超声波辅助提取河豚毒素的研究

摘    要:以月腹刺鲀(Gastrophysus lunaris)卵巢为原料,利用超声波辅助提取河豚毒素(Tetrodotoxin,TTX)。通过单因素实验及二次回归旋转组合设计实验研究液料比、超声温度、超声时间等因素对提取率的影响,确定超声波辅助提取河豚毒素的最佳工艺条件。结果表明:在实验范围内影响河豚毒素得率的各因素大小依次为液料体积质量比、超声温度、超声时间;超声波辅助提取河豚毒素的最佳工艺条件为液料体积(mL)质量(g)比3.03∶1,超声温度39℃,超声时间22 min,该工艺条件下提取河豚毒素得率为63.57μg/g。

Abstract:Gastrophysus lunaris were taken as raw materials and the ultrasonic-assiste extraction technology was used to extract tetrodotoxin(TTX).The single factor experiment and quadratic orthogonal rotation regression combination design were adopted to study the effects of such factors as liquid-solid ratio,ultrasonic temperature and ultrasonic time.The optimal technological condition of TTX extraction from Gastrophysus lunaris by ultrasonic wave was determined.The result showed as follows: The order of factors that influence the TTX extraction rate within the experimental range was liquid-solid ratio the ratio of extracting liquid volume(mL) to sample mass(g)],ultrasonic temperature,and ultrasonic time.The optimal technological parameters of TTX extraction should be liquid-solid ratio 3.03 ∶ 1,ultrasonic temperature 39 ℃ and ultrasonic time 22 min,and under such conditions,and the total TTX extract amount could reach as high as 63.57 μg/g.
