We are investigating the effects of in vivo exposure of prototypical enzyme inducing agents on hepatic biotransformation enzyme expression in largemouth bass (Micropterus salmoides), a predatory game fish found throughout the United States and Canada. The current study targeted those genes involved in biotransformation and oxidative stress that may be regulated by Ah-receptor-dependent pathways. Exposure of bass to beta-naphthoflavone (beta-NF, 66 mg/kg, i.p.) elicited a 7-9-fold increase in hepatic microsomal cytochrome P4501A-dependent ethoxyresorufin O-deethylase (EROD) activities, but did not affect cytosolic GST catalytic activities toward 1-chloro-2,4-dinitrobenzene (CDNB) or 5-androstene-3,17-dione (ADI). Glutathione S-transferase A (GST-A) mRNA expression exhibited a transient, but non-significant increase following exposure to beta-NF, and generally tracked the minimal changes observed in GST-CDNB activities. Expression of the mRNA encoding glutamate-cysteine ligase catalytic subunit (GCLC), the rate-limiting enzyme in glutathione (GSH) biosynthesis, was increased 1.7-fold by beta-NF. Changes in GCLC mRNA expression were paralleled by increases in intracellular GSH. In summary, largemouth bass hepatic CYP1A-dependent and GSH biosynthetic pathways, and to a lesser extent GST, are responsive to exposure to beta-NF.  相似文献

Mo mineralization within the Galway Granite at Mace Head and Murvey, Connemara, western Ireland, has many features of classic porphyry Mo deposits including a chemically evolved I-type granite host, associated K- and Si-rich alteration, quartz vein(Mace Head) and granite-hosted (Murvey) molybdenite, chalcopyrite, pyrite and magnetite mineralization and a gangue assemblage which includes quartz, muscovite and K-feldspar. Most fluid inclusions in quartz veins homogenize in the range 100–350°C and have a salinity of 1–13 eq. wt.% NaCl. They display Th-salinity covariation consistent with a hypothesis of dilution of magmatic water by influx of meteoric water. CO2-bearing inclusions in an intensely mineralized vein at Mace Head provide an estimated minimum trapping temperature and pressure for the mineralizing fluid of 355°C and 1.2 kb and are interpreted to represent a H2O-CO2 fluid, weakly enriched in Mo, produced in a magma chamber by decompression-activated unmixing from a dense Mo-bearing NaCl-H2O-CO2 fluid. 34S values of most sulphides range from c. 0 at Murvey to 3–4 at Mace Head and are consistent with a magmatic origin. Most quartz vein samples have 18O of 9–10.3 and were precipitated from a hydrothermal fluid with 18O of 4.6–6.7. Some have 18O of 6–7 and reflect introduction of meteoric water along vein margins. Quartz-muscovite oxygen isotope geothermometry combined with fluid inclusion data indicate precipitation of mineralized veins in the temperature range 360–450°C and between 1 and 2 kb. Whole rock granite samples display a clear 18O-D trend towards the composition of Connemara meteoric waters. The mineralization is interpreted as having been produced by highlyfractionated granite magma; meteoric water interaction postdates the main mineralizing event. The differences between the Mace Head and Murvey mineralizations reflect trapping of migrating mineralizing fluid in structural traps at Mace Head and precipitation of mineralization in the granite itself at Murvey.  相似文献

Namibia's passive continental margin records a long history of tectonic activity since the Proterozoic. The orogenic belt produced during the collision of the Congo and Kalahari Cratons in the Early Proterozoic led to a zone of crustal weakness, which became the preferred location for tectonism during the Phanerozoic. The Pan-African Damara mobile belt forms this intraplate boundary in Namibia and its tectonostratigraphic zones are defined by ductile shear zones, where the most prominent is described as the Omaruru Lineament–Waterberg Thrust (OML–WT). The prominance of the continental margin escarpment is diminished in the area of the Central and Northern Zone of the Damara belt where the shear zones are located. This area has been targeted with a set of 66 outcrop samples over a 550-km-long, 60-km-broad coast-parallel transect from the top of the escarpment in the south across the Damara sector to the Kamanjab Inlier in the north. Apatite fission track age and length data from all samples reveal a regionally consistent cooling event. Thermal histories derived by forward modelling bracket this phase of accelerated cooling in the Late Cretaceous. Maximum palaeotemperatures immediately prior to the onset of cooling range from ca. 120 to ca. 60 °C with the maximum occurring directly south of the Omaruru Lineament. Because different palaeotemperatures indicate different burial depth at a given time, the amount of denudation can be estimated and used to constrain vertical displacements of the continental crust. We interpret this cooling pattern as the geomorphic response to reactivation of basement structures caused by a change in spreading geometry in the South Atlantic and South West Indian Oceans.  相似文献

Scheelite-mineralized microtonalite sheets occur on the SE margin of the end-Caledonian Leinster Granite in SE Ireland. Scheelite, polymetallic sulphides and minor cassiterite occur in veins in the microtonalites, disseminated throughout the greisened microtonalite sheets and in the adjacent wallrocks. Two major mineralized vein types occur in the microtonalite sheets: (1) Scheelite ± arsenopyrite ± pyrrhotite occur in quartz-fluorite veins, generally without a muscovite selvage; (2) Sphalerite ± chalcopyrite ± pyrite ± galena ± cassiterite ± stannite occur in quartz + fluorite veins with a coarse muscovite selvage and are often intergrown with the muscovite. Quartz-hosted fluid inclusions were examined from representative samples of both vein types using petrographic, microthermometric and laser Raman spectroscopic techniques. Three distinct types of fluid inclusions have been recognized. Primary, vapour rich Type 1 inclusions in quartz from the scheelite-mineralized veins are of H2O-CO2-CH4-N2 ± H2S ± NaCl composition and formed between 360–530 °C. Primary and secondary, liquid-rich Type 2 fluid inclusions in the base metal sulphide-mineralized veins are of H2O-CH4-N2 ± H2S-NaCl composition and formed between 340–480 °C. They also occur as pseudosecondary and secondary inclusions in scheelite-mineralized veins. Late dilute, low temperature H2O-NaCl + KCl fluid inclusions may be related to late-Caledonian convection of meteoric waters around the cooling Leinster Granite batholith. Extensive and economically-important lithium pegmatites have been described from the eastern flank of the large S-type peraluminous Leinster Granite batholith in southeast Ireland. This flank area is also the locus of a major dip-slip ductile shear zone which was active during emplacement of the Tullow Lowlands pluton of the batholith. All the known bedrock occurrences of lithium pegmatite are contained within this marginal deformation zone. Most previous studies have suggested derivation of the lithium pegmatites by fractionation from the Leinster Granite itself, though none are conclusive. More recently, an anatectic model has been proposed whereby the pegmatites were derived by melting of Li-rich sediments adjacent to the batholithic contact. Our investigation has been concerned with a detailed study of the Rb-Sr isotope systematics and trace element geochemistry of the pegmatites and their host granitoids in an attempt to resolve the problem of pegmatite genesis. The results suggest a strong geochemical and temporal coherence between the Leinster Granite and the lithium pegmatites. A model is proposed in which Li enrichment in residual magmas may be linked to the development of muscovite instead of biotite in the Leinster Granite itself. The Li excluded from entry to biotite then accumulates in highly mobile, low-viscosity pegmatitic fluids which migrate into the marginal shear zone of the Tullow Lowlands pluton where they crystallize at considerably lower temperatures.  相似文献

The polybrominated diphenyl ethers (PBDEs) constitute a class of flame retardants whose residues have markedly increased in fish and human tissues during the last decade. In particular, the levels of certain PBDE congeners in salmon have raised concern regarding potential risks associated with dietary PBDE exposures. However, little is known regarding PBDE-mediated cell injury in relevant in vitro cell models. We conducted a comparative study of oxyradical production and cell injury in rainbow trout gill (RTgill-W1) and trout liver cells (RTL-W1) exposed to 2,2',4,4'-tetrabromodiphenyl ether (BDE 47), a predominant BDE residue found in fish tissues such as salmonids. Exposure to low micromolar concentrations of BDE 47 elicited a significant loss in RTgill-W1 and RTL-W1 cell viability as measured by alamarBlue assay. The dose-response of BDE toxicity differed among the two cell lines, with the RTL-W1 liver cells showing greater resistance to toxicity at lower BDE 47 doses, but a more dramatic loss of viability relative to gill cells when challenged with higher (50 microM) doses. The sensitivity of the trout liver cells at higher BDE 47 exposures was reflected by a higher basal production of oxygen radical production by 6-carboxy-2',7'-dichlorodihydrofluorescein diacetate (DCFH-DA) fluorescence that was markedly enhanced in the presence of BDE 47, suggesting an overwhelming of trout liver cell antioxidant defense pathways. Collectively, our data indicate that RTgill-W1 and RTL-W1 liver cells are sensitive to BDE 47-mediated cell injury through a mechanism that may involve oxidative stress. Our data also provide an in vitro basis for potential tissue differences in BDE 47-mediated cell injury.  相似文献

A swarm of Caledonian microtonalite sheets is host to W-Sn mineralization in SE Ireland. Many but not all mineralized sheets are within a regional shear zone which, however, does not appear to have been a major control on mineralization. Scheelite and arsenopyrite are the dominant minerals within and adjacent veins in greisened tonalite. No wolframite occurs. The intrusions are affected by two alterations, a pervasive one and another which is spatially associated with veins. Both are associated with high metal values. Veining is confined to microtonalite sheets and was initiated at least partly by hydraulic fracturing of the intrusions. Stable isotope analyses indicate that mineral deposition occurred over narrow time and temperature intervals from a hydrothermal fluid of magmatic origin. Methane is important in fluid inclusions in quartz veins and the low oxygen fugacity of the mineralizing fluid was probably critical in concentrating metals and in suppressing wolframite formation.  相似文献
