慢性肾衰的进程中伴随着肾脏炎症的发生,研究表明褐藻多糖硫酸酯具有抗炎作用,且对于慢性肾衰的早期治疗具有一定的作用。本文通过脂多糖(lipopolysaccharide,LPS)诱导,建立大鼠肾小球系膜细胞的炎症模型,用来源于海带的高分子质量褐藻多糖硫酸酯F1和低分子质量褐藻多糖硫酸酯F2对细胞进行处理,24 h后MTT法检测各组细胞活力,Griess法检测24 h及48 h后细胞培养液中NO的含量。结果表明,各组间细胞活力无明显差异;脂多糖可以诱导大鼠肾小球系膜细胞NO产量增加,F1和F2均可在一定程度上抑制脂多糖诱导的NO产量的增加,从而减轻细胞炎症反应。剂量为25μg/m L的F1和F2处理细胞48 h后,与模型组相比,F1和F2组细胞培养液中NO含量分别下降22.52%和38.65%,低分子质量褐藻多糖硫酸酯抗炎效果明显高于高分子质量褐藻多糖硫酸酯。  相似文献

诱导型一氧化氮合成酶(iNOS)在生物机体免疫,特别在无脊椎动物免疫中的作用近来得到了广泛的关注,由其催化产生的一氧化氯(NO)除具有已知的神经传导、松弛平滑肌等功能外,还具有抗菌、抗病毒、抗寄生虫等作用。作者通过硝基四氯唑蓝(NBT)法和血细胞形态观察等方法,对中国明对虾(Fenneropenaeus chinensis)血细胞中存在的诱导型一氧化氮合成酶进行了初步鉴定。在此基础上,通过亚硝酸盐法和L-瓜氨酸法对比,研究了感染白斑综合症病毒(WSSV)后中国明对虾血细胞中一氧化氯合成酶的变化情况。结果显示,中国明对虾在感染WSSV后,iNOS活性在12h内有上升趋势,实验36h后酶活性显著下降,至60h后酶活性降至对照组的一半左右。同时,被脂多糖(LPS)诱导的一氧化氮合成酶活性与对照相比也有显著下降。与此对应的是,核酸探针斑点杂交法检测病毒的结果显示:实验36h后在对虾体内能够检测到白斑综合症病毒。对照组中国明对虾血细胞的iNOS在实验过程中基本保持稳定。这说明WSSV在感染中国明对虾初期可以诱导血细胞产生iNOS,但随着WSSV在中国明对虾体内的大量增殖及其对血细胞的破坏,使得iNOS活性显著降低,对虾也趋于死亡。因此,iNOS能够作为反映对虾在病毒感染过程中健康状况的有效指标。  相似文献

The localization of nitric oxide synthases (NOS) is dealed with in the developing gonads of amphioxus Branchiostoma belcheri tsingtauense. It was found by NADPH-diaphorase staining that (1) NOS activity was present in the nuclear membranes of germinal vesicles during the entire period ofoocyte development; (2) NOS was localized in both the nuclear membranes and the perinuclear region of cytoplasm in the vitellogenetic oocytes; (3) NOS was relocated in the cortical layer in the mature egg; (4) NOS activity was present in spermatocytes, but not in the spermatogonia in the middle of October; (5) NOS was detected in both spermatozoa and spermatids as well as spermatocytes during the breeding season. This is the first report on the distribution pattern of NOS in the developing gonads in protochordates. These results suggest a role for NOS in the functioning of the nuclear membranes and yolk synthesis during oogenesis and in cell division and differentiation during spermatogenesis.  相似文献

Microcoleus vaginatus isolated from a desert algal crust of Shapotou was cultured in BG-11 medium containing 0.2 mol l−1 NaCl or 0.2 mol l−1 NaCl plus 100 mg l−1 sucrose, extracellular polymeric substances (EPS) or hot water-soluble polysaccharides (HWP), respectively. Photosynthetic oxygen evolution rates, photosystem II activity (Fv/Fm) and dark respiration of NaCl-stressed cells were enhanced significantly by the added sucrose or EPS under salt stress conditions (0.2 mol l−1 NaCl). Compared with cells treated with salt alone, sodium contents in cells reduced significantly; the content of cellular total carbohydrate did not change, and intracellular sucrose, water-soluble sugar increased significantly following the addition of exogenous carbohydrates. Sucrose synthase (SS) activity of NaCl-stressed cells increased following the addition of sucrose, and sucrose phosphate synthase (SPS) activity of NaCl-stressed cells increased following the addition of exogenous sucrose, EPS or HWP compared with cells stressed with NaCl only. The results suggested that the extruded EPS might be re-absorbed by cells of M. vaginatus as carbon source, they could increase salt tolerance of M. vaginatus through the changes of carbohydrate metabolism and the selective uptake of sodium ions.  相似文献

龙须菜(Gracilariopsislemaneiformis)在我国主要用作鲍饵料和琼胶生产原料,但是南方夏季高温限制了龙须菜的栽培和产业化。抗逆植物激素水杨酸(salicylicacid,SA)与分支酸代谢之间存在着密切的联系。本文以两种分支酸代谢酶——异分支酸合成酶(isochorismate synthase,ICS)和分支酸变位酶(chorismatemutase,CM)为研究对象,分析了23℃(常温)和33℃(高温)条件下添加SA对龙须菜中两种分支酸代谢酶编码基因转录水平的影响。结果表明高温和SA不同程度地刺激了两种分支酸代谢酶转录水平的表达,如在3—12h,SA添加后ics表达量分别提高到常温对照组的2.84—4.76倍和高温对照组的1.25—1.62倍;在24h时,cm表达量分别提高到常温对照组的2.73倍和高温对照组的1.82倍。然后,我们将两种酶的编码基因分别转化到原核表达载体pET28a中,结果得到了多以包涵体形式大量表达的重组ICS和CM蛋白,其最佳诱导条件为0.1mmol/LIPTG在16℃诱导24h,并用镍柱初步纯化了重组蛋白。该研究为阐明温度和外源SA对龙须菜中两种分支酸代谢酶的影响规律,以及为从蛋白水平研究这两种代谢酶提供了资料。  相似文献

采用组织化学、免疫组化和生物化学方法对栉孔扇贝外套膜中的一氧化氮合酶(EC1.14.13.39,NOS)活性进行了研究。组织化学显示结果表明,帆状部上皮细胞呈NOS强阳性,血细胞和神经纤维呈阳性;外套触手内有较多近圆形细胞和大量波浪形神经纤维呈NOS强阳性;边缘膜内有大量神经细胞和神经纤维分别呈NOS强阳性和阳性,外套环走肌束附近和血管内皮周围均有大量近圆形的强阳性细胞分布;中央膜有大量近圆形阳性细胞聚集成团块或分散分布。免疫组化显示表明,血细胞、帆状部和边缘膜上皮细胞呈神经型NOS(nNOS)和内皮型NOS(eNOS)弱阳性,而呈诱导型NOS(iNOS)阳性。生化测定结果表明,总NOS(tNOS)活力和一氧化氮(NO)含量均为中央膜最高,边缘膜次之,外套触手较低,帆状部最低,其中结构型NOS(cNOS)活力也是帆状部的最低,而iNOS活力则是外套触手的最低。栉孔扇贝外套膜可能是其神经系统中NOS的发生中心,NO-NOS体系可在其免疫防御和调节等方面发挥重要的作用。  相似文献

从南极普里兹湾海域海水中分离到一株产海藻糖合成酶的海洋低温细菌BSw10041,革兰氏阴性,杆状,有极生鞭毛,能运动,菌落半透明。进行了常规生理生化和BIOLOG GN2细菌鉴定系统测试,结果表明菌株BSw10041与恶臭假单胞菌(Pseudomonas putida)的表型特征非常相似。为了进一步确定菌株BSw10041的分类学地位,测定了其16S rDNA序列,分析了相关细菌相应序列的同源性,构建了系统发育树,结果表明BSw10041与P.putida的亲缘关系最近。综合上述结果,菌株BSw10041可鉴定为Pseudomonas putida,定名为Pseudomonas putida BSw10041。  相似文献

A high quality cDNA library was constructed from the brown alga Laminaria japonica,with the titer of 1.2×10 5 pfu/ml.The average insert size of the cDNA library is about 1.6 kb.From the cDNA library,591 cDNA clones were randomly selected and sequenced.As a result,574 EST(expressed sequence tag) sequences were generated.All of the 574 ESTs were submitted to the dbEST database section of GenBank with the accession numbers from CX942625 to CX943198.The cDNA library was screened with a α-32 p labeled 453 bp T P S gene probe,which is a partial sequence yielded from Porphyra yezoensis.Four positive cDNA clones were screened and the sequencing data showed that these four cDNA clones covered majority of L.japonica TPS cDNA sequence.After PCR amplification,sequencing and assembling,the entire ORF(open reading frame) sequence of the T P S gene was obtained,which was named LjTPS.LjTPS encodes a protein containing 908 amino acids with a calculated molecular mass of 101 674 Daltons.The LjTPS gene was successfully expressed in E.coli and rice.The LjTPS gene has potential application both in plant breeding to stress tolerance and in deciphering the T P S gene function and mechanism to stress tolerance.  相似文献

The partial ingestion of the placenta by a newly parturient Hooker's sea lion, Phocarctos hookeri (Gray, 1844) (Pinnipedia: Otariidae), was seen on Enderby Island, Auckland Islands, in December 1972.
