单环刺螠染色体核型分析

摘    要:染色体是细胞遗传学的研究基础,为了补充螠虫动物的核型资料,为单环刺螠(Urechis unicintus)的遗传育种和进化分类研究提供基础,以单环刺螠胚胎、幼体及成体体腔细胞、呼吸肠等为材料,经植物血球凝集素(PHA)和秋水仙素处理后,采用低渗和滴片常规方法制备染色体标本,对单环刺螠染色体数目和核型进行了研究。结果表明:单环刺螠成体经0.4 g/L秋水仙素浸泡12 h后,取体腔细胞,经0.075 mol/L KCl低渗45 min,采用热滴片法制片效果最佳。单环刺螠染色体数目为2N=146,核型公式为2N=58M+26SM+20ST+42T,染色体臂数NF=250,未发现异形性染色体和随体。

Abstract:Chromosome studies can provide indispensable basic cytogenetic data. To complement karyotype information and provide a foundation for genetic research on breeding and the evolutionary classification of marine worms, the chromosomal number and karyotype of Urechis unicintus were investigated by a conventional method, including hypotonicity and chromosome dropping after treatment with phytohaemagglutinin (PHA) and colchicine, using embryos, larvae, and some adult tissue including coelomocyte and breathing intestines. The best chromosomal image was obtained from adult coelomocytes by soaking the organism for 12 h in 0.4 g/L of colchicine and subjecting coelomocytes to hypotonic treatment with 0.075 mol/L of KCl for 45 min, followed by the hot drop method. The chromosomal number of U. unicintus is 2N = 146. The karyotype formula is 2N = 58M + 26SM + 20ST + 42T, and the fundamental chromosome number (NF) is 250. No sex chromosome heteromorphism or satellites were identified.
