The response surface methodology (RSM) combined with bioassays was employed to optimize the extraction process of crude fucose-containing sulphated polysaccharides (cFCSP) from Sargassum fusiforme. The central composite design (CCD) was used with four variables, five levels, and four responses. The four variables were pH value of hydrochloric acid solution, extraction temperature (°C), ratio of liquid to raw material (mL g?1), and extraction time (h), respectively. Chemical and bioassay indices were used in combination as the response parameters, which included the yield of cFCSP, fucose content, proliferation rate of spleen cells, and lipopolysaccharide-induced proliferation of splenocytes. The experimental data were fitted to a second-order polynomial equation using multiple regression analysis, and examined using the appropriate statistical methods. The best extraction conditions were as follows: the pH value of hydrochloric acid solution was 3.50; the extraction temperature was 100°C; the ratio of liquid to raw material was 15.00 mL g?1 and the extraction time was 2.50 h. The experimental yield was close to the predicted from the model. The extract could promote spleen lymphocyte proliferation, especially the lipopolysaccharide-induced lymphocyte proliferation in vitro, which suggested that its immunomodulatory effect on B lymphocytes. Therefore, cFCSP extracted from S. fusiforme could be utilized as an immunostimulant in functional foods and pharmaceutical industry in future.  相似文献

The yield criterion parameters of the soil material change with different values of the cohesion and the angle of friction because of sustained rainfall infiltration. Based on the Mohr-Coulomb(M-C) and Drucker-Prager(D-P) yield criteria, some reasonable yield criteria selections were discussed for quantitative analysis of unsaturated soil slope stability. Moreover, a critical point was found at the effective angle of friction equaling to 16.5° by transformation of parameters related to unsaturated soil under sustained rainfall. When the effective angle of friction more than 16.5° through parameter transformation of different yield criteria under natural condition, the calculation result of the safety factor was such that: f(DP1) f(M-C) f(equivalent M-C) f(DP2) f(DP3). While the effective angle of friction less than 16.5°, through parameter transformation, the safety factors were in the following order: f(DP1) f(M-C) f(DP2) f(equivalent M-C) f(DP3). The calculated results from a case study showed that the equivalent M-C yield criterion should be the best at evaluating soil slope stability before rainfall; the DP2 yield criterion should be selected to calculate the soil slope stability at the effective angle of friction less than 16.5° under sustained rainfall. The yield criterion should be selected or adjusted reasonably to calculate the safety factor of unsaturated soil slopes before and during sustained rainfall.  相似文献

In this study, molecular weight controllable degradation of algal Laminaria japonica polysaccharides(LPS) was investigated by ultrasound combined with hydrogen peroxide. Three main factors, i.e., ultrasonic power(A), ultrasonic time(B), and H2O2 concentration(C) were chosen for optimizing parameters by employing three-factors, three-levels BBD. The influence of degradation on structure change and antioxidant activities was also investigated. A second-order polynomial equation including molecular weight(Y) of Laminaria japonica polysaccharides and each variable parameter, i.e., ultrasonic power(A), ultrasonic time(B), and H2O2 concentration(C), was established: Y=20718.67-4273.13A-4000.38B-1438.75C+2333.25AB+1511.00AC+873.00BC+2838.29A2 + 2490.79B2+873.04C2. The equation regression coefficient value(R2 = 0.969) indicated that this equation was valid. The value of the adjusted determination coefficient(adjusted R2 = 0.914) also confirmed that the model was highly significant. The results of selected experimental degradation conditions matched with the predicted value. FT-IR spectra revealed that the structures of LPS before and after degradation were not significantly changed. Antioxidant activities of LPS revealed that low Mws possessed stronger inhibitory than the original polysaccharides. The scavenging effects on superoxide radicals was the highest when IC50 of crude LPS was 4.92 mg mL-1 and IC50 of Mw 18.576 KDa was 1.02 mg mL-1, which was fourfold higher than initial polysaccharide.  相似文献

A feeding trial was carried out to investigate the dietary vitamin E requirement of the oriental river prawn Macrobrachium nipponense (weight of 0.3–0.4 g) and its effect role on antioxidant activity. Prawns were fed with seven levels of vitamin E (0, 25, 50, 75, 100, 200, and 400 mg/kg diet) for 60 days. The results show that dietary vitamin E supplementation could significantly increased the prawn weight (P < 0.05). The activity of superoxide dismutase (SOD) in the hepatopancreas was significantly higher in prawns fed with diets supplemented with ≤75 mg/kg vitamin E than in those fed with diets supplemented with 100–400 mg/kg vitamin E (P < 0.05). The activity of catalase (CAT) in the hepatopancreas decreased significantly as dietary vitamin E supplementation increased (P < 0.05), and no significant difference was detected in glutathione peroxidase (GSH-Px) activity between different dietary groups (P >0.05). The contents of vitamin E in the hepatopancreas and in the muscle increased with increasing dietary vitamin E. There was a linear correlation between the vitamin E level in diet and that in muscle, and between the vitamin E level in diet and that in the hepatopancreas. All the above results indicated that dietary vitamin E can be stored in the hepatopancreas and muscle and lower both the activities of SOD and CAT in the hepatopancreas, suggesting that it is a potential antioxidant in M. nipponense. Broken line analysis conducted on the weight gains of prawns in each diet group showed that the dietary vitamin E requirement for maximum growth is 94.10 mg/kg.  相似文献

Genomic mining has identified a novel microbial alkaline esterase from the Indian Ocean. This esterase was overexpressed in E. coli BL21 (DE3) and further functionally characterized. Under optimal conditions (10 mmol/L substrate, pH 6.0, 2 h at 40 °C), this esterase can hydrolyze racemic methyl mandelate to (R)-methyl mandelate with very high optical purity (e.e. >99%) and yield (nearly 90%). Interestingly, the stereoselectivity of this esterase is opposite to that of two previously reported lipases that can generate (S)-methyl mandelate through the hydrolysis of racemic methyl mandelate. No organic solvents or other additives were required to optimize the optical purity and production of the final chiral product (R)-methyl mandelate, which can potentially simplify the production procedure of (R)-methyl mandelate catalyzed by esterase.  相似文献

Novel preservation condition without ultra-low temperature is needed for the study of pathogen in marine fishes. Freeze-drying is such a method usually used for preservation of terrigenous bacteria. However, studies using freeze-drying method to preserving marine microorganisms remain very limited. In this study, we optimized the composition of protectants during the freeze-drying of Edwardsiella tarda, a fish pathogen that causes systemic infection in marine fishes. We found that the optimal composition of protectant mixture contained trehalose (8.0%), skim milk (12.0%), sodium citrate (2.0%), serum (12.0%) and PVP (2.0%). Orthogonal and interaction analyses demonstrated the interaction between serum and skim milk or sodium citrate. The highest survival rate of E. tarda was observed when the concentration of NaCl was 10.0, 30.0 and between 5.0 and 10.0 g L?1 for preparing TSB medium, E. tarda suspension and protectant mixture, respectively. When E. tarda was frozen at ?80°C or ?40°C for 6 h, its survival rate was higher than that under other tested conditions. Under the optimized conditions, when the protectant mixture was used during freeze-drying process, the survival rate (79.63%–82.30%) of E. tarda was significantly higher than that obtained using single protectant. Scanning electron microscopy (SEM) image indicated that E. tarda was embedded in thick matrix with detectable aggregation. In sum, the protectant mixture may be used as a novel cryoprotective additive for E. tarda.  相似文献

The dinoflagellate Prorocentrum minimum, one of the most widespread red tide causing species, affects marine aquaculture and ecosystems worldwide. In this study, ridgetail white prawn Exopalaemon carinicauda were exposed to P. minimum cells (5 × 104 cells mL?1) to investigate its harmful effects on the shrimp. Antioxidant activities and histological changes were used as indicators of health status of the shrimp. In 72 hours, the mortality of E. carinicauda was not affected, but its antioxidant response and histology were statistically different from those of control. Elevated superoxide dismutase (SOD) and glutathione peroxidase (GPX) activities and depressed catalase (CAT) activity were observed in gill; while increased SOD, glutathione S-transferase (GST), CAT activities and modulated GPX activity were observed in hepatopancreas. Thus, antioxidant activities in gill and hepatopancreas seem to respond differentially to harmful alga exposure. Increased malondialdehyde (MDA) content in early a few hours indicates the damage of the antioxidant defense system. Although MDA content recovered to a low level thereafter, a series of histological abnormalities including accumulation or infiltration of hemocytes, tissue lesions and necrosis were discovered in gill and hepatopancreas. Exposure to P. minimum induced sublethal effects on E. carinicauda, including temporary oxidative damage and histological injury.  相似文献

To study the effect of low temperature stress on hematological parameters and HSP gene expression in the turbot (Scophthalmus maximus), water temperature was lowered rapidly from 18 to 1°C. During the cooling process, three individuals were removed from culture tanks at 18, 13, 8, 5, 3, and 1°C. Blood samples and tissues were taken from each individual, hematological indices and HSP gene expression in tissues were measured. The red blood cell count, white blood cell count, and hemoglobin concentration decreased significantly (P < 0.05) as temperature decreased. Enzyme activities of plasma alanine transaminase and creatine kinase increased as temperature decreased, whereas aspartic transaminase and γ-glutamyl transpeptidase activities displayed no obvious changes above 1°C and lactate dehydrogenase activity increased first and then decreased. Blood urea nitrogen and uric acid levels were highest at 8°C, and creatinine concentration was highest at 3°C. The concentrations of plasma cortisol, cholesterol, and triglyceride all increased significantly (P < 0.05) as temperature decreased. The serum glucose concentration increased first and then decreased to the initial level. The HSP70 mRNA expression showed various patterns in different tissues, whereas HSP90 mRNA expression showed the same tendency in all tissues. Overall, these results indicate that temperature decreases in the range of 8 to 5°C may induce a stress response in S. maximus and that temperature should be kept above 8°C in the aquaculture setting to avoid damage to the fish.  相似文献

Photo-protective functions were investigated in phytoplankton assemblages at Kongsfjorden, Svalbard in spring, using their UV-absorbing compounds (mycosporine-like amino acids (MAAs)), xanthophyll pigments (diadinoxanthin (DD) and diatoxanthin (DT)) and < beta >- dimethylsulphoniopropionate (< beta >-DMSP). The dominant phytoplankton species in the inner bay were dominated by Phaeocystis spp. and nanoflagellates, while the offshore waters were dominated by Thalassiosira spp. In the inner bay, UVabsorbing compounds and xanthophyll pigments exhibited higher ratios of MAA to chlorophyll a (MAA:chl a ratio), and both DD and DT to chlorophyll a (DD:chl a ratio and DT:chl a ratio), respectively. Thus, the photoprotective-pigments such as DD and DT appear to complement MAAs in the natural phytoplankton assemblage. However, the ratio of < beta >-DMSP to chlorophyll a (< beta >-DMSP:chl a ratio) did not show a distinct spatial distribution according to environmental factors or interspecies differences. In this study, we found that photoprotective compounds occurred in a manner dependent on the phytoplankton species composition in Kongsfjorden Bay, where Phaeocystis is the dominant species.  相似文献

We investigated the effects of low temperature (8.5°C) on the growth and feeding rates and feed conversion efficiency of juvenile P. olivaceus with an average initial weight of 3.87 ± 0.06 g (mean ± SE). Fish were exposed to 8.5°C for 0 (control), 1, 2, 3 and 4 weeks, and then to 20°C for 10, 9, 8, 7 and 6 weeks, respectively. Low temperature clearly led to growth depression. The weight of fish exposed to low temperature for 1 week was restored to that of control, while that of fish exposed to low temperature longer was significantly decreased (P < 0.05). During the entire low-temperature period, specific growth rate, feeding rate and feed conversion efficiency of the fish were significantly lower (P < 0.05) than those of control, while in the whole recovery period, specific growth and average feeding rate were markedly higher (P < 0.05) than those of control. At the end of experiment, only the feeding rate of the fish exposed to low temperature for 1 week was not significantly different from that of control (P > 0.05). Feeding rate and feed conversion efficiency were reduced at low temperature in juvenile P. olivaceus. The compensatory growth of juvenile P. olivaceus may therefore be attributed to the improvement of feeding rate. Our results suggested that growth depression occurs when juvenile P. olivaceus are exposed to low temperature for more than one week.  相似文献

Jerusalem artichoke is an economic crop widely planted in saline-alkaline soil. The use of Jerusalem artichoke is of great significance. In this study, the response surface method was employed to optimize the eff ects of processing variables(extraction temperature, p H, extraction time, and liquid-tosolid ratio) on the yield of Jerusalem artichoke pectin. Under the optimal extraction conditions: p H 1.52, 63.62 min, 100°C and a liquid-to-solid ratio of 44.4 m L/g, the maximum pectin yield was predicted to be 18.76%. Experiments were conducted under these optimal conditions and a pectin yield of 18.52±0.90% was obtained, which validated the model prediction. The eff ects of diff erent drying methods(freeze drying, spray drying and vacuum drying) on the properties of Jerusalem artichoke pectin were evaluated and they were compared with apple pectin. FTIR spectral analysis showed no major structural diff erences in Jerusalem artichoke pectin samples produced by various drying treatments. The antioxidant activities of pectin dried by diff erent methods were investigated using in vitro hydroxyl and DPPH radical scavenging systems. The results revealed that the activities of spray dried pectin(SDP) and apple pectin(AP) were stronger than those of vacuum oven dried pectin(ODP) and vacuum freeze dried pectin(FDP). Therefore compared with the other two drying methods, the spray drying method was the best.  相似文献

Seaweeds are one of the largest producers of biomass in the marine environment. It has been well known that marine algae, especially brown algae was a rich source of biogenic compounds with antifouling potential that could be ideal alternatives of tributyltin (TBT). In this paper, antifouling potential of the brown algae Laminaria ‘sanhai’ was explored. Firstly, the dried alga was extracted and the antialgal and antilarval activities were investigated. The EC50 and LC50 values of crude extract of Laminaria ‘sanhai’ against diatom (Skeletonema costatum) and barnacle larval (Chthamalus challengeri) were 8.9 μg mL?1 and 12.0 μg mL?1 respectively. Then, guided by bioassay, the bioactive substances were isolated by liquid-liquid extraction. The antialgal and antilarval activities of isolated fraction were improved with the EC50 value of 7.4 μg mL?1 against S. costatum and LC50 value of 9.7 μg mL?1 against C. challengeri larvae. Identification by IR, Q-TOFMS and GC-MS of the isolated bioactive substances revealed the abundance of fatty acids. These fatty acids, most with 16, 18 or 20 carbon atoms, contained myristic, hexadecanoic, oleic, linolenic, arachidonic and eicosapentaenoic acids. The results indicated that both the crude extract and the isolated bioactive substances had high antialgal and antilarval activities with no highlighted cytotoxicity which made the brown algae Laminaria ‘sanhai’ a promising source of the environmentally friendly antifoulants.  相似文献

The effects of Bacillus subtilis 2-1 from the intestine of healthy sea cucumber on the growth, digestive enzyme activities and intestinal microbiota of juvenile sea cucumber (Apostichopus japonicus) were determined in the present study. Sea cucumber was fed with Sargassum thunbergii powder supplemented with B. subtilis 2-1 at different concentrations varying among 0 (control), 105, 107, and 109 CFU g?1 for 8 weeks. Results showed that the growth performance and intestinal amylase and trypsin activities were significantly increased by dietary B. subtilis 2-1 at 109 CFU g?1 (P < 0.05). However, dietary B. subtilis 2-1 had no significant influence on the lipase activity in sea cucumber (P > 0.05). The polymerase chain reaction denaturing gradient gel electrophoresis and 16S rRNA gene sequencing analysis indicated that dietary B. subtilis 2-1 at 105 and 107 CFU g?1 inhibited most of the Proteobacteria including those in genus Vibrio. Dietary B. subtilis 2-1 at 109 CFU g?1 not only decreased the abundance and species of genus Vibrio, but also increased the intensity of genera Psychrobacter and Bacillus. A specific dosage of dietary B. subtilis 2-1 could increase the growth and modulate the intestinal microbiota of sea cucumber; thus it might be a novel probiotic for keeping the health of sea cucumber.  相似文献

Micro-communities are supposed to have more potential functions of biodegradation of polysaccharides than single strain; however, the intestinal micro-communities involved in the biodegradation of Enteromorpha polysaccharides (EP) were seldom reported. In order to obtain the EP-degrading micro-community, the intestines of Siganus oramin was obtained to isolate the micro-communities, which were enriched by 0.3% of EP as the sole carbon source. A stable micro-community with EP degradative capability was achieved after seven generations of subculture, named H1. Results showed that H1 was able to degrade 75% of EP within 24 hours, and the activity of EP lyases reached 500 U mL?1 in 32 hours. With denaturing gradient gel electrophoresis (DGGE) and 16S rRNA gene clone library analysis, ten bacteria closely related to Marinomonas pontica, Microbacterium sp., Leucobacter chironomi, Cyclobacterium sp., Algoriphagus winogradskyi, Pseudoalteromonas sp. and Vibrio sp. were determined. Furthermore, compared with the DGGE bands sequence and the clone library analysis, the dominant bacteria of the EP-biodegrading micro- community were Pseudoalteromonas sp. and Vibrio sp., with the respective proportion of 38% and 46%, and they should play an important role in EP degradation together with other degrading bacteria in the micro-community H1.  相似文献

Substrate is a critical environmental factor affecting the activity of bivalves. To examine the effect of the substrate component on the growth and survival of juvenile sunray surf clam(Mactra chinensis Philippi), a series of short-term experiments were conducted using a variety of substrates with different ratios of sand to mud. The experimental group cultured without substrate showed poor survival, with all juveniles died after day 20. The juveniles cultured in mud without sand showed a lower survival rate(25.54% ± 0.40% on day 45) than those in other groups. The juveniles cultured in sand without mud, or the mixtures of sand and mud with a ratio of 1:1 and 2:1, respectively, exhibited modest survival at day 45. Maximal weight gain rate(WGR), shell length growth rate(LGR), and specific growth rate(SGR) were observed when the sand concentration was 61.97%, 77.69%, and 64.64%, respectively. As the fast growth and high survival were observed when the sand to mud ratio was 1:1(50% sand) and 2:1(67% sand), a sand concentration of more than 50% is optimal. The optimal concentration of sand in the substrate for rearing juvenile sunray surf clams was 67% which resulted in the fastest growth and highest survival. These results can be used to developing a nursery/farming technique of improving the yield of sunray surf clams.  相似文献

Comparing fat content with physiological status can throw light on the reproduction and feeding behavior of the fish. The biological data of 586 bigeye tuna were collected from the longline fishery in the waters of Western Central Indian Ocean from November, 2012 to March, 2013. The spatial or temporal distribution of the fat content, and the relationships of fat content with gender, round weight, gonadal maturity and fork length were analyzed. A generalized additive model (GAM) was used to analyze the relationships between fat content and fork length (FL), gonadosomatic index (GSI), condition factor (K), and somatic index (SI). Results showed that: 1) the fat content of bigeye tuna was in the range of 3.1%–27.1% with the average 12.8%; 2) there were no significant geographical differences of average fat content (P > 0.05) among 1° squares in general; 3) there were no significant differences (P > 0.05) of the fat content for different genders, months, or gonad maturity stages; 4) there was an extremely significant correlation (P = 0.000) between fat content and FL and GSI. There was no significant correlation (P = 0.051) between fat content and K. There was a significant correlation (P = 0.003) between fat content and SI. The results of this study suggest that the fat content of the matured fish was relatively stable. The survey area was in a spawning region, and the survey period was the spawning season for bigeye tuna.  相似文献

The changes in soil organic carbon(C) mineralization as affected by anthropogenic disturbance directly determine the role of soils as C source or sink in the global C budget. The objectives of this study were to investigate the effects of anthropogenic disturbance(aquaculture pond, pollutant discharge and agricultural activity) on soil organic C mineralization under different water conditions in the Minjiang River estuary wetland, Southeast China. The results showed that the organic C mineralization in the wetland soils was significantly affected by human disturbance and water conditions(P 0.001), and the interaction between human disturbance activities and water conditions was also significant(P 0.01). The C mineralization rate and the cumulative mineralized carbon dioxide-carbon(CO_2-C)(at the 49th day) ranked from highest to lowest as follows: Phragmites australis wetland soil aquaculture pond sediment soil near the discharge outlet rice paddy soil. This indicated that human disturbance inhibited the mineralization of C in soils of the Minjiang River estuary wetland, and the inhibition increased with the intensity of human disturbance. The data for cumulative mineralized CO_2-C showed a good fit(R~2 0.91) to the first-order kinetic model C_t = C_0(1 – exp(–kt)). The kinetic parameters C_0, k and C_0 k were significantly affected by human disturbance and water conditions. In addition, the total amount of mineralized C(in 49 d) was positively related to C_0, C_0 k and electrical conductivity of soils. These findings indicated that anthropogenic disturbance suppressed the organic C mineralization potential in subtropical coastal wetland soils, and changes of water pattern as affected by human activities in the future would have a strong influence on C cycling in the subtropical estuarine wetlands.  相似文献

The aim of the study is to examine the influence of the outplanting time on the cultivation of the kelp Laminaria saccharina in the northwestern Spain, close to its southern limit of distribution in the North Atlantic coast. We compared two L. saccharina cultures outplanted in February and December. We found that the starting time of outplanting affects to the yield and the substantiality value, as well as the growth rate. The yield, the substantiality value and the growth rates of L. saccharina sporophytes obtained for the trial outplanted in December were significantly higher than those in February. These results can be mainly explained by the effect of temperature on the growth of L. saccharina sporophytes during cultivation. On the other hand, our results show that even in the southern limit of distribution of L. saccharina, the cultivation of this species was very profitable in high yield and quality.  相似文献

Lipid oxidation in salt-dried yellow croaker (Pseudosciaena polyactis) was evaluated during processing with commonly used analytical indices, such as the peroxide value (POV), the thiobarbituric acid reactive substances (TBARS) value, and oxidative-relative lipoxygenase (LOX) activity. Additionally, fatty acids were analyzed using gas chromatography-mass spectrometry. Both POV and TBARS increased significantly (P < 0.05) at the rinsing stage. POV reached its peak value of 3.63 meq O2 per kg sample at the drying stage, whereas TBARS constantly increased from 0.05 to 0.20 mg MDA per kg sample. Processing of salt-dried yellow croaker had an extremely significant (P < 0.01) effect on LOX activity. Twenty-six fatty acids were identified. Combined eicosapentaenoic acid (EPA; C20:5n3) and docosahexaenoic acid (DHA; C22:6n3) content varied between (19.20 ± 0.37) mg g?1 and (23.45 ± 1.05) mg g?1. The polyunsaturated fatty acid/saturated fatty acid (PUFA/SFA) ratio in yellow croaker was 0.73–1.10, and the n-6/n-3 PUFA ratio was approximately 0.13–0.20. The contents of most fatty acids varied significantly (P < 0.05) during the different processing stages, and these differences were caused by lipid oxidation. C18:0, C16:1n7, C19:0, and C22:6n3 showed clear changes in principle component one of a principle components analysis. These fatty acids are potential markers for evaluating lipid oxidation in fish muscle because there was a significant correlation between these markers and TBARS and LOX activity (P < 0.05) with Pearson’s coefficients > 0.931.  相似文献
