长蛸的盐度耐受性及盐度胁迫对其血细胞和体内酶活力的影响

摘    要:通过控制暂养水体盐度,用盐度突变与盐度渐变2种方法测定长蛸的盐度耐受性.测定了长蛸血细胞密度、各类血细胞数量比例以及各种酶的活力几组数据,并分析其血细胞与体内酶活力的变化.结果表明:长蛸的生存盐度范围为7.0~30.3,适宜盐度范围为16.3~27.3,最适盐度范围为18.3~24.3,对盐度的适应范围较广,有利于长蛸大规模养殖的开展.在盐度胁迫下,长蛸血细胞密度,各种血细胞数量比例均发生显著改变,长蛸个体也随着胁迫加大变得越来越不适应,出现喷墨、休克、甚至死亡,这表明长蛸的免疫机能降低.无论在低盐度还是高盐度胁迫情况下,受渗透压的影响,长蛸肌体大量吸水和失水,呼吸作用减弱,体内供氧降低,导致长蛸体内有氧呼吸降低和无氧呼吸提高,于是催化有氧呼吸的LDH酶活力降低.同时,细胞内与免疫和消化相关的细胞器由于细胞吸水或失水作用功能受到影响,保护酶(SOD、POD、CAT)、磷酸酶(ACP、AKP)和消化酶(蛋白酶和脂肪酶)的活力均呈下降趋势.由此可见,盐度胁迫对长蛸的影响是显著的.

Abstract:The salinity tolerance of Octopus variabilis was investigated in controlled temporary waters by inducing sudden and gradual changes in salinity.Several indexes were measured,including the density of blood cells,the proportion of blood cells and the activity of various enzymes in vivo.The results indicated that the survival salinity of Octopus variabilis ranged from 7.0 to 30.3,the appropriate salinity ranged from 16.3 to 27.3,and the optimum salinity ranged from 18.0 to 24.3.The adaption to a broad range of salinities favored the large-scale aquaculture of Octopus variabilis.The density of blood cells and proportion of various blood cells changed significantly under salinity stress.Various symptoms of discomfort such as inking,coma and death appeared in individuals in response to increasing salt stress,which indicated a decrease in immune function.Under high or low salinity stress,body cells dehydration or water absorption was caused by a change in osmotic pressure,reduced respiration and oxygen supply in vivo,which led to decreased aerobic respiration and increased anaerobic respiration;therefore,a corresponding decrease in the aerobic respiratory related LDH enzymes occurred.In addition,the functions of organelles related to the immune system and digestion were influenced by dehydration of body cells or water absorption,and the activity of protection enzymes(SOD,POD and CAT),phosphatase(ACP and AKP) and digestive enzymes(protease and lipase) showed a declining trend.Overall,salinity stress had a significant effect on each physiological index of Octopus variabilis.
