扩增仿刺参SSR和ISSR指纹技术的初步研究

摘    要:利用SSR(Simple Sequence Repeats)技术和ISSR(Inter Simple Sequence Repeats)技术对仿刺参(Apostichopus Japonicus)进行了PCR扩增。探索了刺参基因组DNA的提取方法。为了得到更好的PCR扩增结果,对引物浓度、Mg2 浓度、dNTP浓度、模板浓度、Taq浓度和退火温度及循环数等方面进行了研究,从而摸索出一个适合刺参SSR和ISSR分析的反应体系和反应条件,并对PCR反应中的一些特定影响因子进行了分析。

Abstract:SSR technique and ISSR technique were applied to amplifying the genomic DNA of S.Japonicus.We studied a suitable method for DNA extraction.In order to produce clear amplified fragments,the conditions for SSR and ISSR analysis of S.Japonicus were optimized,such as the concentrations of primer,Mg~(2 ),dNTP,DNA and Taq.The reaction systems and reaction conditions suitable for SSR and ISSR analysis were obtained.And the influential factors in the Polymerase Chain Reaction were analyzed.
