红树林海洋细菌的分离鉴定及其活性物质初步分析

摘    要:从广西北部湾红树林海洋淤泥中分离筛选到270株海洋细菌,用喉癌细胞Hep-2的细胞毒作用为筛选模型,用SRB法检测这270株海洋细菌的抑制肿瘤细胞生长活性,其中13株呈阳性反应。依据形态和生理生化反应特征及16SrRNA基因序列分析,确定其中一株为短小芽孢杆菌Bacillus pumilusPLM4。取该菌株液体培养上清液,检测对人喉癌细胞Hep-2的生长抑制率为63.9%,经过凝胶过滤层析分析获得2个活性峰,其中活性高的峰估计平均分子量为116kd。采用经典显色反应法初步判断该活性物质为多糖。

Abstract:Two hundreds and seventy strains of marine bacteria were screened from the sea mud collected in Guangxi from the mangrove of Beibu Gulf.Their growth inhibitory activity to tumor cells were detected by SRB method,and a positive reaction was found in 13 strains among the 270 strains.One strain of the 13 strains was classified to be Bacillus pumilus PLM4 according to its morphological and physiological characters and 16SrRNA gene sequence.The supernatant of the liquid culture of Bacillus pumilus PLM4 could inhibit the growth of tumor cells Hep2,and the inhibition ratio was 63.9%.By gel chromatographical analysis,the supernatant was found to have two active peaks.The molecular weight of the high active peak was about 116kd.This product was determined to be polysaccharide by classical color reaction.
