An isolated fish hepatocyte culture system was developed as a models ystem to investigate the mechanisms of action of environmental contaminants. Hepatocytes were isolated from striped bass (Morone saxatilis) by an adaptation of the two stage perfusion technique of Seglen.1 The system was used to evaluate metal binding protein (MBP) induction in response to cadmium, a primary inducer of metallothionein (MT) in rat hepatocytes. Striped bass hepatocytes appeared to be refractory to the induction of MBP by cadmium, since there was no significant increase in the synthesis of MBP for any of the doses at any of the time points investigated. However, when a similar experiment was performed using rat hepatocytes there was induction of MBP that was related to both dose and time. These comparative experiments indicate that although there are similarities between the hepatocytes of the two species in regard to 35S incorporation into low molecular weight metal-binding proteins, there appear to be significant quantitative differences as well in regard to MBP kinetics. This in vitro model system could potentially be utilized to investigate the toxicological properties of other environmental contaminants.  相似文献

The effects of frictional forces acting on the sidewalls of buried box culverts are presented as determined with finite element method (FEM) and detailed soil modelling. The possibility of reducing earth pressure on deeply buried concrete box culverts by the imperfect trench installation (ITI) method has been contemplated during the last several decades. There have been limited research results published primarily regarding the qualitative aspect of load reduction in ITIs. It was found during the course of this study that significant frictional forces develop along the sidewalls of box culverts and adjacent sidefills in ITIs. Current American Association of State Highway and Transportation Officials provisions do not consider these frictional forces, but they cannot be neglected in ITIs, as their effect is dominant. An optimum geometry for the soft zone in ITIs is presented to maximize earth load reductions. The soil–structure interaction at the box culvert–soil interface was found to have a significant effect on total earth pressure acting on the bottom slab. Predictor equations for earth load reduction rates were formulated for ITIs incorporating the optimum soft zone geometry based on the FEM. Copyright © 2007 John Wiley & Sons, Ltd.  相似文献

An improved scheme for measuring and analyzing the magnetic field of flares is presented in general outline form. The techniques employed during the last solar cycle are reviewed very briefly. The point is made that those techniques were inadequate due to fundamental limitations in the data. A realistic scheme for acquiring the necessary data is then proposed. The scheme is realistic in that it makes only those assumptions which appear to be indeed valid, and it presupposes only instrumental techniques which are presently available. It is suggested that, with this improved data, the structure and evolution of the pre-flare magnetic field can be calculated with sufficient accuracy to form the basis of theoretical models.This research supported by Company funds of The Aerospace Corporation.  相似文献

Between 2000 and 2014, more than thirty geography departments adopted revised or new names, with some entirely dropping geography. Although renaming and rebranding efforts are not new to higher education, the rapid pace at which geography department names have changed raises questions about the discipline's identity and health. We examine the renaming trend within geography programs together with intended and unexpected factors as perceived by faculty. Specifically, we look at the renaming and rebranding trend within the context of four pillars offered by Pattison (1964 Pattison, W. D. 1964. The four traditions of geography. Previous global models of coronal magnetic fields have used a geometrical construction based on a spherical source surface because of requirements for computational speed. As a result they have had difficulty accounting for (a) the tendency of full magnetohydrodynamic (MHD) models to predict non-radial plasma flow out to r 10r and (b) the appreciable magnitude, 3, of B r , (the radial component of B) consistently observed at r 1 AU. We present a new modelling technique based on a non-spherical source surface, which is taken to be an isogauss of the underlying potential field generated by currents in or below the photosphere. This modification of the source surface significantly improves the agreement between the geometrical construction and the MHD solution while retaining most of the computational ease provided by a spherical source surface. A detailed comparison between the present source-surface model and the MHD solution is made for the internal dipole case. The resulting B field agrees well in magnitude and direction with the coronal B field derived from the full MHD equations. It shows evidence of the slightly equatorward meridional plasma flow that is characteristic of the MHD solution. Moreover, the B field obtained by using our non-spherical source surface agrees well with that observed by spacecraft in the vicinity of the Earth's orbit. Applied to a solar dipole field with a moment of 1 G-r 3 , the present model predicts that B r at r 1 AU lies in the range of 1–2 and is remarkably insensitive to heliomagnetic latitude. Our method should be applicable also to more general (i.e., more realistic) configurations of the solar magnetic field. Isogauss surfaces for two representative solar rotations, as calculated from expansions of observed photospheric magnetic-field data, are found to show large and significant deviations from sphericity.  相似文献
