摘    要:于1996年以本实验室培养的随圆小球藻为材料,就其对G418,潮霉素,氯霉素,链霉素,卡好霉素等5种抗生素的敏感性进行了研究。结果表明,小球藻对氯霉素,链霉素和卡那霉素敏感,对潮霉素较为敏感,100μg/ml即0.190mmol/L的用量可抑制固体培养中小球藻的生长;对G418则高度敏感,30μg/ml即0.043mmol/L的G418即可完全抑制固体培养中小藻 的生长,15μg/ml即0.022

Abstract:Much progress has been made for recent years in selection markers of high plants.However, the situation varies for different algae. Researches on Chlamdomynas reinhardtii developedrapidly and there is also a good start in macroalgae, such as Laminaria japonica, but little progresshas been made for Chlorella. Lack of selection marker made the use of gene transferred Chlorellalimited. Ths paper reports a study on sensitivities of Chlorella ellipsoidea to 5 antibiotics in orderto identify suitable selechon marker of Chlorella genetic engineering.Chlorella ellipsoidea cells after 4-5 days' culture were used as material and antibiotics-Hygromycin (Hm), Chloromycehn (Cm), and Streptomycin (Str) and Kanamycin (Km), differentconcentrahons were used for test in solid culture and Genehcin (G418) was used in solid and liquidculture. After 1-5 weeks' culture, the results show that C. ellipsoidea is not sensitive to Cm, Strand Km, 600ug / ml or 1.240mmol / L Km, 600ug / ml or 0.412mmol / L Str and 200ug / ml or0.169mmol/ L Cm cannot inhabit the growth of C. ellipsoidea. However, it is relatively sensitive toHm, 100ug / ml or 0.191mmol / L Hm can completely stop the growth of C. ellipsoidea on solidmedia. Further, C. ellipsoidea was highly sensitive to G418, since 30ug / ml or 0.043mmol / L and15ug / ml or 0.022mmol / L completely inhabited the growth of C. ellipsoidea in solid and liquidcultures, respectively. It suggests that G418 be the most suitable selection regent if using NPTII geneas selection marker for C. ellipsoidea genetic engineering.
