Yield trait improvement of bay scallops following complete diallel crosses between different scallop stocks 总被引:2,自引:0,他引:2
This study describes the complete diaUel hybridization between newly introduced bay scallop stock (W) from Canada and local commercial stock (D) grown under laboratory conditions, in China. Larval survival and growth during all life stages (larvae, spat, and adult) were compared among hybrid (DW, WD) and purebred (DD, WW) populations. Significant heterosis was detected for survival during the larval stage (〉 1% of the mid-parent values). The mean heterosis (Hm) varied in growth throughout the life span. More than 50% of the Hm values were positive and negative in the DW and WD groups, respectively. The influence of maternal effects and mating types (intrapopulation vs. interpopulation crosses) on growth for all life stages was not consistent. Larval survival did not differ significantly (P〉0.05) with maternal effect or mating type. In the harvest stage, shell length (SL), shell height (SH), shell width (SW), and total weight (TW) were larger in the hybrid compared with the inbred groups. Positive Hm values were observed in SL (1.5%), SW (5.8%), and TW (12.3%), and were more significant in the DW groups (6.1%, 4.5%, 6.8%, and 27.2%). These results suggest that hybridization between two geographic populations is a good tool for improving bay scallop growth. 不规则物体体积测量的传统方法有排液法、称重法和排气法等,不仅操作繁琐,而且准确度及精确度低。本文首次提出了排水称重法,利用该方法能够更加简单、快速、准确地测量出物体的体积和质量。用此法对不同海区26个采样点的1100个牡蛎贝壳密度进行了测定分析,结果表明:大部分牡蛎的左壳密度显著小于右壳密度(P0.05);对于不同海区间的牡蛎,贝壳密度存在极显著的差异(P0.001);分布海区相近的牡蛎间,壳的密度也趋于相近;受环境压力相对较大的潮间带上部的牡蛎,贝壳密度也相对较大。本研究为剖析牡蛎贝壳形成机制以及温度、盐度和海洋酸化等环境因素对牡蛎贝壳形成的影响等研究提供了实验数据和参考。 相似文献

采用光镜和电镜方法,观察了吊笼式人工养殖三倍体皱纹盘鲍性腺发育情况,并与二倍体进行比较。结果表明,三倍体鲍性腺发育分期基本与二倍体相同,但雄性腺发育停止在生长期,雌性腺少数可达成熟期,没有发现排放期。三倍体鲍性腺发育较二倍体差,表观看其性腺薄,只包围肝胰腺1/3—1/2,颜色浅:7—10月间检查的86个二倍体个体中成熟期的占24.4%,休止期占11.6%。而83个三倍体个体中,成熟期占1.2%,休止期占72.3%。三倍体成熟卵母细胞直径、核径、线粒体直径、胶膜厚度较二倍体大,但卵黄颗粒、线粒体、粗面内质网等细胞器较二倍体少。 相似文献

皱纹盘鲍(Haliotis discushannai Ino)胚胎RNA分离和cDNA文库构建 总被引:2,自引:0,他引:2
从皱纹盘鲍胚胎样本中分离RNA并构建了cDNA文库。分别收集孵化8、10和12h的皱纹盘鲍胚胎,用抽滤除菌的海水反复悬浮胚胎,将这3个发育阶段的胚胎样本等量混合后用TRIZOL试剂提取总RNA,将提取物用酚氯仿异戊醇再抽提2次,分离获得高质量的总RNA。从总RNA中分离mRNA,构建了皱纹盘鲍混合胚胎的cDNA文库,未扩增胚胎文库的滴度为5.0×106pfu/ml,重组率为94.12%,插入片段均大于400bp,70.6%克隆的插入片段分布在1000—1500bp之间,扩增后的文库滴度为3.06×109pfu/ml。 相似文献

作者采用多重种特异性PCR(multiplex species-specific PCR)技术,研究了巨蛎属(Crassostrea)牡蛎在辽宁沿海的分布。从辽宁沿海的共11个采样点共采集802个牡蛎样本,通过对COI基因的扩增,随机检测了其中的531个牡蛎样本,结果517个个体为长牡蛎(Crassostrea gigas),14个为近江牡蛎(Crassostrea ariakensis),未发现其他巨蛎属牡蛎。结果表明,辽宁沿海有长牡蛎和近江牡蛎等2种巨蛎属牡蛎分布,其中长牡蛎为优势种,分布于潮间带和潮下带,近江牡蛎为稀有种,分布于潮下带,而且在黄海和渤海海域均有分布。 相似文献

根据皱纹盘鲍(Haliotis discus hannaiIno)HSP70和Actin mRNA序列,设计合成引物,建立了皱纹盘鲍HSP70的荧光定量PCR技术平台,并用于检测皱纹盘鲍在热激处理后不同恢复时间HSP70的转录水平变化。结果表明在热诱导后,肌肉组织HSP70的表达呈时间依赖性,存在先升高后降低的趋势,在热诱导后2h,肌肉HSP70基因的表达量明显升高,是对照组(0h)的15倍,随后HSP70的表达量继续升高,在诱导后12h达到最高值,是对照组的21倍。然后表达量开始回落,至热诱导后96h,HSP70表达量降至初始值,表现出一种瞬时表达的趋势。 相似文献
