性范围 ,在 p H7～ 1 1均表现出较高酶活力 ,其最适 p H在 8左右 相似文献

为了明确好氧反硝化微生物在氮元素污染湿地水体原位修复工程中的可行性,采用培养基富集,NO3-BTB平板划线法,从大庆湿地筛选到一株好氧反硝化细菌DQ1,通过生理生化测定和16S rRNA测定(登录号为:JQ669957)鉴定为恶臭假单胞菌(Pseudomonas putida)。菌株DQ1在以硝酸盐为唯一氮源,以柠檬酸为唯一碳源,在30℃的条件下静止培养30 h,总氮(TN)去除率达到91.9%,整个过程中没有检测到亚硝态氮(NO2-—N)的积累;在初始氮源浓度为1 930 mg/L,以柠檬酸、酒石酸钾钠、无水乙酸、葡萄糖和乙醇等为唯一碳源,5d总氮去除率分别为97.4%、97.1%、97.2%、97.6%和98.5%。当pH为6.5、6.95、7.45、8.0和8.5时,5 d总氮去除率分别为93.71%、97.27%、97.59%、97.35%和96.85%;当初始pH为7.45时,总氮去除率最高达97.59%,菌株DQ1在温度20~37℃范围内,5 d总氮去除率分别为98.03%、98.12%、98.23%和98.05%。 相似文献

铬胁迫对旱伞草生理生化特性的影响 总被引：1，自引：0，他引：1
采用水培实验,以不同质量浓度的铬溶液胁迫旱伞草(Cyperus alternifolius),研究其对旱伞草总叶绿素含量、蛋白质含量、游离脯氨酸含量和丙二醛含量的影响,并对旱伞草对铬的去除效果进行了测定。结果表明,铬胁迫使旱伞草的叶绿素含量下降,叶绿素a/b含量比上升;过氧化氢酶活性增强,超氧化物歧化酶活性降低;丙二醛含量先降低后升高;游离脯氨酸先降低后稳定;过氧化物酶活性在低浓度铬胁迫时出现降低,在高浓度铬胁迫时出现升高;蛋白质含量的减少后稳定,由此说明旱伞草对铬胁迫具有一定的抗逆性和耐受性。在培养液中,在铬质量浓度分别为2 mg/L、5 mg/L和10 mg/L胁迫下,六价铬的去除率分别达到了96.88%、92.55%和46.5%;总铬的去除率分别达到了76.4%、73.18%和36.83%。这说明旱伞草对低、中浓度的重金属铬具有较好的去除效果。 相似文献

本文对影响南极细菌S-15-13生长和胞外多糖产量的主要环境因子进行了研究,同时采用16S rDNA序列分析及系统发育分析方法对其进行了分子鉴定。结果表明：菌株S-15-13最佳产糖条件为：培养时间,56h；培养温度,8℃；碳源,1．0%葡萄糖；NaCl浓度,3．0%；pH,6．0-7．0。16S rDNA序列分析和系统发育分析表明,菌株S-15-13属于假交替单胞菌属(Pseudoaltero- monas)。 相似文献
