As seedling culture is a crucial factor for successful cultivation ofGelidium, the authors researched tissue culture technology for producing seedlings. The morphogeny and experimental ecology were observed and studied fully in 2–5 mm isolated tissue fragments. Regeneration, appearance of branching creepers and attaching structure and new erect seedlings production and development were studied. Fragments were sown on bamboo slice and vinylon rope. The seedlings were cultured 20–30 days indoor, then cultured in the sea, where the density of erect seedlings was 3–19 seedlings/cm2, growth rate was 3.84% day. The frond arising from seedlings directly was up to 10 cm per year. The ecological conditions for regenerated seedlings are similar to the natural ones. The regenerated seedlings are suitable for raft culture in various sea areas.  相似文献

分别采用浓度0.1μmol/L、1μmol/L的雌二醇(E2)和塑化剂(邻苯二甲酸二异辛酯,DIOP)对褐牙鲆幼鱼进行连续5周的暴露实验,取鳃、肝、肾进行常规石蜡切片,HE染色,显微镜观察。实验结果显示,在2种试剂的暴露胁迫下,鱼体组织产生不同程度的损伤,随着试剂浓度的增加、暴露时间的延长而变化明显。研究表明,E2可以导致鳃小片弯曲变细,末端弯曲膨大,鳃小片断裂,细胞坏死、脱落;肝细胞排列紊乱,肝细胞肿胀,出现脂肪变性和空泡化;肾小管形态崩解,肾小管变细,肾小管管腔变大,肾小球结构遭到破坏。DIOP可以促使鳃小叶之间发生组织增生、粘连,鳃小叶末端弯曲膨大,出现非组织性空腔;肝细胞出现轻微的细胞脂肪变性和空泡化;肾小管形态遭到破坏,出现管细胞受损或不可见现象。  相似文献

Tissue expression and stock variation of isozymes of stone flounder (Kareius bicoloratus) were analyzed with horizontal starch gel electrophoresis. For the fourteen enzymes assayed, 31 loci were recorded. The results indicated that all the isozymes ex- amined were obviously tissue-specific. The expressions of SOD*, GDH*, G3PDH-2* and ADH-2* were detected only in liver, SDH-1*, MDH-1* and ADH-1* only in muscle, and LDH-B* and LDH-C* only in eyes. In comparison, MDH-2*, GPI-3* and SDH-2* were detected in all tissues examined. Other loci examined were detected in a variety of tissues. Muscle and liver were se- lected to detect the isozyme variation of the two geographic stocks of Qingdao and Weihai, Shandong Province, China. The percent- ages of polymorphic loci (P0.99) were 29.17% and 25.00%, the observed heterozygosities (H0) were 0.028 ± 0.014 and 0.040 ± 0.019, and the expected heterozygosities (He) were 0.039 ± 0.017 and 0.052 ± 0.022 in Qingdao and Weihai stock, respectively. The coeffi- cient of gene differentiation (Fst) and genetic distance (D) between the two stocks was 0.012 and 0.0011, respectively, indicating that the genetic differentiation is low between them. Compared with other species of Pleuronectiformes, both the percentage of polymor- phic loci and the mean heterozygosity of K. bicoloratus were at a middle level.  相似文献

In this study, genes of two distinct tissue inhibitors of metalloproteinases-2 (TIMP-2) from Japanese puffer fish Fugu rubripes, Fugu TIMP-2a and TIMP-2b, were cloned. The open reading frames of Fugu TIMP-2a and TIMP-2b cDNAs are composed of 660 and 657 nucleotides and 220 and 219 amino acids, respectively. Both Fugu TIMP-2s contain 12 cysteine residues, which might form six disulfide bonds as in other animals’ TIMP-2s. Reverse-transcribed polymerase chain reaction analysis showed the mRNAs of Fugu TIMP-2a and TIMP-2b to be expressed in some tissues examined with different expression patterns. These findings suggest that the two distinct Fugu TIMP-2s might perform different functions in Fugu tissues.  相似文献

本文对实验室前期研制的抗中国明对虾(Fenneropenaeus chinensis)血蓝蛋白单克隆抗体进行特性分析,并以该单抗为第一抗体,采用石蜡切片、免疫组化方法,研究了血蓝蛋白在中国明对虾鳃、心、胃、中肠、淋巴器官、卵巢和肝胰脏等组织器官中的分布特点.结果表明:血蓝蛋白在鳃丝小叶边缘的微血腔、心肌束间隙、淋巴器官的淋巴腔中以及肝小管中阳性信号较强;在卵巢卵母细胞之间的血窦、胃内壁环肌层结缔组织的血窦和中肠结缔组织血窦中有阳性信号.结论认为,中国明对虾血蓝蛋白阳性信号大多来自血淋巴而非实质组织,且在血淋巴含量多、血流量大的部位含量丰富,在某些血淋巴无法到达的部位(肝小管内壁)也有血蓝蛋白分布,血蓝蛋白含量的这种组织特异性与各组织器官的功能有着密切的联系.  相似文献

The reproductive style of Diplodus capensis was examined in an unexploited coastal area in southern Angola. The mean length of females was significantly greater than that of males and, although not significant, females dominated the older age classes. The overall sex ratio was 1 male:4.7 females and female dominance increased with size. Histological analysis of gonads revealed that this species is a rudimentary hermaphrodite, with individuals possessing a non-functional bipotential ovotes-tis in the juvenile phase. Histological examination of the gonads of four macroscopically staged ‘intersex’ individuals suggested that they were functional males with a persistent ovarian portion in their gonads from the juvenile ovotestis. The heavily female-biased sex ratio and female dominance in the larger length classes are thought to be an alternative to sex change that is more resilient to exploitation when compared with the protandrous strategy displayed by the South African population of D. capensis.  相似文献

采用单因子实验设计方法, 进行了饲料中添加维生素C对美国红鱼生长(平均增重率、存活率、特定生长率、饲料效率)和免疫反应[血清中溶菌酶活性、血清总补体活性和超氧化物岐化酶(SOD)活性]影响的研究。结果表明: 随着饲料中维生素C添加水平由0提高到100mg/kg时, 美国红鱼的增重率和特定生长率显著提高。以特定生长率为指标, 美国红鱼最佳生长性能的饲料维生素C添加量为104.5mg/kg左右; 饲料中维生素C水平在0—98.9mg/kg时, 美国红鱼肌肉和肝脏中维生素C的积累量随饲料中维生素C的增加而显著增加(P<0.05); 美国红鱼获得最大肝脏和肌肉中维生素C的积累量时饲料中维生素C最低添加量为128.1和104.0mg/kg; 美国红鱼血清溶菌酶和血清总补体活性在维生素C添加水平分别为98.9和198.7mg/kg时达到最高值, 随后变化不明显; 美国红鱼血清SOD活性随维生素C添加水平的增加而显著升高, 并且活性在198.7mg/kg达到最高值, 随后显著下降。综上所述, 美国红鱼维生素C生长和免疫的维生素C适宜添加水平为200mg/kg左右。  相似文献

Oyster extract is an effective bioactivity component. It has abundant nutritional value and antiviral, antitumor and immune defense functions. The role of oyster extract in treating liver injury has been paid more attention. We use Wistar rats to make alcoholic liver disease model through injecting alcohol into rats' stomachs. These rats were randomly divided into five groups: model group, control group, low-dose, middle-dose and high-dose experimental group with a dose of 0.12 g kg-1, 0.40 g kg-1, and 1.20 g kg-1 alcoholic. After nine weeks, serum biomarkers(ALT, AST, TG and TCHO), malondialdehyde(MDA), glutathione(GSH), C3a, C5a, IL-17, TNF-ɑ, anti-MAA-HAS IgG, CD3+, CD4+, CD8+, NK cell activation and zinc content were assessed. The results showed that the serum biomarkers(ALT, AST, TG and TCHO), MDA content, anti-MAA-HSA IgG, serum C3a, C5a IL-17 and TNF-ɑ levels of oyster extract treatment groups were significantly decreased in comparison with model group. On the contrary, GSH showed adverse trend. Serum CD3+, CD4+ and NK cell activation were significantly increased in middle-dose group and high-dose group compared with model group, and there was decrease of CD8+ activity in high-dose group. Plasma Zn level was decreased in model group compared with that in control group. Meanwhile, Mean plasma Zn levels increased dramatically following the dose increase of a given oyster extract.  相似文献

目的 研究局限性脂肪肝发生部位、形态、密度及鉴别诊断方法,提高对该病的认识。资料和方法 筛选40例CT平扫及强化扫描的脂肪肝病例,通过CT值的测量及肝内血管相对密度分级标准,结合强化CT表现进行诊断。结果:40例中29例按肝段分布(72.2%),10例按肝叶分布(22.2%),1例呈球形(2.2%)。40例平扫均示肝血管湮没,10例呈血管反转。强化扫描均示密度轻度升高,其内血管清晰,走行正常。结论:用CT值。血管相对密度变化,结合强化扫描对局限性脂肪肝有较高的诊断价值。  相似文献
