Gene specific primers and DNA probe were designed based on the sequence of 18S rDNA cloned from the red tide alga Thalassiosira rotula. A real-time fluorescent quantitative PCR (RFQ-PCR) method was developed for quantitative detection of T.rotula. The RFQ-PCR assay data showed that the results obtained with the RFQ-PCR quite good agreement with those with the light microscope (LM) counting method, which suggested that the RFQ-PCR could be a useful method for red tide alga detection.  相似文献

在层序地层学研究中,关键是层序划分和对比。而层序划分、对比的关键是层序识别。层序的识别包括层序界面(层序的底界面、初始海泛面和最大海泛面)的识别和构成层序的体系域识别。这里,在众多前人研究成果的基础上,详细研究了层序的测井、地震响应特征,即层序界面和体系域在测井曲线上和地震剖面上的特征。其中,层序底界面在测井上表现为突变的钟型、箱型或侧积式曲线的底界;在地震剖面上表现为剥蚀、顶超、上超、下超;而体系域在测井曲线上的响应为:低水位体系域的海底扇以漏斗形中、高幅的前积式,或钟型中、低幅的后积式模式为特征,陆坡扇成钟型、正向齿形,自下而上幅度由中高幅→低幅,即具后积式测井模式,低水位楔的测井曲线表现为旋回性进积模式特征,其特征表现为锯齿状箱型。海侵体系域的测井曲线呈现向上变细、变深序列,并表现为钟型、正向齿形或齿化状,幅度由高幅变化为低幅,包络线具后积式特征。高水位体系域相应的测井曲线呈现中幅箱形或桶形,不同体系域在地震剖面上的响应特征明显不同。  相似文献

本文在南部Appalachian山区的Blacksubury,Virginia中心地区的两个DEM上评估了高程、坡度、坡向的精度。第一个是美国地调局(USGS)在30m格网7.5分的方形图幅上使用Gestalt Photomapper(GPM)仪器,根据1:40000落叶期的航空像片自动立体相关处理完成的。同一地区的第二个DEM,是利用STX公司设计的专业立体相关技术,处理SPOT全色波段立体像对取得的,其间利用SPOT图像公司高解像力的DEM产品,对于DEM高程、坡度、坡向的精度进行了目视评估,并与野外测量获取的高程、坡度、坡向值进行了比较,USGS用GPM获取的DEM在微观地貌表现方面要好一些,而USGS—GPM和SPOT—STX两个DEM的高程差平均值小于6m,且误差分布中心未显著偏离零值,满足1级精度要求。从SPOT—STX的DEM所导出的,在坡度、坡向的统计上表现出一些显著的大误差,而USGS—GPM的坡度、坡向误差发布中心未显著偏离零值。在坡度、坡向与它们各自的误差之间有显著的大的相关性。本文很少的关于DEM导出的坡度、坡向以及高程的评估之一。读者会注意到这些结论不一定适用于不同的DEM生成方式、算法以及不同地形和植被情形的地区。  相似文献

Recharge estimation for transient ground water modeling   总被引:11,自引:0,他引:11  
Reliable ground water models require both an accurate physical representation of the system and appropriate boundary conditions. While physical attributes are generally considered static, boundary conditions, such as ground water recharge rates, can be highly variable in both space and time. A practical methodology incorporating the hydrologic model HELP3 in conjunction with a geographic information system was developed to generate a physically based and highly detailed recharge boundary condition for ground water modeling. The approach uses daily precipitation and temperature records in addition to land use/land cover and soils data. The importance of the method in transient ground water modeling is demonstrated by applying it to a MODFLOW modeling study in New Jersey. In addition to improved model calibration, the results from the study clearly indicate the importance of using a physically based and highly detailed recharge boundary condition in ground water quality modeling, where the detailed knowledge of the evolution of the ground water flowpaths is imperative. The simulated water table is within 0.5 m of the observed values using the method, while the water levels can differ by as much as 2 m using uniform recharge conditions. The results also show that the combination of temperature and precipitation plays an important role in the amount and timing of recharge in cooler climates. A sensitivity analysis further reveals that increasing the leaf area index, the evaporative zone depth, or the curve number in the model will result in decreased recharge rates over time, with the curve number having the greatest impact.  相似文献

We report new Zr isotope evidence for live (92)Nb (mean life: tau&d1;92Nb=52 Myr) within the early solar system resulting in &parl0;92Nb&solm0;93Nb&parr0;initial approximately 10-3. The meteoritic minerals rutile and zircon have, respectively, very high and very low Nb/Zr ratios and are ideal for exploring the (92)Nb-(92)Zr chronometer. Rutiles exhibit high positive straightepsilon92Zr ( approximately 14-36) while a zircon has a negative straightepsilon92Zr ( approximately -4), as would be expected if (92)Nb was live in the early solar system. The meteoritic rutiles appear to be young, with apparent times of formation of approximately 80-220 Myr subsequent to the origin of the solar system. The initial (92)Nb/(92)Mo for the solar system is broadly compatible with a model of uniform production if the (92)Nb/(92)Mo production ratio for Type II supernova (SNII) sources with neutrino-driven winds is used. Data for all the now extinct p-process nuclides ((92)Nb, (97)Tc, and (146)Sm) are consistent with these isotopes being derived by uniform production from SNII sources and a free decay interval of approximately 10 Myr. Consideration of a range of models indicates that the average p-process production ratio of (92)Nb/(92)Mo needs to be at least in the range of 0.06-0.25.  相似文献

The South China Sea is rich in oil and gas resources. With the increasing exploration of oil and gas resources in the northern South China Sea and the increasing demand for energy in the world, The central‐southern South China Sea have become important constituencies for oil and gas resources. The central‐southern basins of South China Sea can be divided into three types of basin groups, namely, the southern basin group (Zengmu Basin, Brunei‐Sabah Basin), the western basin group (Wan'an Basin, Zhong jiannan Basin) and the Central Basin Group (Liyue Basin, Beikang Basin, Nanweixi Basin and Palawan Basin). At present, the degree of exploration is relatively low, and the source rock has not yet formed a understanding of the system. The main source rock development time, source rock characteristics, hydrocarbon generation potential and control factors of each basin group are still unclear, which seriously restricts the exploration of oil and gas. Based on the sedimentary facies distribution and sedimentary filling evolution, combined with the geoche mical characteristics of the source rocks, the source age, organic matter type, hydrocarbon generation potential and main controlling factors of the basins in the central‐southern basins are discussed. By the detailed research on delta scale, provenance system, paleoclimate conditions, ancient vegetation development and hydroca rbon generation parent material, the main controlling factors of hydrocarbon generation potential of source rocks in each basin group are revealed.  相似文献
