为了研究Mre B在钝顶螺旋藻Spirulina platensis形态建成中的作用,克隆了mre B基因并进行原核表达,对表达的融合蛋白进行了纯化,免疫小鼠制备了Mre B的多克隆抗体,分别用Western blot和免疫荧光技术检测不同形态藻丝体中Mre B的表达和定位,结果显示Mre B表达量在螺旋形和直线形两种藻丝体中无明显差异;免疫荧光定位显示,荧光主要分布于细胞膜下一圈,侧面可观察到荧光呈双股螺旋状分布。实验结果说明,Mre B含量不是导致螺旋藻形态改变的因素,Mre B细胞骨架蛋白参与指导细胞壁肽聚糖的合成,所以有可能通过其双螺旋结构在藻细胞中的不对称分布影响螺旋藻的形态。 相似文献

Based on winter (December to February) daily temperature data of 20 meteorological stations in Ningxia Hui Autonomous Region since 1961, the interannual and interdecadal change trend and the mutation characteristics of the winter average, the maximum, and the minimum temperature of the whole Ningxia region, the Yellow River irrigation area, the central arid zone and the southern mountainous area were analyzed using linear trend, Mann-Kendall test and other methods; Meanwhile,the new characteristics of average temperature of the coldest month, <0 ℃ negative accumulated temperature in winter of the 21st century and impact on agriculture were comparatively analyzed. The results showed that: Firstly, in the whole region and different areas of Ningxia, winter average temperature rose significantly before 2000, the Yellow River irrigation area was the most obvious in winter warming than that in the central arid zone and the southern mountainous area. 1985 was the abrupt change point of winter average temperatures in most areas of Ningxia. In the early 21st century the winter average temperature rose slowly in most areas of Ningxia, even falling in the central arid zone. Secondly, the rise of average minimum temperature was significantly higher than average temperature and average maximum temperature in the winter, indicating that the rise of winter temperature in Ningxia was mainly due to winter minimum temperature. Since the 21st century, the average maximum temperature had changed more dramatically than the average temperature in the winter, dropping in most areas. Winter average minimum temperature in addition to the central arid zone declined slightly after 2002, and continued to rise in most areas of Ningxia.Thirdly, both rise of the average temperature of the coldest month and dramatic reduction of the absolute value of negative accumulated temperature is not only beneficial to the northern planting boundary of winter crops spreading further to north and planting areas expanding with a lower risk of freezing injury, but also beneficial to crop pests increase. 相似文献

We propose an approach for automatic generation of building models by assembling a set of boxes using a Manhattan-world assumption. The method first aligns the point cloud with a per-building local coordinate system, and then fits axis-aligned planes to the point cloud through an iterative regularization process. The refined planes partition the space of the data into a series of compact cubic cells (candidate boxes) spanning the entire 3D space of the input data. We then choose to approximate the target building by the assembly of a subset of these candidate boxes using a binary linear programming formulation. The objective function is designed to maximize the point cloud coverage and the compactness of the final model. Finally, all selected boxes are merged into a lightweight polygonal mesh model, which is suitable for interactive visualization of large scale urban scenes. This paper explores urban temperature in Hong Kong using long-term time series. In particular, the characterization of the urban temperature trend was investigated using the seasonal unit root analysis of monthly mean air temperature data over the period January 1970 to December 2013. The seasonal unit root test makes it possible to determine the stochastic trend of monthly temperatures using an autoregressive model. The test results showed that mean air temperature has increased by0.169?C(10 yr)~(-1)over the past four decades. The model of monthly temperature obtained from the seasonal unit root analysis was able to explain 95.9% of the variance in the measured monthly data — much higher than the variance explained by the ordinary least-squares model using annual mean air temperature data and other studies alike. The model accurately predicted monthly mean air temperatures between January 2014 and December 2015 with a root-mean-square percentage error of 4.2%.The correlation between the predicted and the measured monthly mean air temperatures was 0.989. By analyzing the monthly air temperatures recorded at an urban site and a rural site, it was found that the urban heat island effect led to the urban site being on average 0.865?C warmer than the rural site over the past two decades. Besides, the results of correlation analysis showed that the increase in annual mean air temperature was significantly associated with the increase in population, gross domestic product, urban land use, and energy use, with the R~2 values ranging from 0.37 to 0.43. 相似文献

针对高光谱影像特征提取中地物类别训练样本获取代价较高的情况,在线性判别分析的基础上,结合核方法和半监督学习理论,提出了一种基于核半监督判别分析(KSDA)的高光谱影像特征提取方法。该方法同时利用少量已知类别和大量未知类别样本数据进行模型的学习和训练。通过OMIS高光谱影像数据实验表明:在少量已知类别训练样本的条件下,经KSDA特征提取的样本数据在特征空间中能更好地聚集成团,且类别之间的距离较大,增加了类别之间的可分性,得到了较高的分类精度;同时,提取的特征影像能够较好地区分各种地物类别。 相似文献

针对传统的边缘检测方法将灰度变化较为平缓的区域误判为边缘的问题,提出了基于二阶局部纹理熵的边缘检测方法。利用了二阶局部纹理熵能表示局部区域灰度变化大小的性质,并采用灰度错位压缩法消除灰度值压缩造成的条纹现象。实验表明,本文提出的方法能有效地检测出各类边缘,解决了上述的误判问题;通过不同大小窗口的边缘检测效果比较,建议在采用基于二阶局部纹理熵的边缘检测方法时将窗口设置为3×3。 相似文献

利用济南CINRAD/SA新一代多普勒天气雷达资料,统计分析了2004—2015年约15万km2区域内发生的148个线状中尺度对流系统(linear mesoscale convective systems,简称LMCSs)的多普勒雷达回波特征。主要分析了LMCSs的年和月分布、典型尺度、典型回波强度的统计特征以及初始回波出现时间、位置、LMCSs持续时间、演变过程回波合并特征、移动速度和方向、发展后期回波演变特征、组织类型等。LMCSs存在明显的年际变化,不同年份之间有很大的差别,而每年的6月和7月是LMCSs的高发期;80%的LMCSs是大于50 km的中-β尺度,20%属于中-α尺度,成熟期97.3% LMCSs的最大回波强度在55~70 dBz间;10—22时之间易开始形成LMCSs,14—16时是峰值,凌晨不易形成LMCSs,而LMCSs持续时间在2~18 h之间,6~8 h是峰值;一半的LMCSs在演变过程出现回波合并,合并过程可以分为与孤立对流单体合并、与对流回波群合并和与对流回波带合并三类;地形对LMCSs的触发有重要影响,太行山脉、鲁中山区的北麓和西麓容易触发形成LMCSs。这些研究为认识LMCSs发生、演变、减弱各阶段的特征,进一步提高对LMCSs的实时监测、短时预警水平提供了基础。 相似文献
