黄鳍鲷2个自然群体遗传变异的RAPD分析

摘    要:用RAPD技术对福建和珠江口2个自然黄鳍鲷(Sparus latus)群体进行群体内与群体间的遗传变异分析。在使用的40个随机引物中.有18个引物扩增出清晰稳定的片段,共计93条,大小为200-2500bp,其中多各性片段41条.多态性片段的比例为44.09%。福建和珠江口群体内的相似系数分别为0.8821和0.8785.群体内遗传距离分别为0.1179和0.1215,2个群体间的遗传距离为0.1314。福建和珠江口2个群体内都有较高程度的遗传变异水平,但福建群体的遗传变异水平较珠江口群体的低。用类平均聚类法和邻接法进行聚类分析,结果表明.福建和珠江口群体分别聚成一支,且两者的聚类结果一致。说明福建和珠江口群体已经开始出现了遗传分化,分别属于2个不同的地理群体。

Abstract:Random amplified polymorphic DNA (RAPD) technique was applied to assess genetic variations in two wild populations of Sparus latus (16 individuals). Amplifications with 18 random primers selected from 40 primers gave 93 reproducible and stable bands, ranging from 200 to 2 500bp. There were 41 polymorphic bands. Based on the RAPD data, the intrapopulation similarities of populations in Fujian and Zhujiang River Mouth were 0.882 1 and 0.878 5 respectively, and the intrapopulation genetic distances were (0.117 9) and (0.121 5) respectively. The interpopulation genetic distance was (0.131 4). The results showed that high genetic diversity existed in the two populations, and that of the Fujian population was lower. With the methods of UPGMA (unweighed pair-group method of analysis) and NJ (neighbor joining) on the basis of genetic distances, the two populations assembled into two branches respectively, and with accordant results, indicating that there was obvious genetic differentiation between them and they belonged to two different geographical populations.
