The aim of this study was to examine whether a combination of biochemical, histopathological and toxicogenomic data could be used as a valuable tool for the assessment of biological risk associated with pollutants within the Tamar River and Estuary, S.W. England, U.K. Accordingly, biochemical and histopathological biomarkers (protein carbonyls, lipofuscin, neutral lipids, lysosomal stability [N-acetyl-β-hexosaminidase and neutral red], lysosomal volume, ferric reducing antioxidant power [FRAP] and malonaldehyde [MDA]) and gene expression profiles were assessed in 5 sites from the Tamar River and Estuary (Neal Point, Town Quay, Wilcove, Cremyll Ferry and Whitsand; and a reference site, Trebarwith Strand, N. Cornwall). PAHs were measured in mussel tissue and sediment and metals were measured in mussel tissue only. Data from the biomarkers was integrated into a Mussel Expert System (MES) model to produce a simple assessment of mussel stress. Clear gradients of mussel toxicity were identified by the biomarkers (with the exception of neutral lipids) with the highest impacted animals found furthest up the Tamar, whilst the MES was unable to identify a gradient of effect. Gene expression profiles also indicated a gradient of stress with the greatest number of significantly up- or down- regulated genes found at the uppermost 2 sites. The MES did, however, determine that mussels from all sites, except the reference site, were highly stressed; a conclusion that could not be inferred from the biomarker data alone. It is concluded that the MES is a valuable tool that permits integration and interpretation of complex sets of biomarker data by identifying the biological meaning of biomarker changes.  相似文献

This study addresses the need for women risk communication and highlights the potential role of Women Welfare Associations (WWAs) of Bandung, Indonesia, as risk communicators. A risk communication framework is modeled for women’s risk communication process. A set of indicators in social, institutional, and economic resilience activities (SIERAs), with a scope of 45 activities covering three different disaster periods, were developed to characterize the delivery process of risk information by women in WWAs through their activities at sub-district and ward levels. The data were collected through a questionnaire survey method using the risk communication SIERA approach. Women’s leaders at wards were surveyed concerning their perceptions on these 45 scopes of SIERA, ongoing activities, and their risk information source and dissemination process. Correlation analysis was applied to determine the relationship between the variables such as periods of disaster, types of activities (social, institutional, economic), and attributing factors (location, population, and education institution) in finding variations in risk communication activity that functions for women and communities. Five risk communication processes of WWAs are identified and implemented that work for women in Bandung. When their perceptions and ongoing activities are compared, activities such as dissemination of disaster risk information, conveying early warnings to their peers, and involvement of the local government have been confirmed to match the risk communication plans and implementation of WWAs. These indicate that WWAs’ activities in Bandung implement a certain degree of risk communication that is embedded in their activities. The results confirm that women through their social networks can become active agents of change and thus act beyond their usual domestic roles and responsibilities in order to contribute to the overall enhancement of community resilience.

Mytilus edulis digestive gland microsomes were prepared from indigenous populations sampled from a clean reference site (Port Quin) and an urban-industrial contaminated site (Blackpool) in the UK. Samples were collected in March/April, May, August and December 1998. Western blot analysis was performed using polyclonal antibodies to fish CYP1A and rat CYP2E using partially purified M. edulis CYP as a positive control, to aid identification. CYP1A- and CYP2E-immunopositive protein levels showed different site-specific seasonal variation with higher levels of CYP2E determined in May (P < 0.05). At both sites, lower levels of CYP1A-immunopositive protein but not CYP2E-immunopositive protein were observed in the samples collected in December (P < 0.05). This correlated with lower levels of nuclear DNA damage (Comet assay expressed as per cent tail DNA) observed in December compared to August (P < 0.05).  相似文献

Metallothioneins (MTs) are small, cysteine-rich proteins which detoxify xenobiotic metals such as cadmium (Cd) and mercury (Hg). In crustaceans and mammals they consist of two independent domains which are folded around metal-thiolate clusters. MT clusters of different origins, exhibiting distinct, highly conserved cysteine positions on their sequences, show differences in metal-cysteine coordination and reactivity. Lobster-MT, containing two Cd3 beta domains, is an important model for structure-function relationships among the clusters. The influence of (1) the position of the cysteine residues and (2) steric and electrostatic effects of neighboring amino acids on the folding and stability of MT cluster were investigated. Thus, the native lobster beta C and beta N domains (each having nine cysteines and binding three M2+ ions) and a modified domain Cd3 beta C-->N, in which the cysteines of the C-terminal domain were relocated to match the positions of those in the N-terminal domain, were chemically prepared and characterized. The synthetic native domains (Cd3 beta C and Cd3 beta N) were found to exhibit spectroscopic properties, metal-binding affinities and kinetic reactivity similar to the holo-protein. However, the modified Cd3 beta C-->N domain was unusually reactive and in the presence of Chelex, metal chelation resin, aggregated to a Cd5(beta C-->N)2 dimer, which exhibited unusual structure as observed by its 113Cd-nuclear magnetic resonance. These differences in structure and reactivity demonstrated that the requirements for formation of a stable Cd3S9 beta-cluster are more stringent than simply the sequential positions of the cysteines along the peptide chain and must include interactions involving neighboring, noncysteine amino acids.  相似文献

Abandoned mining operations continue to severely degrade many ecosystems worldwide by releasing acidic water and/or heavy metals into surface and groundwater. Contaminant concentrations in affected streams vary with discharge in patterns that reflect both geochemical reactions and variable mixing of contaminated and non-contaminated waters. However, controls on concentration-discharge (C-Q) patterns remain unclear, particularly for constituents that experience changing solubility across redox and pH gradients. Understanding the C-Q behaviour of contaminants aids in predicting both downstream transport and effects on aquatic life under variable flow. Here, we examined the C-Q behaviours of non-reactive (Na, K, Ca, Mg, Cl) and reactive (Fe, Mn, Al, H+, SO42−) solutes in a stream contaminated with acid mine drainage in northeastern Ohio, USA. Concentration-discharge patterns at the watershed outlet primarily reflected mixing of contaminated baseflow with intermittent inputs of high pH water draining from a passive limestone treatment system into the stream. The treatment system acted as an ephemeral tributary that mitigated contamination in the stream by diluting solutes, raising pH, and driving metal precipitation, but only when flow was present during wet seasons. Consequently, AMD-derived reactive solutes (H+, Fe, Mn, Al) decreased with increasing stream discharge while relatively conservative solutes (e.g., Ca, Mg, K, Na) decreased only slightly or were chemostatic. This study highlights both the unique C-Q patterns of reactive solutes when compared to those of non-reactive solutes and the potential for intermittent streams to control C-Q behaviour in headwater catchments.  相似文献
