Zebrafish (Danio rerio) were exposed with 17β‐estradiol (E2) and nonylphenol (NP) in different concentrations. Gene expression analysis was carried out by two different approaches. First, RT‐PCR experiments were performed for the examination of expression levels of the two marker genes vitellogenin and aromatase. This approach showed a significant increase in the expression of the vitellogenin gene in exposed male fish (500 ng/L 17β‐estradiol and 250 μg/L nonylphenol). This egg yolk protein is usually not synthesized in male vertebrates. A slight decrease of expression of the aromatase gene was observed in exposed female zebrafish. Aromatase is known to catalyze the conversion of androgens to estrogens. Second, DNA microarray experiments were carried out, which allow the simultaneous examination of the expression levels of a great number of marker genes. The microarray experiments resulted in an up‐regulation of vitellogenin up to 850‐fold. In addition, several other genes were identified to be up‐regulated by estrogens, for example the high mobility group box protein ssrp1 (78‐fold) or the chaperonin containing t‐complex polypeptide 1, beta subunit cctb (22‐fold).  相似文献

Radionuclides released to the environment and deposited with or onto snow can be stored over long time periods if ambient temperature stays low, particularly in glaciated areas or high alpine sites. The radionuclides will be accumulated in the snowpack during the winter unless meltwater runoff at the snow base occurs. They will be released to surface waters within short time during snowmelt in spring. In two experiments under controlled melting conditions of snow in the laboratory, radionuclide migration and runoff during melt‐freeze‐cycles were examined. The distribution of Cs‐134 and Sr‐85 tracers in homogeneous snow columns and their fractionation and potential preferential elution in the first meltwater portions were determined. Transport was associated with the percolation of meltwater at ambient temperatures above 0 °C after the snowpack became ripe. Mean migration velocities in the pack were examined for both nuclides to about 0.5 cm hr?1 after one diurnal melt‐freeze‐cycle at ambient temperatures of ?2 to 4 °C. Meltwater fluxes were calculated with a median of 1.68 cm hr?1. Highly contaminated portions of meltwater with concentration factors between 5 and 10 against initial bulk concentrations in the snowpack were released as ionic pulse with the first meltwater. Neither for caesium nor strontium preferential elution was observed. After recurrent simulated day‐night‐cycles (?2 to 4 °C), 80% of both radionuclides was released with the first 20% of snowmelt within 4 days. 50% of Cs‐134 and Sr‐85 were already set free after 24 hr. Snowmelt contained highest specific activities when the melt rate was lowest during the freeze‐cycles due to concentration processes in remaining liquids, enhanced by the melt‐freeze‐cycling. This implies for natural snowpack after significant radionuclide releases, that long‐time accumulation of radionuclides in the snow during frost periods, followed by an onset of steady meltwater runoff at low melt rates, will cause the most pronounced removal of the contaminants from the snow cover. This scenario represents the worst case of impact on water quality and radiation exposure in aquatic environments.  相似文献

Protistan community structure was examined from 6 depths (1.5, 20, 42, 150, 500, 880 m) at a coastal ocean site in the San Pedro Channel, California. A total of 856 partial length 18S rDNA protistan sequences from the six clone libraries were analyzed to characterize diversity present at each depth. The sequences were grouped into a total of 259 Operational Taxonomic Units (OTUs) that were inferred using an automated OTU calling program that formed OTUs with approximately species-level distinction (95% sequence similarity). Most OTUs (194 out of 259) were observed at only one specific depth, and only two were present in clone libraries from all depths. OTUs were obtained from 21 major protistan taxonomic groups determined by their closest BLAST matches to identified protists in the NCBI database. Approximately 74% of the detected OTUs belonged to the Chromalveolates, with Group II alveolates making up the largest single group. Protistan assemblages at euphotic depths (1.5, 20 and 42 m) were characterized by the presence of clades that contained phototrophic species (stramenopiles, chlorophytes and haptophytes) as well as consumers (especially ciliates). Assemblages in the lower water column (150, 500 and 800 m) were distinct from communities at shallow depths because of strong contributions from taxa belonging to euglenozoans, acantharians, polycystines and Taxopodida (Sticholonche spp. and close relatives). Species richness (Chao I estimate) and diversity (Shannon index) were highest within the euphotic zone and at 150 m, and lowest for protistan assemblages located in the oxygen minimum zone (500 and 880 m). Multivariate analyses (Bray-Curtis coefficient) confirmed that protistan assemblage composition differed significantly when samples were grouped into shallow (≤150 m) and deep water assemblages (≥150 m).  相似文献

This study investigates mechanisms and nonlinearities in the response of the Asian Summer Monsoons (ASM) to high-latitude thermal forcings of different amplitudes. Using a suite of runs carried out with an intermediate-complexity atmospheric general circulation model, we find that the imposed forcings produce a strong precipitation response over the eastern ASM but a rather weak response over the southern ASM. The forcing also causes a precipitation dipole with wet conditions over the eastern Tibetan Plateau (TP) and dry conditions over the Bay of Bengal (BoB) and southeast Asia. A moderate increase of precipitation along the southern margin of the TP is also produced. Simulations designed to isolate the causal mechanisms show that thermodynamic interactions involving the tropical surface oceans are far less important than the water-vapour feedback for the transmission of information from the high-latitudes to the ASM. Additionally, we assess the nonlinearity of the ASM precipitation response to the forcing amplitude using a novel application of the empirical orthogonal function method. The response can be decomposed in two overlapping patterns. The first pattern represents a precipitation dipole with wet conditions over the eastern TP and dry conditions over BoB, which linearly increases with forcing amplitude becoming quasi-stationary for large forcing amplitudes (i.e. amplitudes leading to Arctic temperature anomalies larger than 10 °C). The second pattern is associated with increased precipitation over the southeastern TP and is nonlinearly dependent on forcing, being most important for intermediate forcing amplitudes (i.e. amplitudes leading to Arctic temperature anomalies between 5 and 10 °C).  相似文献

This paper presents the basis for the design of streamflow prediction systems for the hydroelectric dams of Rincón del Bonete (Uruguay) and Salto Grande (Uruguay–Argentina). The prediction is made, independently, for each reservoir and each month of the year with two methodologies: data-driven statistical models and hybrid downscaling that includes atmospheric predictors. We determine a set of potential predictors and then fit linear models coupled with variable selection techniques, under the hypothesis of perfectly known predictors. The predictive skill of the schemes outperforms the climatological forecast throughout the year in both reservoirs (except August in Rincón del Bonete). This remains the case even when the forecast lead does not allow for the use of preceding flows as predictors. While in Rincón del Bonete it is not possible to distinguish a period of high predictability, in Salto Grande, there is a robust signal in March–May and October–December.  相似文献

The general relativistic model of Cooperstock and Tieu, which attempts to fit rotation curves of spiral galaxies without invoking dark matter, is tested empirically using observations of the Milky Way. In particular, predictions for the mass density in the solar neighbourhood and the vertical density distribution at the position of the Sun are compared with observations. It is shown that the model of Cooperstock and Tieu, which was so constructed that it gives an excellent fit of the observed rotation curve, singularly fails to reproduce the observed local mass density and the vertical density profile of the Milky Way.  相似文献

Hydraulic fracturing is increasingly being used to produce gas from unconventional resource sites for energy supply. Therefore, concerns about risks of this technology related to human health and the environment have to be addressed. Among the major issues is the potential contamination of surrounding water systems by chemical additives used in fracturing fluids. In this study, the ecotoxicological hazards of fracturing fluids, both, their individual components (chemicals) as well as their mixtures (product) were assessed using a component-based mixture approach. For five exemplary fracturing fluids, 40–90 wt% of the contained substances could unambiguously be defined in their chemical identity. The concentrations used in the applied fluid mixture were considered as (maximum) exposure concentrations. For components with mass fractions between 10 and 74 wt%, the effect concentrations for acute and chronic toxicity of fish, daphnia and algae were retrieved from experimental databases and through predictive modeling. The hazard indices calculated from the ratio of exposure to effect concentration were >1 for all fracturing fluids, using different scenarios. This indicated a hazard from the undiluted fracturing fluids. The assessment framework presented in this study allows for dealing with data gaps and uncertainties in a tiered fashion and in particular accommodates for combined effects resulting from chemical mixtures. It might be employed for ecotoxicological risk assessment of products containing chemical mixtures and optimization of their environmental performance.  相似文献
