We are investigating the effects of in vivo exposure of prototypical enzyme inducing agents on hepatic biotransformation enzyme expression in largemouth bass (Micropterus salmoides), a predatory game fish found throughout the United States and Canada. The current study targeted those genes involved in biotransformation and oxidative stress that may be regulated by Ah-receptor-dependent pathways. Exposure of bass to β-naphthoflavone (β-NF, 66 mg/kg, i.p.) elicited a 7–9-fold increase in hepatic microsomal cytochrome P4501A-dependent ethoxyresorufin O-deethylase (EROD) activities, but did not affect cytosolic GST catalytic activities toward 1-chloro-2,4-dinitrobenzene (CDNB) or 5-androstene-3,17-dione (ADI). Glutathione S-transferase A (GST-A) mRNA expression exhibited a transient, but non-significant increase following exposure to β-NF, and generally tracked the minimal changes observed in GST–CDNB activities. Expression of the mRNA encoding glutamate-cysteine ligase catalytic subunit (GCLC), the rate-limiting enzyme in glutathione (GSH) biosynthesis, was increased 1.7-fold by β-NF. Changes in GCLC mRNA expression were paralleled by increases in intracellular GSH. In summary, largemouth bass hepatic CYP1A-dependent and GSH biosynthetic pathways, and to a lesser extent GST, are responsive to exposure to β-NF.  相似文献

The glutathione S-transferases (GST) are a major group of conjugative enzymes involved in the detoxification of electrophilic compounds and products of oxidative stress. We have previously described the kinetics of hepatic GST conjugation in largemouth bass using a variety of synthetic GST reference substrates. In the present study, we investigated the ability of largemouth bass hepatic GSTs to conjugate 4-hydroxynon-2-enal (4HNE), a mutagenic and cytotoxic alpha-beta-unsaturated aldehyde produced during oxidative injury. Hepatic cytosolic fractions from largemouth bass rapidly catalyzed GSH-dependent 4HNE conjugation, with the rate of GST-4HNE conjugation in bass liver exceeding those of several other mammalian and aquatic species. No apparent sex-related differences in GST-4HNE activity were observed among adult bass. SDS-PAGE and Western blotting analysis of GSH affinity-purified bass liver cytosolic GST revealed the presence of two major GST subunits of approximately 30 and 27 KDa that exhibited slight cross-reactivity when probed with a rat alpha class GST antibody, but not to rat mu, pi or theta class GST. The rapid conjugation of 4HNE by hepatic GST suggests an important role for GSTs in protecting against peroxidation of polyunsaturated fatty acids in bass liver.  相似文献

A thick sequence of late Dinantian (Asbian–Brigantian) carbonates crop out in the Buttevant area, North Co. Cork, Ireland. A mud-mound unit of early Asbian age (the Hazelwood Formation) is the oldest unit described in this work. This formation is partly laterally equivalent to, and is overlain by, over 500 m of bedded platform carbonates which belong to the Ballyclogh and Liscarroll Limestone Formations. Four new lithostratigraphic units are described within the platform carbonates: (i) the early Asbian Cecilstown Member and (ii) the late Asbian Dromdowney Member in the Ballyclogh Limestone Formation; (iii) the Brigantian Templemary Member and (iv) the Coolbane Member in the Liscarroll Limestone Formation. The Cecilstown Member consists of cherty packstones and wackestones that are inferred to have been deposited below fair-weather wavebase. This unit overlies and is laterally equivalent to the mud-mound build-up facies of the Hazelwood Formation. The Dromdowney Member is typified by cyclic-bedded kamaenid-rich limestones possessing shell bands, capped by palaeokarst surfaces, with alveolar textures below and shales above these surfaces. The carbonates of this unit were deposited at or just below fair-weather wavebase, the top of each cycle culminated in subaerial emergence. The Templemary Member consists of cyclic alternations of subtidal crinoidal limestones capped by subtidal lagoonal crinoid-poor, peloidal limestones possessing coral thickets. Intraclastic cherty packstones and wackestones characterize the Coolbane Member, which is inferred to have been deposited below fair-weather wavebase but above storm wavebase. The early Asbian Cecilstown Member has a relatively sparse micro- and macrofauna, typified by scattered Siphonodendron thickets, archaediscids at angulatus stage and common Vissariotaxis. Conversely, macro- and microfauna is abundant in the late Asbian Dromdowney Member. Typical late Asbian macrofossils include the coral Dibunophyllum bipartitum and the brachiopod Davidsonina septosa. The base of the late Asbian (Cf6γ Subzone) is recognized by the first appearance of the foraminifers Cribrostomum lecompteii, Koskinobigenerina and the alga Ungdarella. The Cf6γ Subzone can be subdivided into two biostratigraphic divisions, Cf6γ1 and Cf6γ2, that can be correlated throughout Ireland. Relatively common gigantoproductid brachiopods and the coral Lonsdaleia duplicata occur in the Brigantian units. The base of the Brigantian stage (Cf6δ Subzone) is marked by an increase in the abundance of stellate archaediscids, the presence of Saccamminopsis-rich horizons, Loeblichia paraammonoides, Howchinia bradyana and the rarity of Koninckopora species. Changes in facies at the Cecilstown/Dromdowney Member and the Ballyclogh/Liscarroll Formation boundaries coincide closely with the changes in fossil assemblages that correspond to the early/late Asbian and the Asbian/Brigantian boundaries. These facies changes are believed to reflect major changes in relative sea-level on the Irish platforms. The sea-level variations that are inferred to have caused the facies changes at lithostratigraphic boundaries also brought in the new taxa that define biostratigraphic boundaries. Moreover, many of the Dinantian stage boundaries that are defined biostratigraphically in Great Britain, Belgium and the Russian Platform also coincide with major facies boundaries caused by regressive and transgressive episodes. The integration of detailed biostratigraphic analyses with facies studies will lead to better stratigraphic correlations of Dinantian rocks in northwest Europe. © 1997 John Wiley & Sons, Ltd.  相似文献

Brown bullheads (Ameriurus nebulosus) are a demersal freshwater species that can be found in a number of polluted ecosystems. The purpose of the present study was to determine the overall capacity for in vitro glutathione S-transferase (GST) detoxification by brown bullheads, and to see if bullhead GST catalysis was altered in bullheads from a polluted site. Brown bullhead liver cytosolic GSTs catalyzed the conjugation of 1-chloro-2,4-dinitrobenzene (CDNB) over a large range of substrate concentrations, with apparent Km and Vmax for CDNB at fixed nucleophile (glutathione, GSH) concentrations of 1.8-1.9 mM and 12.1-14.6 mumol CDNB conjugated/min/mg, respectively. Bullhead GSTs were also highly active toward other substrates such as ethacrynic acid (ECA), delta 5-androstene-3,17-dione (ADI), and nitrobutyl chloride (NBC). Initial rate GST catalytic activities toward CDNB, NBC, ECA, and ADI were significantly lower in female bullheads from a contaminated lake (Lake Apopka Marsh) as compared to female bullheads inhabiting a nearby control site (Lake Woodruff). No site differences were observed with respect to male bullhead GST activities. These studies suggest that brown bullheads efficiently carry out GST conjugation of diverse electrophilic substrates. However, bullhead GST catalysis may be compromised in bullheads inhabiting polluted ecosystems.  相似文献

Exploration in the basalt covered areas of the Faroes offshore has always suffered from poor seismic imaging below the basalt. Long offset 2D and 3D seismic data were acquired and a significant improvement in the seismic image below top basalt has been achieved. Deep towing of the source and receiver cables helped by extending the seismic bandwidth towards lower frequencies. Bubble‐tuned rather than conventional peak‐tuned source arrays gave little, if any, incremental benefit. The improvement in the imaging comes primarily from the approach to processing the data. High frequencies (dominantly noise) are filtered out of the data early in the processing to concentrate on the low frequency data. Careful multiple removal is important with several passes of demultiple being applied to the data using both Surface‐Related Multiple Elimination (SRME) and Radon techniques. Velocity analysis is performed as an iterative process taking into account the geological model. Reprocessing legacy 2D surveys, acquired with wide‐ranging parameters, using these processing techniques improved these datasets significantly, indicating that sub‐basalt imaging seems to be more sensitive to processing than to the choice of acquisition parameters.  相似文献

The South African plateau is bordered by passive margin basins preserving the terrigeneous sediment produced during onshore erosion. As such, these basins potentially provide a record of the variation in onshore elevation and relief over time. Here we bring new constraints on the uplift and erosion of the Southern African plateau over the last 150 Ma from the perspective of the stratigraphic architecture of these basins. We review published data to quantify the terrigeneous supply eroded in the drainage area and preserved in the basins. The novel aspect of our approach is the integration of the evolution of the whole domain in sedimentation (i.e. not only the platform) as well as the onshore eroding region.  相似文献

We present an overview of Markov chain Monte Carlo, a sampling method for model inference and uncertainty quantification. We focus on the Bayesian approach to MCMC, which allows us to estimate the posterior distribution of model parameters, without needing to know the normalising constant in Bayes' theorem. Given an estimate of the posterior, we can then determine representative models (such as the expected model, and the maximum posterior probability model), the probability distributions for individual parameters, and the uncertainty about the predictions from these models. We also consider variable dimensional problems in which the number of model parameters is unknown and needs to be inferred. Such problems can be addressed with reversible jump (RJ) MCMC. This leads us to model choice, where we may want to discriminate between models or theories of differing complexity. For problems where the models are hierarchical (e.g. similar structure but with a different number of parameters), the Bayesian approach naturally selects the simpler models. More complex problems require an estimate of the normalising constant in Bayes' theorem (also known as the evidence) and this is difficult to do reliably for high dimensional problems. We illustrate the applications of RJMCMC with 3 examples from our earlier working involving modelling distributions of geochronological age data, inference of sea-level and sediment supply histories from 2D stratigraphic cross-sections, and identification of spatially discontinuous thermal histories from a suite of apatite fission track samples distributed in 3D.  相似文献
