In aquatic environments extracellular enzymes are bound to microbial cells or exist in a free and adsorbed state. Various
filters have been used to fractionate these enzymatic activities, but enzymes may be readily adsorbed onto some materials,
and such adsorption can induce errors in the estimation of enzymatic activity. In this study we examined three filters to
determine the most suitable filter for fractionation when estimating proteolytic enzyme activity in seawater. We found that
the polycarbonate Nuclepore membrane, widely used for size fractionation because of its pore-size accuracy, was the most favorable
for this purpose, even though it adsorbed slightly more enzymes than the low-protein-binding polyethersulfone membrane. We

Organotin compounds are used in a variety of industrial processes therefore their subsequent discharge into the environment is widespread. Bacteria play an important role in biogeochemical transformations acting as natural decontamination agents. Therefore, screening for tributyltin (TBT)-resistant and -degrading bacteria is relevant for the selection of isolates with decontamination ability of these polluted areas. With this purpose, 50 strains were isolated from sediment and water from Ria de Aveiro and their tolerance to TBT, up to 3mM, was evaluated. Generally, occurrence of highly TBT-resistant bacteria was observed, and Gram negative bacteria exhibited more tolerance to TBT than Gram positive bacteria. A memory response was observed when bacteria were progressively exposed to increasingly higher TBT concentrations. One isolate, Aeromonas veronii Av27, highly resistant to TBT (3mM) uses this compound as carbon source and degrades it to less toxic compounds. 相似文献

Abstract– Distributions of organic functional groups as well as inorganic features were analyzed in the Bells (CM2) carbonaceous chondrite using near‐field infrared (NFIR) spectroscopy. NFIR spectroscopy has recently been developed to enable infrared spectral mapping beyond the optical diffraction limit of conventional Fourier transform infrared microspectroscopy. NFIR spectral mapping of the Bells 300 nm thick sections on Al plates for 7.5 × 7.5 μm2 areas showed some C‐H‐rich areas which were considered to represent the organic‐rich areas. Heterogeneous distributions of organic matter as well as those of inorganic phases such as silicates (Si‐O) were observed with 1 μm spatial resolution. The NFIR mappings of aliphatic C‐H (2960 and 2930 cm?1) and structural OH (3650 cm?1) confirm that organic matter is associated with phyllosilicates as previously suggested. The NFIR mapping method can provide 1 μm spatial distribution of organic functional groups and their association with minerals. High local sensitivity of NFIR enables us to find organic‐rich areas and to characterize them by their aliphatic CH2/CH3 ratios. The aliphatic CH2/CH3 ratio of Bells is slightly higher than Murchison, similar to Orgueil, and lower than literature values of IDPs and cometary dust particles. 相似文献

This study examines the tropical cyclone (TC) genesis frequency over the western North Pacific simulated in atmosphere–ocean
coupled general circulation models from the World Climate Research Programme’s Coupled Model Intercomparison Project phase
3. We first evaluate performances of eight models with atmospheric horizontal resolution of T63 or T106 by analyzing their
daily-mean atmospheric outputs of twentieth-century climate simulations available from the Program for Climate Model Diagnosis
and Intercomparison database. The genesis frequency is validated against the best-track data issued by the Japan Meteorological
Agency. Five of the eight models reproduce realistic horizontal distribution of the TC genesis with a large fraction over
the 10°–20°N, 120°–150°E area. These five high-performance models also realistically simulate the summer–winter contrast of
the frequency. However, detailed seasonal march is slightly unrealistic; four of the models overestimate the frequency in
the early season (May–June) while all of them underestimate the frequency in the mature season (July–September). Reasons for
these biases in the seasonal march for the five high-performance models are discussed using the TC genesis potential (GP)
index proposed by Emanuel and Nolan (in Am Meteor Soc, pp 240–241, 2004). The simulated GP has seasonal biases consistent with those of the TC genesis frequency. For all five models, the seasonal
biases in GP are consistent with those in environmental lower-tropospheric vorticity, vertical wind shear, and relative humidity,
which can be attributed to the simulated behavior of monsoon trough. The observed trough migrates northward from the equatorial
region to reach the 10°–20°N latitudinal band during the mature season and contributes to the TC frequency maximum, whereas
the simulated trough migrates northward too rapidly and reaches this latitude band in the early season, leading to the overestimation
of the TC genesis frequency. In the mature season, the simulated trough reaches as far as 15°–25°N, accompanied by a strong
vertical shear south of the trough, providing an unfavorable condition for TC genesis. It is concluded that an adequate simulation
of the monsoon trough behavior is essential for a better reproduction of the TC frequency seasonal march. 相似文献

Precambrian microbial fossils show carbonaceous cellular structure, which often resemble in shape and size cyanobacteria and other prokaryotes. Morphological taxonomy of these minute, simple, and more or less degraded fossils is, however, often not enough to determine their precise phylogenetic positions. Here we report the results of micro-FTIR spectroscopic analyses of well-preserved microfossils in 850 Ma and 1900 Ma stromatolites, together with those of 8 species of extant prokaryotes and 5 of eukaryotes for comparison. These Proterozoic fossils have low CH3/CH2 absorbance ratios (R3/2 < 0.5) of aliphatic CH moieties, suggesting selective preservation of long, straight, aliphatic carbon chains probably derived from bacterial membrane lipids. All the observed R3/2 values of coccoids, filaments and amorphous organic matter resemble lipid fractions of extant Bacteria including cyanobacteria, but not Archaea. The results indicate that Proterozoic microfossils belong to Bacteria, which is consistent with the cyanobacterial origin inferred from morphology. Moreover, the R3/2 value of fossilized cell would reflect chemical composition of its precursor membrane lipid, thus could be a useful new tracer for distinguishing Archaea, Bacteria and possibly Eucarya for fossilized and extant microorganisms. 相似文献

This study determined the factors contributing to the spatial distribution of 14 metal concentrations in the surface sediments of Beppu Bay on the basis of comparisons of the organic geochemical properties and environmental parameters through principal component analysis (PCA) and redundancy analysis (RDA). The results of PCA and RDA showed that the concentrations of V, Cr, Co, and As were closely related to the distances between the sampling sites and the Oita River. This indicated that these metals originated from the river's drainage area. The Mn, Cu, Mo, and Cd concentrations were related to the water depth. These results indicated that the Mo, Cd, and Cu deposition processes were controlled by oxygen depletion, and that these elements accumulated in the deeper parts of the bay under anoxic conditions. 相似文献

In Northeast Asia, the effect of long-range transport of air pollutants is generally pronounced in spring and winter, but can be important even in summer. This study analyzed summer-time atmospheric transport of elemental carbon (EC) and sulfate (SO4 2?) with the Community Multiscale Air Quality (CMAQ) model driven by the Weather Research and Forecasting (WRF) model. The WRF/CMAQ modeling system was applied to regions ranging from Northeast Asia to the Greater Tokyo Area in Japan in summer 2007. In terms of EC, while the model simulated well the effect of long-range transport, the simulation results indicated that domestic emissions in Japan dominantly contributed (85%) to EC concentrations in the Greater Tokyo. In terms of SO4 2?, the simulation results indicated that both domestic emissions (62%) and long-range transport from the other countries (38%) substantially contributed to SO4 2? concentrations in the Greater Tokyo. Distinctive transport processes of SO4 2? were associated with typical summer-time meteorological conditions in the study region. When a Pacific high-pressure system covered the main island of Japan, domestic emissions, including volcanic emission, dominantly contributed to SO4 2? concentrations in the Greater Tokyo. When a high-pressure system prevailed over the East China Sea and low-pressure systems passed north of Japan, synoptic westerly winds associated with this pressure pattern transported a large amount of SO4 2? from the continent to Japan. In addition, although heavy precipitation and strong wind decreased SO4 2? concentrations near the center of a typhoon, peripheral typhoon winds occasionally played an important role in long-range transport of SO4 2?. 相似文献

We measured deuterium excess (d = δD ? 8δ18O) in throughfall, groundwater, soil water, spring water, and stream water for 3 years in a small headwater catchment (Matsuzawa, 0·68 ha) in the Kiryu Experimental Watershed in Japan. The d value represents a kinetic effect produced when water evaporates. The d value of the throughfall showed a sinusoidal change (amplitude: 6·9‰ relative to Vienna standard mean ocean water (V‐SMOW)) derived from seasonal changes in the source of water vapour. The amplitude of this sinusoidal change was attenuated to 1·3–6·9‰ V‐SMOW in soil water, groundwater, spring water, and stream water. It is thought that these attenuations derive from hydrodynamic transport processes in the subsurface and mixing processes at an outflow point (stream or spring) or a well. The mean residence time (MRT) of water was estimated from d value variations using an exponential‐piston flow model and a dispersion model. MRTs for soil water were 0–5 months and were not necessarily proportional to the depth. This may imply the existence of bypass flow in the soil. Groundwater in the hillslope zone had short residence times, similar to those of the soil water. For groundwater in the saturated zone near the spring outflow point, the MRTs differed between shallow and deeper groundwater; shallow groundwater had a shorter residence time (5–8 months) than deeper groundwater (more than 9 months). The MRT of stream water (8–9 months) was between that of shallow groundwater near the spring and deeper groundwater near the spring. The seasonal variation in the d value of precipitation arises from changes in isotopic water vapour composition associated with seasonal activity of the Asian monsoon mechanism. The d value is probably an effective tracer for estimating the MRT of subsurface water not only in Japan, but also in other East Asian countries influenced by the Asian monsoon. Deciduous forest covers vast areas of permafrost under severe dry climate in eastern Siberia. Understanding the water cycle in this forest ecosystem is quite important for climate projection. In this study, diurnal variations in isotopic compositions of atmospheric water vapour were observed in eastern Siberia with isotope analyses of precipitation, sap water of larch trees, soil water, and water in surface organic layer during the late summer periods of 2006, 2007, and 2008. In these years, the soil moisture content was considerably high due to unusually large amounts of summer rainfall and winter snowfall. The observed sap water δ18O ranged from ?17.9‰ to ?13.3‰, which was close to that of summer precipitation and soil water in the shallow layer, and represents that of transpired water vapour. On sunny days, as the air temperature and mixing ratio rose from predawn to morning, the atmospheric water vapour δ18O increased by 1‰ to 5‰ and then decreased by about 2‰ from morning to afternoon with the mixing ratio. On cloudy days, by contrast, the afternoon decrease in δ18O and the mixing ratio was not observed. These results show that water vapour that transpired from plants, with higher δ18O than the atmospheric water vapour, contributes to the increase in δ18O in the morning, whereas water vapour in the free atmosphere, with lower δ18O, contributes to the decrease in the afternoon on sunny days. The observed results reveal the significance of transpired water vapour, with relatively high δ18O, in the water cycle on a short diurnal time scale and confirm the importance of the recycling of precipitation through transpiration in continental forest environments such as the eastern Siberian taiga.
