Artificial seedling production of Sargassum thunbergii is an effective way to relieve pressure on natural resources.In order to improve the utilization of zygotes and reduce the loss of seedlings,studies on the characteristic of the zygotes release,the development of rhizoids,the attachment of germlings,and the influence of jet washing were conducted.Results show that the percent of zygotes released was increased with time in the first 60 h.The capacity of germlings attached to the substratum was significantly increased,especially coincident with the time of the new rhizoids emerged and elongated.The detachment rate of germlings significantly decreased with the delay of starting time of jet washing or the reduction of jet washing velocity.However,the jet washing at any level applied in the experiment could cause considerable loss of germlings within the 20 days after the attachment.Our study provided some parameters to optimize the operation in the early stage of seedling production.  相似文献

Natural seedling regeneration and tree establishment are affected by various environmental factors.In this study,we established eight,eight,six,and four independent forest stands(each stand was further divided into five subplots) respectively based on the altitudinal gradient,stand density,slope location,and slope aspect to investigate the effects of environmental factors on tree seedling regeneration in a pine-oak mixed forest.The results indicated that the seedling density was significantly higher at altitudes of 1,283 m to 1,665 m,whereas the sapling density did not differ with altitudes.The seedling and sapling density decreased significantly at 1,835 m.The seedling densities on the upper slopes were much higher than those on the middle and lower slopes,whereas the sapling density had no difference.The sapling density decreased gradually from the southwest(20°-75°) whereas it increased on the shady slopes to the northeast(40°).The seedling density increased from southwest(20°) to northeast(40°).The seedling and sapling densities increased with the stand density(850 trees ha-1to 1,525 trees ha-1) whereas the sapling density was significantly lower in stands(1,900 trees ha-1).Principal components analysis showed that the slope aspect and stand density had more important roles in tree regeneration in this study region compared with the other two factors.Therefore,our findings suggest that it will be beneficial to keep stands at a moderate density on shady slopes.Appropriate thinning of higher density stands is also expected to promote the natural regeneration of pine-oak mixed forest.  相似文献

The results showed that T. harzianum concentration had significant effects on seed germination, seedling growth, and seedling survival for all the species with different soil types. Seed germination and survival rates peaked at 5% T. harzianum concentration with leaf mold soil and decreased as T. harzianum concentration increased from 5% to 10%. Seedling survival rates of all four species were generally lowest at 0% T. harzianum concentration in all soil types. The height of F. arundinacea and L. cyrtobotrya peaked at 5% T. harzianum concentration whereas that of D. barbatus and P. tricuspidata peaked at 10% T. harzianum concentration. We concluded that 5% T. harzianum concentration with leaf mold soil is appropriate for seed germination and seedling survival rates of most species, thus enhancing seedling establishment. As seedling culture is a crucial factor for successful cultivation ofGelidium, the authors researched tissue culture technology for producing seedlings. The morphogeny and experimental ecology were observed and studied fully in 2–5 mm isolated tissue fragments. Regeneration, appearance of branching creepers and attaching structure and new erect seedlings production and development were studied. Fragments were sown on bamboo slice and vinylon rope. The seedlings were cultured 20–30 days indoor, then cultured in the sea, where the density of erect seedlings was 3–19 seedlings/cm2, growth rate was 3.84% day. The frond arising from seedlings directly was up to 10 cm per year. The ecological conditions for regenerated seedlings are similar to the natural ones. The regenerated seedlings are suitable for raft culture in various sea areas.  相似文献

In our trials, from 2007 to 2008, of mass production of seedlings of Hizikia fusiformis using synchronization techniques, problems of a "dark thalli" phenomenon and epiphytes contamination severely threatened the health of juvenile seedlings. In this investigation, we optimized conditions for improving the growth of juvenile seedlings. Seven string collectors were seeded with zygotes and a series of experiments were conducted including direct exposure to solar irradiance, co-culture with Ulva spp. and treatment with sodium hypochlorite. It was found that direct exposure to solar irradiance (maximum: 1 740 μmol photons/(m2?s)) for 2 h per day could efficiently enhance the growth of young seedlings and simultaneously inhibit the growth of epiphytic algae. In this treatment, 50-day old seedlings could reach an average of 0.44 cm in length and possess up to nine leaflets. However, a single treatment with 18-mmol/L sodium hypochlorite for 10 min severely harmed 15-day old seedlings. In comparison, weekly treatment with 2.2-mmol/L sodium hypochlorite for 10 min brought no apparent harm to seedlings and eliminated epiphytic algae efficiently. However, this treatment significantly increased the detachment rate of seedlings. Inoculating Ulva spp. onto the collector caused a dramatic decrease in the number of seedlings. However, the growth of the remaining seedlings appeared unhampered. All collectors except the control were daily sprayed with a high pressure water jet from the 8th day post fertilization. From the first day to 50th day, no "dark thallus" was observed on any of the seven collectors. We believe that well-timed daily exposure to solar irradiance would favor H. fusiformis in its early growing stages.  相似文献

Sargassum muticum is one of the most abundant and conspicuous native macroalgae species on the northern coasts of China. It often forms large-scale seaweed beds in subtidal zones. This investigation was designed to understand the intraspecific genetic relationships of this alga based on samples collected from four northern coastal sites of China, and to evaluate gamete release and growth capacity in laboratory conditions. The nuclear internal transcribed spacer 2 (ITS2) sequences of 16 samples from four locations were identical. Based on cox3 gene and partial rbcLS operon sequences, intraspecific genetic variability was detected with three and two ribotypes, respectively. Temperature, not irradiance, was shown to significantly affect gamete release and fertilization. Elevated temperature and irradiance enhanced the growth of germlings and vegetative branchlets. Maximum growth rate of germlings was detected at 18-24℃and an irradiance of 60-100 μmol photons/(m^2.s). Under ambient conditions (12-25℃ and 60-125 gmol photons/ (m2.s)), relative growth rate of young branchlets could reach 7.5%/d.  相似文献

Alpine treeline ecotones are harsh environment for tree establishment due to low temperature.Tree establishment at treelines requires favorable climate,suitable microsites,and viable seeds.But most researches have been addressed treeline microclimate and its effects on tree regeneration,the knowledge of seed quantity and quality and its controls on seedling recruitment were limited.We measured seed rain,soil seed bank,seed germination rate and seedling recruitment in natural forests in combination with seed transplanting manipulation to evaluate the controls of seed quantity and quality on seedling recruitment of Abies georgei var.smithii(smith fir) along altitudinal gradient,withfocus on treeline ecotone in the Sygera Mountains,southeastern Tibetan Plateau.Both seed quantity and seed quality of smith fir decreased with increasing altitude and was thereby associated with decline in seed germination rate.Seed quantity and seedling recruitment were better in north-facing slope than in south slope.The treeline ecotone above 4200 m appeared as the threshold altitude to sharply decrease seed quality and seedling recruitment.The emergence and overwintering rates of transplanting seeds from 3600-3800 m also went down remarkably above 4200 m at north-facing slope.It also underpins the fact that treeline ecotone is the bottleneck of seedling recruitment.Our results suggest that seed quantity and quality are the principal limitation of treeline upward advance.This study also provides evidence tosupport stable treeline position in southeastern Tibetan Plateau.  相似文献

The effects of salinity on the copepod, Acartia tonsa in terms of daily egg production rate (EPR), hatching success, fecal pellet production rate (FPR), naupliar development time and survival, sex ratio, and total life span were determined in laboratory conditions through three experiments. In experiment 1, EPR, hatching success, and FPR of individual females were monitored at salinities of 13, 20, 35 and 45 during short-periods (seven consecutive days). Results show EPR was affected by salinity with the highest outputs recorded at 20 and 35, respectively, which were considerably higher than those at 13 and 45. Mean FPR was also higher in 35 and 20. In experiment 2, the same parameters were evaluated over total life span of females (long-term study). The best EPR and FPR were observed in 35, which was statistically higher than at 13 and 20. In experiment 3, survival rates of early nauplii until adult stage were lowest at a salinity of 13. The development time increased with increasing of salinity. Female percentage clearly decreased with increasing salinity. Higher female percentages (56.7% and 52.2%, respectively) were significantly observed at two salinities of 13 and 20 compared to that at 35 (25%). Total longevity of females was not affected by salinity increment. Based on our results, for mass culture we recommend that a salinity of 35 be adopted due to higher reproductive performances, better feeding, and faster development of A. tonsa.  相似文献

Observation of fluid parcel pathways in the Kuroshio Stream revealed a striking crossstream pattern associated with the Kuroshio‘‘ s meanders. In order to understand the observed pattern, a two-dimensional kinematic model of a meandering jet was developed which could be used to examine the relationship between streamfunction patterns and fluid parcel trajectories. The experiments designed to investigate the dependence of the water exchanges between the Kuroshio water and its surroundings on the characteristics of the Kuroshio‘‘s meanders involved the downstream speed,phase speed and the amplitude of other propagating waves. The results suggested that fluid parcels could cross the meandering jet to and fro; and that the water exchanges between the Kuroshio and its surroundings increased with a) increasing meander amplitude, b) decreasing downstream speed, and c) increasing wave phase speed. The results also showed that when the meandering jet was disturbed by other propagating waves, more effective water exchanges could be induced.  相似文献

Ecotoxicity and bioavailability of cadmium(Cd)to the maize(Zea mays L.)and the soybean(Glycine max(L.)Merr.)were investigated by acute toxicity experiment in the laboratory with black soil.Ecotoxicity and bioavailability of Cd were quantified by calculating the median effective concentration(EC50)and bioaccumulation factor(BAF).The measurement endpoints used were seed germination and seedling growth(shoot and root).The results showed that concentrations of Cd in the soil had adverse effect on the growth of roots and shoots.Seed germination was not the sensitive indicator for the ecotoxicity of Cd in the soil,while the growth of roots was the most sensitive measurement endpoint.Absorbability and transport of Cd in plants depended on the test crop species and Cd concentrations in the soil.The maize retains more Cd in its roots,while the soybean transports more Cd to the shoots from roots.  相似文献

The manipulation of the chromosome set for commercially valuable marine animals is important for enhancing aquacultural production. In this study, triploid and tetraploid sea cucumber Apostichopus japonicus were induced by hydrostatic pressure shock, and the conditions of appropriate induction were tested with different starting times, and hydrostatic pressure intensities and durations. The highest rate of triploid induction reached 20% and that of tetraploid was 60%. In consideration of the survival rate and hatch rate, the appropriate treatment for triploid was 55 Mpa of hydrostatic pressure for 5 min at 55 min after fertilization (a.f.), while for tetraploid it was 60 Mpa for 5 min at 61 min a.f. The triploid of the sea cucumber could survive through the pelagic larval stage and attachment stage, and develop like the control group of the experiment. The tetraploid, however, could not survive the attachment stage.  相似文献

Scallop Chlamys farreri was exposed to different concentrations of benzo(a)pyrene (BaP) (0.5 μg/L, 1.0 μg/L, 10.0 μg/L and 50.0 μg/L) for 30 days in seawater. The 7-ethoxyresorufin O-deethylase (EROD) activity was significantly induced, and increased with the increasing BaP concentration. The glutathione-S-transferase (GST), superoxide dismutase (SOD), catalase (CAT), Glutathione peroxidase (GPx) activities increased in short time at low concentration of BaP, and was significantly depressed at high concentrations. Scallop gill was more sensitive to BaP than the digestive gland, and the digestive gland was the main tissue to deal with oxyradicals. The contents of malondialdehyde (MDA) increased with the exposure time and there was a positive correlation (concentration-effect) between the MDA content and the concentration of BaP. The biomarkers determined in this experiment had important roles in detoxification, and showed great potential as biomarkers for oxidative stress. Controlled laboratory experiments designed to simulate field exposure scenarios are particularly useful in ascertaining biomarkers suitable for use with complex contaminant mixtures in the marine environment. Heterotrophic culture of microalgae to develop methods of increasing biomass productivity and storage lipids has brought new insight to commercial biodiesel production. To understand the relationship between heterotrophy and lipid production, the effects of carbon sources on the growth and lipid accumulation of Chlorella sorokiniana GXNN01 was studied. The alga exhibited an increased growth rate in response to the addition of carbon sources, which reached the stationary phase after 48 h at 30°C. In addition, glucose and NaAc had a significant effect on the lipid accumulation during the early-stationary phase. Specifically, the lipid content was 0.237±0.026 g g−1 cell dry weight and 0.272±0.041 g L−1 when glucose was used as the carbon source, whereas the lipid content reached 0.287±0.018 g g−1 cell dry weight and 0.288±0.008 g L−1 when NaAc was used as the carbon source. The neutral lipid content was found to first decrease and then increase over time during the growth phase. A glucose concentration of 20 mmol L−1 gave the maximal lipid yield and the optimum harvest time was the early-stationary phase.  相似文献

【目的】探索不同附着基质对长茎葡萄蕨藻(Caulerpalentillifera)直立枝生长及藻体附着效果的影响。【方法】对长茎葡萄蕨藻直立枝以垂直砂埋、水平砂埋、塑料网网夹、尼龙网网夹及无附着基质(对照组)五种附着方式进行实验,探究长茎葡萄蕨藻直立枝的生长、附着能力、光合特性参数及营养含量的变化。【结果】垂直砂埋处理下,藻体的新生直立枝质量、质量长度比及球状小枝的直径与其他各组相比最优(P <0.05),分别是0.313 3 g、0.086 6 g/mm和1.67 mm,是对照组的7.38、9.62、1.67倍;水平砂埋与对照组相比,藻体附着能力有显著增强(P <0.05),假根数为26,长度为30.77 mm及假根附着砂砾质量为0.566 0 g;除对照组外,各组最大光能转化效率(Fv/Fm)和实际光化学效率Y(II)值呈先下降后上升趋势,24 h后垂直沙埋藻体Fv/Fm和Y(II)最快恢复原始水平,为0.856μmol·m~(-2)·s~(-1)和0.851μmol·m~(-2)·s~(-1),显著高于其余组(P <0.05);垂直砂埋与水平砂埋处理后的藻体,其可溶性糖含量分别比对照组显著降低34.24%和33.93%(P<0.05),而网夹处理后,没有显著差异(P> 0.05)。垂直砂埋处理对藻体总蛋白含量增加有显著促进作用,是对照组的1.26倍(P <0.05)。【结论】长茎葡萄蕨藻直立枝以垂直砂埋附着方式生长最佳,以水平砂埋附着方式附着更牢固。  相似文献

Place attachment is an important motivation for people to spend more time outdoors and to protect landscapes. This study explores visitors’ intention to conserve natural landscapes based on the relationship with their place attachment to National Park landscape. Structural equation modelling (SEM) was used to determine the relationship between landscape conservation and place attachment. A survey with a structured questionnaire was administered to visitors to the seven designated hiking courses of Harz National Park in Germany. The path coefficient of 0.77 revealed that place dependence positively and significantly affected place attachment, whereas place identity did not. Place attachment had a significant effect on both affective appraisals and visiting satisfaction. Higher place attachment led to higher emotional reaction to landscapes on site and higher satisfaction of visiting the park. Among the variables, visiting satisfaction, but not affective appraisals, played a statistically significant mediating role between place attachment and conservation intention. With a path coefficient of 0.86, conservation intention was highly affected by visiting satisfaction. These results suggest that the managers of National Parks should focus on increasing visiting satisfaction based on how visitors are emotionally bonded with their visiting places, in order to enhance the intentions to conserve the landscape of the visitors to National Parks.  相似文献

A filamentous clone of Polysiphonia urceolata was regenerated from segments cut from the fronds of gametophytes. Unlike wild thalli with short virgate branchlets, the clone was filamentous with few branches. Many transparent trichoblasts arose from pericentral cells during the induction culture, but these were seldom observed during normal growth. The trichoblasts were uniseriate, often colorless, and formed lobed rhizoids rapidly when they came into contact with solid substrates. In addition to morphological characteristics, the photosynthetic properties and growth conditions of the clone differed from those of the mother plant. Cross-gradient light and temperature culture experiments revealed that the most favorable conditions for culture of the filamentous clone were 22°C and 95-120 μE/(m2-s) light intensity. The photosynthetic light saturation value for filaments was approx. 100 μE/(m2-s), which is far lower than that of wild thalli. These results could be used to develop techniques for mass cultures of P. urceolata in photobioreactors for production of seed stock or bioactive products.  相似文献
