摘    要:骨条藻(Skeletonema)隶属于硅藻门(Bacillariophyta),是一种分布广泛的广温广盐性藻类,也是我国沿海常见的赤潮生物之一。为确定北部湾涠洲岛5株骨条藻的分类地位,采用PCR克隆了这5株骨条藻核糖体的大亚基基因(LSU)和内转录间隔区(ITS)序列。通过与GenBank数据库中骨条藻属的LSU、ITS序列进行比对,得出的结果一致:5株涠洲岛骨条藻均为热带骨条藻(Skeletonema tropicum)。本研究运用分子标记法,首次确定了北部湾涠洲岛海域存在有热带骨条藻,结果可为涠洲岛与广西北部湾海域浮游植物多样性、赤潮生物组成及赤潮的预警与有效防治提供科学依据。

Abstract:Microalgae in genus Skeletonema belong to phylum Bacillariophyta.These microalgae are the most common marine planktonic diatoms due to their eurythermal and euryhaline features.They are also the most common causative organisms for harmful algal blooms(HABs)along the coast of China.To determine the taxonomy of five Skeletonema strains isolated from Weizhou Island,Beibu Gulf,Guangxi,28 S ribosomal RNA gene(LSU rDNA)and internal transcribed spacer(ITS)sequences of the five Skeletonema strains were cloned by PCR technology.The results showed that all five Skeletonema strains are Skeletonema tropicum by comparing their LSU rDNA and ITS sequences with those downloaded from GenBank.In this study,the molecular marker method was used for the first time to identify Skeletonema tropicum in the waters of Weizhou Island,which will provide more insight into the phytoplankton diversity,the composition of HABs organisms and the early warning and effective control on HABs in Weizhou Island and Beibu Gulf,Guangxi.
