Plasmid DNA was incubated at 25°C with aqueous solutions of dissolved Fe(II), S(-II), and nanoparticulate FeS with a mackinawite structure, FeSm. At ??0.1 mM total dissolved Fe(II) and S(-II), an increase in the proportion of the relaxed plasmid DNA occurs, through scission of the DNA backbone. In solutions where FeSm was precipitated, nanoparticulate FeSm binds to the DNA molecules. In solutions with concentrations below the FeSm solubility product, nicking of supercoiled pDNA occurs. Plasmid DNA appears to be a sensitive proxy for radical reactions. The reactant is proposed to be a sulfur-based radical produced from the iron-catalyzed decomposition of bisulfide, in a manner analogous to the Fenton reaction. This is further supported by experiments that suggest that sulfide free radicals are produced during the photolysis of aqueous solutions of polysulfides. Supercoiling of DNA affects nearly all DNA?Cprotein transactions so the observation of relaxation of supercoiled forms through reaction with FeS solutions has direct implications to biochemistry. The results of this experimentation suggest that genotoxicity in FeS-rich systems is a further contributory factor to the limited survival of organisms in sulfidic environments. Mutations resulting from the interactions of organisms and mobile elements, such as plasmids, in sediments will also be affected in sulfide-rich environments.  相似文献

This paper describes a multi‐proxy palaeoecological investigation undertaken in conjunction with an archaeological survey of the Upper Sangro Valley in the Abruzzo National Park, Central Italy. Despite being a biodiversity hotspot and regarded as a near‐pristine area, the pollen, spore and diatom data all show major changes in the vegetation extending to over 2000 m a.s.l. during the mid to late Holocene. Although there are changes in ecological composition earlier in the Holocene they are different in type and magnitude from the changes which began about 800 cal a BC. The pollen and diatom evidence do not correlate well with regional palaeoclimate data, or on‐site isotopic evidence, but do appear to be related to Samnite (later Iron Age) clearance and upland grazing associated with transhumance and later annexation (and centuriation) of the lower slopes by Roman surveyors. The greatest change in vegetation was during the period c. AD 500–600 and corresponds with the Byzantine–Gothic Wars, and Lombard–Carolingian settlement reorganization into nucleated hilltop settlements which managed upland grazing. This pattern of intensive land use at all altitudes persisted until the early 20th century and only changed following rural depopulation after World War II. These data illustrate how cultural factors had a profound effect on this mountainous region which, in this case, far outweighed the effects of climatic fluctuations which are known to have occurred from both this study area and the region. Biogenic silica contents of sediments on the lower Selenga Delta and Buguldeika saddle in Lake Baikal show distinct fluctuations that reflect changes in diatom productivity, and ultimately, climate. The pattern of the upper 50 m of the section, dating from about 334 ka, is similar to that of the marine oxygen-isotope record, increasingly so as the younger sediments become progressively finer grained and less locally derived with time. The last two interglaciations are marked by biogenic silica abundances similar to those of the Holocene. The equivalent of marine oxygen-isotope stage 3 is distinctly intermediate in character between full glacial and full interglacial biogenic silica values. Following near-zero values during the last glacial maximum, biogenic silica began to increase at about 13 ka. The rise in biogenic silica to Holocene values was interrupted by an abrupt decrease during Younger Dryas time, about 11 to 10 14C ka.  相似文献

A study has been made of the yearly variation of the cosmic ray intensity for the years 1961–67 inclusive using pressure corrected neutron monitor data from both hemispheres to minimize seasonal meteorological effects. An annual wave is found in the data with an amplitude which varied between 0.2 and 1.0 per cent during the period but which had a sensibly constant phase, the time of maximum being in March. These observations, which are shown to be consistent with the observed heliolatitude distribution of coronal 5303Å emission, indicate the existence of a southerly directed asymmetrical gradient of up to 8 per cent perpendicular to the solar equatorial plane. It is found that the cosmic ray intensity at the Earth is controlled by the solar activity in a narrow band of heliolatitudes ±10° or ±20° centred at the heliolatitude of the Earth. Also, the results indicate that there was a phase lag of 1 ± 1 month between solar activity and the resulting changes in the cosmic ray intensity at the Earth giving a radius for the modulating region of ? 10 A.U. during the period of low solar activity considered.  相似文献

The relationship between the cosmic ray intensity and solar activity during solar cycle 20 is discussed. A model is developed whereby it is possible to simulate the observed cosmic ray intensity from the observed number of solar flares of importance 1. This model leads to a radius for the modulation region of 60–70 AU. It is suggested that high speed solar streams also made a small contribution to the modulation of cosmic rays during solar cycle 20.  相似文献

Groundwater bores act as traps. Net samplers are regularly used for sampling this type of trap for fauna. To enable direct comparisons of faunal communities in groundwater bores and stream sediments, stream sediment tubes were built similar to groundwater bores and were sampled with net samplers for fauna. These stream sediment tubes consisted of a tube anchored in the stream sediment, also called interstitial space. To test the efficacy of this trap method in stream sediments, it was compared to another type of trap, Hahn's trap. Faunal communities sampled by a net in the stream sediment tubes did not differ hugely from fauna in Hahn's trap samples. Physical and chemical factors of sampled water in both the stream sediment tubes, the surrounding interstitial sediments and the second type of traps, Hahn's traps, showed that water in both the tubes and Hahn's traps was closely related to interstitial water. The net sampler is inexpensive and easy to handle. It is suggested that sampling stream tubes with nets may be an appropriate method for long‐term monitoring studies.  相似文献

Recent results from a number of redshift surveys suggest that the Universe is well described by an inhomogeneous, fractal distribution on the largest scales probed. This distribution has been found to have fractal dimension, D , approximately equal to 2.1, in contrast to a homogeneous distribution in which the dimension should approach the value 3 as the scale is increased. In this paper we demonstrate that estimates of D , based on the conditional density of galaxies, are prone to bias from several sources. These biases generally result in a smaller measured fractal dimension than the true dimension of the sample. We illustrate this behaviour in application to the Stromlo–APM redshift survey, showing that this data set in fact provides evidence for fractal dimension increasing with survey depth. On the largest scale probed, r ≈60  h −1 Mpc, we find evidence for a distribution with dimension D =2.76±0.10. A comparison between this sample and mock Stromlo–APM catalogues taken from N -body simulations (which assume a CDM cosmology) reveals a striking similarity in the behaviour of the fractal dimension. Thus we find no evidence for inhomogeneity in excess of that expected from conventional cosmological theory. We consider biases affecting future large surveys and demonstrate, using mock SDSS catalogues, that this survey will be able to measure the fractal dimension on scales at which we expect to see full turn-over to homogeneity, in an accurate and unbiased way.  相似文献
