摘    要:硫酸盐有机废水生物处理技术一直是人们关注的热点,该种废水处理的关键是如何在有效地去除COD的同时,高效地去除SO_4~(2-)厌氧生物处理系统中的硫酸盐还原菌(Sulfte-Reducing Bacteria,SRB)在一定的条件下可以有效地将SO_4~(2-)还原而从废水中去除。产酸脱硫反应器是一个复杂的生态学系统,在这个生态系统中,SRB起着最主要的生态学作用,要研究产酸脱硫反应器中的各种群的微生物的生理生态学,是非常复杂而且难以实现的。为了更方便地研究产酸脱硫反应器中的微生物生理生态学,比较好的做法是把在反应器中数量上占优势,并且起主要功能的种群分离出来,对它们进行单独和配合研究。本实验就是利用滚管法和改进的Hungate技术,在COD/SO_4~(2-)=3的稳定阶段,对产酸脱硫反应器中的SRB进行了分离,分离出在数量上占优势的SRB,命名为SRB-ZH07。在厌氧试管和三角瓶中,进行了SRB-ZH07生理生态学研究。pH值对SRB-ZH07的生长和硫酸盐去除率的影响,pH为7.0左右SRB-ZH07的生长状况最好,在34h,菌液的OD_(600nm)达到0.827,而其它pH下的OD_(600nm)...

Abstract:The biological treating technology is always the focus of researchers in the world. The key of this technology is to remove COD and SO_4~(2-) simultaneously and effectively. Sulfate-Reducing Bacteria (SBB) in anaerobic biological treating system can reduce SO_4~(2-) to H_2S so as to remove SO_4~(2-). But acidogenic-desulfate reactor is a complicated ecological system, SRB lead the most important role in this system. To study physiological and ecological characteristic of each microbial group in this system is very difficult, so isolating main functional group,SRB can simplify this research. Preponderant populations of sulfate-reducing bacteria in an acidogenic sulfate-reducing reactor were isolated using improved Hungate technique and tube-rolling method, this sulfate-reducing bacteria was nominated as SRB-ZH07. In anaerobic tube and conical flask physiological and ecological characteristic of SRB-ZH07 were studied. When pH=7.0 SRB-ZH07 grew best, at the culture moment 34h, OD_(600nm) of the culture solution was up to 0. 827, while the highest OD_(600nm) of the culture solution under other pH is 0.08. When pH = 7.0, removal rate of SO_4~(2-) was up to 38.06mg/h at 34h, while the greatest removal of SO_4~(2-) was only 9.62 mg/hr when pH was 3.0,4.0,5.0,8.0. Lactic acid was most suitable to reduce SO_4~(2-) removal rate of SO_4~(2-) was up to 94.24% when lactic is the sole carbon source. Bemoval rate of SO_4~(2-) was up to 28.81% when propionate is the sole carbon source. Glucose was most unsuitable to reduce SO_4~(2-), removal rate of SO_4~(2-) was only 13.6% when glucose is the sole carbon source. The order of carbon source used to reduce SO_4~(2-) was: lactic acid>propionate> butyric acid> acetic acid> ethanol> glucose. When COD/SO_4~(2-) was more than 2, removal rate of SO_4~(2-) was about 80% ; when COD/SO_4~(2-) was less than 2, removal rate of SO_4~(2-) decreased sharply, even lower than 50%. SBBZH07 could grow under aerobic and anaerobic conditions, but only in anaerobic conditions could SBBZH07 remove SO_4~(2-) The growth rate of SRB-ZH07 in aerobic condition was more than that in anaerobic condition. The main metabolic product of SBB-ZH07 metabolizing lactic acid, ethanol, propionate, butyric acid was acetate, so the amount of acetic acid is the index, not reason of the removal rate of sulfate.
