Multixenobiotic resistance mechanism (MXR) in aquatic organisms is mediated by the activity of the P-glycoprotein (Pgp) transporter that binds and actively effluxes different chemicals out of cell. In addition to the Pgp, several other, non-Pgp transport proteins have been recently identified in different human and animal tissues. Given their characteristics and tissue distribution we hypothesized that members of the so-called multidrug resistance-associated protein (MRP) family may be expressed in aquatic organisms. This study attempted to identify MRP related genes in different tissues of several marine and freshwater bivalves (Mytilus galloprovincialis, Dreissena polymorpha, Anodonta cygnea) and fish species (Mullus barbatus, Cyprinus carpio, Salmo trutta). Following an alignment of known MRP1 and MRP2 human sequences, as well as the GenBank available mrp2 sequences from different animals, we determined highly conserved regions and used them to design three pairs of consensus primers. Total RNA was isolated, reverse transcribed to cDNA and the obtained cDNAs were PCR amplified with the corresponding primers. The amplified PCR products were sequenced and their homology compared with Pgp and MRP protein sequences from different species. The expression of MRP related mRNA was clearly identified only in liver tissue isolated from red mullet, with homologies at the protein level ranging from 75% to 76%. Described results clearly pointed at the possibility that at least in the red mullet MXR as a general defense mechanism may be mediated by the activities of at least two different types of transport proteins.  相似文献

Two models of masonry infilled reinforced concrete frame buildings were tested at the shaking table. Models were built in the reduced scale 1:4 using the materials produced in accordance to modelling demands of true replica modelling technique. The first model represented a one‐storey box‐like building and the second one the two‐stories building with plan shaped in the form of a letter H. Models were shaken with the series of horizontal sine dwell motions with gradually increasing amplitude. Masonry infills of tested models were constructed of relatively strong bricks laid in weak mortar. Therefore, typical cracks developed and propagated along mortar beds without cracking of bricks or crushing of infill corners. Data collected from tests will be used in future evaluation, verification and development of computational models for prediction of in‐plane and out‐of‐plane behaviour of masonry infills. The responses of tested models can be well compared with global behaviour of real structures using the modelling rules. The similarity of local behaviour of structural elements, e.g. reinforced concrete joints, is less reliable due to limitations in modelling of steel reinforcement properties. The model responses showed that buildings designed according to Eurocodes are able to sustain relatively high dynamic excitations due to a significant level of structural overstrength. The concentration fluctuations resulting from hazardous releases in the subsurface are modeled through the concentration moments. The local solute exposure concentration, resulting from the heterogeneous velocity field and pore scale dispersion in the subsurface, is a random function characterized by its statistical moments. The approximate solution to the exact equation that describes the evolution of concentration standard moments in the aquifer transport is proposed in a recursive form. The expressions for concentration second, third and fourth central moments are derived and evaluated for various flow and transport conditions. The solutions are sought by starting from the exact upper bound solution with the zero pore scale dispersion and introducing the physically based approximation that allows the inclusion of the pore scale dispersion resulting in simple closed-form expressions for the concentration statistical moments. The concentration moments are also analyzed in the relative and absolute frame of reference indicating their combined importance in the practical cases of the subsurface contaminant plume migration. The influence of pore scale dispersion with different source sizes and orientations are analyzed and discussed with respect to common cases in the environmental risk assessment problems. The results are also compared with the concentration measurements of the conservative tracer collected in the field experiments at Cape Cod and Borden Site.  相似文献

Two major statistical issues can be distinguished in the procedure of wave extreme prediction. The first issue is that predicted extreme values must be based on data collected in a relatively short time. The second issue is extrapolation of the observed data into its extreme region, typically lying well beyond from even the most extreme available observation. The process of extrapolation plays a fundamental role in this area of analysis and therefore it is essential to fit empirically a convenient probability distribution that describes the available data as closely as possible. Determination of extreme values probability distribution parameters by genetic algorithm is applied to improve the methodology of extreme sea state prediction.Illustrative applications of the method are given for a North Atlantic sea environment. The results are presented as crest height maximum values occurring with a given probability or in a design storm that has a specified return period.  相似文献
