南海有毒塔玛亚历山大藻的分子地理标记分析

摘    要:采用聚合酶链扩增反应(PCR)和限制性片段长度多态性分析(RFLP)方法,对1992年5月和1994年12月采自中国南海大鹏湾,大亚湾,香港海域的赤潮有毒甲藻塔玛亚历山大藻8个不同地理株的核糖体小亚基RNA基因(Ss-rDNA)进行分析,并与北美,西欧等地的比较。结果表明,中国南海的塔玛业历山大藻缺少B基因,这与北美等地的塔玛亚历山大藻(包括有毒和无毒株)不同,而与西欧,澳洲等地的有毒和无毒株相似

Abstract:In this study, cultures of eight strains of red the toxic Alexandrium tamarense wereestablished from germinated cysts or vegetative cells isolated from Dapeng Bay, Deya Bay and theHong Kong area in May l992 and December l994, and a method of PCR amplification withindividual cel1s was used to amplify the small subtrit ribosomal RNA genes (Ss-00) of eightstrains for a restriction fragment length Polymorphism (RFLP) assay. PCRRFLP analysis of Ss-rDNAon the red the toxic A lamarense demonstlated that the strains of A lamarense in the South ChinaSea have Ss-rDNA A gene and lack of B gene, which was the same with that of the strains inwestem Japan and Austl-alia (toxic or nontoxic), but different from that of the strains in NorthAlnerican seas. The genetic consistence of toxic A tamarense in China seas suggested that theincrease in the geographic range of A tamarense in ths sea area has Possibly a local origin. Inaddiholl, it was demonstrated that the type of Ss-rDNA could be as a report marker for theidentification and monitoring of the origin and dispersal of toxic species of Alexandrium.
