Polychlorinated biphenyls (PCBs) are known as neurotoxic chemicals and possibly alter animal behavior. We previously reported that PCB-exposure induced abnormal schooling behavior in Japanese medaka (Oryzias latipes). This abnormal behavior might be caused by the functional alteration of central or terminal nervous system. To understand the mechanism(s) of behavioral change by PCB-exposure, we analyzed the gene expression profiles in the brain of medaka exposed to 3,3′,4,4′,5-pentachlorobiphenyl (PCB126) or a PCB mixture (Kanechlor-400: KC-400) using a cDNA microarray that we constructed. Twelve FLF-II strain medaka (six individuals per treatment) were dietary exposed to PCB126 (0.01 μg/g b.w./day) or KC-400 (1 μg/g b.w./day) for three weeks. For the control, six fish were fed a control diet. After the exposure period, fish were dissected, and the brain samples were collected. The samples from control fish were pooled and used as a common reference in the microarray experiment. Microarray data were normalized by the LOWESS method, and we screened the genes whose expression levels were altered more than 1.5-fold. Gene expression profiling showed 97 down-regulated and 379 up-regulated genes in the brain of medaka exposed to PCB126. KC-400 exposure suppressed 15 genes and induced 266 genes in medaka brain. Among these genes, the expression levels of 7 and 188 genes were commonly down- or up-regulated, respectively in both treatment groups. On the other hand, 31 gene expressions were significantly different between PCB126 and KC-400 treatment groups, and three out of 31 genes were received opposite effects. In addition, the microarray data showed that thyroid hormone-responsive genes were up-regulated by PCB-exposure, which may imply that PCBs or their metabolites mimic thyroid hormone effects in the brain of PCB-exposed medaka.  相似文献

The results of subsurface gas monitoring by application of gas chromatography (GC) to the gas composition of bubbles associated with groundwater for seismogeochemical studies are reported. An automated gas monitoring system was used to determine gas compositions in a 500-m borehole at the Haruno Crustal Movement Observation Site (HOS), central Japan during period 1, from December 1999 to December 2000. The average ± two standard deviation (2SD) compositions of gases in this period were He = 82 ± 29 ppmV, H2 = 170 ± 62 ppmV, Ar = 0.05 ± 0.07%, N2 = 50 ± 8%, and CH4 = 45 ± 6%. A new automated gas monitoring system equipped with a micro-GC was installed in the borehole at the HOS, and gas bubbles from the borehole were monitored during period 2, from December 2006 to March 2007. The average ± two standard deviation (2SD) compositions of gases in this period were He = 8 ± 7 ppmV, H2 = 13 ± 15 ppmV, Ar = 0.6 ± 0.3%, N2 = 66 ± 7%, and CH4 = 14 ± 14%. The gas concentration ratios (He/Ar, H2/Ar, N2/Ar, and CH4/Ar) fluctuated significantly over time and repeatedly showed abrupt spike-like increases during period 2. The gas compositions obtained in period 1 and 2 were markedly different. Over the period from 2006 to 2007, the gas bubbles were depleted in He, H2, and CH4 of deep origin, but enriched in Ar and N2 of atmospheric origin. This difference can be interpreted as being due to an irreversible change of the aquifer/gas system. The present deep component in the HOS gas is estimated to have composition He = 63 ppmV, H2 = 37 ppmV, Ar = 0.17%, N2 = 63%, and CH4 = 37%. The new monitoring system is able to analyze the gas composition using a smaller volume of sample gas and with greater precision than the previous system. During the 3-month monitoring period 2, the separation capacity of the capillary column of the micro-GC was sufficiently maintained to determine gas-chromatographic peak areas for the five gaseous species examined. This study confirms that the new monitoring system with micro-GC is promising for continuous subsurface gas monitoring for earthquake prediction studies.  相似文献

Effects of spilled oil on microbial communities in tidal flats were examined by use of a simulator for a tidal flat ecosystem. The simulator is composed of a wave generator, a tide control device, and a tidal flat. Sediment for the tidal flat was obtained at a natural tidal flat in Hiroshima Bay, Japan. After stabilizing the benthic organisms, fuel oil C was added to the surface of the flat at 1 lm(-2). Although the total number of micro-organisms remained at 1.5-3.5 x 10(9) cells g(-1) dry sediment irrespective of the addition of oil, bacterial communities which were analyzed based on the 16S rDNA showed clear changes after the addition of fuel oil C and after a subsequent recovery period. Bacterial colonies were randomly isolated from the oil-supplemented sediment during the experiments, and the isolates were examined for susceptibility to hydrocarbons in order to screen the oil-susceptible bacteria. The proportion of oil-susceptible bacteria in the isolates decreased with the addition of the oil. Oil-susceptible bacteria showed an inability to assimilate petroleum compounds as well as an inhibition of growth. The possibility of using oil-susceptible bacteria as an indicator of bioremediation in tidal flats was discussed.  相似文献

The pharyngeal dental formula ofMylopharyngodon piceus is 4–5 as a rule, and the dentition is asymmetrical. It is difficult to identify each tooth in the larval dentition. In this paper the appearance pattern of tooth germ with development process in this fish is described in detail. The formation pattern of the left dentition is contrasted with that of the right one. In the developmental process, the left pharyngeal dentition lacks teeth at position An3. Thus the left dentition is D-type as designated by Nakajima (1984), while the right one is A-type. We report a new observation of the olivine B-type lattice-preferred orientation (LPO), from the garnet peridotite at Cima di Gagnone, Switzerland. The olivine B-type fabric forms at low temperatures and/or high stress in the presence of water, and is of particular interest because it may be used to explain the trench-parallel shear-wave splitting that is often observed at subduction zones. In conjunction with the olivine B-type fabric, we have found strong orthopyroxene LPO that is identical to those formed under water-free conditions. This suggests that water may not have a significant effect on orthopyroxene fabric. From the olivine microstructure, we determine that a stress of 22 ± 8 MPa was applied during the deformation event that formed the olivine LPO. Using an olivine flow-law, and assuming geological strain-rates, we determine the temperature of deformation to be 800 ± 175°C. This does not preclude an ultra-deep origin for the ultramafic rocks at Cima di Gagnone, but indicates that much of the deformation recorded in the microstructure occurred at modest temperatures.Communicated by T.L. We analyzed responses of soluble reactive phosphorus (SRP), bioavailable phosphate (PO4), particulate phosphorus, turnover time of orthophosphate (Tt), and alkaline phosphatase activity (APA) to varying degrees of nutrient stress. The nutrient stress was evoked by different treatments in concentration and combination of inorganic nitrogen (N) and phosphorus (P), and labile organic carbon (glucose) to mesocosms in experiments carried out in eutrophic southern (Odense Fjord, Denmark) and northern (Tvärminne Archipelago, Finland) coastal zones of the Baltic Sea. Despite seasonal and geographical differences, similar responses were observed in both experiments. Low SRP (<100 nmol l?1), shortT t (<10 h), and increased levels of APA were observed in both N+P balanced and P deficient treatments, while the opposite trend was observed in P replete treatments. The shortestT t and the highest APA were found when glucose was combined with N treatment. Bioavailable PO4 was estimated usingT t and P uptake rates as derived from stoichiometric conversion of carbon based primary and bacterial production. With shorterT t, the PO4 pool declined to <1 nmol-P l?1, whereas the SRP background pool (difference between SRP and PO4) remained relatively constant (c. 50 nmol l?1). APA was inversely related to PO4 but not to SRP. Responses of specific APA and specific affinity for PO4 uptake, which are APA and PO4 uptake rates (inverse ofT t), respectively, normalized to the summed P biomass of phytoplankton and bacteria, responded consistently to the pattern and magnitude of nutrient limitation evoked in our experiments. Our results, together with a literature survey, suggest that both parameters can be useful in examining PO4 availability for the natural phytoplankton and bacteria community in P starved aquatic systems.  相似文献

Fracture aperture is an important transport property in subsurface hydrology because it influences well productivity and the volume of the water resource. Nuclear magnetic resonance (NMR) well logging measures the hydrogen‐bearing fluid molecules in porous or fractured strata, and the NMR signal intensity increases with the amount of fluid in the sensed region of the NMR sonde. Fluid confined in a large fracture of >>0.2 mm in aperture has T2 (i.e. spin‐spin relaxation time) values as long as those of the bulk fluid. The bulk‐fluid porosity (i.e. porosity calculated using this long T2 component in a T2 histogram data) increases linearly with aperture. Therefore, NMR logging enables quantitative estimation of fracture apertures of >>0.2 mm using the bulk‐fluid porosity data if the calibration of the NMR sonde is performed adequately. We applied NMR logging to a borehole in a Holocene andesite lava at Sumikawa, Japan, to estimate the aperture of open fractures within the lava. A test well of 100 m depth and 20 cm diameter, filled with bentonite drilling mud, was scanned with an NMR sonde to obtain a profile of the porosity and the T2 histogram of the andesite. The bulk‐fluid porosity was calculated from the T2 histogram data, as the porosity at which the T2 value is larger than or equal to a threshold T2 of bulk bentonite mud. The bulk‐fluid porosity of a specific inclined fracture responsible for the total loss of circulation at 61.2 m depth during drilling was calculated assuming a threshold or T2 cut‐off of 33 ms, and again for a cut‐off of 100 ms. Calibration of the NMR sensor in a laboratory and measurement of the fracture dip angle by electrical microimaging logging enabled us to estimate the fracture aperture as 1.7 cm, assuming a T2 cut‐off of 33 ms, or 1.6 cm for a T2 cut‐off of 100 ms. The method of aperture determination described in this study is independent of fluid species and lithology, and is applicable to various hydrogen‐bearing borehole fluids (clean water, mud and oil) and geological settings.  相似文献

Effect of water and stress on the lattice-preferred orientation of olivine   总被引:6,自引:1,他引:5  
The influence of water and stress on the lattice-preferred orientation (LPO) of olivine aggregates was investigated through large strain, shear deformation experiments at high pressures and temperatures (P = 0.5–2.1 GPa, T = 1470–1570 K) under both water-poor and water-rich conditions. The specimens are hot-pressed synthetic olivine aggregates or single crystals of olivine. Water was supplied to the sample by decomposition of a mixture of talc and brucite. Deformation experiments were conducted up to γ (shear strain)  6 using the Griggs apparatus where water fugacity was up to  13 GPa at the pressure of 2 GPa. The water content in olivine saturated with water increases with increasing pressure and the solubility of water in olivine at P = 0.5–2 GPa was  400–1200 ppm H/Si. Several new types of LPO in olivine are found depending on water content and stress. Samples deformed in water-poor conditions show a conventional LPO of olivine where the olivine [100] axis is subparallel to the shear direction, the (010) plane subparallel to the shear plane (type-A). However, we identified three new types (type-B, C, and E) of LPO of olivine depending on the water content and stress. The type-B LPO of olivine which was found at relatively high stress and/or under moderate to high water content conditions is characterized by the olivine [001] axis subparallel to the shear direction, the (010) plane subparallel to the shear plane. The type-C LPO which was found at low stress and under water-rich conditions is characterized by the olivine [001] axis subparallel to the shear direction, the (100) plane subparallel to the shear plane. The type-E LPO which was found under low stress and moderate water content is characterized by the olivine [100] axis subparallel to the shear direction, the (001) plane subparallel to the shear plane. Observations by transmission electron microscopy (TEM) and scanning electron microscopy (SEM) show that the dislocations in water-poor samples (type-A) are curved and both b =  [100] and b = [001] dislocations have a similar population. Numerous subgrains are seen in water-poor samples in backscattered electron images. In contrast, water-rich samples (both type-B and type-C) contain mostly b = [001] dislocations and dislocations are straight and sub-grain boundaries are rare compared to those in water-poor samples. These observations suggest that (1) dominant slip systems in olivine change with water fugacity (and stress) and (2) grain boundary migration is enhanced in the presence of water. Seismic anisotropy corresponding to the fabrics under water-rich condition is significantly different from that under water-poor condition. Consequently, the relationship between seismic anisotropy and flow geometry in water-rich regions is expected to be different from that in water-poor regions in which type-A fabric dominates (i.e., the lithosphere). A few cases are discussed including anisotropy in the subduction zone and in the deep upper mantle.  相似文献
